1.Quantification method for MRSI based on Hankel matrix

Objective To study the quantification method based on Hankel matrix,the water suppression method and the metabolite imaging method for magnetic resonance spectroscopic imaging (MRSI) data.Methods Impact of Hankel data matrix on quantification MRSI methods were analyzed to obtain the most efficient Hankel matrix structure.The maximum amplitude method of the water signal peak was proposed for MRSI data water suppression.The interested metabolites information was extracted from MRSI data,and then metabolite image was obtained through bilinear interpolation algorithm.Results The minimum amplitude error and the minimum frequency error were acquired when columns number was 3/4 sampling points.The amplitude,frequency and the damping factor of the simulation data accuracy was 96.94%,99.72% and 95.55% respectively.Hankel lanczos with partial reorthogonalization singular value decomposition (HLSVDPRO) method with 3/4 sampling points was used to form Hankel matrices.The speed of quantification decreased with the increase of sampling points.The error of quantification parameter reached minimum when the number of sampling points was 512.The water suppression degree of simulation data was 99.55% with the maximum amplitude water suppression method.Conclusions The accuracy and the speed of the quantification are promoted with an optimized Hankel matrix structure for the MRSI quantization method.The optimal length of sampling points is 512.The maximum amplitude method can suppress water perfectly.Doctors can detect the presence of tumor regions in human body at the (super) early stage by metabolite information images.

4.Diagnosis and Surgical Operation of the Lateral Patellar Compression Syndrome

Objective To explore the diagnosis and therapies of the lateral patellar compression syndrome.Methods This group included 21 cases(30 knees) of lateral patellar compression syndrome,with the average 45 years old(2 males and 19 females of 36~58 years old).All of the patients had obvious Clinical sings and symptoms,whose auxiliary examination was negative.It was almost as same as the Merchant classificasion of the lateral patellar compression syndrome.We adopt the lateral retinaculum release and debridement cartilage defects and drilling.Results All of the definitely diagnosised patients were reviewed with an average of 4 years follow-up visiting(range from 0.5 to 15 years) after operation.The rate of fineness is 96.7% in accordance with Ye Qibin's synthesis estimation for the ailing knee.Conclusion The lateral patellar compression syndrome is a common disease.The lateral retinaculum release and debridement cartilage defects and drilling is a good therapy with minor injury and significant effects for the severe or recurrent attacking lateral patellar compression syndrome.
5.Determination of Menthol and Methyl Salicylate in Shangtongning Adhesive Plaster by Capillary Gas Chromatography

Objective To establish a capillary gas chromatography method for the determination of menthol and methyl salicylate in Shangtongning adhesive plaster. Methods The internal standard substance of n-Pentadecane was employed. The GC system consisted of a DB-WAX capillary column (30 m?0.32 mm, 0.25 ?m), nitrogen as the carrier gas, column temperature at 130 ℃, FID as the detector at 200 ℃, and with a split ratio of 12∶1. Results The contents of menthol and methyl salicylate in 6 lots of samples were determined and the constituents concerned were separated well under the chromatographic condition. The average recovery of menthol and methyl salicylate were 101.52% (RSD=2.66%) and 100.34% (RSD=3.38%), respectively. Conclusion The method was sensitive, accurate, specific and suitable for the quality control of the preparation.
6.CpG oligodeoxynucletide potently enhances cellular immune responses in suckling mice infected with human rotavirus

