1.Propolis decreases the expression of MMP-9 in fibroblasts and monocyte-derived macrophage in vitro

Objective To explore the potential mechanisms of therapeutic effect of propolis in the treatment of diabetic wound. Methods Human fibroblasts(Fb) and monocytic THP-1 cells were routinely cultured at 37℃ in an environment of 95% air and 5% CO2 in RPMI medium containing 11mmol/l glucose, 15mmol/l HEPES and 5% fetal calf serum. To a portion of culturing THP-1 monocytes phorbol myristate actetate (PMA; 2?10 -7mol/L) was added. After 24 hours when approximately 75% of the monocytes became macrophage which attached to the plate. Propolis was added to a culture system of human fibroblasts(Fb), THP-1 monocyte and THP-1 monocyte-derived macrophage(Mac) with different concentration of propolis(10, 50, 100, 200?g/ml) and different glucose(5mmol/L; 25mmol/L), For each experiment the cells were cultured for 48 hours in the presence of Propolis. Then the media were collected and the protein expression of matrix metalloproternase-9 (MMP-9) were determined by gelatin zymography using a MDPF- labeled gelatin as substrate. The media were loaded onto an 8% SDS PAGE gel containing MDPF labeled gelatin (1%) after electrophoresis, the gels were placed on a long wave ultra violet light box and photographed. The area degraded by the MMPs was observed as dark bands on a light background. The MMPs were quantified by measurement of the band area using the Phoretix_1D_Advanced Digital Science Electrophoresis Documentation and Analysis System. Results the high glucose has a tendency to increase the expression of MMP-9, The propolis can decreased the expression of MMP-9, and has a dependent on doses tendency. F test P
2.Influence of lipopolysaccharide on the expression of matrix metalloproteinases and specific tissue inhibitors of metalloproteinases in fibroblasts

Objective To explore the effect of bacterial endotoxin lipopolysaccharide (LPS) on wound healing by observation of change in expression of matrix metalloproteinases (MMPs) and specific tissue inhibitors of metalloproteinases (TIMPs) in fibroblasts after the challenge of lipopolysaccharide. Methods Human dermal fibroblasts were cultured in RPMI medium containing 10% fetal calf serum (FCS). At cellular confluence the medium was changed, and the cells were cultured under serum free conditions with 0.1% bovine serum albumin (BSA) in the presence of LPS (0, 10?g/ml, 100?g/ml and 1000?g/ml). All cultures were maintained at 37℃ in a humidified atmosphere of 5% CO_2 in air. After 48-hour treatment, the medium was collected for measurement of MMPs by fluorescence labeled gelatin zymography and for measurement of TIMPs by fluorescence labeled reverse gelatin zymography. The expressions of MMPs and TIMPs were quantitatively analyzed with the band's area and density from the gel image using the Phoretix software. Results LPS up-regulated the expression of both MMPs and TIMPs in fibroblasts, and expression of TIMPs was higher than that of MMPs. Compared with base level, MMPs respectively increased 1.3, 1.7 and 1.6 times, TIMP-1 respectively increased 3, 4.5 and 3.6 times, TIMP-2 respectively increased 3, 6 and 5.6 times. Conclusion It seems that LPS showed no damaging effect on fibroblasts in regulation of matrix accumulation in wound healing.
3.Changes in abilities of invasion and metastasis in surviving human lung adenocarcinoma (A549) cells after X-ray irradiation and related mechanism

Objective To investigate the changes in the abilities of invasion and metastasis in surviving human lung adenocarcinoma (A549) cells after X-ray irradiation and the related mechanism.Methods The change in radiosensitivity after X-ray irradiation was determined by colony formation assay.The abilities of invasion and metastasis were evaluated by MTF adhesion assay and Transwell invasion and migration assay in vitro.The mRNA and protein expression of E-cadherin,vimentin,tumor growth factor (TGF)-β1,matrix metalloproteinase (MMP)-2,and MMP-9 was measured by real-time RT-PCR using SYBR Green fluorescence and Western blot.Results The colony formation assay showed that the surviving A549 cells after X-ray irradiation were more resistant to irradiation (ratio of D0 values =0.94).Their abilities of adhesion,invasion,and migration were significantly increased by 1.46 times,1.40 times,and 1.45 times,respectively.In addition,the surviving cells showed enlarged intercellular spaces and had many long pseudopodi.Compared with those in the control group,the mRNA expression levels of vimentin,TGF-β1,MMP-2,and MMP-9 of surviving cells were increased by 1.37 times,2.37 times,1.80 times,and 1.50 times,respectively,the protein expression levels of the above substances were increased by 1.60 times,1.80 times,1.10 times,and 1.20 times,respectively,and the mRNA and protein expression levels of E-cadherin were decreased by 59.4% and 74.6%.Conclusions The surviving A549 cells after X-ray irradiation have significantly increased abilities of invasion and metastasis.This may be due to radiationinduced TGF-β1 expression increase that in turn promotes epithelial-mesenchymal transition and also due to radiation-induced MMP-2 and MMP-9 expression increase.
4.Study on the association between sex hormone-binding globulin and newly diagnosed type 2 diabetes mellitus

