1.Experimental study of injectable intraocular prosthesis in rabbits-Preliminary report

· AIM: To search an easy and simple way for intraocular implantation after the eye evisceration.· METHODS: Fifteen healthy New Zealand rabbits were divided into 5 groups according to the sacrifice time, and each group included 3 rabbits; the left eye received the injection of polymethyl methacrylate (PMMA) bone cement (2g per mL), while the right eye served as control. Under general anesthetia, a 3mm incision was made on the sclera,and the eye contents and pigment tissues were extruded out with fingers. Then, PMMA bone cement (2g per ml) was injected through the scleral incision. Both the operated eye and control eye of the rabbits were enucleated and weighed,The reaction of the operated eye (macroscopically and histopathologically) was noted at frequent interval. The obtained data were then analyzed with ANOVA (SPSS11.5).· RESULTS: There was swelling of eyelids and conjunctiva at the early time after the injection, but no significant difference between the weight of the left and right eyes was noted,Histopathologic examination showed scleral and other tissues necrosis at early period, and then the tissues reaction turned into a great deal of cell proliferation and finally into extensive fibro-connective tissues. Three months after the operation,neovascularization was observed in the cornea of the operated eyes. Histopathologic examination showed formation of fibro-membrane around the intraocular implant,and disappearance of the inflammation.· CONCLUSION: The method of injecting PMMA bone cement (2g per ml) to form an intraocular implant is quite simple and economical; this method is also easy to use clinically.
2.Simultaneous Determination of Sodium Glycerophosphate and Phosphoric Acid in Concentrated Divitamins and Sodium Phosphate Syrup by Quantitative 31P-Nuclear Magnetic Resonance

A 31P-quantitative nuclear magnetic resonance(qNMR)method was established for simultaneous determination of sodium α-glycerophosphate,sodium β-glycerophosphate and phosphoric acid in concentrated divitamins and sodium phosphate syrup.The qNMR experimental conditions were optimized,including hexamethylphosphoramide as internal standard,20%deuterium oxide solution as solvent,zgig pulse sequence,delay time of 30 s,and scan number of 64.The 31P-NMR peaks atδ29.80 of hexamethylphosphoramide,δ0.69 of sodiumα-glycerophosphate,δ0.17 of sodiumβ-glycerophosphate andδ0.03 of phosphoric acid were chosen as the quantitative peaks(pH of the test solution was around 4.8).Method validation was performed in terms of precision(Relative standard deviation less than 0.6%),linearity(Correlative coefficient greater than 0.999),limit of detection(23.58 μg/mL for sodium glycerophosphate and 11.61 μg/mL for phosphoric acid)and limit of quantitation(78.60 μg/mL for sodium glycerophosphate and 38.70 μg/mL for phosphoric acid).The recoveries were 99.8%?103.2%,and relative standard deviations were 0.41%?1.98%.The results showed that the reliability of 31P-qNMR method were suitable for its intended use.Seven batches of concentrated divitamins and sodium phosphate syrups were tested by the established method,of which the total phosphorus content was consistent with that of colorimetry method,but the content of sodium glycerophosphate(Sum ofαtype andβtype)was relatively low,about 82%of the labeled amount.The content of phosphoric acid was high.This method simplified sample pretreatment and had high specificity,and was more suitable for determination and quality control of concentrated divitamins and sodium phosphate syrup.
3.Effects of recombinant human erythropoietin on the immune function of premature rats.