Objective To investigate the effects of CpG oligodeoxynucleotide (ODN) on cellular immune responses in suckling mice infected with human rotavirus (HRV). Methods Forty ICR suckling mice were randomly divided into four groups: control group. HRV infection group, CpG ODN pretreatment group and CpG ODN treatment group. Suckling mice were sacrificed four days after rotavirus challenge. Small intestine and spleen samples were collected under sterile condition. Thedegree of small intestinal mucosal injures was evaluated with standard scoring criteria. The spleen index was calculated and spleen lymphocyte stimulation index was detected by methyl thiazolyl tetrazolium ( MTT) assays. Levels of gamma interferon (IFN-γ), interleukin-4 ( IL-4) in the supernatant of spleen lymphocyte culture were detected by enzyme linked immunosorbent assay (ELISA). T lymphocyte subsets were analyzed by flow cytometry. The data was compared by one way ANOVA. Results The scores of mucosal injures of mice in HRV infection group, CpG ODN pretreatment group and CpG ODN treatment group were 4. 00 ±1. 31, 2. 75 ±1. 28 and 2. 87 ±0. 99, respectively, and the differences among groups were statistically significant (F=ll. 32,P<0. 01). The scores of mucosal injures in CpG ODN pretreatment group and CpG ODN treatment group were both lower than that in HRV infection group (P<0. 05). Compared with control group, spleen lymphocyte stimulation index and the percentage of CD8+ T lymphocytes were higher, the percentage of CD4+ T lymphocyte and ratio of CD4+ /CD8+ T cells were lower, levels of T helper (Th)1 type cytokine IFN-y increased significantly in HRV infection group; while the spleen index, spleen lymphocyte stimulation index, percentages of CD4+ and CD8+ T lymphocytes, IFN-y levels were all increased in CpG ODN pretreatment group and CpG ODN treatment group (P<0. 05). The spleen index, spleen lymphocyte stimulation index, the percentage of CD4+ T lymphocytes, CD4+/CD8+ ratios and IFN-y levels in CpG ODN pretreatment group and CpG ODN treatment group were all significantly higher than HRV infection group (P<0. 05). Conclusion CpG ODN potently enhances cellular immune responses in ICR suckling mice infected with HRV and CpG ODN could induce dominant Th1 response.
7.Establishment of animal model of human rotavirus-induced diarrhea in ICR suckling mice

Objective To establish an animal model of human rotavirus(HRV)-induced diarrhea. Methods Sixty ICR suckling mice were randomly divided into two groups, and each was further divided into 3 subgroups(A, B, C for group Ⅰ, and D, E, F for group Ⅱ);group B, C, E and F were assigned as experimental groups. while group A and D were controls. Mice in group B and C were inoculated with3 × 10~5 PFU and 3×10~6 PFU HRV Wa strain fluid respectively when they were 4-day old. Mice in group E and F were inoculated with 3×10~5 PFU and 3×10~6 PFU HRV Wa strain fluid respectively when they were10-day old. The symptoms, fecal viral excretion, intestinal histopathologies and ultrastructures of animals were observed. One-way ANOVA was used to compare the differences among the groups. Results There were significant differences in the duration of diarrhea and viral excretion, jejunal villous height, the weight at d4 and d7 after inoculation among three subgroups in group Ⅰ(F=204.38, 86.60, 7.18, 18.41 and10.08, P<0.01). Diarrhea was not observed in group Ⅱ, and the differences in jejunal villous height and the weight at d4 and d7 after inoculation among three subgroups were not significant(F=0.16, 0.13 and 1.03, P>0.05). Compared with the group D, the duration of viral excretion was longer in group E and F(F=8. 34, P<0.01). Conclusion Animal model of HRV diarrhea can be established in 4-day-old ICR suckling mice infected with 3×10~6PFU HRV.

