1.Safety of porcine fibrin sealant kit on ocular tissue following intravitreal injection

Background Porcine fibrin sealant kit has been widely used in surgery for clotting and woundssealing.It is thought to be a substitution for inert gas and silicone oil in vitrectomy.But whether it produce toxic effect on retina or not after intravitreal injection is still studying.Objective This study aimed to investigate the retinal toxicity of porcine fibrin sealant kit following intraocular application.Methods Vitrectomy was performed on bilateral eyes of 15 pigmented rabbits.Porcine fibrin sealant kit of 0.5 ml was intravitreally injected in the lateral eyes of the rabbits as the experimental group,and equal volume of BSS was used at the same way in contralateral eye as control group.The inflammatory response of eye was observed under the slit lamp and ophthalmoscopy in 1 day,3,7,15,30 days after injection.Schi(o)tz tonometer was used to measure intraocular pressure(IOP) in 1 day,3,7 days,and retinal function was evaluated by electroretinogram (ERG) before operation and 30 days after operation.B-ultrasonic examination was carried out 30 days after surgery.Animals were sacrificed and eyeballs were obtained for retinal histopathological and ultrastructural examination in 30 days after operation.Results Complications appeared in 7 eyes of the experimental group,including cataract in 3 eyes,proliferation vitreoretinopathy and retinal detachment in 5 eyes,endophthalmitis in 1 eye,however,the cataract in 2 eyes and retina injury in 2 eyes were found in 15 control eyes.On the 30 days after injection,no inflammation was found in the 8 eyes without complication in the experimental group and 11 eyes in the control group.The IOP change was insignificant in different groups and various time points (Fgroup =0.008,P =0.929 ;Ftime =3.600 P =0.075).No significant difference was found in a-wave amplitude between groups and various time points(Fgroup =0.728,P =0.405 ; Ftime =0.516,P =0.482),and so was the b-wave amplitude (Fgroup =0.222,P =0.644 ; Ftime =0.057,P =0.814).On the 30th day after operation,arrangement of the retina was regular and no tissue edema and inflammatory cell infiltration were seen in both groups under the light microscope.Transmission electron microscope revealed that the ultrastructures of retinal cells,photoreceptor and retinal pigment cell were normal with the clear subcellular organelle,intact cellular membrane structure and mitochondria in both groups.Conclusions Domestic porcine fibrin sealant kit does not produce toxic effect on retina after intravitreal injection in rabbit.
2.Effectiveness and safety of bicyclol combined with thymosin in treatment of chronic viral hepatitis B.

OBJECTIVETo analyze the efficacy and safety of bicyclol combined with thymosin in treatment of chronic viral hepatitis B (CHB).
METHODSA total of 135 patients with CHB were randomized into experimental group and control group. The patients in the experimental group received bicyclol orally 75 mg daily and thymosin 20 mg intramuscular injection once every 2 days for 24 weeks and those in control group received bicyclol orally 75 mg daily alone for 24 weeks. The levels of serum aminotransferase (ALT/AST), HBV-DNA, HBeAg /antiHBe were observed.
RESULTSCompared with pre-treatment levels, the serum aminotransferase levels decreased significantly in both groups, but there were no statistically significant differences between them. HBeAg negative conversion rate was significantly higher in the experimental group than in the control group (35.3 percent vs.19.4 percent, P less than 0.05). HBV DNA negative conversion rate was significantly higher in the experimental group than in the control group (36.7 percent vs. 20.9 percent, P less than 0.05). No obvious adverse events which were probably related to the drugs were observed in this study.
CONCLUSIONThe combination of bicyclol with thymosin had better effect in treatment of chronic hepatitis B ias compared with bicyvlol alone.

3.Effect of ASO Blood Stasis Syndrome Serum on Vascular Endothelial Cell Injury and Regulation of Taohong Siwu Decoction on it.

OBJECTIVETo explore the effect of arteriosclerosis obliterans (ASO) blood stasis syndrome (BSS) serum on vascular endothelial cell injury and to study the regulation of Taohong Siwu Decoction (TSD) on it.
METHODSUmbilical vein endothelial cell culture system was established. The serum endothelial cell injury model with ASO BSS was prepared. Low, medium, and high concentrations TSD containing serums were respectively added. The endothelial cell proliferation activity was observed by MTT method. Ultrastructures of endothelial cells were observed under transmission electron microscope. Changes of intracellular calcium ion concentration and the cytoskeleton were observed under laser confocal microscope. Contents of ET, NO, and transforming growth factor beta1 (TGF-beta1) in endothelial cell culture supernatant were detected by ELISA.
RESULTSIn ASO BSS serum group endothelial cell proliferation activities decreased, the cell structure was obviously destroyed, calcium ion concentration increased, contents of ET, NO and TGF-beta1 increased significantly (P < 0.01), and ET/NO ratio was imbalanced. After incubating with TSD drug containing serum, endothelial cell proliferation activities and injured cell structures were obviously improved; ET, NO and TGF-beta1 levels decreased (P < 0.05, P < 0.01), ET/NO ratios approximated to the normal level.
CONCLUSIONThe main mechanism of TSD for treating ASO ASS lied in improving injured vascular endothelial cells and endocrine disorder.

