1.Killing K562 cells by RNA interference compared with imatinib

Objective To compare RNA interference (RNAi) with imatinib in killing K562 cells. Methods Design effective shRNA sequences special for bcr-abl silencing and insert them into the eukaryotic expression vector for RNAi by gene engineering. The recombinant plasmi(ts were then transfected into K562 cells. 48 hours later, the efficiency of transfection was identified by fluorescent microscope, bcr-abl mRNA level was detected by RT-PCR. Another group of K562 cells were treated respectively by imatinib with different concentration. All groups of K562 cells were finally analyzed in apoptosis, cell proliferation and phosphotyrosine-containing proteins. Results Both RNAi and imatinib induced apoptosis, decreased proliferation and reduced phosphotyrosine-containing proteins. Conclusion BNAi can kill K562 cells successfully as imatinib, and it may be a promising way to treat CML patients in clinic, especially for those who fail in imatinib or other chemotherapy.

Objective To evaluate the relationship and bias of the Stago-CT and CA1500 automatic coagulation analyzer.Meth-ods The relationship and bias of PT,APTT,INR,FIB,TT,DD examined by the Stago-CT and CA1500 automatic coagulation ana-lyzer by using NCCLS EP9-A2.Results For the six items(PT,APTT,INR,FIB,TT,DD)the r2 were 0.996 9,0.969 1,0.967 7, 0.955 8,0.972 6,0.949 6,respectively,and the bias were 2.9,0.88,5.22,1.16,3.48,20.3.Conclusion The five items (PT, APTT,INR,FIB,TT)at a good relationship(r2 >0.95)by the Stago-CT and CA1500 automatic coagulation analyzer except for the DD(r2 =0.949 6);The bias of the five items(PT,APTT,INR,FIB,TT)were within in the United States of demanding that a third of the clinical laboratory of CLIA 88′bias,except for the DD.
3.Immune responses of silicotic rats to the antigen of sheep red blood cells.

OBJECTIVETo explore the immune response of silicotic rats to sheep red blood cells(SRBC).
METHODSSilicotic rats were immunized with SRBC by tracheal instillation(Group 1) or intraperitoneal injection (Group 2), and non-silicotic rats were immunized by tracheal instillation as normal control(Group 3). The levels of serum hemolytic index(HC50) were measured on 7, 12, 20, 25, and 32 days after primary immunization and 5, 12, 15 days after the second immunization. Special anti-SRBC IgG was measured with ELISA(A490 nm) on 12, 20, 25, 32 days and 5, 12, 15, 27 days respectively. Delayed-type hypersensitivity(DTH) to SRBC was measured 20 days after second immunization and DTH reaction was determined at 24, 48, 72, and 96 h after administration. Total cell count and cell populations in the bronchoalveolar lavage fluid(BALF), lung associated lymph node(LALN) and spleen weight, special IgG secreted from spleen cells were measured at the end of the experiment.
RESULTSThe HC50 of Group 1(47.4 +/- 1.0, 52.2 +/- 4.6, 31.1 +/- 11.9, 43.8 +/- 3.5, 33.6 +/- 16.8, 49.0 +/- 2.3, 92.9 +/- 20.2, 87.7 +/- 5.2) were statistically higher than those of Group 3(40.4 +/- 10.6, 2.8 +/- 2.5, 0.8 +/- 0.6, 6.6 +/- 5.8, 1.4 +/- 0.1, 36.5 +/- 16.5, 53.0 +/- 33.2, 2.6 +/- 2.2). The special anti-SRBC IgG response in Group 1(1.67 +/- 0.19, 1.98 +/- 0.36, 1.12 +/- 0.50, 1.38 +/- 0.30, 2.75 +/- 0.15, 2.60 +/- 0.28, 2.86 +/- 0.10, 2.50 +/- 0.20) were much stronger than those in Group 3 (0.59 +/- 0.30, 0.56 +/- 0.21, 0.21 +/- 0.16, 0.22 +/- 0.01, 0.81 +/- 0.25, 0.74 +/- 0.25, 0.69 +/- 0.26, 1.38 +/- 0.41). Furthermore, the results of DTH showed positive response and the ratios for diameter of skin rash > 5 mm at 24, 48, 72, 96 h were 16/16, 16/16, 16/16, 15/16 respectively in Group 1, while those in Group 3 were 8/15, 1/15, 1/15, 1/15 respectively. Total cell count in the BALF, LALN and spleen weight, and special IgG secreted from spleen cells in Group 1 were higher too. Group 2 expressed almost of the same but with mild immunologic responses as Group 1.
CONCLUSIONSilicosis-induced extremely strong DTH and over-response of humoral immunity to some antigens may contribute to the likelihood of silicosis complicated with tuberculosis.

