1.Experimental Study on Eqi Compound Prescription Induced Apoptosis of Acute Myelocytic Leukemia HL-60

Objective To explore the apoptotic effect of Eqi compound prescription (contained-herb serum) on cute myelocytic leukemia HL-60 cell, which is relative to intracellular Ca2+, Caspase-3 and Bcl-2. Methods According to serum pharmacology, HL-60 cells were exposed to 10% concentrations of contained-herb serum for 24, 48, 72 and 96 hours respectively. Cells were observed under a fluore-scence microscope. SubG1 DNA was examined by flow cytometer. Intracellular Ca2+ concentration was measured by fura-2 fluorescence load method. Caspase-3 enzymatic activity were measured by colorimetry. Bcl-2 gene expression were measured by LSAB. Results The contained-herb serum could induce apoptosis of HL-60 cells. 2.Therapeutic effect of photodynamic therapy with chlorin e6 and 5-aminolevulinic acid on malignant melanoma in mice

Objective To investigate the effect of photodynamic therapy with chlorin e6 (Ce6) and 5-aminolevulinic acid (5-ALA) on mouse model of malignant melanoma. Methods The mouse model of malignant melanoma was established by injecting 0.1 ml A-375 cells (about 2?10 6 cells) under the right hind leg of BALB/c-6 mice,and that the yellowish white node appears at injection site proves the successful model. Twenty-four of 27 successful mouse models were irradiated at the tumor site with semiconductor laser (wavelength 652 nm) with a total dose of 100 J/cm 2 . Before laser exposure,the mice were treated with 10% 5-ALA by topical compress for 2 h or 7.5 mg/kg Ce6 by intraperitoneal injection for 1 hour or 5-ALA topical application combined with intraperitoneal injection of Ce6 (n=6 in each group). Another six mice as control only underwent PDT. One week after PDT,the mice were killed,the tumor mass was peeled off and weighed,whether the metastasis occurred or not was detected,and the tumor,liver,spleen,lung,kidney were sent to histopathological examination. Results The tumor weight in 5-ALA group,Ce6 group,and the combined group had significant difference as compared with control group (P0.05). The dehydration and scab formation and necrosis could be seen in tumor sites at 1 week after PDT. The cell collapse and necrosis,subdermal thrombosis and cell outline clouding could be observed by histopathological examination. Metastasis of melanoma were found in 5-ALA group,Ce6 group,and the combined group. Conclusion PDT with Ce6 and 5-ALA could kill the malignant melanoma effectively in animal experiment but could not affect the metastasis of melanoma.
3.Impacts on hepatorenal functions and the blood indexes for practitioners occupationally exposed to acrylonitrile

Objective To investigate the health effects on practitioners occupationally exposed to acrylonitrile and to provide scientific bases for the study of toxicological effects of acrylonitrile on human bodies. Methods 465 medical practitioners exposed to acrylonitrile and ( a control group of ) 488 medical practitioners unexposed to acrylonitrile were selected .Age, smoking habits , alcoholic habits and other relevant factors are well-considered in selecting these two groups and all of them had lived in Ningbo for at least three years .Blood samples were collected to measure the level of Alanine aminotransferase ( ALT) , aspartate aminotransferase ( AST ) , alkaline phosphatase ( ALP ) , pancreatic acyl transferase (GGT), total bilirubin (STB), blood urea (UREA), blood uric acid (UA), serum creatinine (SCr), total protein ( TP ) , albumin ( Alb ) , globulin ( Glb ) , ratio of Alb to Glb ( A/G ) , white blood cell ( WBC ) , red blood cell ( RBC ) , platelet ( PLT ) and hemoglobin ( Hb ) .These serum biochemical indices of the two groups were compared to find the differences thereof and to examine the effect of work years on various above-mentioned indices for medical practitioners exposed to acrylonitrile . Results The levels of ALT(t=-2.77,P=0.006), AST(t=-5.74,P<0.001), UREA(t=3.51,P<0.001), UA (t=-3.51,P<0.001)and SCr(t=-7.62,P<0.001)for medical practitioners exposed to acrylonitrile were all significantly higher than those for the control group; however, the levels of ALP(t=18.87,P<0.001), Alb(t=6.92,P<0.001), Glb(t=7.99,P<0.001), A/G(t=11.93,P<0.001), and WBC (t=4.48,P<0.001) were all lower than those for the control group , with the exposure time exhibiting positive correlations with ALT(r=0.564,P<0.001)and AST(r=0.493,P<0.001). Conclusion Acrylonitrile has certain health effects on the hepatorenal function as well as the routine blood test results of medical practitioners occupationally exposed to acrylonitrile .
4.Classification basis and train of thought of functional dyspepsia in disease menu of acupuncture.

