1.Vascular endothelial growth factor 165 promotes proliferation of human adipose-derived mesenchymal stem cells

BACKGROUND:Autologous fat transplantation has been widely used in soft tissue defect repair and cosmetic surgery, and the 1-year transplant survival rate is 20%-80%. Therefore, the establishment of timely and adequate blood supply at early period after transplantation is very important for the survival of transplanted fat tissues.
OBJECTIVE:To observe the proliferation of human adipose-derived mesenchymal stem cel s transfected with vascular endothelial growth factor 165.
METHODS:Human adipose-derived mesenchymal stem cel s were subcultured in vitro. Recombinant adenovirus carrying vascular endothelial growth factor 165 and blank virus were respectively transferred into adipose-derived mesenchymal stem cel s. Cel s cultured normal y served as blank group.
RESULTS AND CONCLUSION:Compared with the control and blank groups, the expressions of vascular endothelial growth factor 165 mRNA and protein were higher in the experimental group (P<0.05). Experimental findings suggest that the recombinant adenovirus carrying vascular endothelial growth factor 165 transferred into adipose-derived mesenchymal stem cel s cannot only maintain the expression of target protein but also obviously promote the proliferation of adipose-derived mesenchymal stem cel s.
2.Establishment of a nude mouse model of glioma orthotopic xenograft with double-fluorescent labeling

Objective To establish a stable and real-time monitorable nude mouse model of orthotopic glioma xenograft.Methods U251 glioma cell line was infected by a lentiviral vector containing green fluorescent protein (GFP) and luciferase (Luc) gene.Cells stably expressing fluorescence of GFP and Luc were sorted by flow cytometry.CCK-8 test and Transwell tumor invasion and migration assay were used to compare the biological features between the cells stably expressing GFP-Luc fluorescence and cells without fluorescence.Then the cells were implanted intracranially in the right caudate nucleus of athymic Balb/c nude mice to establish the tumor model.The growth of intracerebral tumor was monitored over time by a bioluminescence imaging (BLI) system.Hematoxylin-eosin (HE) staining was used to evaluate the histopathological features and tumorigenicity of the transplanted glioma cells in the brain of nude mice.Results U251 glioma cell line with stably expressing GFP-Luc fluorescence and the corresponding orthotopic xenograft model were successfully established.There was no statistically significant difference in the proliferation,invasion and migration abilities between the cells with stably expressing GFP-Luc fluorescence and the control cells.This model showed a high tumor formation rate and stable tumor growth,and takes a moderate time to establish this model.Conclusions Compared with the traditional glioma cells,GFP-Luc-transfected human glioma cells are more feasible for the studies of glioma in vivo.The tumor growth and pathological characteristics in this U251-GFP-Luc glioma model are similar to human glioma,and the growth of this tumor can be real-time monitored.It can be used as an ideal animal model for experimental studies of glioma.
3.Effect of microRNA-155 on regulation of angiogenesis in diabetic rats with cerebral ischemic injury

AIM:To evaluate the effect of microRNA-155(miRNA-155) on the regulation of angiogenesis in diabetic rats with cerebral ischemic injury .METHODS: Adult male Sprague-Dawley rats were randomly divided into 5 groups:sham group, cerebral ischemia group , diabetic cerebral ischemia group , diabetic cerebral ischemia +miRNA-155 inhibitors group and diabetic cerebral ischemia +scramble group .Diabetes model was made by injection of streptozocin and permanent cerebral ischemic model was developed by suture-occluded method .The scores of neurological deficit and infarct volume were estimated at 24 h after cerebral ischemia .miRNA-155 level was detected by real-time polymerase chain reaction.The expression of platelet endothelial cell adhesion molecule-1 ( PECAM-1/CD31 ) and vascular endothelial growth factor ( VEGF) was detected by Western blotting .RESULTS:miRNA-155 inhibitor significantly reduced miRNA-155 levels in the ischemic cortex (P<0.05), improved the scores of neurological deficit , reduced infarction size and up-regulated the levels of CD31 and VEGF (P<0.05).CONCLUSION:miRNA-155 has a critical role in the regulation of angiogenesis in diabetic rats with cerebral ischemia .Down-regulation of miRNA-155 using miRNA-155 inhibitor attenuates brain infarct injury in diabetic rats .
4.Difficulty and skill of laparoscopic right colectomy.