OBJECTIVETo observe the effect of recombinant human erythropoietin (rHuEPO) on immune function of premature rats.
METHODSRHuEPO of 250 IU/(kg.t) or 500 IU/(kg.t) was administered to premature rats every other day for nineteen days. The control premature rats were received normal saline. The changes of hemoglobin (Hb), serum erythropoietin (EPO), red blood cell (RBC) immune function, T lymphocyte proliferative responsiveness, and production of tumor necrosis factor alpha (TNF-alpha) were observed.
RESULTSPremature rats showed lower levels on Hb, RBC immune function, T cell responsiveness and production of TNF-alpha compared with mature rats at birth. The postnatal declines of Hb and RBC immune function were lessened in the treated groups of premature rats, the higher dosage group of 500 IU/(kg.t) was more significant than the lower dosage group of 250 IU/(kg.t). When experiments were over, Hb of control premature rats was (7.72 +/- 0.89) g/dl, Hb of premature rats received 500 IU/(kg.t) was (10.08 +/- 0.90) g/dl (P < 0.01). C3b-R% of control premature rats was (11.00 +/- 0.95)%, C3b-R% of premature rats received 500 IU/(kg.t) was (17.75 +/- 1.04)% (P < 0.01). IC-R% in control premature rats was (12.83 +/- 1.33)%, IC-R% of premature rats received 500 IU/(kg.t) was (10.50 +/- 1.67)% (P < 0.01). The postnatal rise of T cell responsiveness and the production of TNF-alpha in premature rats increased in the treated groups, which was more significant in the higher dosage group of 500 IU/(kg.t) than in the lower dosage group of 250 IU/(kg.t). The OD index of control premature rats was 0.159 +/- 0.014, the OD index of premature rats received 500 IU/(kg.t) was 0.354 +/- 0.050 (P < 0.01). TNF-alpha in control premature rats was (0.270 +/- 0.014) ng/ml, TNF-alpha of premature rats received 500 IU/(kg.t) was (0.415 +/- 0.010) ng/ml (P < 0.01).
CONCLUSIONS(1) Premature rats had lower RBC immune function and T cell responsiveness and underproduction of TNF-alpha at birth. (2) Premature rats had an improvement with the RBC immune function after rHuEPO administration. (3) Premature rats had improvements with T cell responsiveness and TNF-alpha after rHuEPO administration, and there was a positive correction between the RBC immune function and T cell responsiveness with the production of TNF-alpha.

4.The toxic and protective effects of Polygonum multiflorum on normal and liver injured rats based on the symptom-based prescription theory.

The dosage-efficacy/toxicity relationship of the 50% alcohol extracts of Polygonum multiflorum was comparatively investigated on either normal or CCl4-induced chronic liver injury rats, by determining the general condition, serum biochemical indices and liver histopathology, coupled with the factor analysis. The dosages were 10 and 20 g raw materials per kg body weight. Compared with the normal control group, the normal high dose group showed significant increases of the serum alanine transaminase (ALT), total bilirubin (TBIL), high mobility group box 1 (HMGB-1) and interleukin-1β (IL-1β) (P < 0.05 or P < 0.01), as well the frequent incidences of inflammatory cell infiltration, hepatic sinus enlargement and fiber stripes formation in histopathological sections. Compared with the model control group, the model low dose group showed significant declines of serum ALT, aspartate transaminase (AST) and total bile acid (TBA) (P < 0.05), as well the alleviation of vacuoles of hepatocytes, but no amelioration of the inflammatory cell infiltration and fibrous tissue hyperplasia; moreover, the model high dose group showed significant degeneration declines of serum HMGB-1, tumor necrosis factor-α (TNF-α) and IL-1β (P < 0.05, P < 0.01), as well the evident alleviation of vacuoles degeneration of hepatocytes, inflammatory cells infiltration and fibrosis degree. The factor analysis showed that the low dosage treatment had almost neither injuring effect on the normal rats nor protective effect on the model rats; while the high dosage treatment showed observable injuring effect on the normal rats, expressed by the significant increases of the factor-1 (HMGB-1, TNF-α and IL-1β as the main contributors) and factor-2 (TBIL, ALT and TBA as the main contributors) relative to the normal control group. The liver protective effect of the high dosage treatment could be observed with the significant reduction of the factor-1, indicating the effective alleviation of the expression of inflammatory cytokines. In conclusion, it could illustrated the phenomenon of symptom-based prescription theory of Polygonum multiflorum on rat livers: the high dosage of the herb had either an injuring effect on normal rats, or a therapeutic effect on the rats with chronic liver injury.

5.Stress distribution analysis of the surface of condyle during simulated fracture healing of the neck of condyle by miniplate osteosynthesis.

