1.THE OBSERVATION OF IMMUNOMORPHOLOGICAL CHANGES IN THE DRAINING LYMPH NODES OF MICE AFTER IMPLANTATION OF TUMOR CELLS INTO FOOTPAD

The response of murine lymphoreticular tissue with Special reference to regional lymph node after implantation of Ehrlich ascites carcinoma cells was investigated histologically and ultrastructurally. The tumor induced a vigorous histological response in both regional and nonregional lymph nodes that was proliferation of immunoblasts with enlargement of paracortical area in early stage and enlargement, fusion of germinal centers with an increased number of plasma cells in the medullary cords during the late stage of tumor growth.The results suggested that the response of lymph nodes might be mainly related to cell-mediated immune mechanism in the early stage and humoral immunity during the late stage of tumor growth. Regional lymph node gave reaction earlier and more vigorously in participating cell-mediated immunity, nonregional lymph node might be more significantly involved in humoral immunity. It was observed ultrastructurally that sinus histiocyte engulfed tumor cell in which the cooperation with lymphocyte might be necessary. The experimental results indicated that the increased number of immunoblast in paracortex may be considered as an indicator for antitumor cellmediated immunity with an important prognostic significance for host bearing tumor.
2.Exploration in the teaching of forensic medicine for international students

Basic knowledge of forensic science is theoretical and abstract,therefore,confir-matory experiment teaching is often adopted in the experiment teaching. International education has become an important topic in medical colleges and universities. Forensic medicine is a compulsory subject for medical international students. This article discussed the exploration and practice in teach-ing forensic medicine for international students at Wenzhou Medical University,which included faculty constructing and training,encouraging whole English teaching by young teachers,applying heuristic interactive teaching and case teaching,establishing strict attendance and appraisal system and con-ducting comprehensive evaluation on medical teaching quality and teaching management.
3.Effect of double filtration plasmapheresis in refractory rheumatoid arthritis

Objective To evaluate the efficacy of double filtration plasmapheresis (DFPP) in the treatment of patients with refractory rheumatoid arthritis (RA). Methods Eighty-two patients were randomly aesigned,42 to the DFPP group and 40 to the no-DFPP group. All patients previously experienced an incomplete response to 2-3 dis-ease-modifying antirheumatic drugs (DMARDs) and 1-2 nonsteroidal anti-inflammatory drugs (NSAIDs) or predni-sene. All patients received sulphasalazine (SASP,0.75 g three times daily) plus methotrexate (MTX, 10 mg orally once weekly). DFPP was performed once a week for 2-3 sessions. A total of 121 plasmapheresis procedures were per-formed in 42 patients. Control patients did not receive sham DFPP. The efficacy measures recorded one day after the final treatment and latest month in follow up for 12~24 months included the American College of Rheumatology 20% ,50% ,and 70% improvement criteria (ACR20, ACR50, and ACR70), the Health Assessment Questionnaire estimate of disability (HAQ); and the disease activity index. Results Patients in the DFPP group had ACR 20, ACR 50 and ACR 70 improvements of 100% ,92.9% and 81.0%,as compared with the patients in no-DFPP group 17.5% ,0,and 0 (P<0.001). Significant change from baseline was observed in HAQ scores in the DFPP group but not in the no-DFPP group (P<0.001). The changes from baseline in the disease activity scores were significantlygreater than in the no-DFPP group (P<0.001). Conclusion DFPP therapy significantly alters the signs and symp-toms of refractory RA. There are significant increases in physical function and improvement in quality of life.
4.Detection and analysis of specific antibodies of food allergens in patients with allergic rhinitis

