1.Clinical evaluation of preoperative selective intraarterial embolization for nasopharyngeal fibroangioma

Objective To study the practical value of preoperative selective intra-arterial embolization in the surgical treatment of nasopharyngeal fibroangioma. Methods Digital subtraction angiogrophy(DSA) and selective arterial embolization were carried out in 15 cases of nasopharyngeal fibroangioma. The supplying arteries originated from the maxillary and the pharyngeal ascending branches arising from external carotid artery were embolized with gelatin sponge. Results 15 cases underwent operation after selective embolization of blood supplying arteries and the amount of bleeding in operation was thus reduced greatly in 13 cases, while the blood loss during the surgery of another 2 cases was still considerable because both external carotid artery and internal carotid artery supplied the tumor with only embolization of the external carotid artery. Optimum time for the operation was taken on 2-5 days after the embolization. Conclusions Preoperative intra-arteral embolization for nasopharyngeal fibroangioma is a valuable method of choice for the clinical treatment.
2.The diagnostic value of digital subtraction angiography of intracranial meningioma

Objective To evaluate the diagnostic value of digital subtraction angiography (DSA) of intracranial meningioma.Methods DSA was performed in 115 cases with meningioma. The branches of the external carotid artery were embolized in 76 cases. Results The blood supply of the meningioma located at the relatively superficial cerebrum are mainly arising from the middle meningeal artery of the internal maxillary artery and those located at the skull base are mainly from the ophthalmic artery and the cerebral segment of the internal carotid artery. The possible dangerous anastomoses between the extermal and internal carotid arteries and the vertebrobasilar arteries may exist.Conclusions To have a good knowledge of the charactereristics of digital subtraction angiography of meningioma is crucial for endovascular embolization and operation of meningioma.
3.Influence of ginkgolides on expression of iNOS in MCAO rats

AIM: To study the influence of ginkgolides on expression of inducible nitric oxide synthase (iNOS) in rats. METHODS: Middle cerebral artery occlusion(MCAO) model was induced by ligating internal carotid artery of Sprague Dawley(SD) rats.In this model,the behavioral deficits performance of SD rats was analyzed after given ginkgolides and nimo dipin, respectively, and iNOS gene expression changes in MCAO group were investigated. RESULTS:The expression of iNOS was increased in MCAO models and inhibited in ginkgolides groups. CONCLUSION:Ginkgolides have inhibitive efficacy of iNOS on cerebral ischemia and can reduce brain damage.
4.Effect of Fengliao extract on mice with experimental ulcerative colitis

OJECTIVE To investigate the preventive effect of Fengliao extract on ulcerative colitis of mice. METHODS Using the intestinal propulsion rate experiment and senna induced diarrhea model , the intestinal propulsion rate, diarrhea rate and index of diarrhea were observed. Mice were randomly divided into normal group,model group,mesalazine hydrochloride group and Fengliao extract 11.7, 23.4 and 46.8 g · kg-1 group. The mouse colitis model was induced by 4% dextran sulfate sodium. The mice were administraed once daily for 7 d while the disease activity index(DAI)score was calculated and the activity of tumor necrosis factor α(TNF-α),interleukin-1β(IL-1β), myeloperoxidase(MPO) and content of malondialdehyde(MDA) and nitric monoxide (NO) in colon tissue were determined. RESULTS Fengliao extract 46.8 g · kg-1 inhibited the intestinal propulsion rate(P<0.05),reduced the frequency of diarrhea and the diarrhea index(P<0.05). Results of colitis showed that the body mass of mice in the model group was significantly decreased but the DAI score increased compared with normal group(P<0.05). The activity of MPO and the contents of IL-1β,TNF-α,MDA and NO in colon mucosa were increased(P<0.01). Compared with the model group,Fengliao extract 46.8 g·kg-1 decreased the DAI score(P<0.05)while Fengliao extract 46.8 and 23.4 g · kg-1 reduced MPO activity in colonic mucosa and content of IL-1β,TNF-α,MDA and NO in colonic homogenate(P<0.05). CONCLUSION Fengliao extract can significantly improve the DSS induced colitis in mice,which is probably associated with its antispasmodic and anti-diarrheal effect as well as the reduced release of inflammatory mediators and antioxidants.
5.Experimental study on main pharmacodynamics of Kangjun Xiaoyan Tablets

