1.Evaluation of a health education program on iodine deficiency disorders in Jiangsu Province

Objective To evaluate the effects of a health education project on prevention and control of iodine deficiency disorders.Methods From January to June 2011,an investigation was conducted using combined unified questionnaire in 9 counties on target population,and 3 townships were chosen in each project county.Health educational activities were carried out in grade 5 classes in the central primary school.In the meantime,3 villages were chosen in each selected township where the health educational activities in the community were carried out.Before and after the health educational activities,questionnaire surveys on knowledge of iodine deficiency disorders were conducted in each project county.Thirty students in one class of grade 5 in the central primary school were selected,and 15 women of childbearing age near the central primary school were selected too.The health education content included but not limited to:popular science films about the knowledge on endemic disease prevention and public-interest ads were broadcasted by radio,television,newspapers and other media in the project counties,aiming at spreading knowledge on prevention and treatment of endemic diseases.In each of the project township government and village committee locations,posters,banner pieces and slogan suspensions about knowledge on endemic disease prevention were posted or put up.In the project rural hospitals,a special bulletin boards was set up,and in the village clinics (rooms),posters were put up,to carry out a training program for women of childbearing age on endemic disease prevention.In primary schools,students in grades 4 to 6 were given a class about knowledge on prevention and treatment of iodine deficiency disorders,aiming at their transferring the learned knowledge to family members.Results A total of 2678 persons were investigated before and after the health education.After health education,the rate of knowledge(the correct answer rate) on iodine deficiency disorders increased from 69.04% (1920/2781) to 94.08% (2622/2787,x2 =580.63,P < 0.05) among primary school students,and 74.76%(924/1236) to 92.60%(1139/1230,x2 =143.59,P < 0.05) among women of childbearing age.Conclusions The rate of knowledge awareness in the students and the women of childbearing is increased significantly after the health educational activities.Systematic and targeted health education activities are effective measures in prevention and control of iodine deficiency disorders.
2.The expression and significance of the role of differentially expressed gene interleukin 8 of early squamous cervical carcinoma

Objective To determine the correlation between the expression of interleukin 8(IL-8) and lymph node metastases in earlier uterine squamous cervical cancer and to study the mechanism of metastases of earlier squamous cervical cancer. Methods IL-8 mRNA expression was examined with reverse transcription polymerase chain reaction(RT-PCR) and tissue microarray to determine the expression of IL-8 protein. The matrix metalloproteinase 9( MMP-9) protein and microvessel density (MVD) in earlier squamous cervical cancer were decided respectively by antibody against IL-8, MMP-9 and CD-34 associated antigen and Immunohistochemical analysis. Results The cervical cancer with lymph node metastases express IL-8, MMP-9 and MVD at significantly higher level than negtive metastatic lymph node(P =0.00,0.00,0.02). 3.A survey of iodine nutritional status of the crowd in iodine-deficient areas that did not participate in the provincial surveillance program on iodine deficiency disorders in Jiangsu Province

