1.Effect of kinase PLK4 on the proliferation of glioma cells in vitro

Objective:To analyze the correlation of protein kinase PLK4 with the prognosis of patients with malignant glioma and the proliferation of human glioblastoma cells. Methods:The relationship of PLK4 expression to the pathological grade and prognosis was evaluated using CGGA datasets. Designated siRNAs targeting PLK4 were constructed, and the silencing effect was identified by RT-qP-CR and Western blot analyses. The effect of PLK4 on the proliferation of tumor cells was evaluated by MTT and colony formation as-says. Results:The expression of PLK4 increased with pathological grade and was significantly related to the overall survival in patients with glioma and the prognosis of patients with high-grade glioma. MTT and colony formation assay results indicated that the prolifera-tion activity was significantly reduced after depleting PLK4. Conclusion:The expression of PLK4 is related to the prognosis of patients with glioma and can affect the proliferation activity of tumor cells. Therefore, PLK4 could be a potential therapeutic target in glioma treatment.
2.Severe encephalitis caused by infection of Rickettsia felis: a case report

Rickettsia felis is a Gram-negative prokaryotic organism that is obligatorically parasitic in cells. Cat fleas are the main vector of Rickettsia felis. Clinical symptoms of human infection with Rickettsia felis include fever, fatigue, headache, macular papules, and eschar. There are few reports of serious complications or deaths due to infection of Rickettsia felis. A confirmed case of severe encephalitis caused by Rickettsia felis infection was admitted to neurology department of Affiliated Hospital of Jining Medical University on January 29, 2020. After comprehensive treatment of antiviral by acyclovir, mannitol dehydration to reduce intracranial pressure, human immunoglobulin to regulateimmunity, minocycline hydrochloride capsule, levofloxacin mesylate and sodium chloride against Rickettsia felis infection, control of seizure and pulmonary infection, the patient's condition was improved and then discharged. By summarizing the experience and lessons in the treatment of this patient, we hope to remind everyone to strengthen the understanding of Rickettsia felis infection.
3.The clinical significance of cortical laminar necrosis on diffusion-weighted imaging at acute stage of cerebral infarction

Objective To evaluate the clinical significance of cortical laminar necrosis (CLN) on diffusion-weighted imaging at acute stage of cerebral infarction.Methods 41 patients were retrospectively investigated who were diagnosed acute cerebral infarction in area of middle cerebral artery,and they got diffusion weighted imaging ( DWI ),magnetic resonance angiography ( MRA ) and colored ultrasonic inspection of cervical arteries within 48 hours after onset,with exclusion of cardioembolism,and 5 patients also had digital subtraction angiography (DSA).The patients were classified into CLN positive group or negative group.The stenosis or occlusion of ipsolateral MCA,intracranial and extracranial ICA were compared between the two groups.Results 9 patients'DWI had CLN,among whom 7 patients present moderate or more severe stenosis of feeding artery,and 5 patients who only had CLN signal presented severe stenosis or occlusion.32 patients'DWI had not CLN,among whom 12 patients present moderate or more severe stenosis of feeding artery.The CLN positive group had a high incidence of ipsilateral intracranial ICA and(or) MCA moderate or severe stenosis or occlusion(55.6% vs 21.9%,x2 =3.85,P < 0.05 ).The 5 patients who only had CLN signal had ipsilateral feeding artery severe stenosis.A positive correlation was found between CLN and moderate or severe stenosis or occlusion of ipsilateral intracranial ICA and(or) MCA(OR:4.5,95% CI:1.0 ~ 20.2).Conclusion Patients with acute cerebral infarction and CLN on DWI were more likely to suffer from moderate or severe stenosis or occlusion of feeding large artery,and these patients should be evaluated vessel condition.
4.Clinical effects of combined fascia sheath suspension and frontal muscle flap suspension in correction of severe congenital ptosis