OBJECTIVETo explore the classification basis of functional dyspepsia in disease menu of acupuncture, and to provide regular analyzing process for classification of other diseases.
METHODSTaking the control group as the classification basis, and the 5-level literature classification of evidence based medicine as the estimation basis, literatures which complied with the inclusion criteria were examined according to the sequence from high level to low. Evidences of low levels were given up if support can be acquired from documents of high level.
RESULTSAcupuncture is approved to be effective for intervention on functional dyspepsia. Compared to the international accepted medicines such as Cisapride and Motilium, regular acupuncture has better therapeutic effects and less side effects in improving the gastro-power and relieving discomfort sensations.
CONCLUSIONFunctional dyspepsia pertains to grade I of disease menu of acupuncture.

5.Microsurgical treatment of choroids plexus papillomas

Objective To summarize the clinical experiences of clinical surgical treatment of chomid plexus papillomas. Methods The clinical data of 28 patients who were admitced to our department within the last 15 years and definitely diagnosed as choroid plexus papillomas were analyzed retrospectively. Results Choroid plexus papillomas of 28 patients, 22.5 years old on average, were located at the lateral ventricle in 15 cases,the fourth ventricle in 9 cases,the third ventricle in 3 cases and the cerebellopontine angle in 1 case.All the cases had undergone microsurgical treatment,among which 7 underwent preoperative extemal ventricular drainage; total resection was obtained in 24 cases and subtotal resection, in 4 cases; no surgical monality occurred; some cases developed postoperative complications:there was no evidence of tumor recurrence among the 18 cases followed up from 6 months to 5 vears. Conclusiolls Choroid plexus papillomas are mostly benign, occur predominantly in children and have comparatively flavorable prognosis. Satisfactory therapeutic effects could be obtained through active total surgical resection.
6.Clinical features of Symptomatic Rathke's cleft cyst and its surgical treatment

Objective To explore the management of endocrine disturbance related to Symptomatic Rathke's cleft cysts (RCCs) in both pre- and post-operation, as well as the risk factors for the recurrence of RCCs after the operation. Methods The clinical manifestations, radiological and pathologic features, treatment, and surgical outcomes of 11 cases of symptomatic RCCs were reviewed retrospectively. Results Headache recovered in 100% of patients and visual disturbance improved in 83.3%, and Amenorrhea and/or galactorrhea recovered or improved in 66.7% of patients. However, diabetes insipidus and panhypopituitarism did not improve postoperatively. Transcranial surgery was performed in 6 patients and transsphenoidal surgery in 5 patients. No recurrence occurred in transcranial radical resected cases while 2 of 5 subtotally removed cases recurred with an average follow-up of 34.5 months. Inflammation was present in two of the recurrent cases. Conclusion RCCs is a rare pathology with a wide specctrum of clinical and radiological features. Accurate preoperative diagnosis can be difficult. Endocrine disturbance should be evaluated both pre- and post-operatively, and proper hormones replacement therapy is mandatory in these selected patients, Removing as much as possible and multiple biopsies around the cyst wall will be helpful in accurate diagnosis, especially of cases showing atypical pathologic findings, and special attention should be paid to follow-up examination.
7.Sectional anatomy study of the clivus regoin and its three dimensional reconstruction