Objective To investigate the iodine nutritional status of the crowd that did not participate in the iodine deficiency disorders (IDD) survey in iodine deficiency areas in Jiangsu Province so as to provide a basis for prevention and control of IDD.Methods Eight to ten years old children were chosen from the counties(cities,districts) of Jiangsu Province that did not participate in the IDD survey and urine samples of students and salt samples of their families were researched.In each county (city,district) one township (street) was selected according to 5 directions of east,south,west,north,center; in each township one primary school was selected and in each primary school 20 children aged 8-10 were chosen half of the male and female,to collect their urine samples and salt samples at their families.Urinary iodine was measured using arsenic cerium catalytic spectrophotometric method (WS/T 107-2006),and salt iodine was measured using direct titration of universal test method in salt industry (GB/T 13025.7-1999).Iodine nutrition criterion of international health organization recommended staindard:children urinary iodine < 100 μg/L as iodine deficiency,100-199 μg/L as iodine appropriate,200-299 μg/L as iodine more than appropriate,≥ 300 μg/L as iodine excess.Results A total of 7 523 urinary samples of 8-10 years old children were collected from 76 iodine deficiency counties (cities,districts),and the median of urinary iodine was 235.0 μg/L.In all investigated counties (cities,districts),the medians of urinary iodine of 22 counties(cities,districts) were 100-199 μg/L,42 counties(cities,districts) were 200-299 μg/L,and 12 counties (cities,districts) were ≥300 μg/L.A total of 7 523 salt samples were collected,and the median of salt iodine was 29.68 mg/kg; Taizhou City had the highest salt iodine median,which was 31.43 mg/kg; and Lianyungang City,the lowest,25.11 mg/kg.The coverage rate of iodized salt was 98.6%(7 417/7 523); iodized salt qualification rate was 98.6% (7 312/7 417); and consuming rate of qualified iodized salt was 97.2%(7 312/7 523) in 76 iodine deficiency counties (cities,districts).Conclusions The progress in prevention and control of IDD in iodine-deficient areas that did not participate in IDD survey in the past is good in Jiangsu Province; the overall level of iodine nutrition of whole population is adequate.But the median of urinary iodine in some counties (cities,districts) is at a higher level.
4.Effects of saturated and unsaturated fatty acids on proliferation and autophagy of lung cancer cells

Objective To investigate the effects of saturated fatty acids and unsaturated fatty acids on proliferation and autophagy of human lung cancer cells. Methods The lung cancer cells A549 were treated with stearic acid (saturated fatty acid) and doconexent (DHA, unsaturated fatty acid), respectively, in concentrations of 0, 30, 60, 120 and 240μmol/L. MTT test and cell clone formation assay were performed to detect the proliferation of A549 cells. The morphology of A549 autophagy was observed by confocal laser scanning microscopy after A549 cells were treated with stearic acid or DHA for 24 hours. Western blotting assay was used to detect the expression of autophagy-related protein after A549 cells were treated with stearic acid or DHA for 12, 24 and 36 hours, respectively. Results 30-240μmol/L stearic acid or DHA both inhibited the proliferation of A549 cells (P<0.05). Both stearic acid and DHA induced autophagy of A549 cells, meanwhile, down-regulated Phospho-mTOR (ser2481) and up-regulated LC3Ⅱ/LC3Ⅰ of A549 cells (P<0.05). Conclusions Both saturated fatty acid and unsaturated fatty acid can inhibit the proliferation and induce autophagy of lung cancer cells. The mechanisms of autophagy may be related to Phospho-mTOR (ser2481) signaling pathway.
5.Isolation and identification of nucleus pulposus mesenchymal stem cells from tissues removed by percutaneous endoscopic lumbar discectomy

AIM:To explore a novel method to isolate human nucleus pulposus mesenchymal stem cells (hNP-MSCs) in vitro and to identify their biological characteristics.METHODS:The explant culture method was employed to isolate hNP-MSCs from nucleus pulposus tissue obtained by percutaneous endoscopic lumbar discectomy (PELD).The isolated cells were passaged for purification and cultured in vitro followed by morphological observation.The cell proliferation ability was detected by CCK-8 assay.Growth curves of the cells were drawn and surface antigens were detected by flow cytometry.The cells at the 3rd~6th passages were induced for adipogenic, osteogenic and chondrogenic differentiation, and examined by oil red O staining, alizarin red staining and Alcian blue staining.RESULTS:The cells with self-renewal were obtained from nucleus pulposus tissue obtained by PELD.The results of flow cytometry analysis revealed that the cells were positive for CD29, CD44, CD90, CD73 and CD105, but negative for CD34 and CD45.The proliferative capacity was consistent with the growth characteristics of MSCs and multilineage differentiation potential was identified.CONCLUSION:A novel method to efficiently isolate and culture hNP-MSCs,PELD combined with explant culture method,was established, which would promote the study of regenerative medicine based on hNP-MSCs.
6.Correlation of polymorphism of IL-8 and MMP-7 with occurrence and lymph node metastasis of early stage cervical cancer.

