1.Research Advances in G Protein Coupled Receptor-mediated Electroacupuncture Intervention in Rheumatoid Arthritis Pain

G protein coupled receptors (GPCRs) have a marked mediating effect on bodily immunoregulation. Recent years’ studies found that GPCRs plays an important role in the mechanism of the intervening effect of electroacupuncture on rheumatoid arthritis pain. Of them, adrenergic receptors, 5-hydroxytryptamine receptors, adenosine receptors, opioid peptide receptors, dopamine receptors and cannabinoid receptors separately mediate the effect of electroacupuncture on rheumatoid arthritis pain in the central and peripheral nervous systems and the inflammatory region. The exploration of the mechanism by which electroacupuncture increases GPCRs expression is our future research direction.

3.Effects of Long-term High-fat Diet on the Occurrence of Diabetes in Rats

Objective To investigate the effects of long-term high-fat diet and the modern lifestyle of more food and lack of exercise on insulin resistance and islet function. Methods 75 Wistar rats were feed by routine or high-fat diet for 11 months. Some of high-fat rats were induced to diabetic models by injecting 12mg/kg streptozotocin. Islet function was evaluated by intraperitoneal glucose tolerance test. Insulin sensitivity was detected by insulin tolerance test. Results Compared with control group, 2h blood glucose of high-fat group was elevated significantly. Fasting blood glucose of high-fat group was also elevated at eleventh month. Body weight of high-fat group was increased remarkably compared with control group, and the insulin sensitivity of high-fat group was decreased by 50% at forth month. Then insulin sensitivity of both groups was declined greatly. Hypoglycemic effect of 0.5 U/kg insulin at ninth month was similar to that of 0.1U/kg insulin at forth month. Furthermore, no significant difference of insulin sensitivity was observed between high-fat group and control. Conclusions Main course for insulin resistance is the modern lifestyle of more food and lack of exercise. The main effect of high-fat diet on the occurrence of diabetes is damaging islet function.
4.Construction and expression of human disulfide-stabilized Fv fragment gene to Rabies Virus

Objective To realize the construction and expression of human dsFv genes to rabies virus. Methods The genes of dsFv VH and VL were amplified by PCR-based point mutagenesis strategy. Then the genes were cloned into plasmid pET-22b (+). E. coli BL21(DE3) was transformed with the recombinant expression plasmid and the protein was induced in Luris-Bertani medium by addition of IPTG. The inclusion body proteins of VH and VL were denaturalized by GuHCl, and then re-naturalized in refolding solution to form dsFv fragments. Afterwards, we evaluated the antigen-binding activity of the dsFv and its stability as compared with its originative scFv. Results The fragment genes of dsFv to rabies virus were constructed successfully by PCR-based point mutagenesis strategy. Sequence analysis proved that cysteines were introduced into the position 44 aa of VH and position 100 aa of VL. The dsFv VH and VL genes were expressed in PET22b(+)/BL21(DE3). The VH and VL protein folded into the active dsFv antibody fragments which showed specific binding capability to rabies virus. Conclusion We succeeded in achieving the stabilization of the human scFv to rabies virus and obtaining the active human dsFv antibody fragments. This established a solid basis for further study of the biological activaty and clinical application of dsFv to rabies virus.
5.Therapeutic effect assessment of integrated intervention for learning difficulties in children

