1.Significance of the high-risk human papilloma virus detection in the screening and diagnosis of cervical lesions

[Objective]To investigate the significance of the high-risk human papilloma virus (HPV)detection in the screening and diagnosis of cervical lesions.[Methods] The high-risk HPV DNA test results of 797 patients with cervical lesions who all accepted cytology and histopathology test were collected and analyzed retrospectively.[Results]The high-risk HPV DNA positive rates in cervicitis,cervical intraepithelial neoplasia(CIN)Ⅰ,CIN Ⅱ,CIN Ⅲ and cervical cancer were 53.41%(188/352),70.91%(117/165),87.63%(85/97),97.90%(140/143),97.50%(39/40),respectively.The sensitivity and negative predictive value of the high-risk HPV DNA detection for CIN Ⅱ and more serious lesions were 96.66%(318/329),93.29%(153/164),respectively.The detection rate of CIN Ⅱ and more serious lesions in patients with atypical squamous cells of undetermined significance(ASCUS)and positive high-risk HPV DNA was 30.03%(94/313),while the rate in patients with negative high-risk HPV DNA was 1.55%(2/129).[Conclusions] The more serious the cervical lesion is,the higher high-risk HPV DNA positive rate is.It is most closely related with CIN 11 and cervical cancer.The high-risk HPV DNA detection has high sensitivity and negative predictive value for CIN Ⅱ and more serious lesions.The high-risk HPV DNA detection has high negative predictive value in CIN Ⅱ and more serious lesions in ASCUS.

Objective To observe the effect of antisense hypoxia inducible factor-1?(HIF-1?) on (chemosensitivity) of human pancreatic cancer cell line BxPC-3 under hypoxia. Objective To investigate the anti-migratory and anti-invasive effect of somatostatin receptor type 2(SSTR2) gene transfection mediated by adenovirus in human pancreatic carcinoma cell and the mechanisms involved in this effect.Methods The full length human SSTR2 cDNA was introduced into pancreatic cancer cell line BXPC-3 by adenovirus-mediated transfection,and stable expression of RNA and protein of SSTR2 were detected by RT-PCR and Westen-blot.The Matrigel coated Transwell was used to detect the migratory and invasive ability of SSTR2expressing cells,Adv-GFP control cells and mock control cells.Furthermore,the expressions of matrix metalloproteinase-2(MMP-2) and tissue inhibitor of metalloproteinase-2(TIMP-2) were detected by RT-PCR method in these cells.Results The stable expression of SSTR2 was detected in BXPC-3 cells transfected by Adv-GFP-SSTR2.A dramatic decrease of BXPC-3 expressing SSTR2 cell(migrated) through a Matrigel-coated filter was observed,as compared with Adv-GFP control cells and mock control cells(P
4.Expression and significance of EphA2 and E-cadherin in pancreatic carcinoma

Objective To explore the expression and clinical significance of EphA2 and E-cadherin in (pancreatic) cancer tissue and cells.Methods The expression of EphA2 and E-cadherin in 56 pancreatic(carcinomas) and 23 adjacent noncancer tissues were studied by immunohistochemistry,and their relationship to clinicopathological characteristics were analyzed.RT-PCR was performed to explore the expression of EphA2,BXPC-3,PC-3 and PANC-1 in pancreatic carcinoma cell lines.Results In pancreatic carcinomas tissuse showed increased EphA2 expression and reduced E-cad expression which compared with adjacent noncancer tissues.The expression level of EphA2 had a significant positive relationship with tumor differentiation degree,lymphatic metastasis and clinical stage.However,the expression level of E-cadherin had negative relationship with both the tumor clinical stage and lymphatic invasion.Furthermore,a significant negative relationship(between) the expression of EphA2 and E-cadherin was observed.The expressions of EphA2 were higher in high-invasive cell lines PXPC-3 and Panc-1 than in low-invasive cell line PC-3.Conclusions The(expression) and/or abnormal function of EphA2 and E-cadherin may together be involved in the development and progression of pancreatic cancer;the combined measurement of these two proteins may be useful for(determination) of metastatic potency of pancreatic carcinoma.
5.Study on the Effect of Auricular Point Sticking on Sleep Quality of Patients Undergone Liver Transplantation

Objective To observe the effect of auricular point sticking on the sleep quality of patients undergone liver transplantation.Method The sleep quality was assessed by using sleep quality scale. The patients undergone liver transplantation were randomized into two groups, both receiving auricular point sticking, while the insomnia zone was selected in the observation group and non-insomnia zone was selected in the control group, 4 weeks as a treatment course.Result In 4 weeks after the transplantation, there were significant inter-group differences in comparing the scores of unfreshing sleep, difficulty falling asleep, nightmairs, medications, post-insomnia reactivity, and the total score of the self-rating scale of sleep (SRSS) (P<0.05); in 2 months after the transplantation, there were significant differences in comparing the scores of sleep latency, sleep efficiency, sleep disturbance, sleep medications, daytime dysfunction, and the global score of Pittsburgh Sleep Quality Index (PSQI) between the two groups (P<0.05), indicating that the sleep quality in the observation group was markedly improved. Three months after transplantation, there were no significant differences in comparing the sleep indexes between the two groups (P>0.05).Conclusion Auricular point sticking at the insomnia zone can markedly improve the short-term sleep disorder in patients after liver transplantation, and can reduce the dose of sleep medications.
7.Effect of integrin-?1 antisense oligodeoxynucleotides on human pancreatic cancinoma transplanted subcutaneously in nude mice

