1.Alterations in levels of CD8+ CD28- regulatory T cell in the peripheral blood of multiple sderosis patients

Objective To investigate the levels of peripheral blood CD8+ CD28- regulatory lymphocytes and their clinical values in the patients with multiple sclerosis (MS). Method From October 2005 to August 2008, 51 patients with active rehpsing-remitting MS were enrrolled from Department of Neurology of the First Affil-iated Hospital of Wenzliou Medical College. The diagnostic criteria for MS were the 2005 revisions to the "McDon-ald criteria". All the admitted patients received 1 g of methylprednisoione per day intravenously for 5 days, fol-lowed by 60 mg prednisone per day orally for 12 days,and tapered in 6 weeks. Fourteen patients were reevaluated after corticosteroid therapy. Twenty healthy individuals ,as normal controls,matched for age and sex with the MS patients were also enrolled in this study. The percentages of peripheral blood T cells (CD8+ CD28-, CD8+CD28+, CD8+, CD4+ CD8-) were measured by flow cytometric analysis. Parametric statistical analysis were per-formed using standard methods, and linear regression analysis was conducted using Pearson correlation test. Re-sults (1)Compared with controls,the patients with active MS had significantly lower percentage of CD8+ CD28-T cells [(18.48±9.89)% vs. (24.48±4.86)%, P <0.01], and higher percentage of CD8+ CD28+ T cells [(12.23±4.31) % vs. (8.55±3.49) %, P <0.01]. (2)The percentage of CD8+ CD28- T cells was negative-ly correlated with that of CD4+ CDS- T cells (r = -0.488, P < 0.01). (3) After corticosteroid therapy, the per-eentage of peripheral blood CD8+ CD28- / CD8+ CD28+ T cells didn' t significantly decrease or increase in 14 ac-tive MS patients (P > 0.05). Conclusions The decrease of peripheral blood CD8+ CD28- regulatory T cells might be associated with the pathogenesis of MS, and CD8+ CD28- regulatory T cells perhaps played their roles through CD4+ T cells. Corticosteroid therapy could not reverse the levels of CD8+ CD28- T cells.
2.Clinical experience of antegrade radical retropublic prostatectomy in 30 patients

Objective To discuss the experience of antegrade radical retropublic prostatectomy. Methods Thirty cases of prostate cancer were treated by antegrade radical retropublie prostateeto-my. The mean age of patients was 64 years. There were 8 cases of T_1, 15 cases of T_2, and 7 cases of T_3. The surgery included following procedures: Firstly the vesical neck was divided. Then the seminal vesicle and the prostate were freed from above downward. After the urethra was separated from the prostatic apex, the proximal end of urethra was sutured to the new vesical neck. Results All the 30 laparoscopic surgeries were accomplished successfully. The mean operative time was 2.5 hours. The mean blood loss was 350 ml in the operation. No ureteral injury, rectal injury and other severe compli-cation was observed. Histopathologic study showed prostate cancer for all the cases. Four cases had positive surgical margins. All cases were followed up from 6 to 48 months (average 25 months) with-out dysuria and permanent incontinence. No death occurred. Sexual function recovered in 7/13(54%) cases after the operation. Conclusions Antegrade radical retropublic prostatectomy provides low complication, tittle bleeding and positive surgical margins. It is an effective and sale procedure to treat prostate cancer.
3.Clinical diagnosis and management of Wunderlich's syndrome

Objective To investigate the cause, diagnosis and management of Wunderlich's syndrome.Methods Clinical data of 13 cases of Wunderlich's syndrome were reviewed retrospectively. Results There were nine cases of tumorous disease, 2 cases of non-tumorous disease and 2 cases of unknown causes in the group. The most common cause in this series was renal angiomyolipoma. Surgical management was performed in 8 cases. Conservative treatment was adopted in the rest 5 cases. Pathological study confirmed that there were 2 cases of renal cell carcinoma, 4 cases of angiolipoleiomyoma and 1 case of congenital polycystic kidney. Conclusions The cause of Wunderlich's syndrome is complicated, renal neoplastic origin disease appears to be the most common cause. Comprehensive analysis of multiple imaging examinations and clinical features is most important in the etiological diagnosis. The appropriate management depends on the underlying etiological diagnosis and the patient's hemodynamic status. Carefully watching is required for patients who have not been made a definite etiological diagnosis.
4.A survey on the cognition of ICU syndrome by ICU nurses in Guangdong province

