1.Effect of Sodium tanshinone ⅡA sulfonate on activation of extracellular signal-regulated kinase1/2 in angiotensin Ⅱ-induced cardiomyocyte hypertrophy

Aim To observe effects of Sodium tanshinoneⅡA sulfonate(STS) on angiotensionⅡ(AngⅡ)-induced cardiomyocyte hypertrophy and the expression of phosphorylated extracellular signal-regulated kinase1/2 (p-ERK1/2). Methods In the primary culture of neonatal rat cardiomyocytes, as indexes of cardiomyocyte hypertrophy, the total protein was determined by coomassie brilliant blue and protein synthesis rate was measured by [3H]-Leucine incorporation. The expression of p-ERK1/2 was assessed using Western blot and fluorescence microscope. Results ① The total protein and protein synthesis rate stimulated by Ang Ⅱ(1 ?mol?L-1)in the cardiomyocytes increased significantly in contrast to that of control; STS could effectively decrease the increased total protein level induced by Ang Ⅱand markedly inhibit synthesis of protein. ② AngⅡ(1 ?mol?L-1) had the effect of promoting activation of ERK1/2 and then appeared in nucleus rapidly. The translocation process of ERK1/2 induced by AngⅡ was blocked distinctly by STS. ③ Cardiomyocyte pretreated with Ang Ⅱ(1 ?mol?L-1)for 5 min, the p-ERK1/2 protein expression began to increase ,the peak effect was at 10 min. While pretreatment with STS(2, 10, 50 ?mol?L-1) ,Ang Ⅱ-induced increase in p-ERK1/2 were inhibited evidently. ④ In pretreatment of cardiomyocyte with STS in different doses for 30 min, STS was found to be able to inhibit the expression of p-ERK1/2 stimulated by AngⅡ in a dose-dependent manner. Conclusions The results suggested that activation of ERK1/2 might play an important role in cardiomyocytes hypertrophy induced by Ang Ⅱ,and the anti-hypertrophic effect of STS on cardiomyocyte hypertrophy induced by AngⅡ might be associated to its inhibitory effect on ERK signaling pathway.

Aim To investigate the effect of tanshinoneⅡA(TSN) on the hypertrophy induced by angiotensinⅡ(AngⅡ)in the primary culture of neonatal rat cardiomyocytes. Methods Cardiomyocytes of Wistar rats were cultured with pancreatin and preplating technique. The hypertrophy of cardiomyocytes was induced with angiotensinⅡ, and intervened with TanshinoneⅡA and Valsartan. The effect of TSN on cardiomyocytewas evaluated by 3-[4,5-dimethylthiazol-2-yl]-3,5-diphenylformazan (MTT) assay. As the index of cardiomyocyte hypertrophy, protein synthesis rate was measured by ~3H-Leucine incorporation and the cell size was determined by phase contrast microscope. The proto-oncogene c-fos、c-myc and c-jun mRNA expressions were assessed using reverse transcription polymerase chain reaction (RT-PCR). Results Exposure of cultured cardiomyocytes to TSN (5~80 ?mol?L~ -1 ) for 24 h produced marginal cytotoxicity. Additonally, myocyte monolayers continued to contract synchronously in the presence of TSN. With the treatment of cardiomyocytes by AngII for 7 days, the cell size of cardiomyocytes of the group of Ang Ⅱ(28.5?3.8) ?m increased more prominently than the group of control(19.8?1.9) ?m(P

4.Effect of polyethylene glycol 3350 plus electrolytes on the constipation induced by clozapine

Objective To explore the efficacy and safety of Polyethylene glycol 3350 plus electrolyte bulk on the schizophrenia patients with constipation induced by clozapine. Methods 150 cases of schizophrenia inpatient and outpatient with constipation induced by clozapine treatment were selected and randoly dirided into the group of polyethylene glycol,lactulose group and non-intervention group with 50 patients in each group and each patient or their families signed the informed consent. The treatment groups took orally with Polyethyle ne glycol 3350 plus electrolyte 13. 7grams,2 times per day and lactulose oral solution 10ml,3 times per day respectively. The non-intervention group was not given a regular basis laxatives, but with cathartic therapy in demand. The period of experiment was 6 weeks including 2 weeks baseline observation period and 4 weeks treatment. Assessment criteria: an overall assessment of all symptoms of patients and each patient's constipation and safety parameters were assessed. Results After treatment for 4 weeks,the total effective rate in three groups was 90.0% ,68.0% ,38.0% (all P <0.05). The difference was statistically significant. The normal shape of stool rate was 84.0% ,60.0% ,28.0%. PEG group were better than the other 2 groups at baseline in the first defecation time,the average times of bowel movements per week,theutilization of laxatives during treatment, while symptom score improvement in the stool was significantly better than other groups after treatment. The safety parameters including liver and kidney function, lectrolytes, glucose, ECG, symptoms of schizophrenia such as PANSS score had no obvious change in each group after treatment. Conclusion PEG 3350 plus electrolytes was effective and safe in the treatment of clozapine-indueed constipation in chronic schizophrenia.

