1.Expression of PR and Bcl-2,Bax in Ectopic and Eutopic Endometrium in Patients with Endometriosis

Objective:The pathogenesis of endometriosis(EMT)was explored by detecting the relationship of progesterone receptors(PR)and apoptosis-associated factors with EMT.Methods:Immunohistochemical imaging analysis was used to quantify the expression of PR,Bcl-2 and Bax in ectopic and eutopic endometrium in patients with EMT.Results:Expressions of PR and Bcl-2 in eutopic endometrium are higher than those in ectopic endometrium(P<0.05).Expression of Bax in ectopic endometrium is higher than that in eutopic endometrium(P<0.05).There is no statistical difference of PR,Bcl-2 and Bax expression between proliferative and secretary endometrium.Conclusions:Either in proliferative or secretary phase,expressions of PR in eutopic endometrium are higher than those in ectopic endometrium,thus eutopic endometrium is more liable to proliferate in low level continuously,migrate and implant.Expressions of Bcl-2 and Bax in eutopic and ectopic endometrium have no relationship with endometrium cycling.The apoptosis of ectopic endometrium is weakened and its proliferative power is continuously enhanced.
2.The Effect of Bisphenol A and Dibutyphthlate Treated With Rat Liver S9 on Proliferation of MCF-7 Cells

Objective To study the effect of bisphenol A (BPA) and dibutyphthlate (DBP) treated with Rat Liver S9 on proliferation MCF-7 cells. Methods Added the positive control 17?-estradiol (E2) and compounds of BPA and DBP with the effect of Rat Liver S9 to MCF-7 cells proliferation respectively and determinated the quantity of MCF-7 cells with MTT assay. Results The activity of E2 and DBP stimulated MCF-7 cells decreasing significantly with the effect of Liver S9 than without the effect of Liver S9, while BPA stimulated MCF-7 cells increasing significantly with the effect of Liver S9 than that without the effect of Liver S9. Conclusion E2 and DBP have a lower estrogenic activity with the effect of S9 than without the effect of S9. BPA shows a higher estrogenic activity with the effect of S9 than without the effect of S9.
4.EXPERIMENTAL STUDY ON THE CHANGES OF GELATINASE ACTIVITY OF ASCENDING AORTIC ANEURYSMS

Objective To investigate the activity changes of gelatinase in the formation of ascending aortic aneurysm.Methods Thirty five young Wistar rats were divided into two groups:the control group and the experiment group.The rat models induced by ascending aorta banding were made.The ascending aortas were taken after 3-5 months operation,changes of gelatinase activity was observed by gelatin zymography and film in situ zymography.Results Gelatinase activity of ascending aortic aneurysm was significantly increased compared with that of normal ascending aortic aorta.Conclusion Elevation of gelatinase activity may play a significant role in the formation of ascending aortic aneurysm.
5.Injury of ~(125)Ⅰ seeds implants to trachea and esophagus of rabbits

Objective To evaluate the impact of 1125 seeds para-tracheal braehytherapy on regional tissue injury in rab-bit models. Methods 42 rabbits were randomized into 7 groups. Group 1 to 6 belong to study groups (in which 1,4,5 and 6 belong to "dose gradient" subgroup, while 2,3 and 4 to "chronologic" subgroup) , while the last group acts as negative con-trol. The activity of seeds in study group were 0.3 mCi in group 1, 0.5 mCi in group 2 to 5, 0.7 mCi in group 5, and 0.9mCi in group 6. False seeds (0 mCi) were used for the negative control. 4 seeds with equal dosage were implanted between trachea and esophagus in each rabbit under general anesthesia. Seeds arrangement was made according to Paris principle. For the tissue injury evaluation, group 2 was sacrificed by the end of first month post-operatively, group 3 at the end of the second month, and group 4 end of the third month. The rest of rabbits were also sacrificed at the end of the third month. Pieces of adjacent e-sophagus and trachea were sampled from each rabbit. Tissue injury features such as inflammation, edema, congestion or fibrosis as evaluated histologically. Results All rabbits were healthy during study period except 5. Histological analysis revealed that trachea samples from all groups had lymphocytas and plasma cells infiltration as signs of chronic inflammation, hut fibrosis was nut clearly visible. There were no differences between study and control groups with respect to inflammation, edema and con-gestion scores. But in groups which received the highest doses of radiation or sacrificed at 60 d showed more eosinophil infiltra-tion and epithelum degeneration, and statistical significance was reached between these groups and control. Esophageal samples had less histological changes compared with trachea. Conclusion Para-tracheal implantation of ~(125)Ⅰ seeds with therapeutic or higher dosage only induce minor and reversible damage to the regional tissue. This implies that ~(125)Ⅰ implants adjacent to trachea or esophagus are clinically safe.
6.Expression and significance of bcl-2, bax and ER in foci of adenomyosis