OBJECTIVETo find the reasons why patients always have temporomandibular disorders (TMD) after condylar fracture by analyzing the stress distribution change of the condylar surface whose subcondylar fracture ware fixed by miniplate during the whole healing process.
METHODSAnalyzing the stress distribution change of the condylar surface whose subcondylar fracture were fixed by miniplate during the whole healing process by three-dimension finite element method (3D FEM).
RESULTSDuring the whole healing process of the fracture, the miniplate osteosynthesis was helpful to the biomechanics environment rehabilitation of the condylar local, but it still had difference with normal after fixing 12 weeks long.
CONCLUSIONThe difference of stress distribution of condylar surface that fractured and fixed by miniplate with normal may be part of the reason of TMD after the subcondylar fracture miniplate osteosynthesis.

6.Use of angulated radiograph in clinical judgement of obturation quality of molars with multiple root canals.

OBJECTIVETo investigate different angle projection technique for clinical judgment of obturation quality of molars with multiple root canals.
METHODSEighty-seven maxillary first molars with second mesiobuccal canal (MB2) and 105 mandibular first molars were selected. The canals were instrumented by Ni-Ti rotary instruments and obturated with lateral condensation technique. To judge the obturation quality of the root canals, the radiograph was taken at a horizontal angles of 0 and 20 - 30 degrees from distal direction of the tooth after the treatment.
RESULTSFor maxillary first molars, periapical radiographs showed 23.0% of MB2. Distally angulated radiographs showed 81.6% of MB2. For mandibular first molars, periapical radiographs showed 38.1% of mesiobuccal and mesiolingual canals, and distally angulated radiographs showed 90.5% of two mesial root canals.
CONCLUSIONSMost buccal-lingual distributed root canals of the first molar can be shown more clearly by distally angulated radiographs.

7.Bibliometrics study on dominant diseases in modern acupuncture clinic.

OBJECTIVETo find out the dominant diseases in the clinic of modern acupuncture.
METHODSBy means of bibliometrics, clinical acupuncture study literatures from 1978 to 2004, searched from CBM database, were sorted and counted to show the different clinical utilizing quantities and developing trends of different disease groups in the acupuncture clinic.
RESULTSObviously dominant type: nervous system diseases; mature type: motor system diseases; developing type: 3 kind of diseases including psychosis; premature type: diseases related with surgery; steady type: 3 kind of diseases including digestive system diseases (diseases of liver and gallbladder are not included); pre-developing diseases: 5 kind of diseases including otorhinolaryngologic diseases.
CONCLUSIONAmong all these types, obvious advantage type and mature type are the most distinguishing. Developing type has the most significant ascending trend. Premature type has relatively strong developing potentiality.

8.Effect of acrolein exposure on the percentage of CD4⁺CD25⁺ regulatory T cells and expression of transcription factor Foxp3 in asthmatic rats.