4.The mediating effect of health belief on the relationship between health knowledge and health behavior among hypertensive patients with stroke

Objective To explore the mediating effect of health belief on the relationship between health knowledge and health behavior among hypertensive patients with stroke.Methods The questionnaire of Stroke Knowledge Questionnaire (SKQ),Short Form Health Belief Model Scale (SF-HBMS)and health behavior scale for stroke patients (HBS-SP) were used to investigate the health knowledge,health belief and health behavior among 151 hypertensive stroke patients.The structural equation model was established using SPSS 17.0 and Mplus7.0 software.Results The total score of SKQ,SF-HBMS and HBS-SP were (86.23±.14.67),(4.26±0.54) and (3.01 ± 0.46).The Pearson r value for SKQ and SF-HBMS,SF-HBMS and HBS-SP,SKQ and HBS-SP were 0.516,0.603 and 0.449,respectively,P<0.01.Health knowledge had no direct effect on health behavior.but had an indirect effect through health belief=0.338 (P<0.01).The health knowledge has direct and positive effect on the health belief including perceived susceptibility,health motivation,and perceived benefit to health behaviors;the health belief of perceived seriousness to stroke has direct and positive effect on the health behavior,while perceived barrier has direct and negative effect on the health behavior,all P<0.05.Conclusion The health belief has mediator effect between health knowledge and health behavior among hypertensive stroke patients.Stroke education efforts should be targeted at not only health knowledge but also health belief in order to increase patients’health behavior so as to prevent secondary stroke.
5.Transplantation of peripheral purified CD34+ cells in treatment of thromboangiitis obliterans

Objective To evaluate the safety and efficacy of transplantation of purified CD34 + cells (PCCs) in treatment of critical limb ischemia (CLI) caused by thromboangiitis obliterans (TAO).Methods From May 2009 to June 2015,34 TAO-induced-CLI cases underwent PCCs transplantation.None of these patients were eligible for surgical or endovascular revascularization.G-CSF was subcutaneously injected for 5 days before peripheral CD34 + cells were isolated,purified and intramuscularly injected in the limbs.Patients were regularly follow-up.Results Technical success was achieved in all cases.The mean number of transplanted cells was (7.5 ± 2.4) × 105/kg.The follow-up was accomplished in 32 cases,ranging from 6 to 79 months (mean 45 ±24 months),and two patients were lost.Wong-Baker FACES pain rating scale score significantly decreased from 8.0 ±2.0(4-10)to 2.2 ±3.1 (P <0.05) at 1 month.The Peak pain-free walking time improved from (4.0 ± 2.0) min to (13.5 ± 5.3) min (P < 0.05) at 3 months and (19.0 ± 3.1) min (P < 0.05) at 6 months.The ankle-brachial index increased from 0.42 ± 0.20 to 0.50 ± 0.10 (P < 0.05) at 3 months and 0.52 ± 0.11 (P < 0.05) at 6 months,respectively.Transcutaneous partial oxygen pressure rose from (25 ± 11) mmHg to (48 ± 11) mmHg(P < 0.05) at 3 months and (58 ± 10) mmHg (P < 0.001) at 6 months,respectively.Ulcers healed in 21 out of 22 patients at (5 ± 4) months.The overall amputation-free survival rate was 94.1% at 6 months and 91.2% at 48 months.No major adverse events were observed perioperatively or postoperatively.Conclusions Transplantation of PCCs could yield safe,satisfactory and durable treatment outcomes in patients with TAO-induced-CLI.
6.Value of preoperative alanine aminotransferase/aspartate aminotransferase combined with multi-phase CT radiological indicators in predicting clinically relevant pancreatic fistula after pancreaticoduodenectomy