Objective To explore the possible mechanism of sex hormone-binding globulin (SHBG) in diabetes mellitus by detecting the level of serum SHBG in patients with newly diagnosed type 2 diabetes mellitus (T2DM),and analyze the correlation between serum SHBG and insulin resistance.Methods SHBG levels were determined by enzyme linked immunosorbent assay (ELISA) in 30 normal controls (control group) and 60 newly diagnosed T2DM patients(T2DM group).HOMA-IR was calculated.Results The levels of serum SHBG in T2DM group were significantly lower than those in control group [(22.6 ±9.7)mmol/L vs.43.1 ±11.9) mmol/L,P <0.05]; SHBG level was negatively correlated with triglyceride(TG),low density hpoprotein cholesterol(LDL-C),fasting insulin(FINS),hemoglobintHbA1c) and lgHOMA-IR (r =-0.351,-0.361,-0.426,-0.645,-0.483,P <0.05),and positively correlated with high density lipoprotein cholesterol (HDL-C) (r =0.453,P <0.01).When SHBG was analyzed as a dependent variable by multiple linear regression,HbA1c (β =-0.614,P =0.000),lgHOMA-IR (β =-0.473,P = 0.014) and HDL-C (β = 0.234,P= 0.048) were into the equation.Conclusions Patients who have diagnosed T2DM show low level of serum SHBG.HOMA-IR and HbA1c are key indicators of SHBG,suggesting that SHBG may be closely related with T2DM.
5.Effect of lymphocyte active immunotherapy on patients with repeated biochemical pregnancy abortion and psychological intervention

Objective To explore the effect of the clinical result and psychological intervention of lymphocyte active immunotherapy for repeated biochemical pregnancy abortion in the patients with in vitro fertilization and embryo transfer ( IVF-ET) on the psychological reaction of the patients .Methods In the 200 cycles with IVF-ET from March 2012 to December 2012 , 28 cycles were the repeated biochemical pregnancy abortion, and the 28 patients received the lymphocyte active immunotherapy and the effective psychological intervention which can eliminate the concerns of patients before the treatment .Results In 28 patients with repeated biochemical pregnancy abortion , 15 cases were the clinical pregnancy at last , and 13 cases were the full-term delivery.Conclusions Lymphocyte active immunotherapy for repeated biochemical pregnancy abortion in the patients with IVF-ET is safe and effective , and the effective psychological intervention plays a positive role of improving the successful rate .

Objective To explore the application of nursing care of natural IVF /M cycle on patients with poor ovarian response .Methods By the observation of natural IVF/M cycle to standardize the nursing process.Results Fifty patients with low ovarian response used the natural IVF /M cycle.It avoided the adverse reactions of drugs and reduced the anxiety and nervous of patients .Conclusions Patients with low ovarian reaction can gain the eggs by using the natural IVF/M cycle.At the same time, it also can improve the professional level of medical staff and the quality of the nursing care .

Objective To explore the compound compatibility regulation of oral traditional Chinese medicines (TCM) for the prevention and treatment of infantile repeated respiratory infection.Methods The oral TCM compound literatures for the prevention and treatment of infantile repeated respiratory infection were collected from four databases,including China Biology Medicine disc,CNKI,VIP Periodical and Wanfanf Data.The retrieval time was from January 2006 to July 2006.Then the TCM oral compound prescriptions meeting the requirements were obtained by the literature arrangement and performed the terms standardized processing for extracting the information and establishing Excel table.Then the Kaiyuan software package R i386 3.3.0 was used to conduct the drug frequency,association rules analysis and clustering analysis.Results A total of 311 prescriptions were screened out and 208 kinds of traditional Chinese medicine were analyzed.It was found that the higher compatibility frequencies of TCM were atractylodes,radix glycyrrhizae,windproof,radix astragali,radix pseudostellariae,poria cocos,dried tangerine or orange peel,etc.;the strong association rules prompted that modified Yupinfeng Powder composed of core medicines such as astragalus,atractylodes and windproof was the basic formula for prevention and treatment of infantile repeated respiratory infection;the clusters analysis found some TCM societies such as emembranous Astragalus mongholicus-keel-oysters,Atractylodes-Poria cocos-licorice-Astragalus mongholicus-dangshen.Conclusion The drug frequency,association rules analysis and clustering analysis can reveal the compatibility rules of TCM prescriptions for preventing and treating infantile repeated respiratory to provide reference basis for optimizing the clinical prescription and improving the curative effect.

