2.Establishment of sinoatrial node damage model in canine induced by formaldehyde wet dressing of sinoatrial node aera

Objective To explore the feasibility and reliability to build a sinoatrial node damage model in canine induced by formaldehyde wet dressing.Methods Twenty dogs were randomly divided into 4 groups(5 in each group) by means of random number table:2-hour,24-hour,1-week and 4-week groups after wet dressing.The sinoatrial node area of canine was damaged by wet dressing with 20% formaldehyde.Sinus node function was measured before,2 h after wet dressing and corresponding times of each group.Electrocardiogram(ECG) of body surface were recorded synchronically.Results Average wet dressing time was 6.2?2.6(3 to 12)min.Five dogs showed significantly decrease of heart rate(HR)(140?11 vs 89?6 beat/min,P
3.PTEN mRNA expression in proliferative LECs of the rabbit

· AIM: To observe the effects of PTEN and the tumor suppressor gene in proliferative LECs of the rabbit.·METHODS:Forty-two white rabbits were randomly divided into test group (36 rabbits) and control group (6rabbits). The transparent lenses of treated rabbits were operated with extra capsular cortex extraction, and the controls were kept untouched. The rabbits were sacririced at 1d, 3d, 1wk, 2wK, 1mo and 2mo after surgery.Immunohistochemistry was applied to detect proliferative cellular nuclei antigen (PCNA) as well as hybridization in situ and reverse-transcriptase polymerase chain reactions (RT-PCR) were applied to detect phosphatase as well as tensin homology deleted on chromosome 10(PTEN) mRNA in lens equartor.·RESULTS:PCNA expression increased significantly to a high level at 1wk, began to reduce 2wk later, and recovered to the normal level 1 or 2mo after the surgery.PTEN mRNA expressed positively in normal rabbit LECs.The relative PTEN mRNA contents reduced greatly at 1dafter operation and remained low level at 3d. It began to increase slightly at 1wk, kept rising at 2wk and regained the normal expression after 1 or 2mo. There was inverse correlation between the PTEN mRNA and PCNA expression.·CONCLUSION: PTEN mRNA expresses positively in normal rabbit LECs plasma. PTEN participates in LECs proliferation and correlates with lens proliferative conditions.
4.Study on Preparation and in vitro Properties of Mesalazine Thermosensitive Liquid Suppositories

Objective: To prepare mesalazine thermosensitive liquid suppositories and study the in vitro properties. Methods:Poloxamer 407 (P407) and poloxamer 188 (P188) were used as the temperature-sensitive materials, and mesalazine thermosensitive liquid suppositories were prepared by a cold method. The optimal ratio of P407 and P188 was screened using gel temperature as the in-dex. The gel strength and bioadhesive force of mesalazine thermosensitive liquid suppositories were studied, and the in vitro gel erosion and drug release were investigated as well. Results:The optimal ratio of P407 and P188 was 20%: 2. 5%, and the gel temperature was (36.9 ±0.2)℃, the gel strength was (115.1 ±3.2)s and the bioadhesive force was (130.7 ±5.8) ×102 dyne·cm-2. The drug release and gel erosion in vitro were both fitted first-order kinetics, and the two had promising linear relationship, suggesting good sustained-release property based on gel erosion mechanism of mesalazine liquid suppositories. Conclusion:The preparation of mesala-zine liquid suppositories is simple, and the gelation, bioadhesion and sustained-release of the suppositories are promising. Mesalazine liquid suppositories are valuable to be studied further.
5.Advances in Study on Brain-gut Axis Dysfunction in Pathogenesis of Irritable Bowel Syndrome

Brain-gut axis forms a nerve-endocrine-immune network mediating bidirectionaI reguIation pathway,which pIays an important roIe in maintaining homeostasis between centraI nervous system and gut. Brain-gut axis abnormaIities may Iead to imbaIance of the homeostasis and subsequentIy induce the occurrence of IBS. This articIe reviewed the advances in study on brain-gut axis dysfunction in pathogenesis of IBS.
6.Distribution and significance of NGF in cochleae of neonate rats

Objective To investigate the distribution of NGF in the cochleae of neonate rats and to explore the mechanism of NGF in the auditory path. Methods Twenty healthy neonate SD rats aged 5-7 d were selected and their cochlear paraffin sections were labeled with a rabbit anti-NGF polyclonal antibody by SP method. Results The membranous labyrinth of neonate rats appeared similar to triangle and NGF expression was observed in immature corti’s organ, in spiral ganglion cells and in nerve fibers throughout the cochlea. The distribution located in both cytoplasm and nucleus. Conclusion The distribution of NGF was closely related to survival and maturation of spiral ganglion cells and hair cells in rats aged 5-7 d.
7.Construction of Recombinant Adenoviral Vector Carrying Mus-?NGF Gene and Its Expression in the MSCs