4.The Relationship Study between Expressions of P2X5 Receptor and Deficiency-cold Syndrome/Deficiency-heat Syndrome at Various Ambient Temperatures.

OBJECTIVETo detect the expression of the peripheral blood P2X5 receptor at various ambient temperatures, and to explore its relationship with deficiency-cold syndrome and deficiency-heat syndrome.
METHODSSubjects were selected by questionnaire and expert diagnosis, and assigned to the normal control group, the deficiency-cold syndrome group, and the deficiency-heat syndrome group, 20 in each group. 5 mL venous blood was collected at room temperature (25 °C) and cold temperature (-4-5 °C) respectively. Then the expression of P2X5 receptor was relatively quantified by real-time fluorescence quantitative PCR, and compared at room temperature and cold temperature respectively.
RESULTSThe expression of P2X5 receptor in deficiency-cold syndrome and deficiency-heat syndrome groups was lower than that in the normal control group at room temperature (P < 0.05). It decreased more at cold temperature in the deficiency-cold syndrome group than in the normal control group (P < 0.01) as well as in the deficiency-heat syndrome group (P < 0.05). The expression of P2X5 receptor showed no difference in all groups at two different temperatures (P > 0.05).
CONCLUSIONSThe expression of P2X5 receptor was different in different syndrome groups at various ambient temperatures. Ambient temperatures had insignificant effect on the expression of P2X5 receptor of the population with the same syndrome.

5.Correlation research and clinical significance between youth cervical hook and transverse foramen

Objective The correlation analysis was proceeded between youth cervical hook and transverse foramen by using imaging scanning and three-dimensional reconstruction method,which can provide theoretical basis for early diagnosis,treatment and prevention of ju-venile cervical spondylosis.Methods A total of 66 teenagers who had no trauma,neurological symptoms or signs were selected with aged from 6 to 20 years old,who were scanned by multi slice spiral CT,ranged from C1 to T1 .The original data in DICOMformat to import 3D re-construction software was related index measurement and subsequent statistical analysis.Results There was a correlation between hook height,base width,base length of luschca joint and transverse diameter,longitudinal diameter.Conclusion Hook vertebral meridian of lus-chca joint and transverse foramen has a close contact.Factors such as fracture and hyperplasia of luschca joint,vertebral artery of transverseforamen can be oppressed that cause corresponding vertebral artery type of cervical spondylosis.
6.Correlation of uncinate process and various vertebral body structures in adolescents:three-dimensional reconstruction based on CT images

BACKGROUND:With the change of modern living habits, the incidence of cervical disease and cervical related diseases is increasingly at younger age. Based on this feature, the discussion of the relationship between the cervical spine and vertebral body as wel as regular feature can provide theoretical basis for early diagnosis, prevention and treatment of cervical spondylosis. OBJECTIVE:To analyze the correlation structure of the cervical vertebra uncinate process with the vertebral body and intervertebral foramen among adolescents. METHODS:A total of 66 cases without injuries, neurological symptoms or signs of 6 to 20 years old were scanned with multi-row thin-slice spiral CT from C1-T1. The original data were loaded in DICOM format into three-dimensional reconstruction software for measurement and statistical analysis. RESULTS AND CONCLUSION:(1) In addition to uncinate process height and vertebral body height, the uncinate process spacing, uncinate base width and diameter between the vertebrae and a uncinate process base radius vector length and vertebral bodies were positively correlated (P<0.05). (2) Uncinate high uncinate base long uncinate base width and height between the foramen in addition to the uncinate process had a positive correlation with the length of the substrate (P<0.05). (3) In conclusion, there is a certain correlation in juvenile cervical uncovertebral joint between the uncinate process and vertebral body and intervertebral foramen. With the growth of age, cervical activity is increased and the uncinate hyperplasia, trauma, and fracture can oppress spinal nerve within the intervertebral foramen to cause corresponding nerve root type of cervical spondylosis.
7.Immune effects of mutated Hepatitis B Virus precore-core DNA vaccines in mice

Objective To observe the immune effect of DNA vaccines encoding mutated HBV pre-c/c gene(VE2,VFA) in mice. Methods Three kinds of plasmid VEC(DNA vaccines encoding HBV pre-c/c gene),VE2 and VE4 were injected into the thigh muscles of different group of BALB/c mice. Blood and splenocytes from mice were isolated at 4 weeks after immunization. We also have mouse groups immunized with three of these plasmid combined with IFN-γ gene plasmids. The anti-HBc and anti-HBe antibody in peripheral blood in mice were detected by enzyme linked immunusorbent assay( ELISA),antigen-specific cell immune responses were detected by CTL test and enzyme linked immunospot assay( ELISPOt). Results We found that anti-HBe titers of VE2 and VE4 immunizing groups are higher than VEC group( P < 0.05). We also observed that VE2 and VE4 could induce stronger antigen-specific immune responses than VEC and when combined with IFN-γ plasmid,the antigen-specific immune responses are stronger than those without combination immunization in mice (P < 0.05). Conclusions The DNA vaccine VE2 and VE4 could induces stronger antigen-specific immune responses than VEC,and when combined with IFN-γ plasmid,the antigen-specific immune responses are improved in mice.
8.Treatment of displaced radial head fractures by internal fixation with absorbable pins.

