OBJECTIVETo investigate the protective effect of melatonin on residual hair follicle cells of scald rats at early stage.

METHODSEighteen male Sprague-Dawley rats were randomly divided into scald group, treatment group, sham group , with 6 rats in each group. The rats in scald group and treatment group were subjected to 30% TBSA partial thickness scald on the back, and were resuscitated with balanced solution after 1 hour, while those in sham group were immersed in water at 37 degrees C for 25 s to simulate scald, and did not receive fluid replacement. Rats in treatment group were intraperitoneally injected with 10 mg/kg melatonin solution at 1 minute, 8 hours and 12 hours after scald, while those in sham group and scald group were given equal volume of 1% alcohol sodium-isotonic saline instead. Tissue samples were harvested at 6, 12 and 24 post scald hours (PSH) for determination of MDA and GSH levels. Apoptosis of residul hair follicle was detected by TUNEL method and immunohistochemistry of caspase-3.

RESULTSThe level of MDA in scald group at each time point was much higher than that in sham group (P < 0.01) and treatment group (P < 0.05), and it peaked at 12 PSH. The changes in GSH were just opposite to that of MDA. Under fluorescence microscope, the residual hair follicle cells were blue, and the apoptotic cells appeared green. The apoptosis rate in scald group at 6, 12, 24 PSH was obviously higher than that in sham (P < 0.01) and treatment groups (P < 0.05), which was (20.2 +/- 3.4)% vs (4.3 +/- 2.3)% vs (10.9 +/- 3.2)%, (31.2 +/- 3.6)% vs (5.1 +/- 2.5)% vs (19.1 +/- 3.7)%, (22.4 +/- 2.7)% vs (4.1 +/- 2.4)% vs (13.1 +/- 3.4)%, respectively. The score of caspase-3 positive cell in scald group was higher than those in sham group (P < 0.01) and treatment group (P < 0.05).

CONCLUSIONSThere is obvious correlation between oxidative stress and apoptosis rate of hair follicle cells in rats with partial thickness scald. Early administration of melatonin may have anti-apoptosis ability for residual hair follicle cells by attenuation of oxidative stress.

The reform included setting up flexible program,formulating objective and detailed rotary guide,making the best of the resources,implementing medical humanities education based on the reality and forming unique cultivation system and mode. These measures can be taken as references for medical education reform in mainland.

Sodium houttuyfonate (SH) is a derivative of effective component of a Chinese material medica, Houttuynia cordata, which is applied in anti-infection of microorganism. But, the antimicrobial mechanisms of SH still remain unclear. Here, we firstly discovered that SH effectively inhibits the three types of virulence related motility of.Pseudomonas aeruginosa, i.e., swimming, twitching and swarming. The plate assay results showed that the inhibitory action of SH against swimming and twitching in 24 h and swarming in 48 h is dose-dependent; and bacteria nearly lost all of the motile activities under the concentration of 1 x minimum inhibitory concentration (MIC) (512 mg x L(-1) same as azithromycin positive group (1 x MIC, 16 mg x L(-1)). Furthermore, we found that the expression of structural gene flgB and pilG is down-regulated by SH, which implies that inhibitory mechanism of SH against motility of P. aeruginosa may be due to the inhibition of flagella and pili bioformation of P. aeruginosa by SR Therefore, our presented results firstly demonstrate that SH effectively inhibits the motility activities of P. aeruginosa, and suggest that SH could be a promising antipseudomonas agents in clinic.

Objective To explore the necessity of opening nephrostomy tube for patients with septic shock follow-ing the indwelling double-J stent of post-percutaneous nephrolithotomy (PCNL),and provide reference for the treatment of septic shock after PCNL.Methods 60 patients with septic shock after PCNL in a hospital from January 1,2015 to December 30,2016 were chosen,patients were randomly divided into clipping nephrostomy tube group (clipping group,n =30) and opening nephrostomy tube group (opening group,n =30),clinical data of two groups of patients were collected and analyzed.Results After 24-hour treatment,heart rate (HR),mean arterial pressure (MAP),oxygen saturation (SpO2),serum lactate (Lac),and hourly urine volume all improved in both groups of patient compared with pre-treatment(all P＜0.05);HR,MAP,SpO2,Lac,and hourly urine volume in opening group after 24-hour treatment were all significantly different from clipping group (all P＜0.05).Levels of serum procalcitonin (PCT) and C-reactive protein (CRP) in two groups after 3-day treatment decreased significantly compared with pre-treatment (both P＜0.05);PCT and CRP levels in opening group after 3-day treatment were both significantly lower than clipping group (both P＜0.05);cure rate of two groups were both 100.00％,hospitalization time and extubation time in opening group were both shorter than clipping group,and cost was less than clipping group,difference were all significant(all P＜0.05).Conclusion Opening nephrostomy tube on the basis of indwelling double-J stent is necessary for patients with septic shock after PCNL.