Objective To investigate the expression and signification of B-cell lymphoma/leukemia-2(bcl-2),bax and ER proteins in epithelial cells in different depth of adenomyosis.Methods Expression and correlation of bcl-2,bax and ER proteins were detected by immunohistochemistry staining in 36 adenomyosis cases with superficial (< 1/2 depth of penetration) and deep (≥ 1/2 depth of penetration)ectopic endometrium and eutopic endometrial tissues matched with 30 cases with benign ovarian tumor,uterine septum,pelvic floor dysfunction without adenomyosis as controls.Results (1) The expression of ER protein in superficial and deep ectopic tissues (5.04 ±0.24,4.91 ±0.16) were found significantly lower than 6.06 ± 0.36 in eutopic tissues (P < 0.01) and higher than 3.70 ± 0.58 in control group (P < 0.05).There was no statistical difference of ER expression in superficial and deep ectopic endometrium (P > 0.05).(2) The level of bcl-2 protein of 5.6 ± 0.4 in superficial and 6.0 ± 0.3 in deep myometrium of ectopic tissues were significantly higher than 3.6 ± 0.4 in eutopic tissues and 1.9 ± 0.4 in control group (P < 0.01).(3) The level of bax protein of 3.50 ± 0.28 in superficial and 4.80 ± 0.29 in deep myometrial ectopic and 4.43 ± 0.37 in eutopic tissues were significantly lower than 6.18 ± 0.65 in control groups (P <0.05).The expression of bax in superficial myometrium is significantly lower than deep myometrium in ectopic tissues (P <0.01).(4) The positive correlation between the expression of ER and bcl-2 protein at superficial myometrium of ectopic tissues (r =0.720,P < 0.01).And there was no significant correlation with the expression of ER and bax protein at superficial myometrium (r =0.008,P > 0.05).As well as,there was not significant correlation with the expression of bcl-2,bax and ER protein at deep myometrium (r =0.089,r =-0.023,P > 0.05).The expression of bax protein in ectopic endometrium was positive correlation with the depth of adenomyosis penetration (r =0.736,P < 0.01).There was positive correlation between the expression of ER and bcl-2 protein at normal endometrium (r =0.453,P < 0.05).And there was negative correlation between the expression of ER and bax protein at control group (r =-0.514,P =0.05).Conclusion The bax protein expression of ectopic endometrium in deep adenomyosis was higher than superficial adenomyosis.
7.Influence factors of formation of collateral circulation in patients with chronic occlusive coronary ;artery disease

Objective To study the possible influencing factors in the formation of coronary collateral circulation in patients with chronic total occlusion (CTO). Methods Patients were enrolled having at least 1 major coronary artery angiography revealed as CTO of 144 patients. According to the Rentrop classification, patients with grade 0 and grade 1 filling were catogorized as insufficient collateral circulation group (n=72) and patients with grade 2 and grade 3 filling as collateral circulation group (n=72). Serum biomarkers and insulin-resistance by HOMA model were also studied in all patients. Results In the insufficient collateral circulation, BMI,TC,ApoB, lipoprotein a, fasting insulin HOMA-IR,HOMA- beta, CRP was significantly higher than the well collateral circulation group and ApoA-Ⅰ, ISI lower than the well collateral group ( all P ﹤0. 05 ) . Bivariate correlation alaysis showed. Rentrop score, BMI, TC, ApoB, lipoprotein a, fasting insulin, HOMA-IR,HOMA- beta and CRP are positively correlated to the formation of collateral circulation ( P ﹤ 0. 05 ); ApoA-Ⅰ and ISI were negatively correlated ( P ﹤0. 05 ) . Logistic regression analysis after calibration with weight, ApoA-Ⅰ and HOMA-beta factors, lipoprotein a ( OR 7. 575,P=0. 009), TC (OR 2. 154,P =0. 001) were found to be the independent factors of coronary collateral circulation formation. Conclusions TC, lipoprotein a, obesity, CRP, and HOMA-IR are correlated with the formation of coronary collateral circulation and may predict formation of collateral circulation in patients with CTO.
8.Outcomes of combined wavefront-guided and aspheric LASIK for myopia and myopic astigmatism