Objective To investigate the effects of apolipoproteinA1 (apoA1) on levels of cholesterol, cholesteryl ester (CE), and expression of ATP-bindiag cassette transporter A1 (ABCA1) in human acute monocytie leukemia cell line (THP-1) macrophage-derived foam cells.Methods The cultured THP-1 cells were induced into foam cells by exposing first to phorbol myristate acetate (PMA, 50 ng/ml) for 48 h, and then to oxidized-low density lipoprotein (ox-LDL, 50μg/ml) for 48 h.Under treatment of apoA1 in different doses (5, 10, 15 and 20 μg/ml) and one simple dose (10 μg/ml) for different time (6, 12 and 24 h), THP-1 macrophage-derived foam cells were incubated to observe the expression of cholesterol and ABCA1.The concentrations of cellular total cholesterol (TC), free cholesterol (FC) and CE were determined by oxidization enzymatic methods.Oil red O dyeing experiment was used to show the cellular lipid droplets in the cells.The expression of ABCA1 was tested by immunofluorescence method.Reverse transcription-polymerase chain reaction was applied to investigate mRNA expression of ABCA1.Results The THP-1 cells turned into typical foam cells after treated with PMA (50 ng/ml) for 48 h, and ox-LDL (50 μg/ml) for 48 h.apoA1 could lower the levels of TC, FC and CE in THP-1 macrophage-derived foam cells in a dose-dependent and a time-dependant manner, apoA1 could increase the expression of ABCA1 protein in THP-1maerophage-derived foam cells without up-regulation of mRNA.Antibody of ABCA1 could up- regulate the expression of ABCA1.Conclusions apoA1 may decrease the levels of cholesterols in THP-1 macrophage-derived foam cells, by promoting the expression of ABCA1 and the reverse cholesterol transport of high density lipoprotein.
9.Design and application of biodegradable coronary stents:what will be brought by further innovations in materials science?

BACKGROUND:With the social and economic development and aging, coronary heart disease has become the primary cause of death endangering human life and health, while cardiovascular intervention (especial y stenting) in the treatment of cardiovascular disease is at a growing status. OBJECTIVE:To describe the development, research progress and latest clinical application of heart interventional materials as wel as advantages and disadvantages of biodegradable stents, and meanwhile to prospect for its future development and improvement. METHODS:A computer-based online search was conducted in PubMed for English language publications from January 1999 to April 2014 using the key words of“stent, scaffold, bioabsorbable, bioresorbable, biodegradable, biocompatibility, material properties”in English. RESULTS AND CONCLUSION:The new ful y biodegradable stents are considered as another major progress in the cardiac intervention, which brings a new gospel for patients with coronary heart disease. In the initial stage of implantation, biodegradable stents exhibit the same mechanical support as bare metal stents;after implantation, anti-proliferative drugs are released to prevent thrombosis and restenosis, and the stent is completely degraded within a specified period, which significantly reduces the late-stage and very late-stage thrombosis and the risk of in-stent restenosis. However, long-term safety and efficacy of biodegradable stents need further studies. Innovation of stent design and materials science is the key to overcome the current shortcomings of biodegradable stents.
10.Cardiac collagen metabolism in murine viral heart diseases

AIM: To investigate the dynamic alteration of cardiac collagen metabolism in mice with acute,chronic myocarditis and dilated cardiomyopathy(DCM).METHODS: BALB/c mice infected with coxsackievirus B_3 were used to establish animal models of acute,chronic myocarditis and dilated cardiomyopathy,while uninfected animals were also prepared and served as controls.After verification of models by histopathological methods and echocardiography,serum concentration of aminoterminal propeptide of type Ⅲ procollagen(PIIINP),aminoterminal propeptide of type Ⅰ procollagen(PINP) and carboxyterminal propeptide of type I procollagen(PICP) in each group of mice were detected by enzyme linked immunosorbent assay(ELISA).The expression of matrix metalloproteinase 1(MMP1) and its tissue inhibitor(TIMP-1) were determined by Western blotting analysis.The MMP-1 activity was also detected.RESULTS: Marked myocardial fibrosis was observed in all groups of CVB_3-infected mice.Reparative fibrosis,promotion of synthesis and degradation of cardiac collagens were presented in heart tissue of acute myocarditis mice. Both reparative and reactive fibrosis,enhanced synthesis and lightened degradation of collagen were present in chronic myocarditis,while reactive fibrosis and excess collagen synthesis were confirmed in DCM.Expression and activity of(MMP-1) was progressively decreased.TIMP-1 showed unchanged.The ratio of MMP-1/TIMP-1 was progressively descended.CONCLUSION: Collagen metabolism was special in different phase of viral heart diseases,which may play different roles in the progression and prognosis of these kinds of disease.