[ABSTRACT]OBJECTIVETo investigate the relationship between food allergen specific IgE, IgG and allergic rhinitis, and to explore the significance of food allergen in the diagnosis and prevention of allergic rhinitis.METHODSTen kinds of food specific IgE were detected by Western blot and 14 kinds of food specific IgG were detected by ELISA in 2860 allergic rhinitis patients. The positive results of specific IgE and specific IgG were retrospectively analyzed.RESULTSThere was no difference in positive rate of allergen in patients with different gender. Total positive rate of food specific IgE was 68.5%. The specific IgE positive rate of crab was 31.5%, shrimp 27.6%, milk 21.3%, fish assemblages 19.2%, freshwater fish assemblages 18.3% and soybean 17.2%. With the growth of age, the specific IgE positive rate of peanut increased, but eggs decreased. The total positive rate of food allergen specific IgG was 81.5%. The specific IgG positive rate of egg was 59.4%, milk 38.2%, codfish 32.6%, soybean 22.8%, rice 19% and shrimp 12.7%. With the growth of age, the specific IgG positive rate of crab increased. Specific IgE and IgG of soybean, milk, crab, shrimp and fish were correlated well.CONCLUSIONThe detection of specific food allergen antibodies is helpful for the diagnosis of allergic rhinitis.
5.Instability of anthocyanin composition under different subculture conditions during long-term suspension cultures of Vitis vinifera L. var. Gamay Fréaux.

The instability of secondary metabolite production is a ubiquitous problem in plant cell culture. In order to understand the instability in plant cell culture, we investigated anthocyanin accumulation in suspension cultures of Vitis vinifera, as a model system, in our laboratory. Not only the anthocyanin contents but also its composition exhibited instability along with the long-term subculture. New methods were developed to indicate the instability of plant cell culture. Both the definition of instability coefficient (delta) and the application of factor scores were the first time in this field. To examine the effects of culture conditions on instability of anthocyanin biosynthesis, different subculture cycles and inoculum sizes had been investigated. Subculture cycle and inoculum size were both environmental cues driving the instability. Compared with subculture cycle, inoculum size was more effective in working on the instability of anthocyanin accumulation. Among all the conditions investigated in our study, (6.5 d, 2.00 g), (7 d, 2.00 g), (7.5 d, 2.00 g), (7 d, 1.60 g) and (7 d, 2.40 g), the condition of 7 d-subculture cycle together with 1.60 g-inoculum size was the best one to keep the stable production of anthocyanins.

6.Mechanism of Dan Hong Treatment of Cervical Spondylotic Myelopathy

Objective To investigate the therapeutic mechanism of Dan Hong in a rat model of cervical spondylotic myelopathy (CSM).Methods Thirty Sprague-Dawley rats were divided into an experimental group (n=10),aCSM modelgroup (n=10),and acontrol group (n=10).The CSM model group received a Dan Hong injection,while the control and experimental groups received an injection of an equivalent volume of physiological saline.The motor function of the rats was assessed before administration and at 4 weeks after administration,at which time the rats were sacrificed.The expression of cytokines and bcl-2 was detected by ELISA and Westem blotting.Results Four weeks after treatment,the number of oblique plates in the experimental group increased.Additionally,the expression of cytokines (IL-6 and TNF-α) in the CSM model group was higher and the expression of bcl-2 was lower.Conclusion Dan Hong injection can reduce the inflammatory response and reduce cell apoptosis in CSM.
7.Interaction between implants and natural teeth in patients with severe periodontitis:a retrospective study

