OBJECTIVE To develop the strategies for preventing nosocomial infection in the department of(laboratory).METHODS The current situation in nosocomial infection management in department of laboratory was analyzed.RESULTS There were some problems in training knowledge of nosocomial infection,preparing(equipment) and supplies for prevention,implementing prevention measures,and cultivating good work habit.(CONCLUSIONS) It is very important to strengthen nosocomial infection management for preventing efficiently(nosocomial) infection in the department of laboratory.
2.Prevention of Nosocomial Infections in Laboratory Departments of Large Scale Hospitals:Current Situation and Measures

OBJECTIVE To analyze the reasons of nosocomial infection in laboratory departments, and then advance corresponding measures to overcome them. METHODS The current situations in laboratory department between domestic large scale hospitals and overseas hospitals were compared, especially paying attentions to those parts involving in management system and precautionary measures. RESULTS There were a lot of shortcomings existed in the supervision of nosocomial infections in laboratory departments; many measures should be taken to increase the management level. CONCLUSIONS To reform and improve the system of nosocomial infection control and prevention, and establish an effective and systematic alerting and prevention system will benefit all kinds of the hospitals.
3.in vitro Expansion and Differentiation of Cord Blood Hemotopoietic Cells

Objective: To explore the role of human hematopoietic growth factors (HGFs) in proliferation and differentiation of cord blood CD34+ cells. Methods: Human cord blood mononuclear cells (MNC) were cultured with different combinations of HGFs (including rhIL-1,rhIL-3, rhIL-6, rhG-CSF, rhGM-CSF and rhSCF). The expansion folds of MNC, the changes of cellular surface markers by FACS, and CFU-GM of hematopoietic cells were observed. Results: The total nucleated cells expanded by 44 folds after cultured with 6 cytokines for 20 days. The number of CFU-GM increased by 14 .74 folds after cultured in liquid for 8 days, but decreased heavily after 18 days. The total number of CD34+ cells increased by 127. 79 ~ 196 .40 folds at 6 to 8 day, but decreased to 101.51 folds at 18 day. Conclusion: Human hematopoietic growth factors can increase the expansion of cord blood CD34+ cells and CFU-GM significantly.
4.Relationship between plasma content of Cu,Zn,Mn,Se and enzymatic activities of SOD and GSH-Px following severe burn injury

Objective To study the relationship of concentraions of trace elements and the SOD, GSH-Ps activities in plasma following severe burn injury, and provide witness for clinical treatment of trace elements disorder in burned patients. Methods The plasma and urine from 67 severely burned patients in our hospital were collected on day 2, 4, 6, 8, 10, 15, 20, 30 after hospitalization. Trace element levels were determined by atomic absorption spectrophotometry. The vital force of SOD and GSH-Px were measured in plasma with the kits provided by Nanjing Jiancheng bioengineering research institute. Results The ion concentration of Cu in serum got higher and that of Zn got lower after burn, but that of Mn, Se was of no significance. The concentration of Cu, Zn, Mn, Se in urine were increased. The enzymatic activities of T-SOD and Cu, Zn-SOD as well as GSH-Px decreased from the early period to the metaphase. Conclusion For a great deal of oxygen free radicals released by tissues, SOD and GSH-Px were consumed and their concentration decreased in early stage after burn. Due to the trace elements released into blood by destroyed tissues and their increased excretion from urine, tissues could not generate adequate antioxidase, resulting in weak ability against inflammation and poor tissue repair. Our result provides a theoretical clue for the replenishment of trace elements for severely burned patients in early and middle stage and offer a clue to pathology and etiological treatment of burn.
5.Clinical Observation on Treatment of 124 Cases of Insomnia with Moxibustion Therapy

Objective To observe clinical therapeutic effect of treating insomnia by moxibustioning Balhui (DU20)and Sisbencong(EX-HN 1) . Methods 276 cases of insomnia were divided into treatment group and control group. The treatment group was treated by moxibustioning on Baihui (DU 20) and Sishencong(EX-HN 1); while the control group was treated by moxibutioning on Zusanli (ST 36). Evaluate the therapeutic effects and PSQI index of the two groups. Results Clinical symptoms got improvement in the both groups. The treatment group was better than the control group in terms of therapeutic effect of the (P<0.05) and the improvement of PSQI (P<0.01). Conclusion Moxibustioning on Baihui(DU 20)and Sishencong (EX-HN 1) has a good therapeutic effect for insomnia.
6.Rehabilitation interventions for improving activity and participation ability of the earthquake injured

Objective To study the effects of rehabilitation interventions on the earthquake injured to reduce the incidences of disability,to improve the recovery of overall function of the wounded in the earthquake and to guarantee their return to family and society.Methods The rehabilitation team·work provided one-month comprehensive intervention on 92 fracture patients after earthquake and assessed with scale of participate and activities ability,before,after and in the middle of intervention.Results The ability of understanding and communication,moving body,taking care of oneself,getting along with others,undertaking daily activities.social participating and the scores from the scale of participate and activities ability improved significantly(P<0.05).Conclusion Rehabilitation interventions with team-work can effectively enhance the abilities of the earthquake injured to participate in activities and improve quality of life.
7.Detection of Pseudomonas aeruginosa with Piezoelectric Immunosensor Microarray:An Experimental Research

