Objective To observe the abnormality on behavioral ability of transgenic mice for HRX-EEN fusion gene.Methods Transgenic mice for HRX-EEN fusion gene(transgenic group,n=12)and C57/BL mice(control group,n=12)were tested in hidden platform training(day 1 to day 4)and probe trial testing(day 5)in Morris water maze in which ability of spatial learning and retention was assessed.Results In hidden platform training,the latencies of transgenic group were longer than those in control group,and significant differences were observed between the two groups for day 2,3 and 4(P

5.Value of Different Indexes in Evaluating Insulin Resistance in Obese Children

Objective To explore the effect of different indexes on evaluating insulin resistance in obese children. Methods Oral glucose tolerance test (OGTT) was performed in obese children (n= 61) and age - matched normal volunteers( n= 23) Serum glucose and insulin levels were determined at 0,30,60,120,180 min after OGTT, insulin resistance was estimated by homeostasis model assessment of insulin resistance (HOMA-IR). 8.Establishment of rabbit model of juvenile nonalcoholic steatohepatitis.

OBJECTIVETo establish a rabbit model of juvenile nonalcoholic steatohepatitis (NASH) for further study.
METHODSTwenty-eight New Zealand rabbit pups were fed with a high-fat diet (standard diet+10 % lard+2 % cholesterol) for 8 or 12 weeks as the two model groups, and 10 rabbits were fed with standard diet as the controls. Liver tissue samples were collected for Heamatoxylin-Eosin staining and pathological examination.
RESULTTypical histological hepatic lesions of NASH were observed in both model groups. Compared with control group, model groups showed a significant increase in serum ALT, AST, TG, TC levels (P <0.01), and decrease in serum adiponectin, IL-10 levels (P <0.05), meanwhile there was no significant difference between two model groups. TC and the degree of liver fatty infiltration were independent determinants of serum adiponectin level by stepwise multiple regression, beta=-1.33, P=0.006 and beta=-0.97, P=0.038, respectively, R square equal to 0.294.
CONCLUSIONThe juvenile steatohepatitis rabbit model has been established and the level of adiponectin can partly reflect the severity of liver steatosis.

9.Relationship between sleep duration and metabolic syndrome in obese children

Objective To investigate the relationship between sleep duration and metabolic syndrome in obese children.Methods Sleep time was recorded and metabolic syndrome (MS) diagnostic criteria as blood pressure,body mass index (BMI) were measured among 311 obese children.The children were divided into 4 groups by sleep time while normal-weight children served as the control group.A disorderly multi-classification logistic regression method was used.Results Statistically,significant differences were found in the levels of BMI,blood pressure,fasting blood glucose and postprandial 2 h glucose and the incidence rates of abnormality on MS,blood pressure,fasting insulin,and BMI≥30 kg/m2 between the group with daily sleep time less than 8 h and the group with daily sleep time no less than 10 h (P<0.05).Date from logistic regression analysis revealed that the reduction of sleep time was the risk factor of MS in obese children.Conclusion Insufficient sleep could greatly contribute to the occurrence of MS in obese children.
10.Diagnostic value of the ratio of ligamentum flavum thickness to oblique diameter of lumbar canal in patients with ligamentum flavum hypertrophy

Objective To evaluate the diagnostic advantage of the ratio of ligamentum flavum(LF) thickness to oblique canal diameter(TODR)measured on CT images in patients with lumbar canal stenosis. Methods Seventy-one patients underwent CT and MRI examinations respectively,and they were divided into two groups,the positive group and negative group,according to the presence or absence of dural sac notch caused by the LF on bilateral parasagittal MR images.Meanwhile,50 volunteers without any symptom in the lumbar region or legs were examined by CT.TODRs were measured at the L3—S1 levels of the inferior margin of the intervertebral disc on transverse CT images,respectively.The results were further analyzed with the positive findings on MR images,clinical symptoms and physical examination,so as to find out the statistical correlation between them.Results LF thickness was(3.01?0.72)mm and TODR was 0.19?0.04 in the negative group,(3.94?0.84)mm and 0.28+0.06 in the positive group,and(3.16? 0.85)mm and 0.19?0.04 in the control group.There was significant difference between positive group and negative group or control group for LF thickness(P0.24, the sensitivity,specificity and positive predictive value were 74.8%,89.6% and 73.6% respectively.
