1.Research on the IgE, EOS, LTE-4 content and its clinical significance with the inacute phase of pediatric asthma

Objective To explore the clinical significance of total serum immunoglobulin ( IgE) , peripheral blood eosinophils ( EOS) and urinary leukotriene E4 ( LTE-4 ) detection on pediatric asthma clinical symptoms of the inacute phase .Methods 90 cases with pediatric asthma clinical symptoms of the inacute phase from March 2014 to March 2015 were selected as experimental group, and divided into the general asthma group(n=32), the moderate asthma group(n=31), the sereval asthma group(n=27) and another 30 health children as control group.Give each patients children doses of β2 agonists (LABA) as monotherapy, and inhaled corticosteroids (ICS) used as combination therapy.The use of LABA is 3-4 times/d.After three months' treatment, clinical efficacy were evaluated, and IgE, EOS, LTE-4 content were detected.Results After 3 months' treatment, IgE, EOS, LTE-4 contents in three experiment groups were significantly decreased (P<0.05), improved to the basic condition compared to control groups.The clinical efficacy among three groups had no significant differences.Conclusion IgE, EOS, LTE-4 levels detection of patients with pediatric asthma clinical symptoms of the inacute phase had evaluation significance, can be a good indicator of pediatric asthma health recovery in patients of the inacute phase.
2.Application of the revised trauma score in patients with multiple trauma emergency

Objective To study the effect of revised trauma score (RTS) in the rescue of patients with multiple trauma emergency application. Methods According to the admission time, 56 multiple trauma patients were assigned into the control group, and another 58 into the observation group. The control group was treated with traditional pre-hospital and hospital emergency care. The observation group with pre-hospital emergency care and hospital based on the RTS results. The two groups were compared in terms of effective treatment time and complications. Result The effective rescue time of the observation group was significantly shorter than that of the control group (P<0.001) and the complication rate was significantly lower than that of the control group (P<0.05). Conclusion Based on the integrated emergency rescue measures on the basis of RTS scores, accurate condition judgment can be made in a short time so that consistent care can be given to the patients and the success rate can be improved , and the occurrence of complications can be reduced and the rescue success rate can be increased.
3.Analysis of 120 cases of reduction of postpartum bleeding in cesarean section by misoprostol

Objective To observe the effects of misoprostol on the reduction of postpartum bleeding in cesarean section.Methods With oxytocin as control,the amount of bleeding during and two hours after cesarean section was observed.Results The mean amount of bleeding was (232.0±46.6) ml in misoprostol group and (385.0±74.7) ml in oxytocin group respectively.The difference between the two groups was significant (P<0.01).Conclusion Misoprostol is more effective to improve uterin contraction than oxytocin and is better in the reduction of postpartum bleeding with safety and convenience.
4.Influence of asarone injection for the pulmonary surfactant proteins and oxygen supply state of newborns with pneumonia

Objective To observe the influence state of asarone injection for the pulmonary surfactant proteins and oxygen supply state of newborns with pneumonia. Methods 72 newborns with pneumonia were selected as the study object, and they were randomly divided into control group (36 cases) and observation group (36 cases), the control group were treated with conventional treatment of pneumonia, the observation group were treated with asarone injection on the treatment method of control group, then the pulmonary surfactant protein indexes, oxygenation function indexes and blood gas analysis of two groups before and after the treatment were respectively detected and compared. Results The pulmonary surfactant protein indexes, oxygenation function indexes and blood gas analysis of two groups before the treatment all had no significant differences, while the pulmonary surfactant protein indexes, oxygenation function indexes and blood gas analysis of observation group at first, second and fifth day after the treatment were all significantly better than those of control group, all P<0.05, there were all significant differences. Conclusion The asarone injection can significantly improve the pulmonary surfactant proteins and oxygen supply of newborns with pneumonia, so its application value in the treatment of newborns with pneumonia is higher.
5.CT and MRI Diagnosis of Nasopharyngeal Angiofibroma

