Objective To explore the correlation between lens and angle closure in acute angle-closure glaucoma(AACG).Methods Forty eyes of 40 cases of first attack of AACG in one eye were included in present study.Forty controls(people with no glaucoma)match in age and gender were selected.The other eye of 40 AACG patients was defined as fellow eye group.Anterior chamber depth(ACD),lens thickness(LT),lens position(LP)and chamber crowd ratio(CCR)were compared among the 3 groups(40 each):attacked eye group,fellow eye group and the controls.Results Significant differences among the 3 groups(attacked eye group,fellow eye group and the controls)were found in LT(5.43?0.37,5.36?0.33 and 4.84?0.65mm,respectively,P
2.Clinical internship and innovation of psychiatry from the perspective of medical undergraduates

Objective To evaluate the current methods for psychiatry clinical internship and needs for innovation from the perspectives of medical undergradutes.Methods Independent interview on patients and their relatives as well as social psychological interview were conducted for five-year psychiatry majors in clinical internship (n =72).Self-made questionnaire was used to investigate the teaching feedback concerning the practice effect and teaching method tendency.Data were analyzed by t test and P < 0.05 signifies statistically significant differences.Results Medical students considered the clinical internship as the main source of psychiatry knowledge (rising from 29.4 % (20/69) to 36.2% (25/69),t =4.67,P <0.01).Respective interviewing with patients and their families and then doing focus-group discussion was the most acceptable method for medical undergraduates:82.6%(57/69) of medical undergraduates thought this method helped expose defects of clinical skills and prejudice against psychiatry; 54.3% (37/69) of medical undergraduates thought this method was more beneficial to acquire psychiatry knowledge.The second acceptable method was the demonstration of a typical case by teacher:27.3% (19/69) thought this method helped expose defects of clinical skills and prejudice against psychiatry; 27.2% (19/69) thought this method was more beneficial to acquire psychiatry knowledge.62.0% (43/69) of medical undergraduates ranked the characteristics of the symptomatology and core symptoms differentiation as the first place.49.0% (34/69) considered the training of interviewing skills with patients as the second important in the clinical training.91.3% (63/69) expected to see more kinds of mental disorders.65.2 % (45/69) expected to learn more about the interviewing skills.Conclusions It is necessary to pay more attention to the method of doing respective interviewing with patients and their families and then making focus-group discussion.Meanwhile training of interviewing skills in the clinical internship of psychiatry should be enhanced.
3.Effects of corneal plastic surgery on stereopsis and dynamic regulation of myopia and the mechanism of controlling myopia progression

Objective To investigate the effect of corneal plastic surgery on stereopsis and dynamic regulation of myopia and the mechanism of myopia.Methods From March 2015-March 2016,83 cases (166 eyes) myopia children in our hospital were subjected to corneal plastic surgery.The naked eye distant visual acuity,naked eye near visual acuity,naked eye near stereoscopic sharpness,dynamic adjustment function [Near point of accommodation (NP),adjusted sensitivity (AF),negative relative adjustment (NRA),positive relative adjustment (PRA)] and intraocular pressure,anterior chamber depth,central corneal thickness and axial length of children before wearing glasses,wearing glasses 6 months later,wearing glasses 12 months later were compared.Results The distant and near visual acuity,AF and PRA of naked eyes after 12 months wearing glasses were higher than those after 6 months wearing glasses,and all the above indexes were higher than those before wearing glasses,with statistically significant difference (P < 0.05).After 12 months wearing glasses,the acuity of naked near stereopsis,NP and NRA were lower than those after 6 months wearing glasses,and all the above indexes 6 months after wearing glasses were lower than those before wearing glasses (P < 0.05).There were no significant differences in intraocular pressure,anterior chamber depth,central corneal thickness and axial length between children before wearing glasses,6 months after wearing glasses and 12 months after wearing glasses (P > 0.05).Conclusions Corneal plastic surgery can significantly correct the visual acuity of children,improve their stereovision,control the progression of myopia,improve the ocular adjustment function of children,and its safety is high.
4.Comparison of human and Drosophila atlastin GTPases.

