1.THE OBSERVATION OF IMMUNOMORPHOLOGICAL CHANGES IN THE DRAINING LYMPH NODES OF MICE AFTER IMPLANTATION OF TUMOR CELLS INTO FOOTPAD

The response of murine lymphoreticular tissue with Special reference to regional lymph node after implantation of Ehrlich ascites carcinoma cells was investigated histologically and ultrastructurally. The tumor induced a vigorous histological response in both regional and nonregional lymph nodes that was proliferation of immunoblasts with enlargement of paracortical area in early stage and enlargement, fusion of germinal centers with an increased number of plasma cells in the medullary cords during the late stage of tumor growth.The results suggested that the response of lymph nodes might be mainly related to cell-mediated immune mechanism in the early stage and humoral immunity during the late stage of tumor growth. Regional lymph node gave reaction earlier and more vigorously in participating cell-mediated immunity, nonregional lymph node might be more significantly involved in humoral immunity. It was observed ultrastructurally that sinus histiocyte engulfed tumor cell in which the cooperation with lymphocyte might be necessary. The experimental results indicated that the increased number of immunoblast in paracortex may be considered as an indicator for antitumor cellmediated immunity with an important prognostic significance for host bearing tumor.
2.Clinical Observations on the Treatment of Senile Skin Pruritus with Yin-reinforcing and Blood-nourishing Drink Plus Electroacupuncture

Purpose: To investigate the clinical effect of Yin-reinforcing and Blood-nourishing Drink plus electroacupuncture in the treatment of senile skin pruritus. Methods: Eighty patients were randomly divided into two groups. The treatment group was treated with Yin-reinforcing and Blood-nourishing Drink plus electroacupuncture and the control group, with Western medicine. Results: The total effective rate was 100.0% in the treatment group and 67.5% in the control group (P<0.01). Conclusion: This therapy has a better effect on senile skin pruritus.
3.Biosynthesis of indigo and indirubin by whole-cell catalyst designed by combination of protein engineering and metabolic engineering.

The phenylacetone monooxygenase, isolated from Thermobifida fusca, mainly catalyzes Baeyer-Villiger oxidation reaction towards aromatic compounds. Met446 plays a vital role in catalytic promiscuity, based on the structure and function of phenylacetone monooxygenase. Mutation in Met446 locus can offer enzyme new catalytic feature to activate C-H bond, oxidizing indole to finally generate indigo and indirubin, but the yield was only 1.89 mg/L. In order to further improve the biosynthesis efficiency of the whole-cell catalyst, metabolic engineering was applied to change glucose metabolism pathway of Escherichia coli. Blocking glucose isomerase gene pgi led to pentose phosphate pathway instead of the glycolytic pathway to become the major metabolic pathways of glucose, which provided more cofactor NADPH needed in enzymatic oxidation of indole. Engineering the host E. coli led to synthesis of indigo and indirubin efficiency further increased to 25 mg/L. Combination of protein and metabolic engineering to design efficient whole-cell catalysts not only improves the synthesis of indigo and indirubin, but also provides a novel strategy for whole-cell catalyst development.

4.Clinical efficacy of botulinum toxin type A injected by BellaVita in treatment of focal hyperhidrosis

Objective To explore the new way of administration and clinical effect of botulinum toxin A in the treatment of focal hyperhidrosis.Methods The clinical efficacy was observed in 132 sites of 28 patients with focal hyperhidrosis,and the degree and range of focal hyperhidrosis were determined by the minor iodine-starch test.50 U of botulinum toxin A was injected in unilateral axillary,palms and soles with BellaVita instrument and 30 U for forehead.Each patient was followed-up in 1 week,2 weeks and every month after injection for 8 months.According to the results of the minor iodine-starch test the objective effect and evaluation score were obtained,and the comprehensive effect evaluation score was calculated with the objective effect evaluation score and the subjective effect evaluation score in each follow-up.Results The comprehensive effect evaluation score before injection of botulinum toxin A was 1.34±3.94,and that after injection was 23.21±9.44 for 1 week,92.41±11.95 for 1 month,98.21±5.60 for 2 months,95.98±5.94 for 3 months,and 86.61±10.17 for 4 months,respectively.Compared with that before injection,the difference was statistically significant (P <0.05).The effect decreased slowly after 4 months of injection,and the efficacy was maintained for 8 months (4.46±6.98);compared with that before injection,the difference of the clinical efficacy was not statistically significant (P >0.05).Based on the comprehensive effect evaluation scores,the differ ence of the clinical efficacy was not statistically significant between 1 week and 6 months after injection (P>0.05).Conclusions The clinical effect of botulinum toxin A injected by BellaVita is prompt and effective for focal hyperhidrosis.
5.Merkel cell carcinoma polyomavirus-positive primary cutaneous Merkel cell carcinoma: two case reports