Objective To understand the cognition of ICU syndrome by ICU nurses in Guangdong province. Methods Through questionnaire filling in, this investigation was made on the first hatch of ICU nurses dispatched to Hung Kong for ICU specialized training by Guangdong province. Information collected included the nurses' personal information and their knowledge about ICU syndrome. Results The sub-jects' knowledge about ICU syndrome was scanty, and was not related to their years of work, professional ti-tles, academic degrees or administrative duties. The overwhelming majority of the subjects had not paid at-tention to the disease in clinical work. Most departments had not formulated guidelines whatsoever on nurs-ing for ICU syndrome. In assessing the ICU syndrome, the vast majority of nurses relied on their clinical experience, with none of them relying on the confusion assessment method for the intensive care unit (CAM-ICU). Only very few of the subjects acquired some knowledge about the syndrome during their study at colleges. Conclusions The ICU nurses in Guangdong province should attach importance to ICU syn-drome and strengthen the study of related knowledge in their work and study. And it is essential to popular-ize CAM- ICU and to build a standard assessment system and work out the intervention measures for ICU syndrome.
5.Observation of the clinical effect of extracapsular cataract extraction and implantation of intraocular lens combined with vitreoretinal surgeryby samall incision

Objective To observe the clinical effect of extracapsular cataract extraction and implantation of intraocular lens combined with vitreoretinal surgery by samall incisionto to treat cataract merge vitreoretinal disease.Methods A retrospective study was managed on 18 cases(18eyes). Eyesight and complications after operation was study. Result Follow-up period was from 3 months to 18 months( the 8 months on average). The cases of corrected visual acuity less than 0. 02 was 2 eyes, among 0.02 to 0. 1 was 4 eyes, among 0. 2 to 0.3 was 8 eyes, then more than 0.3 was 4 eyes. The complications of surgery included tunicae uveae reaction, cornea reaction, ocular hypertension, vitreous hemorrhage,retinal detachment. Conclusion Extracapsular cataract extraction and implantation of intraocular lens combined with vitreoretinal surgery by small incision was safe and effective to treat Cataract combined with vitrcoretinal disease. The main factor affected the eyesight retrieval was posterior segment pathology.
6.The impact and mechanisms of ursolic acid on Hep-2 cells invasion

Objective To study the effect of ursolic acid on human laryngeal carcinoma Hep‐2 cells invasion and to explore its mechanism .Methods Methabenzthiazuron(M TT ) was used to observe the proliferation of Hep‐2 cells treated with various con‐centrations of ursolic acid at different time .Boyden chamber was used to compare ursolic acid effect on cell invasion .Western blot was used to the measure the NF‐κB protein expression .Gelatin zymography was used to the activity of MMP‐9 and MMP‐2 .Results Ursolic acid inhibited human laryngeal carcinoma Hep‐2 cells growth ,its effect showed dose dependent and time dependent (P <0 .01) .Boyden chamber results revealed that after the ursolic acid treatment Hep‐2 cells ,invasion ability showed a concentration de‐pendent decreased (P< 0 .01) .Western blot results revealed that ursolic acid showed a concentration dependent decreased on human laryngeal Hep‐2 cells NF‐κB protein expression(P< 0 .01) .Gelatinases spectrum revealed that ursolic acid a concentration depend ‐ent decreased on human laryngeal Hep‐2 cells MMP‐9 and MMP‐2 cells activity (P< 0 .01) .Conclusion Ursolic acid inhibit human laryngeal cancer Hep‐2 cell proliferation and invasion ,its mechanism is related to down‐regulate MMP‐9 and MMP‐2 activity and the NF‐κB protein expression .
7.GC Fingerprints of Essential Oil from Schizonepeta Tenuifolia Briq

Objective: To establish the GC fingerprints of essential oil for Schizonepeta tenuifolia Briq. to control the quality. Methods:The temperature of the feed inlet and detector was both 250℃, the carrier gas was nitrogen, and the flow rate was 2 ml· min-1 . The essential oil from ten batches of Schizonepeta tenuifolia Briq. was analyzed by GC-MS to determine the characteristic peaks and the similarity was studied as well. Results:The GC fingerprints of essential oil from Schizonepeta tenuifolia Briq. was established, and totally 13 characteristic peaks were calibrated with high similarity. Conclusion: The method is simple, precise and reliable. The established fingerprints can be used as one of the quality control index for essential oil from Schizonepeta tenuifolia Briq. .
8.Hyaluronic acid mixed with BMP-2 gene transfected bone marrow derived mesenchymal stem cells for repair of diaphyseal defects in rabbits