Objective To investigate the effect of integrin-?1 antisense oligodeoxynucleotide(ASODN) on(human) pancreatic cancinoma transplanted subcutaneously in nude mice.Methods The models of human(pancreatic) cancinoma transplanted subcutaneously were established in nude mice,then divided randomly into 3 groups and different treatment was given respectively(control group,random oligodeoxynucleotide group and ASODN group).After treatment,the weight of nude mice and tumor volume were observed,and the tumor growth inhibitory rate and the tumor response rate were calculated.The expressions of integrin-?1 mRNA and protein in tumor tissue were determined by RT-PCR and Western-blot.Results The tumor growth inhibitory rate in the random oligodexynucleotide group and the ASODN group was 4.75% and 72.70%,respectively.The tumor decrease rate of the ASODN group was 10.91%.The expression level of integrin-?1 mRNA and protein was decreased in the ASODN group compared with other 2 control groups. Conclusions Our findings suggest that integrin-?1 antisense oligodeoxynucleotides result in marked inhibition of human pancreatic(cancinoma) growth in nude mice.It may be a novel treatment approach for human pancreatic carcinoma.
8.Investigation on water fluoride content and water-improving defluoridation projects in endemic fluorosis areas in Jining City, Shandong Province in 2005

Objective To investigate water fluoride content and the current status of water-improving project in Jining,Shandong Province in 2005.Methods Water fluoride content and the status of water-lmprovlng project were havestigated in Bencheng Ditrict,Liangshan and Jiaxiang Couties of Jining City.The fluoride content in drinking warer was assessed by F-ion selective electrode.Results Water fluoride contents of 671 fluorosis villages were determined.One hundred and seventy villages had water fluoride content less than or equal to 1.00 mg/L(accounting for 25.34%)and 501 villages water fluoride content higher that 1.00 mg/L(accounting for 74.66%),with the highest being 11.33 mg/L.Four hundred and ninty.seven projects were surveyed,in which 161 Projects were discarded (accounting for 32.39%),334 projeets worked well(accounting for 67.20%),and 2 pmjects worked fitfully(accounting 0.40%).Two hundreds and eighty-one projects had water fluoride content≤1.00 ms/L and 55 projects had water fluofide cantent>1.00 mg/L(aceounting for 26.82%),with the highest being 4.28 ms/L.Conclusions The situation of endemic fluorosis control is still serious in Jining City.Countermeasures for endemic fluorosis must be carried out as sppm as possible and surveillance of water-improving and defluoridation projects must be strevIsthend.
9.Effect of vincristine on the myoelectric activity and motility of the small intestine in rats

Objective To investigate the effects of vincristine on myoelectric activity and motility of the small intestine in conscious rats and its mechanism. Methods Seventy-two SD rats were divided into six groups. The rats in control group were received 0.9% NaCl solution (n=18). The rats in group B were injected with vincristine and subdivided into 0.25 mg/kg(n=6) ,0.5 mg/kg(n=6)and 0.75 mg/kg groups. The group C and D was false operation (n=6)and false operation plus injection with 0.75 mg/kg of vincristine(n=6), respectively. The group E and F was subdiaphragmatic vagotomy plus 0.9% NaCl (n=6) or subdiaphragmatic vagotomy plus 0.75 mg/kg of vincristine (n=6), respectively. The myoelectric activity and motility of the small intestine were recorded. The frequency and area under the curve of slow wave, the periodicity of the migrating myoelectrie complex (MMC) and the duration of MMC Ⅲ were analyzed. The expressions of the myenteric neurons and interstitial cells of Cajal were evaluated by immunofluorescence stain. Results The myoelectric activity in 0.25mg/kg group was not different from the controls, but it changed in 0.5 mg/kg and 0.75 mg/kg groups which correlated with the time of vincristine injection. The irregular spike activity arose and accompanied with disruption of the MMC at (51±14.27) minutes,but recovered at (78.33±13.08) minutes. The periodicity was shorter in 0.5mg/kg [(343.17±142.93)s]and 0.75 mg/kg groups [(302.67±66.67)s] compared with controls [(740.22± 98.92) s, F=31.325, P<0.01]. Three days after vincristine administration, the area under the curve of slow wave decreased to (2.56±0.30) mV·s in 0.5 mg/kg group and (2.57±0.56) mV·s in 0.75 mg/kg group compared with the controls ((4.10±0.80) mV·s , F = 11.442, P<0.01). The intestinal propulsive rate was lower in 0.75 mg/kg group compared with the controls [(33.59±1.43) vs(60.34±2.41)%,t= 23.36, P<0.01]. The expression of the interstitial cells of Cajal was less than those in controls. Three days later, the area under the curve of slow wave in F group was less than that in controls. Conclusions Vincristine provokes alterations of myoelectric activity and motility of the small intestine. The early vincristine-induced escalating in myoelctric activity of the small intestine is through vagus nerve pathway. The decreased motility is contributed to the damage of the interstitial cells of Cajal.
10.Genotyping analysis of a polymorphic G-954C of NOS2A in diabetic retinopathy with cystoid macular edema

AIM: To analyze the genotype of the allele distribution of a polymorphic G-954C within the 5 upstream promoter region of the nitric oxide synthetase 2A gene (NOS2A) in samples of diabetic retinopathy in patients with cystoid macular edema in the mainland of China.METHODS: Eighty-nine patients with diabetic retinopathy and cystoid macular edema and 90 healthy controls were enrolled in this study. Nest polymerase chain reaction (PCR)was performed, and restriction endonudease digestion and gene fragments sequence were examined to detect the genotype of NOS24 G-954C.RESULTS: The genotypes of the sample population of 89 cases and 90 healthy controls were all detected as GG.CONCLUSION: The distribution of G-954C of NOS2A polymorphism are at a lower frequency in China, with little relevancy to the frequency of diabetic retinopathy combined with cystoid macular edema.