With increase in the incidence of right colon cancer, the proportion of laparoscopic right colon resection is increasing. Though the advantage of laparoscopy in minimal invasiveness has been widely accepted, its procedure still possesses certain difficulty. In this article, we shared the experience of laparoscopic right colectomy, including entering the correct Toldt's place, management of ileocolic and middle colic vessels and the problems of D3 lymph node dissection. Basic surgical techniques and several difficulties are discussed here, which may be helpful for beginners.

5.A case report of primary renal fibrosarcoma

A patient with intermittent right lumbar pain for more than one month and aggravating for one week was reported. The right laparoscopic radical nephrectomy was performed, and fibrosarcoma of the right kidney was diagnosed. The patient refused further radiotherapy or chemotherapy and was discharged on the 11th after surgery. One year after operation, the patient underwent chest CT, and subdural tumor of right lobe of liver was detected. A surgery was performed in another hospital, and liver metastasis from right renal fibrosarcoma was diagnosed. He was alive four months after operation.
6.Biphasic insulin aspart 30 plus metformin in patients with type 2 diabetes inadequately controlled on basal insulin therapy:An evaluation of efficacy and safety

Objective To evaluate the efficacy and safety of biphasic insulin aspart 30 (BIAsp30)plus metformin in type 2 diabetes subjects switching from basal insulin plus oral antidiabetic drugs (OAD)Methods During 16 weeks, multiple-center, open-label, and single-arm study including 2 weeks of screening period,4 weeks of run-in period,and 16 weeks of treatment period were carried out. Subjects with type 2 diabetes mellitus inadequately controlled on basal insulin therapy with or without oral antidiabetic drugs were switched to twice-daily BIAsp30 plus metformin with dose titration to achieve fasting plasma glucose target. Results Of the 293 Chinese subjects exposed to trial drugs [age: ( 54.0±9.6 ) years, diabetes duration: ( 8.54±5.49 ) years, body mass index: (24.89±3.28)kg/m2, baseline HbA1c: 8.16% ±0.89%], 122 were previously treated with basal insulin analogues and 169 with human basal insulin. At end of the trial ,the mean reduction of HbA1 c was 1.30% ±0.96% (P<0. 01 ). The proportion of patients achieved HbA1c<7.0% and HbA1c ≤6.5% was 60.4% and 38.9% respectively. 8-point plasma glucose measurements showed significant improvements at all the time points examined ( all P<0. 01 ) ,and the average value of all 8 points measured decreased from ( 10.53±2.58 ) mmol/L atbaseline to (7.79± 1.58 ) mmol/L at the end of treatment ( P<0. 01 ), reduced by 2.76 mmol/L. Postprandial glucose increments were significantly reduced after breakfast ( -1.73 mmol/L,P<0.01 )and dinner ( -1.28 mmol/L,P<0.01 ), while no significant reduction was observed after lunch ( -0.09 mmol/L, P = 0. 734 5 ). No severe adverse effect and no major hypoglycemia were reported. The overall hypoglycaemia rate was 2.68 events/subject year. The average weight gain was (0. 76 ±0. 14 )kg (P<0. 0l ). Conclusion Twice-daily BIAsp30 plus metformin is effective and safe to type 2 diabetic subjects inadequately controlled on basal insulin treatment.BIAsp30 treatment should be considered for type 2 diabetic subjects who have unsatisfactory response to previous basal insulin treatment.
7.Discussion on standardized implementation of laparoscopic radical lymphadenectomy for distal gastric cancer.