Objective To study the anatomical characteristics of sectional clivus region and to provide anatomical data for the operation of the clivus regoin.Methods Continuous thin sections(transverse position and coronal position)from two head specimens were performed with freezing milling technique.Observed the anatomical relationship and anatomical characteristics of the clivus region,and finished the three dimensional reconstruction based on the data of sections.Results Totally obtained 320 transverse sections and 232 coronary sections.The related structures were described in 4 typical sections,and three dimensional reconstruction of the clivus region were produced.Conclusion Combinationof section anatomy and three dimensional reconstruction can stereoscopically display the anatomical characters of the clivus re -gion.The three dimensional models could continuously and dynamically display the anatomic structures.
8.Diagnostic value of echocardiography for cardiac tumors in infancy and childhood.

OBJECTIVEThe present study was designed to evaluate the clinical manifestations, surgical findings, pathologic types and treatment of cardiac tumor and to analyze the echocardiographic characteristics of the cases.
METHODSRecords of 19 patients with cardiac tumors confirmed by operations and pathology at Fuwai Cardiovascular Hospital in Beijing, China between Jan, 1983 and Dec, 2003 were reviewed. Clinical and echocardiographic data of all patients were analyzed.
RESULTSThe median age of patients was 7 +/- 5 years, ranging from 5 months to 14 years. There were 8 male and 11 female cases. The surgical findings revealed that 57.9% (11 cases) of cardiac tumors were found in left heart, 36.8% (7 cases) in right heart, 5.3% (1 case) in two ventricles. The pathological study revealed that 17 cases (89.5%) were diagnosed as primary cardiac benign tumors including myxoma in 10 cases (52.6%), rhabdomyoma in 4 cases (21.1%), fibroma in 2 cases (10.5%) and lipoma in 1 case (5.3%). Two cases were diagnosed (10.5%) as cardiac malignant tumors including a primary cardiac rhabdomyosarcoma and a metastatic epithelioid sarcoma. By using echocardiography 11 cases were diagnosed as myxomas and rhabdomyoma (11/19, 57.9%), 8 cases were diagnosed as space occupying lesions of the heart or myxoma (8/19, 42.1%).
CONCLUSIONSMyxomas are the most common heart tumors seen in infancy and childhood, followed in frequency by rhabdomyomas, fibromas and lipomas. Surgery is recommended for patients with refractory and severe clinical symptoms.

9.Role of MUC2 gene in the regulation of rat intestinal barrier function by probiotics.

OBJECTIVETo investigate MUC2 expression in rat colons induced by probiotics and its effects on the inhibition of E.coli K1 (E44) penetration of the intestinal barrier by probiotics.
METHODSSD rats were subjected to intragastric administration of probiotics, E44, or probiotics +E44 on a daily basis for 7 days, and MUC2 expression in the colons was determined by RT-PCR. MUC2-targeted shRNA (shRNA MUC2) and scrambled shRNA plasmids (shRNA NC) were respectively transfected into Lovo cells, and the efficiency of MUC2 knockdown was determined using qRT-PCR. Competitive exclusion assay was used to evaluate the effects of the probiotics against E44 adhesion and invasion.
RESULTSIntestinal MUC2 mRNA expression was up-regulated in the rats after intragastric administration of probiotics, while E44 administration caused significantly lowered MUC2 expression. MUC2 expression was down-regulated (by 66.7%) by transfection with shRNA MUC2 in Lovo cells as compared with the negative control and mock control cells. The inhibition of E44 adherence and invasion by probiotics was significantly attenuated in transfected Lovo cell culture (in which the relative adhesion and invasion rates of E44 were 56.64% and 66.64%, respectively) as compared with those in the control group.
CONCLUSIONThe up-regulation of MUC2 in rat colons can be one of the mechanisms of the probiotics in antagonizing the translocation of the pathogenic bacteria. Silencing MUC2 expression causes attenuated inhibitory effect of the probiotics on E. coli K1 penetration across human intestinal epithelial cells.