Objective To investigate the risk factors for clinically relevant postoperative pancreatic fistula(CR-POPF)after pancreaticoduodenectomy(PD),and to establish a predictive model for early identification of CR-POPF.Methods A total of 244 patients who underwent PD in General Hospital of Northern Theater Command from January 2019 to October 2023 were collected,and based on strict inclusion and exclusion criteria,179 patients were finally enrolled in this study.According to the presence or absence of CR-POPF,these patients were divided into non-CR-POPF group with 120 patients and CR-POPF group with 59 patients.Univariate and multivariate logistic regression analyses were used to determine the independent risk factors for CR-POPF,and a nomogram model was established based on such factors.The receiver operating characteristic(ROC)curve was used to assess the predictive performance of the model,the calibration curve was used to evaluate the calibration degree of the model,and the clinical decision curve and the clinical impact curve were used to analyze and validate the clinical application value of the model.The chi-square test or the Fisher's exact test was used for comparison of categorical data between groups;the independent-samples t test was used for comparison of normally distributed continuous data between two groups,and the Mann-Whitney U test was used for comparison of continuous data with skewed distribution between two groups.Results Among the 179 patients,59(33.0%)developed CR-POPF.The multivariate Logistic regression analysis showed that alanine aminotransferase/aspartate aminotransferase(odds ratio[OR]=2.221,P=0.004),main pancreatic duct diameter(OR=0.276,P=0.022),the distance between the peritoneum and the anterior pancreatic neck(OR=1.034,P=0.027),and extracellular volume fraction(OR=0.001,P=0.005)were independent risk factors for CR-POPF.Based on the above four independent risk factors,a nomogram was established to predict CR-POPF after PD,with an area under the ROC curve of 0.837,a sensitivity of 0.932,and a specificity of 0.725.The decision curve and the clinical impact curve also showed that the nomogram had good clinical practicability.Conclusion Preoperative clinical indicators combined with multi-phase CT have a good performance in predicting CR-POPF after PD,which can be used to early identify patients at high risk of pancreatic fistula before surgery and provide further guidance for clinical work.
7.Psychometric properties of Chinese version of post-event processing inventory trait in high school students

Objective:To Chinesize Post-Event Processing Inventory Trait (PEPI-T) and verify the reliability and validity of its application in high school students.Methods:First, the PEPI-T was translated and pre-measured, then the wording according to the pre-measurement results was adjusted and back-translated.Then, the Chinese version of PEPI-T(PEPI-T-C) was finalized.In total, 947 adolescents aged from 15 to 19 were recruited to complete Social Anxiety Scale for Adolescents, Short Mood and Feelings Questionnaire, General Self-Efficacy Scale and PEPI-T-C, which were used as validity indicator.Results:Confirmatory factor analysis results showed that the PEPI-T-C still followed the three dimentions of self-judgement, frequency and intensity of the original scale and showed good model fit indices (χ 2/ df=2.59, RMSEA=0.04, CFI=0.97, TLI=0.95, SRMR=0.03). The internal consistency coefficients for PEPI-T-C was 0.78, while the three subscales was (0.44-0.75). PEPI-T-C and its subscales were significantly correlated with social anxiety and depression ( r=0.22-0.43, all P<0.01). The subscale of intensity showed significant correlation with self-efficacy( r=-0.11, P<0.01). PEPI-T-C and its subscale of self-judgement and frequency showed no significant correlations with self-efficacy( P>0.05). Conclusions:PEPI-T-C showed an acceptable reliabilities and validities in Chinese high school students, and would be used to assess their post-event rumination.Future research should examine the psychometric properties of PEPI-T-C in other Chinese adolescents’ samples.