OBJECTIVEThis work was intended to investigate the effect of acrolein fog exposure on the ratio of CD4⁺CD25⁺ regulatory T cells (Treg) and expression of transcription factor Foxp3 in asthmatic rats.
METHODSSixty 6 - 8 weeks male Wistar rats were randomly divided into 4 groups according to random number table (15 rats for each group) which were control group (animals were treated with saline), aerosolized ovalbumin (OVA) exposure group, acrolein exposure group and combined OVA and acrolein fog exposure group, respectively. The rats were exposed to air or/and to saline or OVA aerosol for 6-8 weeks respectively.24 h after the last challenge, 4 ml of peripheral blood and lung tissue were collected from each rat. The percentage of CD4⁺CD25⁺ T cells was determined by flow cytometry analysis. The concentration of interleukin-4 (IL-4) and γ-interferon (IFN-γ) in peripheral blood and lung homogenates were measured by ELISA. The protein expression of Foxp3 in the lung was detected by Western blotting.
RESULTSThe percentage of CD4⁺CD25⁺T cells in aerosolized OVA group ((6.23 ± 1.11)%) was significantly lower than that in the normal saline group ((9.97 ± 1.23)%) (P < 0.01). The percentage of CD4⁺CD25⁺ T cells ((3.26 ± 0.84)%) in OVA combined acrolein fog exposure group was remarkably lower than that in the aerosolized OVA exposure group and in the normal saline group (P < 0.01). IL-4 in both plasma and lung ((22.57 ± 4.34), (0.86 ± 0.12) ng/L) was significantly increased in the OVA exposed rats compared with the normal saline group ((11.57 ± 2.86), (0.31 ± 0.10) ng/L) (P < 0.01). Further remarkable increase in IL-4 of both plasma and lung tissue was observed in the group exposed to both OVA and acrolein ((34.32 ± 6.21), (1.45 ± 0.32)ng/L) compared with the aerosolized OVA exposure group and the normal saline group (P < 0.05). γ-IFN of plasma and lung tissue in OVA exposed group ((59.67 ± 20.12), (0.56 ± 0.17) ng/L) was significantly decreased compared with the normal saline group ((151.74 ± 56.68), (1.54 ± 0.21) ng/L) (P < 0.01), and a further remarkable decrease in IFN-γ of plasma and lung tissue was observed in the group exposed to both OVA and acrolein ((10.12 ± 2.57), (0.49 ± 0.10) ng/L) compared with the aerosolized OVA exposure group and the normal saline group (P < 0.05). Protein expression of Foxp3 in the aerosolized OVA group (8.07 ± 0.24) was lower than that in the normal saline group (10.25 ± 0.31) (P < 0.01), while the protein expression of Foxp3 in OVA combined acrolein fog exposure group (6.38 ± 0.32) was lower than that in the normal saline group and the aerosolized OVA exposure group (P < 0.01).
CONCLUSIONThe number of CD4⁺CD25⁺ Treg cells and the expression of Foxp3 were likely to be altered by acrolein fog exposure, which might play an important role in acrolein induced Th1/Th2 imbalance in asthmatic rats.

9.Analysis of clinical features and prognosis of 98 patients with mantle cell lymphoma

Objective:To study the clinical features, therapeutic effects, survival time, and prognosis of patents with mantle cell lymphoma (MCL). Methods:Clinical data of 98 MCL patients admitted from January 2005 to December 2013 were retrospectively an-alyzed. Results:The median age was 61 years old, and the male-to-female ratio was 2.9∶1. Among these cases, 85 (86.8%) were in Ann Arbor stageⅢ-Ⅳ, 46 (46.9%) had bone marrow involvement, 25 (25.5%) had digestive tract involvement, and 53 chose R-CHOP as first-line treatment. The expected 3-year overall survival (OS) of these patients was only 61.4%. A total of 14 cases were treated with R-CHOP followed by ASCT. The expected 5-year OS was 92.3%, and the OS of the ASCT group was significantly higher than that of the R-CHOP group (75.5 months vs. 43.6 months, P=0.039). Elevated ESR,>60 years old, increased LDH level, B symptoms, and Ki-67≥25% were poor prognostic factors. Conclusion: Most patients with MCL were elder adults with bone marrow involvement. R-CHOP followed by ASCT had better clinical efficacy than conventional chemotherapy in the treatment of MCL.
10.An analysis on the prognostic factors of community -acquired pneumonia among children

Objective To explore the prognostic factors of community -acquired pneumonia (CAP)among children. Methods A total of 783 children with CAP admitted to hospital from 2009 to 2012 were selected.Related information including baseline data was extracted from the hospital health records and analyzed by univariate and multiple logistic regression analysis.Results The logistic regression analysis showed that obnubilation (OR =5.866,95%CI =2.020 -17.033),dysphoria (OR =1.420,95%CI =1.038 -1.942),multi -infection(OR =1.376,95%CI =0.993 -1.907) and polypnea(OR =1.026,95%CI =1.004 -1.049)were the independent risk factors that influenced the prognosis of patients with community -acquired pneumonia.Conclusion More attentions should be paid to CAP children with the symptoms of obnubilation,dysphoria,multi -infection and polypnea.