Objective To compare the therapeutic effect of combined fascia sheath suspension and frontal muscle flap suspension in the treatment of congenital severe congenital ptosis.Methods 46 cases (58 eyes) with congenital severe upper eyelid prolapse who were treated from March 2015 to March 2016 in Eye Hospital of Hebei Province were selected.The patients were divided into two groups:combined fascial sheath suspension group and frontalis muscle flap suspension.Comparisons of postoperative patients with eyelid height,eyelid radian,corneal exposure with eyes closed,eyelid activity,complication of the situation were carried out.And patient satisfaction survey was conducted.Results The effective rate of combined fascial sheath suspension group was 93.10%,the complication rate was 6.9%,and the patient satisfaction rate was 91.22±0.37,significantly better than the frontal muscle flap suspension group of patients (75.86 %,24.14 % and 75.05 ± 0.28,respectively).The time of closure of the eyelid was shortened,the complications were less,and the eyelid had better activity.The difference between the two groups was statistically significant (P<0.05).Conclusions Combined fascia sheath for congenital severe upper eyelid droop is significant,with fewer complications and a higher rate of satisfaction,which is worth clinical promotion.
5.Single- and double-bundle posterior cruciate ligament reconstruction under arthroscopy: a prospective cohort study

BACKGROUND:Some studies have shown that the double-bundle posterior cruciate ligament reconstruction is not superior to the single-bundle posterior cruciate ligament reconstruction, and stil has some deficiencies difficult to overcome. Which is better, double-bundle reconstruction or single-bundle reconstruction? There is no uniform conclusion. OBJECTIVE:To perform a prospective cohort study on the clinical efficacy and safety of autologous single- and double-bundle posterior cruciate ligament reconstruction. METHODS:Totaly 81 patients with posterior cruciate ligament injury were randomly divided into single-bundle reconstruction group (n=41) and double-bundle reconstruction group (n=40). The knee stability, Lysholm score, Tegner score, hospital stay, operation time, fever days and number of puncture cases were compared between the two groups before and 24 months after reconstruction. RESULTS AND CONCLUSION:Compared with the single-bundle reconstruction group, the knee stability was significantly worse in the double-bundle reconstruction group (F=4.362,P=0.000); the operation time, hospital stay and number of puncture cases were also higher in the double-bundle reconstruction group (P < 0.05). At 24 months after reconstruction, the Lysholm and Tegner scores were both increased significantly in the two groups (P < 0.05), but there was no difference between the two groups (P > 0.05). These findings indicate that both single- and double-bundle reconstruction under arthroscopy is safe and effective treatment for posterior cruciate ligament injury, but the double-bundle reconstruction is not recommended as the preferred surgical procedure because of longer time and larger trauma.
6.Comparative study on image diagnosis and pathological results of non-epithelial ovarian tumors

Objective To improve the imaging diagnosis and differential diagnosis of non-epithelial ovarian tumor.Methods 34 patients with non-epithelial ovarian tumor confirmed by pathology were analyzed retrospectively.The image features of each patients were analyzed and compared with surgical pathology.Results The results exhibited 13 cases of teratoma,6 cases of ovarian granulosa cell tumor, 6 cases of ovarian fibroma, 5 cases of thecoma, 2 cases of dysgerminoma,and 2 cases of endodermal sinus tumor.All kinds of non-epithelial ovarian tumors had its imaging manifestations and clinical features.Conclusion Non-epithelial ovarian tumors have certain characteristic CT and MR performance,and combining the onset age,clinical symptoms and biochemical examination,can make the qualitative diagnosis.
7.Imaging diagnosis of primary laryngeal amyloidosis

Objective To investigate imaging findings of primary laryngeal amyloidosis in order to improve the diagnosis of this disease.Methods Findings of plain CT and contrast enhanced CT scanning of three cases of laryngeal amyloidosis confirmed by biop-sy were investigated.Results Of the three cases,one case was located,two cases was diffused.The pharyngeal stenosis,bronchus in-vasion and calcification were showed in one case respectively.All the three cases showed mild homogenous enhancement similaryly. Of the three patients,one case was hospitalized by hoarseness and a serious sense of obstruction,and two cases by feeling pharyngeal pain.Conclusion This disease is rare ,and difficult differentiated from laryngeal carcinoma,but a correct diagnosis can be obtained by combining the CT findings with clinical manifestation.
8.Study on therapeutic effect of Cefoselis sulfate versus Cefepime on stroke-associated pneumonia in elderly patients

