1.Effects of ACh on Voltage-dependent Outwardly Rectifying Potassium Currents in Isolated Outer Hair Cells in Cochlea of Guinea Pig

Objective To observe the effect of ACh on outwardly rectifying potassium currents in isolated outer hair cells(OHCs) with variant length and to analyze effects of ACII on kinetics of the currents. Methods The whole cell patch-clamp technique was used. Results 100 ?mol/L ACh induced greater effects on outwardly rectifying potassium currents in short OHCs. The maximal amplitude of the outward currents increased by 34.8% in the presence of 100 ?mol/L ACII when conditioning potential was 50 mV. ACh effects on the peak currents were greater when compared to the effects on the steady-state currents and changed the shapes of the currents. ACII caused a hyperpolarizing shift of zero current potential of 5 mV. Kinetics of the currents were changed by 100 ?mol/L ACh which induced significantly more hyperpolarizing than control, with increased potential sensitivity of activation, V1/2(Ach) =(-52.38 ? 3.98) mV, SACh =(40 ? 4.14) mV(n= 5). Conclusion ACh increased the conductance of outwardly rectifying K + channel in a voltage dependent manner and caused a hyperpolarizing shift of activation potential. Effect of ACh was to hyperpolarize the membrane potential of OHCs from rest.
2.Chitinase Gene Overexpression of Chaetomium cupreum in Pichia pastoris by Multi-site Mutations

By using gene SOEing PCR, Arg (CGC) in the expression fragment of chitinase gene (chi58) from Chaetomium cupreum was mutated synonymously to Arg (AGA), which is a bias code of Pichia pas-toris yeast. The expression plasmid pPIC9K-chi58A was constructed and transformed into GS115 strain through electroporation. The chitinase activity reached 101.71 U/mL?3.33 U/mL after induced with 120 h, which was the 3 fold of the original strain (31.83 U/mL?4.85 U/mL). Additionally, recombinant yeast showed good genetic stability after 10 cycle. SDS-PAGE analysis suggested that the weight of protein was 58 kD.
3.Clinical analysis of 48 patients with metachronous bilateral primary breast cancer

ObjectiveTo evaluate the characteristics,treatment and prognosis of metachronous bilateral primary breast cancer(mBPBC). MethodsClinical materials of 48 patients were performed with retrospective study. ResultsIn 48 patients,43 cases taken both sides modified radical mastectomy and 5 cases taken lumpectomy and axillary dissection in the second side.All patients were treated with chemical treatment and treated with radiotherapy in accordance with lymph node metastasis.Besides a part of patients were treated with endocrine therapy in accordance with the expression of estrogen receptor(ER)and progesterone receptor(PR).The follow-up time was 6 ~ 48 months and the average time was 21.9 months.No died and 5 cases found distant metastasis. ConclusionmBPBC should be diagnosed earlier,taken appropriate treatment and followed up actively,so it would have a better therapeutic effect.

8.Application of comprehensive experiment evaluation indicators to the fundamental nursing assessment

Objective To evaluate the comprehensive experiment in fundamental nursing by using the indicator system and analyze the teaching effect.Methods 450 nursing graduates of grade 2011 were divided into 56 groups and comprehensive experiments were graded by using the indicator system in each group.Results The four indexes in level one scored (18.08±1.50),(19.99±2.31),(28.18±3.35),(18.16±1.73) points and accounted for 90% 、80% 、81%、91% respectively.The total points' scorings and proportions were (84.41±5.84) points and 84% respectively.Some deficiencies were found to be improved during evaluation.Conclusions Nursing comprehensive experiment evaluation indicator system can assess comprehensive experiments well,and the related competencies of nursing graduates need to be improved.
9.The investigation and evaluation of bone mineral density in type 2 diabetic women

Objective To observe the bone mineral density(BMD)in type 2 diabetic women and discuss the methods for evaluation of BMD.Methods BMD was measured by dual-energy X-ray absorptiometry(DEXA)at skeletal regions of supine and lateral spine,left femur neck and left hip in 484 type 2 diabetic women and 868 healthy women.The relation of BMD of healthy women in skeletal regions with age changes was found to fit a cubic regression model.Results The peak BMD of lateral spine and hip appeared at 25-34 and 35-44 years old respectively.BMD in diabetic women was not different from age-matched healthy women.The BMD values of the different age stages in diabetic women showed no difference(P>0.05)as compared with healthy women,and decreased with the age increase.The numbers of damaged bone sites were increased with the age increase.The BMD difference values between peak value and values of different age stages showed no difference between diabetic and healthy women groups(P>0.05).Conclusion The changes of BMD in type 2 diabetic women show no difference from age-matched healthy women.And in both diabetic and healthy women,BMD decreases with age increasing.
10.Expression of relaxin receptor in placental tissues of normal pregnancy and pre-eclampsia

Objective To study the location and level of relaxin receptor in placenta tissues of preeclampsia and normal pregnancy,and the relationship of relaxin receptor with pre-eclampsia.Methods Twenty-six placenta tissue samples from pregnant women with severe preeclampsia(study group),and 20samples from normal pregnancies(control group)were obtained.We detected the expression of relaxin receptor by immunohistochemistry and the expression of relaxin receptor mRNA by RT-PCR.Results In the placenta of control and preeclampsia groups,the leucine-rich repeat-containing G protein-coupled receptor(LGR7)was positively expressed.Relaxin receptor was located in the membrane of trophoblast cells.There were both strong signals on cytotrophoblastic cells and syneytiotrophoblast.The level of relaxin receptor in the control group was 0.912 ±0.003.and 0.625±0.037 in the preeclampsia group.Thedifference between the two groups was obvious(P<0.01).In the control group,the level of relaxin receptor mRNA was 0.776±0.021;in the preeclampsia group,it was 0.393±0.075.The difference was obvious(P<0.01).Conclusion Decrease in the expression of relaxin receptor at placenta is related with the occurrence and development of preeclampsia.
