Objective To investigate the possible mechanism of the biologic ethology effects of NGF and SP(substance P)on primary cultured adrenaline medullary chromaffin cells(AMCC).Methods To establish primary cultured AMCC by means of enzyme digestion and purify the cells by means of isopycnic gradient centrifugation and differential plating. To observe the morphological and ultrastructural changes after the addition of NGF and SP,and detect the concentration of adrenaline and noradrenaline in serum by ELISA.Results Confervaceous processes could be observed after 2 d of addition of NGF to the culture and the processes strenched longer as days went by the observation of electron microscopy.there are some drumstick-like and villiform processes in the cell membrane and some vesiculation be observed near the cell membrane of the primary cultured AMCC cells after the addition of NGF.The bioblast was abundant but the structure was not clear in the intracytoplasm and the concentration of adrenaline were decreased(P

Objective To establish the rapid purification of Coxsackievirus A16 using ultracentrifugation .And To prepare and i‐dentify the neutralizing monoclonal antibody against CA16 .Methods The CA16 culture supernatant was harvested and then con‐centrated by 100K capsule .The concentration of CA16 was purified by cesium chloride ultracentrifugation .Purification of CA16 were identified by transmission electron microscopy .BALB/c mice were immunized with inactivated CA16 .Spleen cells were harves‐ted and fused with SP2/0 myeloma cells ,hybridoma cell strain secreting mAb against CA16 were objected to screening .Character‐ization of the prepared mAb were analyzed by ELISA and microneutralization assay .Results The purified CA16 method of cesium chloride gradient ultracentrifugation was established ,TEM analysis was showed that CA16 particles have icosahedral structure ,the diameters of the viral particles were approximately 20-30 nm .Two hybridoma cell strains secreting mAb against CA16 were ob‐tained ,the subtypes of two mAbs were IgG2a ,the binding titers of Anti/CA16/5 and Anti/CA16/10 were 103 and 104 respectively . Neutralizing titer of the two mAbs were 1∶256 and 1∶1 024 respectively .Conclusion Establishment method of cesium chloride gradient ultracentrifugation was performed to purify CA16 ,the two mAbs with neutralizing ability to against CA16 may become ap‐plication of treatment and vaccine .

Microbiology 2008;0(12):-
This paper investigated contamination situation of Listeria monocytogenes(Lm). To compare dif- ferent selectivity enrichment broth for detectable effect of Lm and compare detectable effect in different samples by using different methods, furthermore, choose the best enrichment broth for specific food. One hundred and thirty five random samples from raw meat, aquatic product, fruit and vegetable, quick-frozen food in Baoding. Applied LB enrichment broth, EB enrichment broth, new modification FDA enrichment broth and Fraser enrichment broth before separated by PALCAM selective agar, then identified by interna- tional standard method after PCR. Results: Four methods showed that there were 23 Lm positive, detected 5 Lm by LB method, 6 Lm by Fraser method, 5 Lm by EB method and 7 Lm by new modification FDA method. The total detectable rate of four methods had no large specificity, but to specific kind of food was different.

Objective To investigate the dynamic changes of interleukin-1 receptor-associated kinases (IRAK-4) in hypoxic neurons and explore their role in regulation of inflammatory reaction. Methods The B35 cells exposed to hypoxia of 3% O2,5% CO2 and 92% N2 were cultured for 1,3,6, 12,24,48,72 and 96 hours respectively. Then, mRNA and protein expressions of IRAK-4 were detected by RT-PCR and Western blot, the expression of IRAK-4 in the cells were observed by laser scanning con-focal microscope (LSCM), and the concentration of IL-6 was measured by ELISA method. Results After hypoxia, the mRNA and protein expressions of IRAK-4 were increased at one hour, reached the peak at six hours (P<0.05), kept at a high level at 12 hours (P<0.05) , but decreased gradually to the normal oxygen level at 24 hours (P < 0.05) and to below the normal oxygen level at 48 hours (P < 0.05). Immunofluorescence results showed that the fluorescence intensity of IRAK-4 was gradually increased with time. The changes of IL-6 in the supernatant were positively correlated with protein expression of IRAK-4 (r =0.84, P < 0.05 ). Conclusions Hypoxia can increase the expression of IRAK-4 at transcription and translation levels in a certain period of time, which may participate in down-stream inflammatory reaction and lead to increase of IL-6 expression.