6.Effects of Dexmedetomidine on Renal Function of Patients after Valvular Heart Surgery

Objective To investigate the effect of dexmedetomidine on postoperative renal function of patients with rheumatic heart disease following valvular heart surgery. Methods Fifty patients following valvular heart surgery were randomized into experimental group and control group,with 25 cases in each group. The patients in experimental group receiveddexmedetomidine 0.5 μg/kg infusion 15min before induction, and then 0.5 μg/(kg·h) infused to the end of surgery, while those in control group received the same amount of saline in the same way. Blood from central venous was collected before surgery, at the end of surgery, 24 h and 48 h after surgery in order to detect Src, BUN and Cys C and the length of ICU duration was also recorded. Results Src and Cys C of the patients in both groups obviously elevated 24h after surgery when compared with pre-operation , while BUN elevated at both 24 h and 48 h after surgery(P < 0.05). Compared with those in control group, Src, BUN and Cys C were lower in experimental group ( P < 0 . 05 ) , and the length of ICU duration was shorter (P < 0.05). Conclusion Dexmedetomidine could improve the postoperative renal function after valvular heart surgery and lead to a better prognosis.
7.Study on expression and clinical significance of ARD1 in nasopharyngeal carcinoma

Objective To detect and explore the expression of ARD1 and its clinical significance in the nasopharyngeal in-flammatory tissue ,nasopharyngeal carcinoma group and its subgroups .Methods Expression of ARD1 in nasopharyngeal carcinoma (56 cases) and nasopharyngeal inflammatory tissue (20 cases) were detected by immunohistochemical staining SP ,the correlation between the expression of ARD1 and age ,gender ,histological grade ,TNM clinical stage and tumor metastasis were analysed . Results The positive expression rate of ARD1 were 10 .00% (2/20) ,55 .35% (31/56) in the nasopharyngeal inflammatory tissue and nasopharyngeal carcinoma ,respectively .The expression level of ARD1 in nasopharyngeal carcinoma was significantly higher than in the nasopharyngeal inflammatory tissue ,the difference was significant (P< 0 .05) ;expression of ARD1 in the nasopharynge-al carcinoma was correlated with the histological grade of nasopharyngeal carcinoma(P< 0 .05) and the expression was increased in poor differenciation tissue .But there was no statistical difference between the expression of ARD1 and the patient′s age ,gender , TNM clinical stage ,tumor metastasis(P> 0 .05) .Conclusion The expression of ARD1 is high in nasopharyngeal carcinoma ,and has a closely correlation with diffferentiation level of tumor ,which suggested that ARD1 may be involved in the the occurrence and development of nasopharyngeal carcinoma .However ,further research needs to be done for its mechanism in the nasopharyngeal car-cinoma .
8.Detection of sex hormone levels in children with hypospadias

Objective To investigate the differences in sex hormone levels between hypospadias and circumcision groups.Methods Fifty cases of circumcision and 137 cases of hypospadias the dihydmtestosterone (DHT) value was tested with radioimmunoassay,and testosterone was tested with lightimmunoassay.Results DHT value was (64.51 ±32.10)pg/ml in circumcision group,and (46.72 ±28.94)pg/ml in hypospadias group (P <0.05).DHT value in hypospadias type Ⅰ,Ⅱ,Ⅲ,and Ⅳ were (50.20 ±32.90)pg/ml,(46.63 ±25.67)pg/rnl,(51.60 ±32.16)pg/ral,and (39.02 ±26.32)pg/ml,respectively (P =0.29).The differences between circumcision and hypospadias groups were statistically significant (P =0.00).Luteinizing hormone (LH) and testosterone (T) in children with hypospadias were significantly lower than those in children in the circumcision group (P < 0.05).No statistically significant difference was found between two groups in follicle-stimulating hormone (FSH),estradiol (F2),and prolactin (PRL) (P > 0.05).No statistically significant differences were found in FSH among all types of hypospadias (P > 0.05).Conclusions Inadequate secretion of T or activity insufficiency and functional deficiency of 5 alpha reductase (SRD5A) are likely found in children with hypospadia.Inadequate secretion and low T value might be found in LH-T shaft in children with hypospadia.The normal T value in some children with hypospadia does not show that androgens produced during pregnancy are normal.
10.Effects of breath holding in air on cardiac responses in man.

Healthy male subjects were asked to hold their breath in air at the tidal inspiratory level, and time-dependent changes in mean values of several parameters of cardiovascular function were studied. The heart rate increased abruptly after the beginning of breath-holding and then decreased slightly with time. The stroke volume (SV) remained significantly low until the end of breath-holding, so that the cardiac output was decreased in parallel. With regard to the systolic time interval, the totai eiectromechanical systole and the left-ventricular systolic time (LVET) were not significantly changed, whereas the pre-ejection period (PEP) was markedly prolonged. Therefore, ratios of PEP/LVET and SV/LVET were increased and decreased with time, respectively. As the systolic, and in particular the diastolic blood pressures were elevated, the pulse pressure was reduced. Ratios of the diastolic and systolic times in relation to the heart rate at rest, during breath-holding and during recovery after breath-holding were all proved to be described by a single regression curve. However, the ratios obtained for cycling exercise at various heart rates showed a marked shift above the curve. These results clearly indicate that cardiac performance is lowered during breath-holding in air.