Objective:To evaluate the clinical outcomes after implant restoration in the posterior region of severe periodontitis patients and to investigate the factors of natural tooth affecting the implant from the perspective of improving natural periodontal health, which may provide a reference for clinical practice.Methods:Fifty-three patients with severe periodontitis who visited the Department of Periodontology at the Affiliated Stomatological Hospital of China Medical University from June 2014 to June 2023 and completed posterior implant treatment with single crown were included, among which were 16 males and 37 females, aged (52.2±8.0) years old, with a total of 136 implants, 135 adjacent natural teeth in the edentulous area. We retrospectively compared the changes of probing depth (PD), bleeding on probing (BOP) and tooth mobility (TM) before and after implant placement. Besides, we explored the effects of the natural periodontal status on PD, BOP and marginal bone loss (MBL) of the implant at the last follow-up examination by univariate analysis and multivariate analysis.Results:Fifty-three patients were followed up for (44.5±14.1) months in average, with longest interval of (8.3±2.7) months. The PD of adjacent natural teeth in the edentulous area improved from 4.3 (3.6, 4.6) mm before implantation to 3.6 (3.2, 4.0) mm in the last review ( P<0.01), while the proportion of BOP (+) improved from 69.6% (94/135) before implantation to 46.7% (63/135) in the last review ( P<0.01). The proportion of teeth with mobility≥Ⅱ decreased from 15.6% (21/135) to 5.9% (8/135) ( P<0.01). The percentage of natural teeth with PD≥4 mm in the same segment improved from 21.0% (13.3%, 26.0%) before implantation to 18.0% (12.0%, 25.0%) in the last review ( P<0.05). The BOP (+)% improved from 29.0% (24.0%, 35.0%) before implantation to 23.0% (18.0%, 31.0%) in the last review ( P<0.05), and the number of teeth with mobility≥Ⅱ decreased from 0.0 (0.0, 1.0) to 0.0 (0.0, 0.8) ( P<0.05). The functional tooth unit score of full natural teeth increased from 8.0 (6.0, 10.0) points before implantation to 12.0 (12.0, 12.0) points in the last review ( P<0.01). PD≥4 mm % increased from 11.0% (6.0%, 25.0%) before implantation to 13.0% (3.0%, 21.0%) in the last review ( P<0.05) and there was no significant differences in BOP (+)% [(17.0±9.7) % vs (14.6±7.2) %, P>0.05]. The number of teeth with mobility≥Ⅱ decreased from 1.0 (0.0, 1.8) to 0.0 (0.0, 0.8) ( P<0.05). Conclusions:Under the premise of regular supportive care, implant restorative treatment in the posterior region of severe periodontitis patients is helpful to improve the PD, BOP and TM of remaining natural teeth. Besides, the stages and grades of periodontitis at initial diagnosis can affect the PD and BOP of implants.
8.Optimization of drying methods of Compound Scutellaria baicalensis extract powder

OBJECTIVE To optimize the drying methods of Compound Scutellaria baicalensis extract powder. METHODS Compound S. baicalensis extract powder was prepared by atmospheric pressure drying, decompression drying and spray drying respectively, and high-performance liquid chromatography (HPLC) fingerprint of the extract powder was established. The physical fingerprint of the extract powder was established by 13 secondary indexes, such as particle size, particle size distribution and bulk density. HPLC fingerprints and physical fingerprints of 3 kinds of extract powders were compared by similarity evaluation method; through the conversion of the secondary indexes, five primary indexes of homogeneity, stacking, compressibility, fluidity and stability were calculated, and the physical properties of 3 kinds of extract powder were evaluated. The compressibility parameters of 3 kinds of extract powder such as index of parameter (IP), index of parametric profile (IPP) and index of good compression (IGC) were calculated by the secondary index to evaluate the compression formability of the powder. RESULTS The similarity means of HPLC fingerprint of the extract powder obtained by atmospheric pressure drying, decompression drying and spray drying were 0.74, 0.90 and 0.94, respectively, and the similarity means of physical fingerprint were 0.74, 0.83 and 0.92, respectively. The overall similarity of spray drying extract powder was higher. The results of physical fingerprint analysis showed that the physical properties of the extract powder obtained by different drying methods were different. The 3 kinds of extract powder showed poor stability but good stacking. The homogenity and compressibility of extract powder by spray drying and decompression drying were better than those by atmospheric pressure drying, but the fluidity was worse than that by atmospheric pressure drying. The results of further compression formability analysis showed that the IP of spray drying extract powder was 0.54, IPP was 5.23, IGG was 5.03, and the spray drying extract powder could be directly pressed after adding a small amount of lubricant. CONCLUSIONS There are differences in HPLC and physical fingerprints of Compound S. baicalensis extract powder obtained by different drying methods.