By the bioactivity technology of Low-Temperature Preparation of Anatase and Rutile Layers on porous Titanium Substrates, CPTi(Commercially pure titanium Phi4.0 mm x 20 mm) was processed and treated as experiment specimens while CPTi was treated as control specimens. Experiment specimens and control specimens were implanted into the holes (4.0 mm in diameter) in rabbit's right and left tibias respectively. After implantation for predetermined periods of 4,8,12, 16 weeks, the specimens were taken out with bone tissues, and were examined by a press-out tester to evaluate the shearing force between the implant and the bone tissue. It is found that the shearing force between the experiment specimen and the bone is more significantly higher than that between the control specimen and the bone, and the bonding time is shorter, the stabilization time is faster. This study has laid down a good foundation for the titanium metal's innovative applications in clinical orthopaedics.

OBJECTIVETo study change and the value of diagnosis of harmonic imaging ultrasonic integrated backscatter (IBS) on the very early diagnosis of acute myocardial infarction (AMI).

METHOD72 patients were divided 2 groups, A group 30 cases with AMI at the very early stage of acute myocardial infarction (in 2 hours), B group 42 cases with typical acute myocardial infarction (in 2 - 12 hours) were examined by IBS and the cyclic variation of integrated backscatter (CVIB) with HP-5500 ultrasonic system in the segment of myocardial infarction and no myocardial infarction.

RESULTSIn A group the IBS of the segment of AMI are very higher than that of the segment of no AMI [(18.8 +/- 3.4) dB vs (8.3 +/- 1.2) dB, P < 0.01], the CVIB are lower [(6.3 +/- 0.7) dB vs (7.6 +/- 1.1) dB, P < 0.01]. But in the meanwhile there are no obvious changes in ECG. In B group the IBS of the segment of AMI is obvious higher than the normal person and no infarction segment in the same heart [(22.2 +/- 4.1) dB vs (8.3 +/- 1.2) dB, (21.1 +/- 3.2) dB vs (8.7 +/- 0.9) dB, P < 0.05], but CVIB is obvious lower than the normal person and no infarction segment in the same heart [(5.6 +/- 0.8) dB vs (7.6 +/- 1.1) dB, P < 0.05; (5.8 +/- 0.7) dB vs (9.3 +/- 0.9) dB, P < 0.01], the changes of the ultrasonic is coincidence with ECG. The change of IBS are very obvious just as in ECG.

CONCLUSIONThe result demonstrate that ultrasonic tissue characterization with harmonic imaging integrated backscatter can be used for diagnosis the very early stage of AMI, and can judge the segment range of AMI and function of the whole heart.

This study was aimed to investigate the effects of apogossypolone (ApoG2) on proliferative inhibition and apoptotic induction of multiple myeloma cells and its mechanism. The effects of ApopG2 on cell growth, cell viability, cell cycle and cell apoptosis were determined by Hoechst 33258 staining, DNA ladder formation and subdiploid peak analysis respectively. Cleavage of caspase-3 and caspase-9 was analyzed by colorimetric assay. Expression of BCL-2 and BCL-XL was detected by flow cytometry. The results indicated that the ApoG2 inhibited multiple myeloma cell proliferation in dose-and time-dependent manners, with IC(50) value to both U266 and Wusl cells at 0.1 and 0.2 micromol/L at 48 hours after treatment. ApoG2 effectively inhibited the proliferation of multiple myeloma cells, the IC(50) value in U266 cells and Wusl cells (at 48 hours) were 0.1 micromol/L and 0.2 micromol/L respectively. ApoG2 could induce the apoptosis of cells of myeloma in a time-dependent manner.The typical apoptotic morphological changes were observed under transmission electron microscope, while DNA ladder formation and remarkable peak of subdiploid cells appeared. ApoG2 could arrest the myeloma cells in G(2) phase, increasing from 9.7%(0 micromol/L) to 19.6% (10 micromol/L) in U266 cells and 9.8%(0 micromol/L) to 31.7% (10 micromol/L) in Wusl cells. ApoG2 could induce increase of caspase 9 and caspase 3 activity and down-regulate the expression of BCL-XL in U266 and Wusl cells, as well as the expression of BCL-2 in Wusl cells. It is concluded that ApoG2 has significant effect of antiproliferation and induction of apoptosis on multiple myeloma cells in vitro, ant its mechanisms may involve in down-regulation of BCL-2/BCL-XL and in change of cell cycle.