The IL-8 and MMP-7 genes participate in the carcinogenesis of many malignancies, but the role of both genes in cervical cancer is not fully elucidated. The aim of this study was to determine the frequency of IL-8 and MMP-7 gene mutations and to assess their effects on the risk of early stage cervical cancer and lymph node metastasis. The clinical stage and histological grade of cervical cancer were also studied. The peripheral blood from the patients with early stage cervical cancers and normal controls was collected and the DNA was extracted. The incidence of IL-8 and MMP-7 gene mutations was assessed by using tetra-primer amplification refractory mutation system polymerase chain reaction (ARMS PCR) and restriction fragment length polymorphism (RFLP). The data were statistically analyzed by x2 test. The results showed that: (1) The genotype frequency of IL-8 -251AT and TT was significantly higher in the cervical cancer group than in the normal control group (OR=2.290 and 2.619 respectively, P=0.001), and it was also higher in the lymphatic metastasis group than that without metastasis (OR=2.917, P=0.035); (2) The frequency of MMP-7 -181G/G genotype was significantly higher in the cervical cancer group and in the lymphatic metastasis group (P<0.05); (3) The incidence of IL-8 mutation was two times higher in IIa cervical cancer group than in Ib1 and Ib2 cervical cancer group (P=0.006). For the MMP-7 gene, there was statistically significant difference in the incidence of mutation between the Ib1, Ib2 and the IIa (P=0.000); (4) Different histological types and different grades of cervical cancer had different incidence of mutations, statistically. It was suggested that there was significant difference in the genotype of IL-8 -251TT and MMP-7 -181GG polymorphism between the cervical cancer group and the lymph node metastasis group. Moreover, individuals with IL-8 T allele or MMP-7 G allele carriers were at significantly higher risk of cervical cancer, particularly the early (IIa) and medium, poorly differentiated cervical cancer (G2+G3).
7.Influence of vitamin D3 on proliferation and apoptosis of PANC1 cells through hedgehog signaling pathway

Objective To investigate the role of vitamin D3 in anti-pancreatic cancer.Methods After treatment of different concentrations of vitamin D3 on PANC1 cells (25,50,75,100μmol/L),MTT assay was used to detect the growth inhibition rates of PANC1 cells,the early apoptotic rates of the cell were detected by flow cytometry,PTCH and Gli-1 mRNA expression were detected by RT-PCR method,and cells without treatment were used as control.Results The vitamin D3 inhibited the proliferation of PANC1 cells in a dose-dependent manner,the highest inhibition rate was at 48 hours.After 48 hours,the control group,25,50,75,100 μmol/L vitamin D3 groups' inhibition rates were 0,16.1%,18.8%,31.8% and 39.4%,the differences among these groups were statistically significance (P < 0.05).After 24 hours,the control group,25,50,75,100μmol/L vitamin D3 groups' early apoptotic rates were (5.89 ±0.57)%,(6.06 ±0.44)%,(16.21 ± 1.62)%,(16.94± 0.91)% and (20.96 ± 0.98)%,early apoptotic rates were inhibited in a dose-dependent manner,and the differences was statistically significance (P < 0.05).After 24 hours,the control group,25,50,75,100μmol/L vitamin D3 groups' PTCH mRNA expression were 0.117 ± 0.009,0.104 ± 0.011,0.069 ± 0.011,0.052 ± 0.009 and 0.056 ± 0.007,meanwhile the Gli-1 mRNA expressions were 0.323 ± 0.007,0.312 ± 0.015,0.299 ± 0.015,0.233 ± 0.007 and 0.175 ± 0.014,all in a declining trend with the increase of concentration,and the difference was statistically significant (P < 0.05).After 75 μmol/L vitamin D3's intervention in 0,12,24,36 and 48 hours,the expression of PTCH mRNA were 0.142±0.008,0.127± 0.009,0.111± 0.010,0.115± 0.003 and 0.102± 0.007,meanwhile the expression of Gli-1 mRNA were 0.341 ± 0.011,0.317 ± 0.017,0.320 ± 0.018,0.226 ± 0.011 and 0.191 ±0.010,all in a declining trend with time,and the difference was statistically significance (p<0.05).Conclusions Vitamin D3 can effectively inhibit the proliferation of PANC1 cells and promote its apoptosis,and these effects may be related to blocking of hedgehog signaling pathway.
8.Comparative analysis of seven marine biological source of mineral drugs.

