1.Effects of pulmonary resection on perioperative right ventricular function

Objective To investigate the changes of perioperative right ventricular function after pulmonary resction. Methods 45 cases of pulmonary resection were divided into four groups.Group Ⅰwas wedge resection(n = 10), GroupⅡ was lobectomy(n = 19), Group Ⅲ was double lobectomy(n=7)and Group Ⅳ was pneumonectomy(n=9). The changes of CVP, PAMP, PEP/ET, Sa, VTIs, RVSP, Ea/Aa, and Tel were evaluated by traditional ultrasonic and tissue Doppler ultrasonic techniques at different time inter- vals (pre-operation, 5-7 days orland 1 month post-operation), Results Compared with the pre-operation CVP, the indexes of all groups have no significant changes post-operatively. Wedge resection didn' t obviously influence right cardiac after load and systolic function/diastolic function. No evidet changes detected in Tei pre-and post-operatively. However, the indexes (PAMP, RVSP and PEP/ET ratio) reflecting right cardiac afterload evidently increased at post-pneumonectomy or lobectomy group5- 7 days post-opera- tive. All these reflect the decrease of Tei, which was more obvious in pneumonectomy than in lobectomy group. Right cardiac after- load, systolic/diastolic function and Tel index recovered to pre-operative level 1 month post-operation in single lobectomy group. But the above indicators, especially the Tel, were still high in double lobectomy and pmeunonectomy groups 1 morth post-operation.Tei index is positively correlated with PAMP and is weakly correlated with PET/ET ratio and Ea/Aa ratio. Conclusion Pulmonary wedge resection doesn't evidently influence right cardiac function. However, right cardiac diastolic function evidently decreases temporarily at lobectomy group. The systolic function and diastolic funetions decrease after double-lobectomy and pneumonectomy and it's more evident in pneumonectomy group. Though the right cardiac afterloads of lobectomy, double lobectomy and pneumonectomy groupa all increase significantly post-operativlye, only the former recover to pre-operative level 1 month after surgery.

Objective To investigate the inhibitory effects of overexpression of PTEN on renal epithelial-mesenchymal trans-differentiation induced by TGF-β1,and the signaling transduction mechanism.Methods HKC cells were transfected with GFP-PTEN via lipofectAMINE2000.The efficiency of transfection was detected by fluorescence microscope.The expression of PTEN protein and mRNA in the translected cells was detected by Western blot and RT-PCR respectively.The experiment was divided into four groups:normal group,TGF-β1 stimulation group,GFP-PTEN+TGF-β1 group and empty vector+TGF-β1 group.The expression of E-cadherin,a-SMA,Akt and p-Akt was detected by Western blot.Results Most ceils transfected with GFP-PTEN expressed GFP.The expression of PTEN protein and mRNA was strongly increased when HKC cells were transfected with GFP-PTEN(all P<0.05).In both TGF-β1 stimulation group and empty vector+TGF-β1 group,the expression level of E-cadherin was lower(all P<0.05),while that of p-Akt and a-SMA was higher than in normal group(both P<0.05).The expression level of p-Akt and a-SMA in GFP-PTEN+TGF-β1 group was Iower(both P<0.05),while that of E-cadherin was higher than in TGF-β1 stimulation group and empty vector+TGF-β1 group(both P<0.05).The expression of Akt was similar in the four groups.Conclusion Overexpression of PTEN can inhibit renal epithelial-mesenehymal trans-differentiation induced by TGF-β1 through suppressing the activation of PI3K/Akt signal pathway.
4.The study of associated reconstruction using MV linear accelerator and cone-beam CT.

In this paper, we proposed a new scan mode and image reconstruction method, which combines the data from both the linear accelerator and the cone-beam CT to reconstruct the volume with a limited rotation angle and low sampling rate. The classical filtered backprojection method and the iterative method are utilized to reconstruct the volume. The reconstruction results of the two methods are compared with each other with a relavant anlysis given here.