Adoptive cell therapy with chimeric antigen receptor (CAR)-engineered T cells (CAR-Ts) has emerged as an innovative immunotherapy for hematological cancer treatment. However, the limited effect on solid tumors, complex processes, and excessive manufacturing costs remain as limitations of CAR-T therapy. Nanotechnology provides an alternative to the conventional CAR-T therapy. Owing to their unique physicochemical properties, nanoparticles can not only serve as a delivery platform for drugs but also target specific cells. Nanoparticle-based CAR therapy can be applied not only to T cells but also to CAR-natural killer and CAR-macrophage, compensating for some of their limitations. This review focuses on the introduction of nanoparticle-based advanced CAR immune cell therapy and future perspectives on immune cell reprogramming.
9.Incidence of adverse reactions to whole blood donation in China: a Meta-analysis

【Objective】 To systematically evaluate the characteristics of adverse reactions to blood donation(ARBD) among whole blood donors in China, so as to provide scientific basis for the prevention and control of adverse reactions related to blood donation. 【Methods】 PubMed, EMbase, CNKI, VIP and WanFang Data were retrieved to collect research literature concerning ARBD among whole blood donors in mainland China with observation time≥6 months. The retrieval time was set from the time of database establishment to June 2020. The literature was supplemented by reference tracing and manual retrieval. Two reviewers independently screened the literature, extracted the data, and evaluated the methodological quality of the included studies according to the inclusion and exclusion criteria. Then Review Manager 5.3 software was used for Meta analysis. 【Results】 A total of 36 literature were included, involving 3 769 415 blood donors and 38 403 cases of ARBD. Meta-analysis results showed that the overall incidence of ARBD among whole blood donors was 1.36% [95%CI (1.19, 1.54)], and 1.52% [(95%CI (1.26, 1.78)] in the eastern, 1.26% [(95%CI (0.87, 1.70)] in the central, and 0.76% [(95%CI (0.74, 1.21)] in the western China when stratifying by geographic distribution, with statistically significant differences (P<0.01). The incidence of ARBD in male and female whole blood donors was 1.19% [(95%CI (0.99, 1.39)] and 1.26% [(95%CI (1.05, 1.48)), respectively, which was more commonly reported by women than by men (P<0.01). The incidence of ARBD was 2.17% [(95%CI (1.82, 2.52)], 0.98% [(95%CI (0.82, 1.14)] and 0.76% [(95%CI (0.61, 0.91)] in the age groups of 18~24, 25~40, and 41~55, respectively, and the incidence in donors aged 18~24 years was significantly higher than that in other two age ranges (P<0.01). The incidence of ARBD in first-time and repeat blood donors was 1.57% [(95%CI (1.35, 1.79)] and 0.63% [(95%CI (0.56, 0.70)), respectively(P<0.01). The incidence of ARBD with blood donation <400 mL and 400 mL was 1.10% [(95%CI (0.90, 1.30), P<0.001] and 0.87% [(95%CI (0.71, 1.03), P<0.001], respectively(P<0.01). 【Conclusion】 In order to improve the effectiveness of ARBD monitoring, it is necessary to unify the standards for ARBD, strengthen the knowledge training of ARBD, and accelerate the information construction to explore the national ARBD monitoring mode.

Following the principles of evidence-based medicine,in accordance with the structure and drafting rules of standardized documents,based on literature research,according to the characteristics of chronic cough in children and issues that need to form a consensus,the TCM Guidelines for Diagnosis and Treatment of Chronic Cough in Children was formulated based on the Delphi method,expert discussion meetings,and public solicitation of opinions.The guideline includes scope of application,terms and definitions,eti-ology and diagnosis,auxiliary examination,treatment,prevention and care.The aim is to clarify the optimal treatment plan of Chinese medicine in the diagnosis and treatment of this disease,and to provide guidance for improving the clinical diagnosis and treatment of chronic cough in children with Chinese medicine.