Formation of the endoplasmic reticulum (ER) network requires homotypic membrane fusion, which involves a class of atlastin (ATL) GTPases. Purified Drosophila ATL is capable of mediating vesicle fusion in vitro, but such activity has not been reported for any other ATLs. Here, we determined the preliminary crystal structure of the cytosolic segment of Drosophila ATL in a GDP-bound state. The structure reveals a GTPase domain dimer with the subsequent three-helix bundles associating with their own GTPase domains and pointing in opposite directions. This conformation is similar to that of human ATL1, to which GDP and high concentrations of inorganic phosphate, but not GDP only, were included. Drosophila ATL restored ER morphology defects in mammalian cells lacking ATLs, and measurements of nucleotide-dependent dimerization and GTPase activity were comparable for Drosophila ATL and human ATL1. However, purified and reconstituted human ATL1 exhibited no in vitro fusion activity. When the cytosolic segment of human ATL1 was connected to the transmembrane (TM) region and C-terminal tail (CT) of Drosophila ATL, the chimera still exhibited no fusion activity, though its GTPase activity was normal. These results suggest that GDP-bound ATLs may adopt multiple conformations and the in vitro fusion activity of ATL cannot be achieved by a simple collection of functional domains.

5.Effects of nicorandil on the expression of hypoxia-inducible factor 1α in lung tissue of one-lung ventilation

Objective To investigate the effects of nicorandil on hypoxia-inducible factor (HIF)-1α mRNA and protein in lung tissue of one-lung ventilation.Methods Twenty-four clean New Zealand white rabbits were randomly divided into sham group (group S) (two-lung ventilation+thoracotomy),negative control group (group C) (one-lung ventilation + thoracotomy + saline),nicorandil group (group N) (one-lung ventilation+ thoracotomy+ nicorandil) and antagonist group (group J) (one-lung ventilation + thoracotomy + nicorandil + glibenclanide) equally.The implementation of mechanical ventilation depended on self-made double-lumen endotracheal tube after intravenous induction through ear marginal vein.Intravenous maintenance medicine was infused by trace injection pump after anesthesia induction.The implementation of thoracic surgery was simulated through one-lung and two-lung ventilation by auscultation,bubble test and direct observation.Group S was given anaesthesia only,no one-lung ventilation group S,the other three groups had single lung ventilation,and the drug was injected before the operation.Group N was infused nicorandil 100 ptg· kg-1 · h-1 before the implementation of single lung ventilation for 1 h.Group C was injected with the same amount of normal saline.Group J was intravenous infusion of glibenclamide 75 μg· kg-1 · h-1 and nieorandil 100μg · kg-1 · h-1 the implementation of single lung ventilation for 1 h.Then wet and dry weight ratio(W/D) and superoxide dismutase (SOD) activity were measured after non-ventilatory lung was processed and preserved.The expression of HIF-1α protein of non ventilatory lung tissue was detected by Western-blot in the four groups.The transcription of HIF-1α mRNA was detected by real-time quantitative real-time PCR (qRT-PCR) in all groups.Results W/D in groups C and J were significantly higher compared with that of groups S and N (P<0.05).The activity of SOD in groups C and J was significantly lower compared with groups S and N (P<0.05).The expression of HIF1α protein and transcription of HIF-1α mRNA in groups C,N and J were significantly higher than those in group S,and that of group N was significantly higher than those of groups C and J (P<0.05).Conclsion Nicorandil has a protective effect on the collapse and inflation of non-ventilatory lung in rabbit with one-lung ventilation,reducing oxidative stress by SOD,acting on mito KATP and coming into play by up-regulation of HIF-1α.
6. Expression of Fermintin family homologous protein 2 in non-small cell lung cancer and its clinical significance

Objective:
To investigate the expression of Fermintin family homologous protein 2 (FERMT2) in non-small cell lung cancer and its clinical significance.
Methods:
Seventy-two patients with non-small cell lung cancer were collected at Xinxiang Central Hospital, Henan Province, from January 2015 to January 2017.There were 48 male and 24 female patients, the age ranged from 37 to 78 years (mean 58 years). The expression of FERMT2 in tumor samples and para-cancerous tissues were detected by immunohistochemistry. Protein and mRNA expression of FERMT2 were detected by Western blot and real-time fluorescence quantitative PCR, respectively. Western blot method was also used to detect integrin-related protein expression, including integrin beta 1 (CD29), vascular cell adhesion molecule (VCAM1), and mobile related protein-1 (MRP1).
Results:
Immunohistochemistry showed that the positive rates of FERMT2 expression were 81.9%(59/72)in carcinoma tissue and 15.4%(11/72) in para-cancerous tissues, and the difference was statistically significant (
7.Changes of intestinal flora and the mechanism of NLRP3 inflammasome in elderly mice with cognitive dysfunction induced by sevoflurane anesthesia

