1.Advances in research on inhibitors based on targets: IDO1 and TDO

Indoleamine 2,3-dioxygenase 1 (IDO1) and tryptophan 2,3-dioxygenase (TDO) catalyze the initial and rate limiting step in the catabolism of tryptophan, which is related to tumor immune tolerance and poor prognosis in patients. In this regard, two enzymes have become important therapeutic targets for tumor immunotherapy. So far, nine IDO1 inhibitors and three IDO1/TDO dual inhibitors have entered clinical trials. This review summarizes the research progress of IDO1 inhibitors, TDO inhibitors and IDO1/TDO dual inhibitors from the perspective of medicinal chemistry.
2.Application of Thermal Shift Assays in Purification of Recombinant TBEV Serine Protease

Thermal Shift Assays was applied for high-throughput screening optimized buffer system used in the gel filtration chromatography for purification of recombinant TBEV serine protease. The optimization included kinds of salt and pH. It was optimized new buffer remarkably improved the quality of the purified TBEV serine protease by increasing its monomer from 75% to 99%. This result may improve future crystal studies of this protein.
3.Survey of a study on the real-time control of medical quality in hospitals

Objective To explore advanced and effective methods of medical quality control. Methods A perspective method, together with literature summary, process analysis, statistics and computer network techniques, was employed. Results A model of real time control of medical quality via element management, process management, and monitored control management was set up; methods of real time control of quality and expenses for disease entities in hospitals were defined; the effect of clinical paths on real time control of the quality of hospitalization was ascertained; and ways of real time control of the quality of medical records were determined. Conclusion Real time control of medical quality is a new, effective way of monitoring medical quality.
4.Preparation and vitality detection of protoplast in Salvia miltiorrhiza Bunge.

We prepared protoplasts from Salvia miltiorrhiza Bunge suspension culture cells. Then, the protoplasts' vitality and functions were tested by fluorescein diacetate staining method and Fluo-3/AM flourescent probe. The optimal condition of protoplast isolation was Cellulase R-10 1.5%, Pectinase Y-23 0.3%, Macerozyme R-10 0.5%, 40 r/min 12 h, 600 r/min 5 min, and the protoplasts yield was 1.1x10(6) cells/g FW, the vitality was more than 95% by using fluorescein diacetate staining method. It has been confirmed that calcium fluorescent probe Fluo-3/AM can be successfully loaded into protoplasts.

6.Determination of ATP, ADP and AMP in Cardiac Muscle of Rats with Heart Failure by HPLC

Objective:To establish the determination method for ATP, ADP and AMP in the myocardial tissue of rats and research the changes of ATP, ADP and AMP in heart fallure rats. Methods: A Thermo Hypersil C18 (250 mm × 4. 6 mm,5 μm) column was used. The solution of 0. 68% potassium dihydrogen phosphate and 0. 116% soduim hydroxide was applied as the mobilie phase; The detection wavelength was at 254 nm. The SD rats randomly divided into 2 groups received saline and adriamycin by intraperitoneal in-jection for 6 weeks, respectively. The heart tissue was withdrawn for homogenate. The contents of ATP, ADP and AMP in rat heart tis-sue were determined by HPLC. Results:The calibration curves were linear from 0. 625μg·ml-1 to 40μg·ml-1 for ATP, ADP and AMP. The limit of detection and limit of quantification were 0. 625μg·ml-1(r>0. 99). The precision of RSD was less than 9. 50%. The accuracy was 93. 2%-108. 0%. The method stability (RSD) was from 3. 0% to 14. 0%. The content of ATP in heart fallure mod-el group induced by adriamycin was significantly lower than that in saline group,while that of ADP were much higher than that in saline group(P<0. 01) and AMP. Conclusion:The HPLC method for the content determination of ATP, ADP and AMP in myocardial tissue meets the requirements of the determination of biological samples, which can be used to study the effects of anthracycline-based antitu-mor drugs in heart fallure rats.

Objective: To study the cellular immunoregulatory effect of Huangqi through intravenous injection in patients with chronic hepatitis B. Methods: In treated groups, light and mild chronic hepatitis B (15 patients in each group) were treated with Huangqi and Ganlixin. Objective: To investigate the chemopreventive effects of pioglitazone (exogenous PPAR? ligand) on rat colon aberrant crypt foci, a rat carcinogenesis model induced by dimethylhydrazine (DMH),and to compare pioglitazone with sulindac (a NSAID). Methods: Thirty two, 8 week old, female Sprague Dawley rats were randomly divided into four groups ( n =8 each). Group 1 rats were injected with DMH alone (120 mg?kg -1 , single subcutaneous injection). Group 2 rats were injected with saline alone. Group 3 rats were pre treated with sulindac (320 mg?kg -1 ) for 7 days before DMH initiation. Group 4 rats were treated with pioglitazone (100 mg?kg -1 ). The animals were killed at the end of the experiment (week 5) and the colons were stained with methylene blue. The aberrant crypt foci (ACF) of the colonic mucosa were assessed. Results: In Group 1 rats (DMH only), the average numbers of ACF/colon and AC/colon were (182?93) and (263?198), respectively. In Group 2 (saline group) rats, no ACF were found. In Group 3 (sulindac group) rats, the average numbers of ACF/colon and AC/colon were (91?49) and (140?69), respectively. 9.AngioJet rheolytic thrombectomy for the treatment of acute deep venous thrombosis of lower extremity

Objective To discuss the efficacy and complications of using AngioJet rheolytic thrombectomy in treating acute lower extremity deep vein thrombosis (DVT).Methods The clinical data of 22 patients with acute lower extremity DVT,who were treated with AngioJet rheolytic thrombectomy during the period from February 2015 to August 2016,were retrospectively analyzed.The improvement of clinical symptoms and the thrombus clearance rate were calculated to evaluate the curative effect.The procedure-related complications were documented.Results The clinical symptoms were relieved immediately after operation in all 22 patients.The thigh circumference difference between the affected side and the healthy side decreased from preoperative (4.5±0.6) cm to postoperative (1.0±0.4) cm,the difference in change was statistically significant (P<0.05).The mean used dose of urokinase was (0.18±0.03) million unit and the average duration of thrombolysis was (4.2±0.7) hours.Complete removal of DVT (>90%) was achieved in 19 patients,most removal of DVT (50%-90%) in 2 patients,and partial removal of DVT (<50%) in one patient.After treatment,6 patients developed transient hemoglobinuria,which was relieved after hydration with fluid infusion on the same day.No serious complications such as pulmonary embolism or hemorrhage occurred.Conclusion For the treatment of acute lower extremity DVT,AngioJet rheolytic thrombectomy is safe and effective with less complications.
10.The study on duodenogastric reflux after cholecystectomy

Objective To observe the alteration of duodenogastric reflux after cholecystectomy so as to explore the basis for diagnosis and therapy. Methods Intragastric bile reflux during 24 hours was assessed using ambulatory bilirubin monitoring and 24-hour pH monitoring techniques in 20 cholecystectomy patients, 10 cholelithiasis patients and 15 healthy volunteers. Results Bile reflux and alkaline reflux of cholecystectomy patients did not increase compared with those of cholelithiasis and the control. Conclusion Duodenogastric reflux of patients receiving postcholecystectomy patients does not increase.