BACKGROUND:Children with learning difficulty often present with cognitive impairments and imbalanced development of intelligence accompanied by emotional and behavioral problems.OBJECTIVE:To evaluate the therapeutic effects of integrated interventions in multiple aspects for children with learning difficulties.DESIGN:A case-control study of children with learning difficulties.SETTING:Department of Pediatric Internal Medicine, Union Hospital Affiliated to Fujian Medical University.PARTICIPANTS:This study involved school-age children of 6 to 12years old with learning difficulties who came to the department specialized in pediatric psychology for consultation from July 1998 to July 2001. The diagnosis of learning difficulties conformed to the diagnostic criteria for special learning disability in ICD-10 with IQ > 70 as an essential condition. Children with emotional disorders, mental deficiency, hyperactivity,visual and auditory dysfunctions and organic cerebral diseases were excluded. The diagnosis was further verified by a professor specialized in neuropsychology. The children voluntarily participated in the cognitive training, behavioral intervention and comprehensive trainings of senses and those who fulfilled the one-year treatment were included in this study. The ratio of therapists to the children was 1:2, and each session of treatment lasted for 90-120 minutes given 2-3 times a week, and a treatment course consisted of 24 sessions. The children were supposed to complete 4 consecutive treatment courses. Totally 31 children completed the treatment courses, including 24 boys and 7 girls with the mean age of(7.7±1.2)years who had an average IQ of 87.5±8.6.grated interventions for one year consisting of cognitive training, behavioral intervention and comprehensive training of the senses. Raven's Deductive Reasoning test and Beery Visual-Motor Integration Test were use to examine the deductive reasoning ability and visual-motor integration of the children, and visual attention test and auditory attention test, as well as sequential and reverse-sequence memory tests were employed to test their attentions. Achenbach children behavioral scale was applied to evaluate the children's behavioral problems, and Conners' questionnaire for children's behavioral problem was used to measure their attention-deficit/hyperactivity disorder (ADHD) index.MAIN OUTCOME MEASURES:Changes in the deductive reasoning ability, capability of visual-motor integration, attention and psychological behaviors after one year of intervention.RESULTS:All the 31 patients children entered the result analysis with no loss. After one year of integrated intervention, the children had significantly improved scores for Raven's Deductive Reasoning Test (87.51 ±8.64 vs 90.03±8.70, t=-9.610, P < 0.001) and Beery Visual-Motor Integration Test (6.75±1.74 vs 8.27±1.94, t=-9.002, P < 0.001). Obvious improvement was also seen in the visual attention test (15.90±20.11 vs 2.74±3.67), scope of auditory attention and memory (7.70±1.60 vs 11.23±1.23for sequential memory and 3.52±0.93 vs 6.97±0.16 for reversed-sequence memory, P < 0.01). The total scores for ADHD assessment declined to different extents at 3 and 6 months during the treatment and upon the completion of the treatment (22.65±3.53 vs 18.87±4.14 at month 3, 5.65±3.92at month 6 and 12.65±3.36 at one year, t=7.35-15.36, P < 0.01). The scores for 9 psychobehavioral items in Achenbach children behavioral scale obviously declined were also reduced significantly during the therapy,and 87% (26/31) of the children had obviously improved academic performance.CONCLUSION:Interventions integrating cognitive training, behavioral intervention and comprehensive training of the senses may produce good and persistent effect on the cognitive and emotional symptoms of children with learning difficulties, resulting in obviously improved visual, auditory and brain functions.

9.Correlation study of high-risk type HPV infection with mycoplasma,chlamydia infection among 524 sterile female patients in northwest area

Objective To explore the correlation of high-risk type HPV infection with mycoplasma,chlamydia infection in sterile female patients.Methods The cervical specimens from 524 sterile female patients in northwest area were performed the laboratory detection of high-risk type HPV,mycoplasma and chlamydia.The correlation among these 3 kinds of pathogen was analyzed.Results In 524 sterile female patients,the sequence of the infection rate of 3 kinds of pathogen from high to low was mycoplasma,high-risk type HPV and chlamydia.The positive rate of high-risk type HPV in the patients with simple mycoplasma positive,simple chlamydia positive and double positive of mycoplasma and chlamydia was significantly higher than that in the patients with myco-plasma and chlamydia negative,the difference had statistical significance(P <0.05).Conclusion Certain correlation exists between the high-risk type HPV infection with mycoplasma and the chlamydia infection in sterile female patients.So it is suggested that in the pathogenic microbiological detection,such patients in clinic need to simultaneously perform the high-risk type HPV detection in addition to completing the laboratory detection of mycoplasma and chlamydia.