Objective:To investigate changes of intestinal flora and the mechanism of NLRP3 inflammasome in elderly mice with cognitive dysfunction induced by sevoflurane anesthesia.Methods:Eighteen fourteen-month-old male SPF grade C57BL/6J mice were randomly divided into control and sevoflurane groups, with 9 mice in each group. The mice of sevoflurane group inhaled 3% sevoflurane for 2 hours daily for three days. Fecal samples were collected post-exposure 24 hours for 16S rRNA sequencing. Morris water maze was then used to test the cognitive ability. Western blot was used to detect the expressions of synapse-associated proteins, NLRP3 inflammasome-related proteins of hippocampus, and NLRP3 inflammasome-related proteins of colon. Golgi staining was used to observe the number of dendritic spines in the hippocampus. qPCR was used to detect the expression of inflammatory cytokines IL-1β, IL-18, TNF-α mRNA in mice colon and hippocampal tissues.Results:(1) The Morris water maze test showed that the escape latency of the sevoflurane group was longer than the control group, but there was statistical difference only on the fifth day ( P<0.05). In the spatial exploration test, escape latency of the sevoflurane group was higher than that of the control group((49.50±9.99)s, (18.67±7.63)s, t=6.005, P<0.001), and platform crossing frequency was less than that of the control group((0.83±0.75)times, (2.33±1.03)times, t=2.87, P=0.017). (2) Western blot and Golgi staining results showed that the expression of hippocampal synaptic-related proteins and the number of dendritic spines in the sevoflurane group were significantly reduced compared with those in control group (all P<0.05). (3) 16S rRNA sequencing showed significant β-diversity difference between the two groups ( P<0.05). Compared with the control group, potential pathogens that p_Desulfobacterota and g_Desulfovibrio increased significantly in the sevoflurane group (both P<0.05), and beneficial bacteria that p_Verrucomicrobiota and g_Akkermansia decreased significantly (both P<0.05). (4) Compared with the control group, the results of qPCR showed increased expression of inflammatory cytokines TNF-α, IL-1β mRNA in the colon and hippocampal tissues of the sevoflurane group (all P<0.05). Western blot results showed increased expression of NLRP3 inflammasome-related proteins in the colon and hippocampal tissues of the sevoflurane group (both P<0.05). Immunofluorescence results showed the higher fluorescence intensity of ASC in the DG region of the hippocampus of the sevoflurane group compared with the control group ( P<0.01). Conclusion:The cognitive dysfunction model induced by sevoflurane in elderly mice shows neuroinflammatory reactions and synaptic damage, which may be related to intestinal microbiota imbalance and activation of NLRP3 inflammasome.
8.Deformation of the Cupula of a Semicircular Canal Based on a Biomimetic Physical Model

Objective To explore the biomechanical responses of the cupula of the human semicircular canal to three basic rotational perception processes.Methods A one-dimensional visual semicircular canal model was successfully fabricated using three-dimensional printing and hydrogel physical cross-linking technologies,and the response deformation of the cupula was explored by applying constant angular velocity,constant angular acceleration,and sinusoidal oscillation stimulations.Results The time constant of the biomimetic semicircular canal model was stable at approximately 3 s and close to the human time constant.The displacement deformation of the ampullary cupula was proportional to the angular acceleration applied.Under sinusoidal oscillation stimulation of 0.07-5.00 Hz,the gain of the semicircular canal increased from 1.54 um/° rises to 42.34 μm/°,but the phase difference decreased from 109.72° to 11.27°.Conclusions The biomimetic semicircular canal model prepared in this study can accurately simulate the working mechanism of the human semicircular canal and is expected to play a role in mechanism research and disease diagnosis of the human vestibular semicircular canal.
