3.Clonality of Large Regenerative Nodule Accompanied by Hepatocellular Carcinoma.

In order to clarify the preneoplastic nature of large regenerative nodules without dysplastic change, we analysed the clonality of hepatocellular carcinomas (HCCs) and large nodules, diameter > or =0.5 cm, of cirrhotic liver by X-linked human androgen receptor (HUMARA) gene assay, using the principle of random X chromosome methylation and inactivation in female. Ten cases of HCC and 5 cases of large nodules without dysplasia from 9 female patients were selected. All the tumors, large nodules and paired normal control cells were selectively microdissected from deparaffinized hematoxylin and eosin stained slides. Genomic DNA was isolated and digested with HhaI. Polymerase chain reaction(PCR) amplication of the HUMARA locus was performed using 32P-a-dCTP containing PCR mixtures. The PCR amplified products were separated by gel electrophoresis and analysed by autoradiography. Nine HCCs from 8 patients were monoclonal and 1 case was polyclonal and the remaining 1 case was not polymorphic at the HUMARA locus. The HCC case which showed polyclonality contained many inflammatory cells. All the large nodules were polyclonal by HUMARA assay. These results suggest that all or most of the cells composing the large regenerative nodules without dysplasia are polyclonal. This assay may be informative for the differentiation between regenerative and preneoplastic nodules in cirrhotic liver and the size of nodule may be not important in hepatocarcinogenesis.

Letterer-Siwe disease occurred in a 3 year 7 month old male child showing severe emaciation, hepatosplenomegaly, otitis media, hemorrhagic and impetiginous crusted patches on the body, and seborrheic dermatitis like skin rashes in the scalp. Also showed are severe anemia, thrombocytopenia, bronchopneumonia and multiple bony defects in skull. The child expired in a month after admission.

7.The Effects of SCH-T2 Seaweed Extract.

BACKGROUND: Melanocytes synthesize melanin pigment by the action of specific enzyme tyrosinase. Melanogenesis inhibitors such as ascorbic acid, kojic acid, arbutin, hydroquinone have been developed for use in cosmetic preparations for the skin hyperpigmentation,but they are still unsatisfactory to efficacy and tolerance. OBJECTIVES: In this study, the inhibitory effects of seaweed extract(SCH-T2)on tyrosinase activity and melanogenesis were investigated with B-16 melanoma cell line. METHODS: The patients were randomly assigned to one of three groups;control, diazepam, and ketorolac plus diazepam groups. Pain intensity was assessed 5 times at every ten minutes from the beginning of the peeling using visual analog scale(VAS). RESULTS: The seaweed was added with 21 organic solvents and extracted during 12 hours at 20-To evaluate the inhibitory effects of SCH-T2 on tyrosinase activity following to extraction time and temperature, this study was done at 4, 8, 12, 24, 72 hours and 4, 10, 20, 30, 40℃. Also, the effects of whitening agents (kojic acid, arbutin, licorice extracts,SCH-T2)on tyrosinase activity were compared by measuring the IC50,the concentration of the compound at which half of the original tyrosinase activity is inhibited. RESULTS: 1.The inhibitory effects of SCH-T2 on tyrosinase activity were high at 60-80% ethanol as an extraction organic solvent and showed increase in proportion to concentration following to extract concentration, but showed little differences following to extraction time and temperature. 2.SCH-T2 has relatively stronger inhibitory effects on tyrosinase activity than arbutin and licorice extracts. 3. The inhibitory effects of SCH-T2 on melanogenesis following to extract concentration were greatly increased at more than 20㎍/ml. CONCLUSION: This study showed that SCH-T2 seaweed extract has strong inhibitory effect on tyrosinase activity and melanogonesis. So, the results of this study suggest that SCH-T2 seaweed extract can be used as a new whitening agent.

9.The Study for the Outflow Of Aqueous Humor after Injection of Dye into the Anterior Chamber.

The aqueous bumor makes an important role to maintain intraocular pressure. The production and outflow of aqueous humor controls the intraocular pressure. The accurate understanding of the pathway of aqueous outflow and the rate of outflow is essential to the study of glaucoma. The purpose of this study is to investigate the pathway of aqueous outflow after injection of dye into the anterior chamber, the duration of staining and the degree of delay of aqueous outflow after the use of viscoelastics. Three dyes. Gentian Violet, Methylene Blue and Viscoelastics mixed with Gentian Violer were injected into the anterior chambers of 20 eyes of 10 rabbits. The eyes were enucleated at 1 hour, 2 hour, 3 hour, 4 hour after injection of Gentian Violer and Methylene Blue. After injiection of viscoelastics mixed with Gentian violet, the eyes were enucleated at 2 hour, 4 hour, 6 hour, 8 hour, and 9 hour. The enucleated eyes were performed for frozen section and the fragments were observed by light microscopee. Gentian violet and Merhylene Blue were stained in the pathway of the pectinate ligaments, trabecular meshwork, intrascleral plexus and the pathway of the capillary network of iris and suprachoroidal space. The disappearance of injected dye was ended between 3 hour and 4 hour. And the viscoelastics mixed with dye was removed between 8 hour and 9 hour. This means that viscoelastics make the outflow of aqueous delay.
