2.Predictive and intelligent control of sevoflurane closed circuit anesthesia

Objective To evaluate the efficacy of servo controlled sevoflurane closed-circuit anesthesia and the feasibility of the predictive and intelligent control.Methods One hundred and forty-three ASA Ⅰ-Ⅲ patients (90 males, 53 females) aged between 3-77 yrs undergoing elective surgery were studied. Anesthesia was induced with intravenous fentanyl 2-3 ?g ?kg-1 , midazolam 0.12 mg?kg-1 and vecuronium 0.1 mg?kg-1 . After tracheal intubation the tracheal tube was connected to the servo-controlled closed-circuit system composed of IBM computer, O2 mass flow controller and electrically controlled sevoflurane injection pump and multifunctional monitor. The fresh gas flow of O2 = [body weight (kg)3/4 ? 10 + 20] ml?min-1 . The end-expired sevoflurane concentration was maintained at 1.3 MAC by predictive and intelligent control.Results The average wash-in time to reach the target concentration was (5.2 ?2.4) min. The O2 flow rate was(0.22?0.04) L?min-1 . The cumulative uptake of sevoflurane was 5.16 ml, 7.74ml, 9.17ml, 11.08ml, 12.57ml, 13.00ml, 14.18ml, 15.60ml, 18.56ml and 24.6 ml at 30, 60, 90, 120, 150, 180, 210, 240, 300 and 420 min respectively. The uptake rate of fluid sevoflurane was equivalent to (0.2673e-0.0598t + 0.2269e-0.0597t + 0.1150e-0.002t) ml?min-1 . Conclusion The servo controlled sevoflurane closed-circuit system can effectively control the pre-set end-tidal sevoflurane concentration in spite of the influence of multiple factors and is safe and effective.
3.A RNA Isolation Method Suitable for a Wide Range of Materials

High levels of RNase,polysaccharides and polyphenol compounds make isolation of high quality RNA difficult. Thus it is presented an effective RNA extraction method based on the nuclease adsorbent macaloid,poly vinyl pyrrolidone,and high concentration of KAc and ethylene glycol monobutyl ether,which has successfully extracted high-quality RNA from many materials difficult to RNA isolation,such as RNase-rich rabbit liver,plant and microbial tissues rich in polysaccharides,lipids and polyphenol compounds. This method was found to be better than the ones in common use-Trizol and Guanidinium isothiocyanate,the yield of which was at least three time higher. Furtherly,small RNA was enriched from total RNA sample from rice seedling through by repeat deposit which deals with high concentration of LiCl,PEG8000 and NaCl. The small RNA gained was confirmed to be used for following molecular biological research by RT-PCR with the primers designed on osa-mir-156 sequence from rice miRNA.
4.Correlation between adult lifestyle and ankle-brachial blood pressure

Objective: To explore the relationship between lifestyle and ankle-brachial blood pressure. Methods: Ankle-brachial blood pressures of 2121 adults from all over China were measured with self-developed KD-991 portable ankle-brachial blood pressure simultaneous detecting instrument. They were divided into smoking group and no smoking group, heavy drinking group and moderate drinking group, exercise group and lack of exercise group and comparisons of abnormal ankle-brachial index (ABI, ≤0.9 or≥1.3) rate between paired two groups were performed. Results: Rate of abnormal ankle-brachial index (ABI, ≤0.9 or≥1.3) in smoking group (15.3%), heavy drinking group (13.6%) and lack of exercise group (10.5%) were significantly higher than those of no smoking group (7.9%), moderate drinking group (7.5%) and exercise group (7.4%) respectively, P<0.05 all. Conclusion: Unhealthy lifestyle is related to abnormal ankle-brachial blood pressure and it is very good to prevent occurrence of atherosclerosis , early discover and effectively control its development using ABI to guide people to change unhealthy lifestyles.
5.Analysis of the curative effect of one-stage bilateral total hip arthroplasty

Objective To study the factors influencing the curative effect of one-stage bihteral total hip arthroplasty in a sequential procedure.Methods Thirty-six patients were indicated for one-stage bilateral total hip arthroplasty from February 1998 to March 2008.All cases were made with Smith-Peterson incision.Results The operative time was(3.2±1.2)h.The volume of blood transfusion during operation was (620 4±120)ml.All the 36 patients were followed up for(22.0±6.0)months.Except for 1 case of deep venous thrombosis of lower limbs,1 case of limb length discrepancy,there were no severe complications,such as postoperative infection,pulmonary embelism,dislocation,loosening or subsiding of components.Postoperative Harris scoreg of the joint function[(88.0±3.6)points]was significantly improved compared with their preoperative Harris scores[(28.0±4.2)points](P<0.05).Conclusions One-stage bilateral total hip arthroplasty is an effective method.However.the satisfactory effects can only be achieved by strict case selection,sufficient preoperative preparation,attention to operation procedures,prevention of complications and active recovery after operations.