OBJECTIVETo study the effect of internal fixation with absorbable pins on treatment of displaced radial head fractures.
METHODSFrom May 1999 to May 2004, 16 patients with displaced radial head fractures (Mason types II and III) were treated with internal fixation by absorbable pins. The duration of follow-up averaged 22.6 months (12-58 months). The outcome was assessed on the basis of elbow motion, radiographic findings and the functional rating score delineated by Broberg and Morrey.
RESULTSAll fractures healed within 10 months without avascular necrosis of radial head. The mean elbow flexion loss was 15 degrees (0 degrees-35 degrees), and pronation and supination decreased by 10 degrees (0 degrees-30 degrees) on average compared with those of the contralateral elbow. Five patients had an excellent result, 6 a good result, and 3 a fair result according to the criteria of Borberg and Morrey.
CONCLUSIONSInternal fixation with absorbable pins is an effective method in treating displaced radial head fractures. It can maintain the biomechanical stability of forearm, improve the elbow function and avoid second operation.

9.Chemical constituents from the male anthotaxy of Populus tomentosa Carr.

Objective To study the chemical constituents of the male anthotaxy of Populus tomentosa Carr.Methods The compounds were isolated and purified by ODS,silica gel,Sephadex LH-20 column chromatography and recrystallization.Their struc?tures were identified based on the physiochemical properties and spectroscopic data. The antioxidant capacities of some compounds were assayed by a rapid ABTS method.The antitumor activity was tested by the MTT assay.Results Fourteen compounds were isolat?ed from the ethyl acetate part of 70% ethanol extract of the male anthotaxy of P.tomentosa Carr.,and they were identified as pinocem?brin(1),dillenetin(2),chrysoeriol(3),naringenin(4),isosakuranetin(5),apigenin(6),kaempferol(7),apigenin-7-O-β-D(-6″-p-coumaroyl)-glucoside(8),tremuloidine(9),catechol(10),3,3′,4,4′-tetrahydroxybiphenyl(11),coumaric acid(12),ursolic acid (13),and betulonic acid(14),respectively. The results of the ABTS assay showed that 2,6 and 7 could scavenge ABTS+free radicals with the total antioxidant capacity of 0.61,0.14,0.46 respectively.The screening results of antitumor activity in vitro showed that the half inhibitory concentration(IC50)of compounds 1,4 and 5 for adenocarcinomic human alveolar basal epithelial cells A549 was 26.04 mg/L,43.45 mg/L and 24.01 mg/L,for human hepatocellular liver carcinoma cells HepG2 were 13.50 μg/ml,23.80 μg/ml and 9.13 μg/ml,and for human colon carcinoma cells HCT116 was 36.11 mg/L,48.95 mg/L and 32.25 mg/L.Conclusion Compounds 2,3,11,13 and 14 were isolated from the Populus genus for the first time,2-5,10,11 and 13-15 were isolated from the plant for the first time.Compounds 2,6 and 7 displayed the antioxidant activity to a certain extent.Compouds 1,4 and 5 showed varying de?grees of inhibitory effects on tumor cells in vitro.
10.Survey of the awareness of Keshan disease and its influencing factors on the residents in a Keshan disease area, Fuyu County of Heilongjiang Province

Objective To explore the inhabitants' knowledge about Keshan disease(KD) in endemic area in Heilongjiang Province and analyze its influencing factors. Methods Yongjin Village, a national KD monitoring site in Fanrong Township of Fuyu County in Heilongjiang Province,was selected as an investigating spot. Specially designed questionnaire was applied in the investigation, which included three aspects, demographical information (gender, age, educational level, etc.), KD-related knowledge survey (KD prevention measures including a healthy diet supplementing selenium, preventing microbial infection, inducers such as climate change, fatigue, cold, mental shock, overeating) and KD controlling measures (take prescribed medicine, dietetic regulation and regular exercise, etc.) Results Seventy-senven point six percent(272/352) knew the disease by the name in the endemic area, while only 29.7% (81/272)and 30.8% (84/272)were aware the preventive measures and inducers , respectively. The awareness rates of KD preventive measures and inducers among the KD patients were 6/17 and 9/17 respectively. The influencing factors of the awareness rate of KD name were gender, vocation and the state of education (χ2 value were 9.838,9.878,12.462, all P < 0.05). However, factors influencing the awareness rates of preventive measures were gender and the state of education(χ2 value were 7.400,20.251, both P < 0.05). Conclusions The awareness rates of KD preventive measures and inducers axe low in the endemic area. The major factors influencing the awareness rate of KD are gender and the educational degree.