AIM: To evaluate the clinical outcomes of combined wavefront - guided and aspheric laser in situ keratomileusis ( LASIK ) to correct myopia and myopic astigmatism.
METHODS: Prospective study. Forty-five patients ( 62 eyes ) with myopia and myopic astigmatism were randomly dividied into two groups:the wavefront-guided and aspheric LASIK group and the wavefront- guided LASIK group. Safety, efficacy, predictability, ocular higher order aberrations(HOAs), and contrast sensitivity under mesopic condition were compared at 6mo postoperatively.
RESULTS: Both platforms had equal safety, efficacy and predictability. At 6mo after operation, total HOAs, spherical aberration and coma increased in both groups(P<0. 01 ). The changes of total HOAs and spherical aberration in wavefront-guided and aspheric group were significantly less than those in wavefront-guided group ( P<0.05 ) , while the change of coma between two groups was not statistically significant ( P = 0. 657 ). Contrast sensitivity in the wavefront-guided and aspheric group recovered to preoperative levels under mesopic conditions at all spatial frequencies ( P > 0. 05 ), while contrast sensitivity in the wavefront-guided group recovered to preoperative levels at all spatial frequencies(P>0. 05).
CONCLUSION: Wavefront-guided and aspheric LASIK induced less HOAs and associated with better mesopic contrast sensitivity compared with wavefront - guided LASIK.
9.Application of endoscopically subcutaneous mastectomy and immediate mammary prosthesis reconstruction

Objective To study the feasibility of endoscopically subcutaneous mastectomy and immediate mammary prosthesis reconstruction.Methods From December 2006 to October 2007,9 breast cancer patients underwent endoscopic skin sparing mastectomy and immediate implanting breast reconstruction,with preoperatively performed systemic TE chemotherapy two to six times.Whether or not the nipple-areola complexes were preserved depended on the results of frozen pathological examination.Results Of 9 patients,bilateral skin sparing mastectomy were performed in two patients,and others underwent single lateral masteetomy with the nipple-areola complexes,at the same time sentinel lymph node biopsy was done in 8 cases of all.Combined level Ⅰ and Ⅱ axillary dissections were carried out vvia the sarne incisions underthe axillaries in 7 patients,and 2 patients spared axillary dissections.8 of them got satisfactory results,one patient did not.The postoperation complications included subcutaneous seromas 1 case,epidermic nipple necrosis 1 case and bleeding of skin flap 1 case.No local reeurrence occurred during the follow-up ranged 1~10 months.Conclusion It is technically safe and feasible that endoscopically subcutaneous mastectomy with immediate mammary prosthesis reconstruction.The technique can minimize skin incision With little trauma,and offers a greater esthetic advantage tomostpatients.
10.Video-assisted subcutaneous mastectomy and immediate reconstruction

Objective To explore the feasibility and cosmetic results of video-assisted subcutaneous mastectomy and immediate reconstruction. Methods Two patients who suffered from breast diseases and need to excise the breasts received a video-assisted total subcutaneous mastectomy and immediate reconstruction in our center. Results Both patients felt satisfied on the cosmetic result. According to the operative experience of these two patents combined with review, the key point of the operation was summarized as follows. The incisions on the anterior axillary line were small and concealing. External retraction method was used to establish the working space. Video-assisted operation was combined with euthyphoria. Blunt dissection was combined with electrical surgery unit and ultrasound knife to isolate the breast skin flap. After breast glandectomy was completed, silica gel prosthesis was put into the interspace behind ectopectoralis. Pressure dressing for 3 weeks was done to prevent the prosthesis upward displacement. Conclusion The video-assisted subcutaneous mastectomy and immediate reconstruction is a safe and feasible operation method with the better cosmetic result.