Objective To explore the therapeutic effect of Cefoselis sulfate and Cefepime on the stroke-associated pneumonia in elderly patients.Methods 96 cases of stroke-associated pneumonia with positive pathogen were divided into two groups treated with Cefoselis versus Cefepime respectively.Body temperature,leukocyte count,arterial partial oxygen pressure,the serum level of C-reactive protein (CRP) and bacterial clearance rate were observed before and after the treatment.The cefoselis sulfate group (n =48) or the cefepime group (n =48) was treated with 2.0 g Cefoselis sulfate or 2.0 g Cefepime every 12 hours for 7 to 10 days.Results In post-treatment versus pretreatment,both two groups showed that clinical symptoms and inflammatory indicators were improved,and the patient's body temperature,white blood cell count and neutrophil cell count,hs-CRP and arterial partial oxygen pressure were significantly decreased (all P<0.05).Arterial partial oxygen pressure was better in post-versus pre-treatment (P<0.05).The total effective rate was 87.5% in Cefoselis sulfate group and 83.3% in cefepime group (P > 0.05).The bacterial clearance rate was higher (89.6%) in Cefoselis sulfate groups than in cefepime groups (67.9%) (P<0.05).In posttreatments of Cefoselis sulfate groups versus Cefepime groups,white blood cell count and neutrophil cell count,hs-CRP and arterial partial oxygen pressure showed no statistically significant differences (all P > 0.05).Conclusions Compared with Cefepime,Cefoselis has the similar anti-infection efficacy in the treatment of stroke-associated pneumonia.While,the bacterial clearance rate and the total effective rate are better in cefoselis group than in Cefepime group.
9.The study of coronary artery morphology and left heart function of resident population in plateau

Objective To explore the features of coronary artery morphological changes and left ventricular function indicators of resident population in plateau .Methods 50 plateau healthy adults and 50 plain‐and Dalian‐healthy volunteers completed coronary CTA examination .We measured the pipe diameter of left main coronary atery and right coronary artery and their branches ,respec‐tively .Furthermore ,SV and EF value of left ventricular cardiac function indexes and the wall thickening of free ventricular wall at left ventricular end systolic were measured .Results The average diameter of proximal right coronary of plain‐and Dalian‐healthy volun‐teers thicker than plateau group ,with statistical difference ;Other coronary artery branch did not exhibit any significant between‐group differences ;The two groups did not show significant between‐group differences with heart function indexes .Conclusion The body circulation system have adaptability and compensatory changes of resident population in plateau ,and their coronary artery mor‐phology and left ventricular function are as similar as plain people .
10.Cloning,expression and bioinformatics analysis of cathepsin B of Echino-coccusgranulosus

Objective To clone and express cathepsin B gene of Echinococcus granulosus(EgCatB)and analyze EgCatB protein by using bioinformatics tools and online databases. Methods The total RNA of E. granulosus was extracted and reverse?ly transcribed into cDNA as the template sequence for PCR. The EgCatB gene was cloned by using the In?Fusion PCR cloning method and expressed by a wheat germ cell?free system,and then the recombinant protein was identified by Western blotting. The signal peptide,transmembrane helices and subcellular location of the EgCatB sequence were predicted by the online soft?ware SignalP 4.1,TMHMM sever v. 2.0 and TargetP 1.1 respectively. Subsequently,the homologue sequence and conserved sites were aligned by using BLASTP and GeneDoc software. Finally,the structures and the glycosylation modification site of the EgCatB encoding protein were analyzed and predicted in turn by ProtParam,SMART,Predictprotein,Swiss?model,NetOGlyc 4.0 and NetNGlyc 1.0 approaches. Results The EgCatB gene was successfully amplified from cDNA of E. granulosus and ex?pressed in the soluble fractions. The molecular weight of the expressed protein was estimated 35 kDa. The bioinformatics analysis revealed that EgCatB was a classical secreted protein containing a Pept_C1 domain. The homology analysis indicated that the amino acid sequence of EgCatB was highly conserved in the active enzyme sites. The protein structure prediction showed a cata?lytic active center was formed through Gln106,Cys112,His282 and Asn302. It was found that there were nine O?glycosylation sites in the EgCatB sequence,but no N?glycosylation sites. Conclusions The EgCatB gene is cloned and expressed successfully,and the recombinant protein is analyzed by bioinformatics approaches and structure predication. The study provides useful informa? tion for further functional study of the EgCatB protein.