Objective To evaluate osteogenetic effectiveness of hyaluronic acid (HA) mixed with adenovirus mediated human bone morphogenetic protein-2 gene(Adv-hBMP-2) transfected bone marrow derived mesenchymal stem cells(BMSCs) of rabbits in the repair of radial shaft defects of rabbits. Methods The BMSCs were collected from bone marrow of rabbits, cultured and transfected by Adv-hBMP-2 in vitro. The radial shaft defects (1.5 cm) models of 20 rabbits (40 sides) were created and then respectively injected the cells and/or materials according to the following four groups (n=10): Ⅰ, Adv-hBMP-2 transfected BMSCs+HA; Ⅱ, Adv-hBMP-2 transfected BMSCs; Ⅲ, HA; Ⅳ, control. Results The bone defects of 6 out of 8 sides in the group Ⅰ were repaired completely and partial bone marrow cavities were revasculized. Only 3 sides were repaired completely in the group Ⅱ and no side was repaired completely in the groups Ⅲ and control. There were statistic significance between each groups in X-ray scores. In the groups ofⅠand Ⅱ, much woven callus formed in the sites of the bone defects in the 4-8th week after injection. Partial bone marrow cavities were revasculized in the 12th week after injection; while in the groups of HA and control, the sites of bone defects were mainly full of fibrous tissue and no healing happened. Both ends of the bone defects were hardened. There was significant difference statistically in the new trabecular bone area in the groups ofⅠand Ⅱ. The maximum vertical loading and elastic modulus were significant differenct in the group ⅠandⅡ. The ratios of average maximum vertical loading/normal value of radial shaft in the groups ofⅠandⅡwere 75.86% and 45.45%, respectively. Conclusion The tissue-engineered bone made by HA mixed with Adv-hBMP-2 transfected BMSCs can repair the bone defects in the radial shaft of rabbits. HA is a safe, effective carrier for tissue engineering.
9.The role of Tianlongchuankeling in modulation of the ?-smooth muscle actin induced by rhTGF-?_1 and the involved signal transduction

Objective To investigate the role of Chinese traditional medicine-Tianlongchuankeling in modulation of the transdifferentiation of sub-epithelial fibroblasts into myofibroblasts induced by rhTGF-?1,and the involved mechanism of the signal transduction.Methods Human primary bronchial sub-epithelial fibroblasts(HPBFs)were incubated with either Tianlongchuankeling or its main components of Qingtiankui,Danfuzi and Faxia respectively before being treated with rhTGF-?1(10 ?g/L)for different time.Total protein of cells were collected at different timelapse(10min,30min respectively).Phosphorylated MAPKs for p38,ERK1/2,phosphorylated smad2 and the expression of ?-SMA were detected by using Western blotting.Total RNA was extracted at 20 h and 72 h and subjected to RT-PCR for evaluation of the expression of ?-SMA gene.Results The induction of ?-SMA in HPBFs by TGF-?1 was down-regulated significantly in the cells treated with Tianlongchuankeling or Danfuzi.The phosphorylation of ERK1/2 under TGF-?1 stimulation was inhibited significantly in Tianlongchuankeling and Danfuzi group.No difference of the level in phosphorylation of p-38 and smad2 was found among groups.Conclusion Tianlongchuankeling has the capability to suppress the induction of ?-SMA by TGF-?1 through down-regulation of the phosphorylation of ERK1/2,thereby preventing the transdifferentiation of sub-epithelial fibroblasts into myofibroblasts,which may be responsible for the treatment of airway hyperresponsiveness in asthma.Danfuzi,one of main components of Tianlongchuankeling,may make a major contribution to this process.
10.An in vitro study of inhibitory activity of prothrombin to calcium oxalate crystallization

Objective To study and evaluate the inhibitory role of ?-carboxyglutamic acid(Gla)in the Gla domain of prothrombin in the formation of calcium oxalate crystals. Methods The addition of morpholine and formaldehyde to the solution of prothrombin caused the conversion of 8 Gla to ?-methyleneglutamic acid (8?-MGlu)or 2 Gla to ?-methyleneglutamic acid(2?-MGlu) ,the inhibitory activity of which to calcium oxalate crystallization was detected with seeded crystallization technique. Results The conversion of Gla to ?-MGlu leads to the decreased inhibitory activity of prothrombin to calcium oxalate crystallization,the inhibitory index being decreased from normal 14.2 to 12.8 of 2?-MGlu or 5.0 of 8 ?-MGlu prothrombin respectively. Conclusions Gla in prothrombin may play an important role to inhibit the formation of calcium oxalate calculi.