Laparoscopic radical gastrectomy for gastric cancer has been widely applied in clinical practice, and its indications have been extended from early gastric cancer to advanced gastric cancer. It is acknowledged that laparoscopic radical gastrectomy is technically challenging because of the complexity of anatomy, rich blood supply, and extensive lymph node dissection. This paper primarily intends to share the experience of laparoscopic radical D2 gastrectomy for distal gastric cancer with details of choosing the location of Trocar, surgical approaches and the sequence of lymph node dissection. All the surgeries were performed at Department of General Surgery and Gastrointestinal Surgery, Guangdong General Hospital. The finding suggests that a correct laparoscopic Trocar placement is the foundation of adequate surgical field visualization. Under most circumstances, the observation hole should be around 2 cm below the umbilicus and the operating hole should be close to the bilateral clavicle midline. Furthermore, proper surgical approach and sequence of lymph node dissection are the prerequisites for successful laparoscopic radical D2 gastrectomy, as well as the reassurance of dissecting lymph node safely and comprehensively. The position of surgical team adopted in our center is that the surgeon stands to the left of the patient, with laparoscope operator stands in between patient's legs while the first assistant positions himself opposite the surgeon on the right side of the patient. This position correlates to the rules of sequential lymph node dissection, which is "from left to right", "from proximal to distal" and "from inferior to superior". Therefore, it is conductive to inferior and superior pylorus region dissection and it can effectively prevent subsidiary-injury. In our center, the procedure of lymph node dissection has been standardized: the initial step is to undergo station 4sb dissection and greater gastric curvature clearance; then change the patient's position to clean the sub-pyloric lymph node region and cut off the duodenum by linear stapler; followed by the clearance of inferior region of the pylorus and the upper margin of the pancreas; in the final step, the first and the third groups of lymph node dissection is performed. Although varied surgical approaches and sequences of lymph node dissection are applied in different hospitals, the techniques required for laparoscopic D2 radical gastrectomy for gastric cancer are sophisticated and advanced in general. Radical lymph node dissection is complicated, urging surgeons to familiarize themselves with the anatomy of gastric peripheral vascular system and characteristics of lymph node drainage. By designing and implementing effective strategies, such as formulating a regular team, positioning surgical team reasonably, changing a patient's posture during operation, choosing an appropriate surgical approach and following a logically sequence of lymph node dissection, surgeons can standardize the complete surgical procedure, which ultimately reduces bleeding during surgery and shortens the operative time.
8.Expression and Action Mechanism of miR-98 in Colon Cancer Tissues

Objective To investigate the expression and action mechanism of miR-98 in colon cancer tissues. Methods Tumor tissues and adjacent tissues were collected from 40 patients with colorectal cancer. The expression of miR-98 in tumor tissues and adjacent tissues was detected by real-time PCR. miR-98 was overexpressed or silenced in cells,and the effects on proliferation,cell cycle,and apoptosis were analyzed using MTT,flow cytometry,and Hoechst 33258 assays. Results Real-time PCR showed that the expression of miR-98 in tumor tissues was lower than that in adjacent tissues (P = 0.022). The survival rate of patients with lower miR-98 expression was shorter than that of patients with higher miR-98 expression. The MTT assay showed that miR-98 overexpression inhibited the proliferation of HCT116 cells. Hoechst 33258 staining showed that the overexpression of miR-98 could inhibit the cell cycle and promote the apoptosis of HCT116 cells. Conclusion miR-98 can inhibit the proliferation and promote the apoptosis of colon cancer cells. The expression of miR-98 is closely related to the survival of patients with colon cancer.
9.A case report of renal leiomyoma with concurred eosinophilic adenoma

A female patient who was admitted to the hospital on March 4, 2020 due to the right kidney mass for 4 days by physical examination. Ultrasound examination revealed a single space-occupying lesion in the right kidney, and further examination of the abdominal enhanced CT and three-dimensional reconstruction showed two lesions in the right kidney. The retroperitoneal laparoscopic partial nephrectomy was carried out. The pathological diagnosis were renal leiomyoma and renal eosinophiloma, respectively. After 1 year follow-up, no recurrence or metastasis was found.
10.Effect of different light of LED light quality on growth and antioxidant enzyme activities of Ganoderma lucidum.

OBJECTIVETo study the effect of light quality on growth, antioxidant enzyme activities of Ganoderma lucidum mycelium.
METHODG. lucidum mycelium was cultured under different light qualities by light emitting diodes (LED). The growth G. lucidum mycelium was observed and antioxidant enzyme activities was determined in different growth periods.
RESULTUnder the red LED, the blue LED and dark condition (CK), the mycelium grew faster than that under other light qualities. The white LED resulted in a largest increase in the amount of the mycelium and always kept the activities of CAT high level. Major fluctuations of POD activities emerged under the green LED, while enhanced severely in the late phase. Under the yellow LED, the activities of SOD appeared high level. However, SOD activities on dark (CK) raised obviously in late period. At the late stage, the content of mycelium polysaccharides was significant higher than that under the blue LED.
CONCLUSIONThe light quality could influence the growth and metabolism of G. lucidum mycelium.