5.Effect of different pacing sites on ventricular synchrony evaluated by gated blood pool SPECT

Objective To compare the effect of right ventricular outflow tract (RVOT) and right ventricular apex (RVA) pacing on ventricular systolic synchrony using gated blood pool SPECT (GBPS).Methods A total of 50 patients implanted with pacemaker due to high degree or complete atria-ventricular block were enrolled in the study. Twenty-three patients were RVOT paced ( Group A, n = 23) and 27 were RVA paced (Group B, n=27). Twenty-four patients with malignancy, normal echocardiographic findings and no history of cardiac diseases were scheduled for pre-chemotherapy evaluation of cardiac structure and function and were enrolled as control group ( Group C, n = 24). All patients underwent GBPS imaging and the values of phase angle (PS), mean phase of each wall, standard deviation (SD) of mean phase of each wall, lateral-septal motion delay of left ventricle ( LV Sep-Lat Delay), septal-right ventricular (RV) delay of LV ( LV Sep-RV Delay) and LV-RV Delay were acquired. The parameters of ventricular systolic synchrony among the three groups were compared using one-way ANOVA. Results The mean phase of LV lateral wall in Groups A and B were significantly higher than that in Group C: Group A (120.50 ±40.58) ms; Group B (103.23±28.34) ms; Group C (84.63 ±22.38) ms (F=7.72, P <0.05). There was no significant difference between Groups A and B ( t = 1.30, P > 0.05 ). The mean phase of RV in Group A was significantly larger than those in Groups B and C: Group A ( 137.05 ± 39.27) ms, Group B ( 100.85 ± 23.79) ms,Group C (59. 13 ±30.52) ms (F=35.55, P<0.05). PS, SD and LV Sep-Lat Delay in Groups A and B were significantly higher than those in Group C: (85.73 ± 12.00)°vs (89.85 ± 15.61 )°vs (58.95 ±9.87)°, (27.68±10.66) ms vs (26.15 ±13.02) ms vs (15.63 ±8.35) ms, (25.06±34.23) ms vs (2. 62 ± 60. 31 ) ms vs ( - 23.66 ± 31.39) ms, F = 41.54,8.55,6.81, all P < 0.01 ), however, there was no significant difference between Groups A and B ( t = 0. 68, 0.68, 1.30, all P > 0.05 ). LV Sep-RV Delay and LV-RV Delay were significantly different among the three groups ( LV Sep-RV Delay: Group A (57.60 ±56.77) ms, Group B (6.36 ±61.88) ms, Group C ( -41.89 ±35.78) ms; LV-RV Delay:Group A (47.36 ±42.59) ms, Group B ( 3.08 ± 38.81 ) ms Group C ( - 26.50 ± 20.99 ) ms, F = 20. 32,25.38, both P < 0.01 ). Conclusion Both RVA and RVOT pacing increase the segmental phases detected by GBPS, causing inter- and intra- ventricular asynchrony compared with patients without pacemakers.
6.Endovascular embolization for intracranial aneurysms in patients with Moyamoya disease: a report of 8 cases

Objective To observe the treatment effect of endovascular embolization on intracranial aneurysms combined with hemorrhagic moyamoya disease (MMD). Methods The clinical features and outcomes of 8 patients with intracranial aneurysms combined with MMD, performed endovascular embolizafion in our hospital from January 2002 to February 2010, were retrospectively analyzed. The aneurysms in 6 patients with major artery aneurysm were successfully embolized with Guglielmi detachable coil, and the other 2 (the peripheral artery type) with n-butyl cyanoacrylate (NBCA). Results In the 6 patients performed embolization with Guglielmi detachable coil, 3 got complete embolization, 2 vestigital of the aneurysm neck, 1 partial embolization and all the parent arteries were unobstructed. In the 2 patients performed embolization with NBCA, no imaging was noted on the parent arteries and aneurysms, and no cerebral infarction appeared in the distal blood supply region.Conclusion For MMD patients with aneurysms, we can treat the aneurysms with endovascular embolization safely and effectively.
7.Application of modified quadruple stapling technique in radical proximal gastrectomy for gastric carcinoma.