7.Effects of Platelet Activating Factor on Murine Splenocytes Proliferation and Production of IL-2, NKCF and CSF

Platelet activating factor (PAF) is a biologically active membrane phospholipid mediator. In this study we investigated the effects of PAF on the lymphocyte proliferation, production of IL-2, natural killer cytotoxic factor (NKCF) and colony stimulating factor (CSF) by BALB/c mice splenocytes in vitro. The results showed that PAF inhibited lymphocyte proliferation at concentrations of 10-10~ 10-7 mol/L. CTLL-2, YAC-1 and munne bone marrow cells were used for the assays of IL-2, NKCF and CSF, respectively. Production of IL-2, NKCF and CSF from ConA stimulated murine splenocytes was significantly inhibited by PAF at 10-7 mol/L. Inhibition of spleen cells proliferation and secretion may play an important role in the pathological effects of PAF.
8.Shuxuening injection combined with routine therapy in treating patients with active ulcerative colitis: an analysis of efficacy .

OBJECTIVETo observe the therapeutic effect and relevant mechanism of shuxuening Injection (SI) in treating patients with active ulcerative colitis (UC).
METHODSTotally 91 patients with active UC were randomly assigned to 2 groups, 44 in the control group and 47 in the treatment group. Patients in the control group received routine treatment, while patients in the treatment group additionally received intravenous injection of SI (15 mL), twice daily for 14 days in total. Colonoscopy was performed before and after treatment. The therapeutic effect was assessed by Mayo scoring system and the grading of activities evaluated by Baron endoscope. Serum levels of IL-6 and TNF-α were detected by ELISA. The activity of SOD was detected by xanthine oxidase method. The content of MDA was detected by thiobarbituricacid (TBA). Besides, 20 healthy subjects were recruited as the healthy control group.
RESULTSTotally 82 patients completed the study (40 in the control group and 42 in the treatment group). There was no statistical difference in serum levels of IL-6, TNF-α, SOD, MDA, the Mayo score and endoscope grading between the two groups before treatment (P >0. 05). Compared with the healthy control group, serum levels of IL-6, TNF-α, MDA significantly increased (P <0.01), and the serum SOD level decreased (P < 0. 05) in the treatment grup and the control group before treatment. Compared with before treatment in the same group, serum levels of IL-6, TNF-α, MDA, the Mayo score and endoscope grading all decreased in the treatment group and the control group after treatment (P <0. 01, P <0. 05). Compared with the control group after treatment, serum levels of IL-6, TNF-α, MDA, the Mayo score and endoscope grading all decreased (P <0.01, P <0.05), the serum SOD level increased (P <0.05) in the treatment group after treatment. The serum SOD level was obviously negative correlated with serum levels of IL-6, TNF-a, Mayo score, and endoscope score (r = -0. 621, -0.638, -0. 509, -0.787, P <0.01). The serum MDA level was obviously positive correlated with serum levels of IL-6, TNF-α, Mayo score, and endoscope score (r =0.711, 0. 882, 0. 525, 0. 639, P <0.01).
CONCLUSIONSI could improve inflammatory injury and clinical symptoms of patients with active UC, and its mechanism might be associated with antioxidant and scavenging oxygen free radicals.

9.Studies on Fatty Oils in Yolk Oil Processed by Different Methods

Objective:To investigate the influence of different processing methods on fatty oils in yolk oil. Method: The contents of fatty oils and fatty acids were determind by ROESS GOTTILE and GC respectively, and the physicochemical properties of fatty oils were identified by pharmacopoeial method. Result and Conclusion: The yolk oils processed by daking and dry distillation are almost similar in the contents of fatty oils, type of fatty acids, acid value, iodine value, saponification value, relative density and refractive index, while obvious differences were found in the products processed by chloroform extraction.

Objective To investigate the correlation between the expression of STAT3 and MMP-2 in human pancreatic cancer,and to probe the mechanism by which STAT3 signal pathway regulates the expression of MMP-2 in pancreatic cancer cells.Methods Immunohistochemistry was used to detect the expression of STAT3,phosphorylated STAT3(p-STAT3)and MMP-2 in pancreatic cancer tissues of 34 cases and normal pancreatic tissues of 10 cases.Correlation between the expression of STAT3、p-STAT3 and MMP- 2 were statistically analyzed.Human pancreatic cancer cell lines SW1990 was cultured.AG490,an inhibitor of the upstream Janus kinase(JAK)of STAT3 was added into the culture medium.Electrophoretic mobility shift assay(EMSA)was used to detect STAT3 DNA-binding activity in SW1990 cells.Western blot was used to detect the expression of STAT3,p-STAT3 in SW1990 cells.In addition,the protein and mRNA expression of MMP-2 in SW1990 cells were determined by Western blot and RT-PCR,respectively.
