Objective To study the effects of exercise on blood pressure (BP) and aorta gaseous molecules of spontaneously hypertensive rats (SHR),so as to explore the role of gaseous molecules in exercise-induced changes of hypertension. Methods Sixteen male SHR were randomly divided into the SHR control (SC) group and SHR training (ST) group.Eight healthy male Wistar rats were used as normal control (WC) group. All the rats were fed normal foodstuff. The ST group was subject to 90-minute moderate swimming exercise once daily,6 times a week, for a total of 8 weeks, while the SC and WC groups were give no special intervention. The blood pressure and the contents of aorta NO,CO and H2S were examined before and at the end of the 4th,5th and 8th weeks of the exercise. Results After 8-week,a within-group comparison showed that the systolic BP (SBP) and diastolic BP (DBP) was significantly elevated in SC group as compared with the baseline BP[(198.07 ± 7.27) vs (159.91 ±6.48) mmHgin SBP,(132.75 ±11.93) vs (103.75 ±3.69) mmHg in BBP](P<0.05),while that the BP remained without significant changes in the ST group [(164.85 ± 3.73) mmHg for SBP and (103.20 ± 7.63)mmHg for DBP] after 8-week of exercise.A between-group comparison showed that BP values measured at the end of 4th,5th and 8th weeks post-exercise in the ST group were significantly lower than those in the SC group (P < 0.05).It was also shown that,at the end of 8th week post-intervention,the levels of aorta NOS,NO,HO,CO,CSE and H2 S of SC group was significantly lower than those of the WC group (P < 0.05),and the levels of NOS,HO,CSE in the ST group were significantly lower than those in the WC group (P < 0.05).Conclusion Moderate exercise can help relieve hypertension in SHR,and the gaseous molecules might synergistically mediate the effect of exercise in lowering the BP.

To compare the difference of total phenol of magnolia solid dispersion prepared by different methods. Hot melt extrusion, solvent evaporation method, and fusion-cooling method were used to prepare total phenol of Magnolia accessory solid dispersion, Plastone S-630 and HPC. The drug dispersion state in the prepared solid dispersion was evaluated with DSC and X-ray diffraction; FT-IR method was used to analyze the possible connections between drug and accessories. Finally, accelerated stability-in vivo dissolution test was use to compare the stability differences between these three processes. The results of DSC and X-ray diffraction showed that all of the drug in solid dispersion processed by three processes can exist in amorphous form; FT-IR results also could not distinguish the difference between the three processes; accelerated stability-in vivo dissolution test showed the stability of solid dispersion prepared by HPC was better than Plastone S-630, and the same kinds of materials solid dispersion prepared by hot melt extrusion showed a better stability than the other two processes.

Objective To compare the clinical efficacy between continuous and intermittent androgen deprivation in high risk prostate cancer. Methods Sixty-four patients with high risk prostate cancer were treated from January 2008 to April 2009,36 cases who accepted goserelin and bicalutamide were taken as intermittent hormonal therapy (intermittent treatment group),while 28 cases who accepted bilateral orchiectomy in addition to flutamide were regarded as continuous hormonal therapy (continuous treatment group). The comparison of tumor specific mortality, time of prostate specific antigen (PSA) to nadir, tine to PSA recurrence, serum testerone and quality of life score were assessed between the two groups. Results In continuous treatment group and intermittent treatment group,follow-up period was (26.4 ± 10.3) and (28.1 ± 8.7) months, the time of PSA to nadir was (3.8 ± 2.1 ) and (4.0 ± 3.6) months, the time to PSA recurrence was (20.1 ± 12.3) and (24.5 ± 14.6) months, respectively. There was no significant difference between the two groups. At the time of 18,24 and 30 months after therapy, serum testerone was 0.85,0.88,0.89 μg/L in continuous treatment group, while 1.21,1.36,1.48 μg/L in intermittent treatment group, respectively (P < 0.05 ).Similarly, quality of life score was 38.7,40.5,39.8 scores in continuous treatment group, while 49.2,51.4,52.3 scores in intermittent treatment group at the time of 12,18 and 30 months after therapy, respectively (P < 0.05 ).Conclusions Clinical efficacy could not been found between continuous and intermittent endocrinic therapy of prostate cancer.During intermittent, quality of life seems to be better and increases in accordance with serum testerone recurrence at given time.

OBJECTIVETo study the overexpression of Sox9 gene on rabbit bone marrow mesenchymal stem cells for repairing articular cartilage injury in vivo.
METHODSRabbit bone marrow mesenchymal stem cells (BMSCs) were transduced with lentivirus vector containing Sox9 gene and then cartilage specific molecule was detected by RT-PCR in vitro. Total 48 knee joints of 24 mature New Zealand white rabbits were randomly divided into 3 groups according to different defect treatment. After animals anesthesia,a full-thickness cylindrical cartilage defect of 4 mm diameter and 3 mm deep was created in the patellar groove using a stainlesssteel punch. Meanwhile, the transfected cells were implanted to repair the rabbit model with full-thickness cartilage defects. Cartilage defects tissue was observed with light microscope, electron microscope, HE and immunohistochemistry staining to assess the repair of defects by the complex at 6 weeks or 12 weeks after the implantation.
RESULTSAt 3 days after the transfection, Sox9 gene expression was highest and Sox9 gene expression decreased with the increase of time. At 3 days after the transfection, the expression of collagen type II began and reached the peak at 14 days. It showed that the bone marrow mesenchymal stem cells went into chondrogenic differentiation after transfected by Sox9 gene. Histological observation showed that at 6 weeks after the operation, the defects in the experimental group was filled with hyaline like cartilage tissue, 12 weeks after operation, the defects of cartilage and subchondral bone had satisfactory healing. Both at 6 and 12 weeks postoperatively, the defects were filled with fibrous tissues in control groups. Meanwhile, immunohistochemical staining of sections with type II collagen antibodies showed the proteins in the regenerated tissue stained positive for type II collagen and stronger than the control groups. The histological scoring system indicated that the cartilage repair of experiment groups were better than the two control groups with statistical significances.
CONCLUSIONOverexpression of Sox9 gene on rabbit bone marrow mesenchymal stem cells (BMSCs) promote the repair of cartilage defect.

Objective To establish the xenotransplanted tumor model of Craniopharyngioma in chick chorioallan?toic membrane (CAM) and detect the angiogenesis ability, microvessel density (MVD) and cell proliferation of the xeno?graft. Method Craniopharyngioma tissues from surgical craniopharyngioma patients were transplanted on the CAM. An?giogenesis assay was performed and the MVD and PCNA were evaluated using immunohistochemistry following the trans?plantation. Results The tumor formation rate of adamantinomatous craniopharyngioma (ACP) and squamous papillary cra?niopharyngioma (SPCP) was 47.14% and 43.33%, respectively. There was no significant difference in tumor formation rate between ACP and SPCP(χ2=0.123,P=0.726). The CAM angiogenesis, MVD and expression of PCNA were higher in ACP than in SPCP. The expression of PCNA was positively correlated with MVD (Pearson r=0.639,P<0.001) and CAM assay score (Spearman r=0.490,P=0.001 ) in CP. Conclusion The model of human craniopharyngioma can be es?tablished in the CAM. The angiogenesis of the xenograft in the CAM can be evaluated and the craniopharyngioma xeno?graft of CAM possesses a new blood circulation and cell proliferation ability.