OBJECTIVETo investigate the feasibility and safety of modified quadruple stapling technique in radical proximal gastrectomy for gastric carcinoma.
METHODSMedical records of 55 consecutive patients who underwent radical proximal gastrectomy for gastric cancer were analyzed retrospectively. Twenty-eight patients (modified group) undergoing modified quadruple stapling technique were compared to 27 patients (traditional group) who underwent traditional approach during the same period.
RESULTSThere was no perioperative mortality. All the patients had negative pathological resection margin. The mean operative time in the modified group was significantly shorter than that in the traditional group [(158±31) min vs. (195±42) min, P<0.05]. There were no immediate complications such as stricture, bleeding or leakage at the anastomosis, gastroparesis, and wound infection. Postoperative recovery did not differ between the two groups (P>0.05). During the follow-up (range: 3 months-2 years), 2 (7.1%) patients in the modified group and 2 (7.4%) in the traditional group developed reflux esophagitis (P>0.05) and anastomotic inflammation occurred in 2 cases (7.1%) for the modified group and 8 (29.6%) for the traditional group (P<0.05).
CONCLUSIONModified quadruple stapling technique is a feasible and safe method in radical proximal gastrectomy.

8.The oral immune efficacy of recombinant lactobacillus casei expressing CSFV E290 peptide and it elicited specific CTL response.

UNLABELLEDThe gene encoding classical swine fever virus (CSFV) T cell epitope E290 peptide was synthesized by PCR, cloned into the expression vector pPG-VP2 and named pPG-VP2-E290. The recombinant plasmid was electrotransformed into Lactobacillus casei 393 generating pPG-VP2-E290/L. casei 393. Specific anti-CSFV E290 peptide immunoglobulin G (IgG) antibody was detected by indirect ELISA in the serum of BALB/c mice and rabbits immunized with recombinant strain by oral administration. The CTL of E290 was analyzed with lymphocytes taken from the immunized mice, and the immunized rabbits were attacked with CSFV to validate the protective function of E290 antibody induced.
RESULTThe recombinant expression system constructed with L. casei 393 in this study show a good immunization property and could elicit the mice and rabbits to produce high anti-E290 antibody levels. Furthermore, E290 peptide antibody could elicit specific CTL response, and restrain attack of CSFV to rabbits.

9.Clinical analysis of therapeutic effect of zoledronic acid combined with radiotherapy for metastatic bone cancer.

OBJECTIVETo evaluate the efficacy of zoledronic acid combined with local radiotherapy for limited metastatic bone cancer.
METHODSForty-five patients with limited bone metastatic cancers were randomly divided into two groups: 23 in the combination group who received intravenously administration of zoledronic acid and local radiotherapy, the other 22 in the radiotherapy alone group who underwent local radiotherapy only.
RESULTSThe response rate of pain relief was 91.3% in the combination group versus 86.4% in the radiotherapy alone group, without statistically significant difference between two groups (P > 0.05). However, the recalcification rate was significantly higher in the combination therapy group (52.2%) than that in radiotherapy alone group (22.7% P < 0.01), and the proportion of patients with new bone metastasis formation was significantly lower in the combination group (13.0%) than that in the radiotherapy alone group (40.9%, P < 0.05). The common side-effects were transient pyrexia and nausea.
CONCLUSIONZoledronic acid combined with local radiotherapy is effective in relieving pain, improving bone recalcification and reducing the formation of new bone metastasis.

10.Production and Characterization of Monoclonal Antibodies of Shrimp White Spot Syndrome Virus Envelope Protein VP28

BALB/c mice were immunized with purified White spot syndrome virus (WSSV).Six monoclonal antibody cell lines were selected by ELISA with VP28 protein expressed in E.coll in vitro neutralization experiments showed that 4 of them could inhibit the virus infection in crayfish.Westernblot suggested that all these monoclonal antibodies were against the conformational structure of VP28.The monoclonal antibody 7B4 was labeled with colloidal gold particles and used to locate the VP28 on virus envelope by immunogold labeling.These monoclonal antibodies could be used to develop immunological diagnosis methods for WSSV infection.