OBJECTIVE To study the main bacteria and their drug-resistance of postoperative infections after liver transplantation.METHODS The distribution and drug-resistance profile of 156 strains of bacterial isolates from(various) specimens in 68 liver transplantation inpatients were retrospectively analyzed last year.RESULTS The(incidence) of infections after liver transplantation was 67.6 %.The major bacteria were Klebsilla pneumoniae((17.3%)),Staphylococcus aureus(14.1%),Candida albicans(11.0%),and Pseudomonas aeruginosa((11.0%)).The bacteria had the character of multidrug-resistance and high drug-resistance.The most effective(antibiotics) to Gram-negatives and Gram-positives were still the carbopenems and glycopeptide.CONCLUSIONS(Infection) is a major complication after liver transplantation.Prevention,early diagnosis and treatment of the(infection) are very important.
8.Inhibition of resveratrol on proliferation of different breast cancer cell lines

Objective To investigate the effects of resveratrol (Res) on the proliferation,cell cycle and apoptosis of three breast cell lines DU4475,MDA-MB-23land MCF-7 with different estrogen receptor (ER) expressions.Methods Res was added to the drug treatment group at 6.25,12.5,25,50,100 and 200 μmol/L,respectively.Three observation periods at 24,48 and 72 hours were set up respectively.MTT assay was used to detect the effects of Res on proliferation of breast cells.Cell cycle and apoptosis were analyzed by flow cytometry (FCM).The concentrations of drugs were 0,12.5,25,50 μmol/L.The results were analysed by the statistical software SPSS17.0.Results After Res intervention,the proliferations of three cell lines were inhibited to different extent.After 48 and 72 hours of Res,inhibitions of Res with different concentration were significant different (F =15.26,P < 0.05).Inhibition rates of DU4475 and MDA-MB-231 with ER-negative were higer than that of MCF-7 with ER-positive.FCM results prompted that these three kinds of cells were blocked in S phase after 48 hours treatment of 12.5-50 μmol/L Res and the block percentages had significant difference (F =34.81,P < 0.05).The percentages of S phase for DU4475 and MDA-MB-231 arresting were higher than that of MCF-7.For DU4475,the apoptotic and necrosis percentage were higher than that of MCF-7 at 100 μmol/L (t =16.082,P <0.05).Conclusion Res can inhibit proliferation,induce cell cycle changing and apoptosis of DU4475,MDA-MB-231 and MCF-7 cells.The inhibitions of Res on DU4475 and MDA-MB-231 are better than that of MCF-7 with ER-positive.
9.A study on the pathogenic characteristics and traceability of Vibrio cholera strains circulated in Hubei province in 2012

Objective To investigate the pathogenic characteristics of Vibrio cholera strains isola-ted from Hubei province in 2012 , and to identify the source of infection by analyzing their genetic correla-tions.Methods The biochemical identification , toxin gene detection and drug susceptibility test were car-ried out to analyze a total of 35 Vibrio cholera strains isolated from three epidemic areas .Comparison of ge-nomic DNA fingerprints and cluster analysis among isolates of Vibrio cholera was conducted by using pulsed-field gel electrophoresis ( PFGE ) .Results All of the 35 strains were Vibrio cholera O139 , of which 71.42%were toxic strains.The drug resistance rates of Vibrio cholera strains to tetracycline, cotrimoxazole and rifampincin were 57.14%, 88.57%and 80.00%, respectively.Analysis of genomic DNA fingerprints of the isolates showed highly similar with similarity values ranging from 80%-100%.Most of the strains iso-lated from the same epidemic area fell into the same one cluster with 100% homology in genome Only a strain isolated from turtle in Jingzhou area was belong to a different cluster .Conclusion The Vibrio cholera O139 strains were the dominant strains causing the outbreaks of cholera in Hubei province in 2012 .Most of them were toxigenic strains .A large majority of the strains had developed resistance to tetracycline , cotri-moxazole and rifampincin , but all strains showed high susceptibility to ceftriaxone and imipenem .Vibrio cholera strains isolated from the same epidemic area were mainly belonged to the same one cluster , sugges-ting the same source of infection .However, the strains varied among different epidemic area .Follow-up in-vestigations of three outbreaks of cholera in this study were all associated with food infection .Therefore , more attention should be paid to food sanitation and safety measurement .Although a non-toxigenic strain iso-lated from turtle was not associated with the epidemic of cholera , surveillance for seafood and aquatic prod-ucts would still be necessary .
10.Percutaneous nephrolithotripsy with pneumatic and ultrasonic power for treatment of pediatric renal cal-culi

Objective To assess the safety and efficacy of percutaneous nephrolithotripsy withpneumatic and ultrasonic power in pediatric renal calculi. Methods The clinical data of 44 patientstreated with combination of pneumatic and ultrasonic power during pereutaneous nephrolithotripsywere retrospectively analyzed. The 44 patients had 49 renal calculi. The patients were all under 14years old. The average age was 11 years (range 7-14 years). There were 39 unilateral and 5 bilateralcalculi. Among the 44 patients,metabolic disturbance occurred in 19 cases (43.2%),anatomical dys function occurred in 15 cases (34.1%),urinary tract infection occurred in 14 cases (31.8%). Ante grade percutaneous access was established under ultrasound guidance,a combination of pneumatic andultrasonic lithotripsy were used. The effect was evaluated by postoperative KUB and ultrasonic. Re suits The access was successfully established in all patients. Complete stone clearance was achievedin 36 kidneys in phase Ⅰ,stones from 9 kidneys were completely removed with second lithotripsy.Leftover stone in 2 kidneys were treated by ESWL. Open surgery was performed in 2 kidneys due toexcessive bleeding. The operative time ranged from 52 132 min,average time was 79 min. Two pa tients needed blood transfusion. No severe complications occurred in all patients. Thirty seven pa tients were followed up for 3 18 months. The renal function was not worsened and hydronephrosiswas not aggravated in these patients. Conclusion The percutaneous nephrolithotripsy with pneumatic and ultrasonic power is a safe,effective treatment for pediatric renal calculi.