Objective To construct the adenoviral vector carrying Mus-?NGF gene and to study the expression of the gene in the marrow mesenchymal stem-cells(MSCs).Methods Mus-?NGF cDNA was cloned from mice glandulae maxillaries whole mRNA by reverse-transcription polymerase-chain-reactions.The Mus-?NGF cDNA was positively cloned into the adenoviral shuttle vector pAdtrack carrying cytomegalovirus promoter(CMV)and tag-protein(GFP),and then homologous recombined into competent BJ5 183 cells with the adenoviral backbone plasmid pAdeasy-1.The recombinant adenoviral plasmid was identified by restriction enzymes and transfected into HEK293 cells to package and amplify recombinant adenoviral particles which was capable of infecting and would express ?NGF and GFP proteins.After the adenovirus infected the MSCs,the expression was observed via fluorescent microscrope and detected using immunocytochemical method.The secretion of NGF was detected by WB.Results The recombinant adenoviral vector carrying Mus-?NGF gene was constructed and confirmed by restriction endonuclease enzyme analysis.The relucent green fluor-light would be observed in the MSCs.IHC showed that Mus-?NGF gene was exactly transcripted and expressed in the MSCs,and the secretion of NGF was confirmed.Conclusion The exogenous gene,such as ?NGF,an be imported into the MSCs efficiently by recombinant adenoviral vector,and can be expressed and secreted successfully in the cells.It improves foundation for the celluar transplant and gene intervention of MSCs expressing NGF in inner ear.
8.Gasless Laparoscopic Surgery for Benign Ovarian Tumors during Pregnancy:a Report of 45 Cases

Objective To investigate the feasibility, safety and effect of gasless laparoscopic surgery in the treatment of benign ovarian tumors during pregnancy. Methods Clinical data of 45 pregnant women with benign ovary tumors undergoing gasless laparoscopic surgery from January 2006 to June 2013 were studied retrospectively.Most of the cases were given oophorocystectomy. Adnexectomy was performed for the large sized lesions, lacking of or remaining few normal ovarian tissue, or ovarian cyst torsion. Results All the 45 cases were successfully treated by gasless laparoscopic surgery.No perioperative and postoperative complications were observed.There were 42 cases receiving ovarian tumor resection while 3 cases receiving simple salpingo-oophorectomy.The mean operation time was (40.7 ±14.9) min (25-90 min), the mean blood loss was (27.3 ±16.6) ml (10 -80 ml), and the mean hospitalization time was (4.5 ±1.3) days (3-8 days).At pre-, peri-, and post-operative time, the blood pH value, blood pressure carbon dioxide (PaCO2), and blood oxygen partial pressure (PaO2) showed no statistical difference (F=0.00,P=0.999;F=2.21, P=0.114;F=0.60,P=0.555), and blood bicarbonate (HCO3-) and blood oxygen saturation (SpO2) showed statistical difference but no clinical significance (F=14.96,P=0.000;F=9.45,P=0.999), all of which were in the normal range.The fetal heart rates were all in the normal range.Pathological diagnoses were ovarian mature cystic teratoma in 25 cases (55.6%), serous cystadenoma in 6 cases (13.3%), mucinous cystadenoma in 4 cases (8.9%), mesosalpinx cyst in 3 cases (6.7%), endometrial cyst in 2 cases (4.4%), and luteal cyst in 5 cases (11.1%).No spontaneous abortion was found in post-operational follow-up.Term delivery was obtained in 43 cases, with normal fetal weight and the Apgar scores.Two cases gave up the pregnancy in the postoperative period. Conclusion Gasless laparoscopic surgery for benign ovarian tumors during pregnancy is safe and feasible.
9.Study of postoperative visual quality and dry eye and biomechanical stability of patients with myopia and astigmatism treated with different corneal refractive surgeries

AIM: To study the visual quality, dry eye and biomechanical stability of patients with myopia and astigmatism after different corneal refractive surgeries. ·METHODS: A total of 986 patients with myopia and astigmatism were selected as the research object in our hospital from July 2015 to July 2016, according to the operation mode of the selection of the research object, the 986 patients were randomly divided into small incision lenticule extraction ( SMILE) group, femtosecond laser in situ keratomileusis ( FS-LASIK ) group, sub-bowman-keratomileusis ( SBK ) group and laser-assisted in situ keratomileusis ( LASIK ) group. The postoperative visual quality was determined by comparing the diopter, uncorrected visual acuity ( UCVA ) , best corrected visual acuity ( BCVA) and high-order image difference of 25d, 90d. The postoperative dry eye condition was determined by comparing the postoperative tear secretion test ( Schirmer Ⅰ test ) , tear film rupture time ( BUT ) and fluorescence staining ( FS) . The biomechanical stability of the patients was determined by comparing the corneal hysteresis ( CH ) and corneal resistance factor ( CRF ) values of the four groups. ·RESULTS: The diopter, UCVA, BCVA and high-order aberration comparison of FS-LASIK group, SBK group and LASIK group between before and after surgery, showed no significant difference ( P>0. 05 ); on diopter, BCVA, UCVA, there was no significant difference between before and after surgery in SMILE group (P>0. 05), but statistical significance difference on high order aberration (P<0. 05). The BUT and FS value of the four groups decreased obviously after operation, and the difference was statistically significant (P<0. 05). In LASIK group SIt after operation significantly decreased, with statistically significant compared with that before operation ( P <0. 05). After operation, CH and CRF of the four groups decreased with significant differences (P<0. 05). ·CONCLUSION: SMILE, FS-LASIK, SBK and LASIK are equally safe, effective and stable in the treatment of myopia and astigmatism.