The marine biological source of mineral drugs recorded in Chinese Pharmacopoeia (2010 version) mainly including pearl, nacre, clam shell, common oyster shell, ark shell, cuttle bone, and sea-ear shell are widely used in clinical. Calcium carbonate and a small amount of protein are the main components in this type of drugs. In this paper, a systematical and comparable study were carried out by determination of calcium carbonate by EDTA titration method, the crystal of calcium carbonate by X-Ray powder diffraction and the total amino acids (TAAs) of the hydrolyzed samples by ultraviolet spectrophotometry method. As a result, the crystal structure is calcite for common oyster shell, mixture of calcite and aragonite for nacre and sea-ear shell, aragonite for the other drugs. The content of calcium carbonate ranged from 86% to 96%. Cuttle bone has the highest amount of TAAs among the seven drugs which reached 1.7% while clam shell has the lowest content of 0.16% on average. In conclusion, an effective method was developed for the quality control of marine mineral drugs by comprehensive analysis of calcium carbonate and TAAs in the seven marine mineral drugs.

9.Analysis of iodized salt monitoring results in high iodine county and non-excessive iodine county in Jiangsu Province in 2011

Objective To investigate the supply of iodized salt in non-excessive iodine counties and iodine-free salt in excessive iodine counties at household level in Jiangsu Province so as to provide a basis for the prevention and control of iodine deficiency disorders(IDD).Methods According to the National Iodine Deficiency Disorders Monitoring Program(Trial),county(city,district) was taken as a elementary sampling unit in Jiangsu Province.Townships(towns) and administrative villages were selected by systematic sampling and random sampling in every county and households were chosen by random sampling to collect their edible salt samples.The salt iodine content in non-and excessive iodine regions was detected by direct titration method and semi-quantitative method,respectively.Results All 30 840 salt samples were collected from 106 non-excessive iodine counties,and qualified iodized salt was 30 303 copies,iodine-free salt 199 copies.Weighted by the population of counties,the rate of iodine-free salt was 0.71%,the coverage rate of iodized salt accounted for 99.29%,out of which,98.93% was qualified and the consuming rate of qualified iodized salt was 98.23%.All 1296 salt samples were collected in 5 counties with excessive water iodine content and the coverage rate of iodine-free salt was 98.99% (1283/1295).Conclusions The national targets for preliminary elimination of IDD have been achieved in regions of nonexcessive and excessive iodine of Jiangsu Province.But it still should be strengthen the management work of iodinefree salt in excessive iodine counties and iodine saft in non-excessive iodine counties.
10.Clinical observation of elderly patients with coronary heart disease undergoing coronary artery bypass grafting through descending thoracic aorta

Objective To investigate the clinical effectiveness of coronary artery bypass grafting through descending thoracic aorta in elderly patients with coronary heast disease and to decrease the post-operative complication.Methods Thirteen elderly patients underwent coronary bypass surgery with minimally invasive direct coronary artery bypass (MIDCAB).Age range from 70 to 82 years with a mean of(72.1?6.0)years.Patients suffered from multi vessel disease.Many minimally invasive techniques of“Y”blood vessel graft anastomosis,anastomosis of blood vessel graft to descending aorta,minimally invasive direct,thoracoscope assist were used.Results All patients were survived.The mean duration of intubation was (6.9?0.9) hours.The average ICU stay was (2.5?0.5)days.No patients received blood transfusion.During the short-term follow-up(3 to 14 months) patients had no complaint of angina,Conclusions The technique of“Y”blood vessel graft anastomosis,descending aorta blood vessel graft,minimally invasive direct and thoracoscope assist in combination with coronary artery bypass grafting is a safe and cost-effective new procedure for elderly patients with multi-coronary artery disease.
