1.Bone defect repair using allogenic ECM combined with chondrocytes in rabbits

[Objective]The characteristics and mechanism of the bone defect repair with the using allogenic extracellular cartilage matrix(ECM)were studied.[Method]Forty New Zealand white rubbits were divided into three groups at random:A and B on the right side,the defect was covered with the complex of ECM and autologous mesenchymal stem cells(MSCs),the ECM in the groups A and B,respectively.The defects on the left side served as controls correspondingly.The rabbits were killed at the 2nd,4th,6th,10th week.Samples were taken for radiological and histological studies.[Result]In the experimental sides the bone defects were healed.New bone appeared in the way of intramembranous ossification and entochondrostosis.[Conclusion]It is suggested that the complex of ECM and autologous MSCs has the effects on guiding bone regeneration and preventing from nonunion.
2.The role of peripheral nerve TNF? content in the development of neuropathic pain produced by chronic constriction injury of peripheral nerve in rats

Objective To investigate the changes in TNF? content of sciatic nerve induced by chronic constriction injury (CCI) of sciatic nerve to determine the role of TNFa in the development of neuropathic pain. Methods Eight-four female health SD rats weighing 250-300g were anesthetized with sodium barbiturate. Unilateral sciatic nerve was exposed and ligated at the middle of thigh. Three ligatures (chromic catgut 4.0) were placed around the sciatic nerve and tied. The distance between the two ligatures was about 1 mm. Sham operation was performed on the contralateral thigh. The sciatic nerve was exposed and mobilized but not ligated. The thermal nociceptive threshold was determined by measuring the withdrawal latency of hindpaw placed on a 58℃ hot plate on days 0.5, 1, 3, 5, 7, 9, 11, 13 and 14 after surgery. Animals were sacrificed on days 0.5, 1, 3, 7, 10 and 14 after surgery. Sciatic nerves were removed from both thighs and frozen at - 80℃ for determination of TNF? content. Sciatic nerve from healthy animals was used as control. The percentage of maximal possible response (% MPR) , was determined for each group (CCI, sham operation, control) % MPR= (new withdrawal latency- average baseline latency)/( 15-average baseline latency) . The distribution of TNF? between supernatant and sediment was also determined. Results The average baseline nociceptive threshold (withdrawal latency) was (7.9?0.2)s. There was significant different in %MPR between the two hindpaws on days 1, 3, 5, 7, 9 and 11 after surgery. The TNF? content of sciatic nerve from healthy rats was (40.62? 0.24) pg/mg protein. The TNF? content of the ligated sciatic nerve was elevated abruptly in 12h after ligation, then abruptly declined to a plateau but was still significantly higher than that of sham-operated side on days 1 and 3. There was no significant difference in TNF? content of sciatic nerve between control group and sham-operation group. The relative content of TNF? content in the sediment of ligated sciatic nerve gradually increased and reached the peak on day 7 and then gradually decreased. Conclusion The TNF? content of peripheral nerves plays an important role in the development of neuropathic pain. Membrane-combined TNF? is involved in the process of nerve repairing.
3.Clinicopathologic study and review of alveolar soft part sarcoma

Objective To investigate the clinicopathologic features and differential diagnosis of alveolar soft part sarcoma(ASPS).Methods A case of ASPS was studied by HE stain,light microscopic observation,histochemical stain and immunohistochemical stain,and discussed the disease synthetically with the literature.Results The patient was female,23 years old.The lesion was located in the deep skeletal muscles of left buttocks,microscopically,which consisted of round to polygonal tumor cells,showing alveolus and nest arrangement,arounded by stroma containing sinusoidal capillaries,and there were abundant crystals in cytoplasm of the tumor cells,prominent nucleoli and fewer nuclear fission images in the tumor cells.PAS stain was stongly positive.Immunohistochemical stain showed positive expression of myoglobin in tumor cells,and negative expression of Myosin,Desmin,SMA,Vimentin,S-100 protein,EMA and Syn in them.Conclusion ASPS is a rare malignant soft tissue neoplasm,its histogenesis is uncertain,and diagnosed by pathological examine and treated mainly by operation.
4.Research on distribution of gastrointestinal peptide glucagon-1 of positive cells in tree shrews

Objective To observe tree shrews gastrointestinal glucagon like peptide 1 ( glp-1 ) immune response positive cells ( EG cells) distribution and the difference of the digestive tract of rats.Methods Immunohisto chemical SABC method to detect tree shrews diges-tive tract (except for oral cavity, pharynx) glp-1 cells positive content.Results EG cells distributed in tree shrews duodenum,ileum and colon,glandular epithelial cells of jejunum,positive material is distributed in the cytoplasm.Tree shrews stomach,rectum did not see EG cell distribution.Ileal immune dyeing results show positive,EG cells of the digestive tract,only the ileum and colon distribution glp-1 more posi-tive, tree shrews compared with the rats in the digestive tract glp-1 significant difference (P<0.01).Conclusion Gastrointestinal tree shrew in duodenum,ileum and colon,jejunum have glucagon positive cells.The rat only have positive cells in the ileum and colon.According to the mechanism of the glp-1 and the distribution of tree shrews gastrointestinal glp-1,tree shrews atherosclerosis ( atherosclerosis,AS) AS the model is not easy to make may be associated with the role of glp-1.
5.Operative Technique and Curative Effect Analysis of Minor-Incision Cholecystectomy

