1.Effects of ACh on Voltage-dependent Outwardly Rectifying Potassium Currents in Isolated Outer Hair Cells in Cochlea of Guinea Pig

Objective To observe the effect of ACh on outwardly rectifying potassium currents in isolated outer hair cells(OHCs) with variant length and to analyze effects of ACII on kinetics of the currents. Methods The whole cell patch-clamp technique was used. Results 100 ?mol/L ACh induced greater effects on outwardly rectifying potassium currents in short OHCs. The maximal amplitude of the outward currents increased by 34.8% in the presence of 100 ?mol/L ACII when conditioning potential was 50 mV. ACh effects on the peak currents were greater when compared to the effects on the steady-state currents and changed the shapes of the currents. ACII caused a hyperpolarizing shift of zero current potential of 5 mV. Kinetics of the currents were changed by 100 ?mol/L ACh which induced significantly more hyperpolarizing than control, with increased potential sensitivity of activation, V1/2(Ach) =(-52.38 ? 3.98) mV, SACh =(40 ? 4.14) mV(n= 5). Conclusion ACh increased the conductance of outwardly rectifying K + channel in a voltage dependent manner and caused a hyperpolarizing shift of activation potential. Effect of ACh was to hyperpolarize the membrane potential of OHCs from rest.
2.Chitinase Gene Overexpression of Chaetomium cupreum in Pichia pastoris by Multi-site Mutations

By using gene SOEing PCR, Arg (CGC) in the expression fragment of chitinase gene (chi58) from Chaetomium cupreum was mutated synonymously to Arg (AGA), which is a bias code of Pichia pas-toris yeast. The expression plasmid pPIC9K-chi58A was constructed and transformed into GS115 strain through electroporation. The chitinase activity reached 101.71 U/mL?3.33 U/mL after induced with 120 h, which was the 3 fold of the original strain (31.83 U/mL?4.85 U/mL). Additionally, recombinant yeast showed good genetic stability after 10 cycle. SDS-PAGE analysis suggested that the weight of protein was 58 kD.
5.Clinical analysis of 48 patients with metachronous bilateral primary breast cancer

ObjectiveTo evaluate the characteristics,treatment and prognosis of metachronous bilateral primary breast cancer(mBPBC). MethodsClinical materials of 48 patients were performed with retrospective study. ResultsIn 48 patients,43 cases taken both sides modified radical mastectomy and 5 cases taken lumpectomy and axillary dissection in the second side.All patients were treated with chemical treatment and treated with radiotherapy in accordance with lymph node metastasis.Besides a part of patients were treated with endocrine therapy in accordance with the expression of estrogen receptor(ER)and progesterone receptor(PR).The follow-up time was 6 ~ 48 months and the average time was 21.9 months.No died and 5 cases found distant metastasis. ConclusionmBPBC should be diagnosed earlier,taken appropriate treatment and followed up actively,so it would have a better therapeutic effect.
6.Application of red blood cell distribution width in the early diagnosis of patients with chronic heart failure

Objective To investigate the clinical significance of red blood cell distribution width in the early diagnosis of patients with chronic heart failure.Methods Seventy eight cases of chronic heart failure patients from November 2011 to November 2013 in our hospital were recruited in this study,and were divided into three different heart failure groups (Ⅱ,Ⅲ,and Ⅳ) according to the degree of heart failure; 40 healthy cases were used as control group.The red blood cell distribution width was measured among four groups.The relationship of the red blood cell distribution width and chronic heart failure was analyzed.According to the red blood cell distribution width data,patients were grouped into < 13.1%,13.1% ~ 14.5%,and > 14.5% groups.The eventual fatality rates were compared among three groups with different red blood cell distribution width.Results (1) The red blood cell distribution width was (11.63 ±0.71)%,(13.11 ±1.08)%,(14.31 ±1.52)%,and (16.83 ±0.71)% in the healthy group,andⅡ,Ⅲ,and Ⅳ heart failure groups,respectively.The red blood cell distribution width was gradually increased with the degree of heart failure,with a statistically significant difference among four groups (P < 0.05).(2) The mortality was 0 (0/32),14.8% (4/29),and 41.2% (7/ 17) in different red blood cell distribution width groups (< 13.1%,13.1% ~ 14.5%,and > 14.5%),respectively.The mortality was gradually increased with the increased red blood cell distribution width,with a statistically significant difference among three groups (P < 0.05).Conclusions Red blood cell distribution width is of great importance to detect disease degree of chronic heart failure and can guide one to effectively evaluate disease processes and therapeutic effect.
7.Identification and Antimicrobial Resistance Analysis of Mycoplasma from Cervical Secretion

OBJECTIVE To study infection and drug resistance of Mycoplasma from female genital tract.METHODS Mycoplasma in cervical secretion from inflammed female genital tract were identified by cultivation,and the sensitivities to antibiotics were also performed.RESULTS Mycoplasma were detected in 318 specimens among 588 patients with genital tract inflammation(54.1%),among which Ureaplasma urealyticum(Uu) accounted for 312(98.1%),and Mycoplasma hominis was 6(1.9%).The resistance to 12 kinds of antibiotics indicated that the sensitivities to 8 kinds of antibiotics were higher than 80% and the sensitivities to erythromycin,spiramycin and tetracycline were the lowest.CONCLUSIONS The infectious rates of Uu are on big rise in female genital tract with inflammation and the resistant strains increase continuously.
8.Analysis of the Exam Outline and the Test Questions in the 7th Edition of Licensed Pharmacist Examina-tion Pharmacy Expertise(Ⅱ)

OBJECTIVE:To investigate the direction of examination reform of licensed pharmacists,and to provide a basis for setting up the courses for pharmaceutical major in higher vocational college. METHODS:By the method of literature analysis and comprehensive analysis,examination points and their distribution,the 7th edition of Pharmacy Expertise (Ⅱ) was analyzed in-depth in terms of the change of exam outline,score distribution and the contents of test papers,correlation between teaching ma-terials and scores of test papers,key and difficult points. RESULTS:The exam outline of the 7th edition of Pharmacy Expertise (Ⅱ)was changed from 7 systems into 16 chapters;each chapter contained 3 parts:pharmacological action and clinical evaluation, medication monitoring and clinical application of commonly used drugs;there were most examination points in pharmacological ac-tion and clinical evaluation which were important points. The quantity of test questions was reduced,but choice questions were add-ed,mainly on medication monitoring. The 5th,8th,10th chapters accounted for the most points in the test papers,and ratios of page numbers to points in chapters 1-2,4,9,and 14-16 were all lower than average level. The pharmacological action and clinical evaluation were key points,and the clinical application of commonly used drugs were difficult pints,which emphasized on practi-cal skills in pharmaceutical care. CONCLUSIONS:Licensed pharmacist examination is closely related to clinical practice,which re-flects the characteristics ofexamination based on practiceandpriority to practice,and provides reference for setting up pharma-cy courses.

10.Investigation and analysis of the current human resources in clinical laboratories of hospitals in Ningxia province

Objective To investigate the current human resources and continuing education of clinical laboratories in Ningxia's hospitals and identify problems to resolve.Methods A questionnaire survey was made to study the current human resources in clinical laboratories of hospitals in Ningxia,and the data obtained were analyzed.Results There were 676 laboratory staff employed in 75 clinical 5.1.In terms of education,the ratio their titles ranging from postgraduate,graduate,college Differences in their continuing education are significant statistically.Conclusion Imbalance is found in the distribution of academic titles,education and age groups in such clinical laboratories,with a shortage of those of high academic title and education,and great gaps in continuing education.