Objective To estimate the value of detection of Merkel cell carcinoma polyomavirus (MCPyV)in the diagnosis of Merkel cell carcinoma (MCC).Methods Two cases of MCC were studied using light microscopy and immunohistochemistry.PCR was performed to detect DNA sequences encoding MCPyV large T antigen(LT)and viral protein 1 (VP1) in paraffin-embedded tissue specimens from the two patients with MCC,five patients with T cell lymphoma,two normal human controls,as well as in two T cell lymphoma cell lines MAC 1 and MAC2.DNA sequencing was also carried out.Results Both of the patients with MCC were male.The patient 1 presented with a mass in the right anterior shin for more than one year,and the patient 2 had a mass in the left knee for more than six months.Skin examination revealed densely distributed pink nodules in the right anterior shin,with confluence into an indurated plaque which measured 10 cm × 8 cm with superficial erosion,exudates and crusts and was surrounded by multiple irregularly sized erythematous nodules with limited mobility in the patient 1,as well as a royal blue,hard,poorly marginated nodular mass measuring 5 cm × 4 cm in the left medial knee with limited mobility in the patient 2.Pathological manifestations were similar in the two patients.Tumor cells were uniform with large hyperchromatic nuclei,eosinophilic and sparse cytoplasm,and fine chromatin.Mitotic figures were easily seen.Immunohistochemistry revealed that the tumor cells stained positive for pan-cytokeratin,synuclein (Syn),neuron-specific enolase (NSE),chromogranin (CgA),CK20,and Ki-67 (> or =60%),but negative for S100 protein,HMB45,CD34,thyroid transcription factor 1 (TTT-1),CK7 and leukocyte common antigen (LCA).MCPyV DNA was detected in both MCC specimens,but absent in the other skin specimens or T cell lymphoma cell lines.Conclusions MCC has distinctive clinical and pathological appearance.Immunohistochemistry and detection of MCPyV DNA sequences using PCR may be beneficial to the definitive diagnosis of MCC.
6. Effects and mechanism of allogeneic platelet rich plasma on collagen synthesis in wound healing

Objective:
To investigate the effects and mechanism of allogeneic platelet rich plasma (PRP) on collagen in wound surface at different time.
Methods:
A total of 50 clean 7-week rats were selected for this study, including 10 rats for platelet-rich blood plasma preparation, 20 rats for PRP group and 20 rats for control group, 0.1 ml allogenic PRP and 0.1 ml saline were smeared respectively on wound surfaces of PRP and control group, wound regeneration and healing were examined. Cellular and histological morphology alteration was observed via Masson staining, type Ⅰ and type Ⅲ collagen protein and mRNA expression level were detected by Western blot and real-time PCR. T test was applied for comparison between two samples and one-way ANOVA was utilized for comparison between two groups.
Results:

7.Mechanism of Dan Hong Treatment of Cervical Spondylotic Myelopathy

Objective To investigate the therapeutic mechanism of Dan Hong in a rat model of cervical spondylotic myelopathy (CSM).Methods Thirty Sprague-Dawley rats were divided into an experimental group (n=10),aCSM modelgroup (n=10),and acontrol group (n=10).The CSM model group received a Dan Hong injection,while the control and experimental groups received an injection of an equivalent volume of physiological saline.The motor function of the rats was assessed before administration and at 4 weeks after administration,at which time the rats were sacrificed.The expression of cytokines and bcl-2 was detected by ELISA and Westem blotting.Results Four weeks after treatment,the number of oblique plates in the experimental group increased.Additionally,the expression of cytokines (IL-6 and TNF-α) in the CSM model group was higher and the expression of bcl-2 was lower.Conclusion Dan Hong injection can reduce the inflammatory response and reduce cell apoptosis in CSM.
8.Study on molecular mechanism of etomidate by promoting expression of miR-142-3p to reduce hypoxia-induced neuro-inflammatory response and cell apoptosis in PC12 cells

Objective:To investigate whether etomidate affects inflammatory response and apoptosis of PC12 cells induced by hypoxia by regulating miR-142-3p.Methods:PC12 cells were pretreated with different doses(2,6,12 μmol/L)of etomidate to establish hypoxia model;PC12 cells that transfected with miR-142-3p mimics or inhibitors were pretreated with 0 or 12 μmol/L of etomidate to establish hypoxia model.Cell viability,apoptosis and protein(CyclinD1,Cleaved-caspase-3)expressions were detected by CCK-8 method,flow cytometry and Western blot,respectively.ELISA was used to detect levels of inflammatory factors TNF-α,IL-1β,IL-6.Expression of miR-142-3p was detected by RT-qPCR.Results:Etomidate increased hypoxia-induced PC12 cells activity and expres-sion of CyclinD1 protein and miR-142-3p,while decreased cell apoptosis rate,Cleaved-caspase-3 protein expression and levels of inflammatory factors TNF-α,IL-1β,IL-6(P<0.05).Up-regulation of miR-142-3p increased activity and expression of CyclinD1 pro-tein of hypoxia-induced PC12 cells,while decreased cell apoptosis rate,Cleaved-caspase-3 protein expression and levels of inflamma-tory factors TNF-α,IL-1β,IL-6(P<0.05).Down-regulation of miR-142-3p reversed effects of etomidate on hypoxia-induced PC12 cell activity,apoptosis and expressions of inflammatory factors(P<0.05).Conclusion:Etomidate can reduce inflammatory response and apoptosis of PC12 cells induced by hypoxia,and its mechanism may be related to the up-regulation of miR-142-3p expression in cells.