Objective To investigate the value of CT and MRI in diagnosing nasopharyngeal angiofibromas.Methods 18 cases of nasopharyngeal angiofibromas were retrospectively analyzed including the location, density ,signal intensity, extension of tumors and changes of surrounding bones.Results 16 cases located in the area around sphenopalatine foramen. 6 cases exhibited homogeneous density on CT scans(6/11 cases) and 12 cases displayed inhomogeneous signal intensity on MR scans(12/14 cases). All the Fourteen MRI examinations revealed vice signal by vessels, inhomogeneous enhancement and extensive invasion along the foramina or fissures.Conclusion MRI not only determines the location and traces out the extension of nasopharyngeal angiofibroma more exactly, but also displayes its diagnostic signal fashion. Namely MRI has greater value in diagnosing this tumor than CT.
6.Effects of co-grafts mesenchymal stem cells and nerve growth factor suspension in the repair of spinal cord injury.

To investigate effect of the transplantation of mesenchymal stem cells (MSCs) in combination with nerve growth factor (NGF) on the repair of spinal cord injury (SCI) in adult rats, spinal cord of adult rats (n= 32) was injured by using the modified Allen's method. One week after the injury, the injured cords were injected with Dubecco-modified Eagles medium (DMEM, Group I), MSCs (Group II), NGF (Group III), and MSCs plus NGF (Group IV). One month and two months after the injury, rats were sacrificed and their injured cord tissues were sectioned for the identification of the transplanted cells. The axonal regeneration and the differentiation of MSCs were examined by immunocytochemical staining. At the same time, rats were subjected to behavioral tests by using the open-field BBB scoring system. Immunocytochemical staining showed that axonal regeneration and the transplanted cells partially expressed neuron-specific nuclear protein (NeuN) and glial fibrillary acidic protein (GFAP). At the same time, significant improvement in BBB locomotor rating scale (P<0. 05) were observed in the treatment group. More importantly, further functional improvement were noted in the combined treatment group. MSCs could differentiate into neurons and astrocytes. MSCs and NGF can promote axonal regeneration and improve functional recovery. There might exist a synergistic effect between MSCs and NGF.

8.The clinical significance of advanced oxidation protein products in patients with systemic lupus erythematosus complicated with atherosclerosis praecox

Objective To estimate the degree of oxidative stress and atherosclerosis praecox in patients with systemic lupus erythematosus (SLE), and emphasize on exploring AOPPs' clinical significance in premature atherosclerosis in SLE. Methods The levels of AOPPs, Hey, MDA, SOD, baPMV and CIMT were detected by ELISA and spectropholometry in 44 non-menopausal female SLE patients and 31 healthy middle-aged women respectively, and baPWV. The results were compared with AOPPs of the two groups. Then each group was stratified based on disease duration (≥5 years or <5 years) and the disease activity(active and inactive) in SLE patients. The patients' TC, TG, LDL were analyzed. T test, t' test and Pearson correla-tion were selected. Results The levels of AOPPs, Hcy, MDA in SLE patients were significantly higher than those of the healthy controls (P<0.05). The activities of SOD were lower than controls (P<0.05). The levels of AOPPs, Hcy, MDA, SOD had statistical significance between SLE patients disease duration ≥5 years or <5 years, active or inactive groups. There were two cases with CAP in patients with SLE (more than 5 years disease duration),while there wasnone in healthy controls. The levels of baPWV and CIMT in patients with SLE were higher than healthy controls (P<0.05), and had statistical significance in SLE patients disease duration more than or less than 5 years (P<0.05). The oxidative stress targets (AOPPs, Hey, MDA, SOD) had significant correlation with the level of baPWV and CIMT (P<0.01). The level of serum AOPPs had significant positive correlation with the levels of Hcy, TC, TG and LDL (P<0.01~0.05). Conclusion SLE patients have increased oxidative stress , and significantly higher prevalence of atherosclerosis than healthy controls. The disease duration and oxidative stress play important roles in the duration of atherosclerosis. AOPPs probably involves in the accelerated atherosclerosis of SLE patients. It may be a predictor for SLE complicated with atherosclerosis praecox.
9.Construction of immortalized human precartilaginous stem cell lines