Objective To discuss the operative technique and curative effect of minor-incision cholecystectomy.Methods The clinical data of 672 patients with application of mini-cholecystectomy from June 2001 to June 2009 were analyzed.Perioperative management and operative technique were emphasized.Results Six hundred and fifty-two cases (97.0%) were cured with mini-cholecystectomy and 20 cases (3.0%) with incision lengthened.Operation time was (40.0?10.0) min.One case with hemorrhoea during operation was cured by interventional embolotherapy.Bile duct injury was found in 1 case during operation,and adopted suture with T tube.There were no infection of incisional wound or death in this study.Conclusion On the basis of skillful conventional cholecystectomy,by controlling indication and improving operative technique,it is an economical and safe way to perform minor-incision cholecystectomy.
6.Analysis of screening and therapeutic effect of congenital hypothyroidism in Zhongshan

Objectives To summarize screening and therapeutic effects of congenital hypothyroidism (CH) in Zhongshan. Methods The thyroid stimulating hormone (TSH) concentration in dried heel blood samples on iflter paper was detected using time-resolved fluorescence immunoassay. The cases of positive screening tests were called back for further examination of venous blood TSH concentration using chemiluminescence method. Fifty-four children with permanent CH treated routinely for 2 years (CH group) and 120 age-gender matched health children (control group) were recruited. The physical development (height, body weight) was monitored. The neurodevelopment and temperament type were tested using Pediatric Nneuropsychological Development Assessment and Children's Temperament Scale respectively at 6 and 24 months after birth. Results Two hundred eight-five thousand two hundred forty-two neonates were screened. One hundred and forty cases were confirmed and the incidence rate was 1/2037. There was no statistical difference in length-for-age z score (LAZ) and weight-for-age z score (WAZ) between CH and control group (P>0.05). The neurodevelopment in CH group was normal, but gross motor development was worse than that in control group (P<0.05). The temperament type and distribution had statistical difference between CH and control group (P<0.05). The percentage of the dififcult type and central dififcult type was increased in CH group as compared to control group, especially in the activity, adaptability, reaction intensity and perseverance (P<0.05). Conclusions The physical and neurodevelopment are nearly normal in patients with CH after early supplementation, but the psychological behavior problems need to be focused on in the process of intervention.
7.Effect of Scalp Acupuncture on Cognitive and Motor Functions of Stroke Patients in Recovery Stage

Objective To observe the effect of scalp acupuncture on cognitive and motor functions of stroke patients in recovery stage.Methods80 stroke patients with cognitive disorder were randomly divided into the scalp acupuncture group (n=41) and control group (n=39). All patients of two groups were treated with routine therapy and rehabilitation training, but the cases of the scalp acupuncture group were added with scalp acupuncture. The cognitive function, motor function and activities of daily living (ADL) of all patients were assessed by delitescence and amplitude of event-related potential (ERP) P300, Fugl-Meyer Assessment (FMA), Barthel Index (BI) respectively before and 3 months after treatment.ResultsAfter treatment, P300 delitescence of the patients in the scalp acupuncture group shorten to 38 ms in average while in the control group shorten only to 17 ms, the therapeutic effect of the scalp acupuncture group was superior to that of the control group ( P<0.05). The scores of FMA and BI of the patients in the scalp acupuncture group were also better than those in the control group ( P<0.05).ConclusionScalp acupuncture can efficiently improve the cognitive function of stroke patients in the recovery stage, and promote the recovery of motor function and ADL.
8.Study on the problems of the public health emergency system and countermeasures to complement the shortcomings in Suzhou

Objective To meet the needs of public health emergency management in the new era, to improve the problems exposed in the public health emergency system after the outbreak of COVID-19, and to provide the government with a list of the gaps in the construction of the public health emergency system. Methods Several methods, including literature review, observation (discussion and questionnaire survey), and comparative and descriptive research, were used to conduct comprehensive analysis through the combination of induction and deduction. The evaluation questions were graded and were translated into some recognition indexes (averages). The golden section method was used for the recognition standard of evaluation problems, and U test method was used for statistical test. Results Five aspects of problem evaluation and suggestions were determined, including early warning, emergency plan, emergency management department, linkage between systems and epidemic situation judgment. The recognition indexes were 0.916, 0.905, 0.571, 1.000 and 1.190, respectively, all of which met the recognition standard, and there was no significant difference between them(P=0.357). The corresponding measures and suggestions were as follows: early warning trigger mechanism, modular emergency plan, government public health emergency agencies, collaborative emergency drill system of various industries, and expert epidemic situation judgment mechanism. Conclusion It is determined that the proposed measures to complement the weaknesses of public health emergency system in Suzhou are suitable for the locality and objectivity after the three steps of literature review, management staff discussion and professional questionnaire survey. The present study provides an objective basis for the health administrative department to propose the public health emergency system construction project to the government.
9.Bladder microenvironment actuated proteomotors with ammonia amplification for enhanced cancer treatment.

Enzyme-driven micro/nanomotors consuming in situ chemical fuels have attracted lots of attention for biomedical applications. However, motor systems composed by organism-derived organics that maximize the therapeutic efficacy of enzymatic products remain challenging. Herein, swimming proteomotors based on biocompatible urease and human serum albumin are constructed for enhanced antitumor therapy via active motion and ammonia amplification. By decomposing urea into carbon dioxide and ammonia, the designed proteomotors are endowed with self-propulsive capability, which leads to improved internalization and enhanced penetration in vitro. As a glutamine synthetase inhibitor, the loaded l-methionine sulfoximine further prevents the conversion of toxic ammonia into non-toxic glutamine in both tumor and stromal cells, resulting in local ammonia amplification. After intravesical instillation, the proteomotors achieve longer bladder retention and thus significantly inhibit the growth of orthotopic bladder tumor in vivo without adverse effects. We envision that the as-developed swimming proteomotors with amplification of the product toxicity may be a potential platform for active cancer treatment.