BACKGROUND: The precartilaginous stern cells are limited regarding in vitro proliferative capacity, but the immortalized cell lines can provide a large number of stable immortalized cells, and simian virus 40 large T antigen gene (SV40Tag) is one of gene fragments which are commonly used and effective in vitro immortalized ceils. OBJECTIVE: To construct human immortalized precartilaginous stem cells (IPSCs) using human precartilaginous stem calls induced by SV40LTAg gane. METHODS: The human immortalized precartilaginous stem calls were isolated from aborted fetus and purified with enzyme digestion and immunomagnetic beads screening method. By using liposome-mediated gene transfection technology, plasmid pCMVSV40T/PUR containing SV40Tag was transfected in primary embryonic precartilaginous stem cells, while non-transfected cells sewed as negative controls. Positive clones were cultured to observe the cell morphology and the passage recovery, to calculate cell survival rata and population doubling time, to drew call growth curve. Immunofluorescence cytochemistry was used to detect the expression of IPSCs fibroblast growth factor receptor 3, the expressions of SV40Tag and fibroblast growth factor receptor 3 in the human precartilaginous stem cells were determined by RT-PCR. RESULTS AND CONCLUSION: Morphology of human IPSCs seemed coincidence with primary human precartilaginous stem cells. The survival rate of human IPSCs was not influenced by subculture, freezing and recovery, but the survival rate was descended in the human precartilaginous stem cells at the 6~(th) and 10~(th) passages (P < 0.01). Compared with cells at the 6~(th) and 10~(th) passages, the proliferation of human IPSCs was greater, with short population doubling time and high growth rate (P < 0.01). The immunofluorescence showed that fibroblast growth factor receptor 3 was positive in human IPSCs at the second passage, and the RT-PCR results of fibroblast growth factor receptor 3 revealed a specific amplification band at 400 bp,.while that of SV40Tag revealed at 560 bp. No band was seen in the primary cells. It is indicated that SV40Tag human IPSCs can be constructed successfully using immunomagnatic bead screening technology and liposome transfection technique.
10.Study on the Pharmacokinetic-pharmacodynamic Model of Nisoldipine Controlled-release Patches in Spon-taneously Hypertensive Rats

OBJECTIVE:To establish the pharmacokinetic-pharmacodynamic(PK-PD) model of Nisoldipine controlled-release patches(NCRP)in spontaneously hypertensive rats(SHR). METHODS:SHR were randomized into a patch(NCRP)group and a tablet(Nisoldipine tablets)group,with 6 rats in each group. The microdialysis probes were implanted in SHR. Each rat was given 5 mg nisoldipine. Plasma microdialysate was collected within 36 h after administration. HPLC was adopted to determine the plasma concentration of nisoldipine,and WinNonlin 5.3 was employed to calculate Pharmacokinetic parameters. With heart rate and blood pressure as pharmacodynamic indexes,PK-PD model study was conducted. RESULTS:Vs. nisoldipine tablets,NCRP has con-trolled release effect. The relationship between NCRP drug effect and effect-site concentration met the Sigmoid-Emax model. The main parameters of the PK-PD model for heart rate and systolic blood pressure were as follows as Emax of (2.65 ± 0.06) and (10.71 ± 0.87),EC50 of (83.65 ± 35.25) and (1.29 ± 0.26) ng/ml,γ of (0.83 ± 0.91) and (1.2 ± 0.35),Keo of (0.37 ± 0.53) and (0.91±0.24)h-1. CONCLUSIONS:PK-PD model of NCRP in SHR has been established successfully.