AIM: To study the mostly pharmacological effects of Kangjun Xiaoyan Tablets (Flos Lonicerae, Radix Stemonae, Radix et Rhizonia Rhei, etc.) METHODS: The pharmacological functions of KJXYT were observed by measuring its antibacterial, anti-inflammatory, antipyretic and immune etc. RESULTS: KJXYT had remarkably protected the mice from the infection with streptococcus pneumococcus and staphylococcus aureus; had dramatic anti-inflammatory effect on the inhibited feet edema of rat induced by egg white and the ear edema of mice induced by dimethylbenzene; and had obvious effect on fever of rats caused by dried yeast; besides, and strengthened the phagocytosis of mice’s reticuloendothelial system. 6.Effects of acacetin on T47D cell proliferation

Aim To investigate the effect of acacetin on cell proliferation and the influence of acacetin on estrogen receptor expression in vitro.Methods The proliferation rates and the cell cycle changes of acace-tin-treated T47D cells were measured by sulforhodam-ine B(SRB)assay and flow cytometry,respectively. Moreover,the mRNA expressions of estrogen receptor-alpha(ERα),estrogen receptor-beta(ERβ)and pro-liferating antigen(Ki67)were determined by quantita-tive real time PCR (qPCR).Western blot was em-ployed to detect the ERαand ERβprotein expression. Results Acacetin significantly promoted the prolifera-tion and increased the amount of cells arrested in S and G2 /M phase under the concentration of 0.001 ~1 0μmol·L -1 .Ki67 mRNA level and the ERαprotein level in T47D cells were remarkably upregulated after acacetin treatment.To clarify which estrogen receptors played a role in acacetin induced the proliferation of T47D cells,the combination treatment of acacetin and ERαinhibitor (MPP)/ERβ inhibitor (PHTPP) was employed.We found that MPP could reverse the cell proliferation,the cell arrested in S and G2 /M phase and the increased Ki67 mRNA level induced by acace-tin.PHTPP also alleviated the T47D cell proliferation induced by acacetin,whereas no significant changes were found in cell cycle and Ki67 mRNA level.Con-clusion Acacetin stimulates the cell proliferation of T47D cells in the concentration from 0.001 μmol · L -1 to 1 0 μmol·L -1 ,which is mainly mediated by ERα.
7.Subtype analysis and feature detection of peripheral blood Vγ9Vδ2T cells in hepatocellular carcinoma patients

Objective To investigate the percentage,mature classification and Immune killing function of Vγ9Vδ2T cell in peripheral blood of HCC patients.Methods Peripheral blood mononuclear cells (PBMCs) were isolated from HCC patients (n =25) and healthy donors (n =20) by discontinuous density gradient centrifugation.Proportion,mature and differentiate subtypes and IFN-γ and CD107a expressing of the delta 2 T cells were detect by using flow cytometry,δ2Tcell were selectively cultured with zoledronate and human IL-2.After 12-14 days cells were collected and tested for the second time.Results While the percentage of Vγ9Vδ2Tcell of total T cell in peripheral blood of HCC patients is lower than healthy people before culture (t =4.505,P < 0.001),after augmentation in vitro the proportion increased significantly (t =8.782,P < 0.001),to a level similar to healthy group (t =1.644,P =0.109).There was no statistically significant difference when differentiation subtypes of patient's Vγ9Vδ2Tcell were compared with healthy group before culturing (all P > 0.05),after culture the proportion of Tn,Tcm and Temra decreased [t(Tn) =2.081,t(Tcm) =2.478,t(Temra) =2.953,all P < 0.05],and the proportion of Tem,Tem+ Temra increased [t(Tem) =12.6,t (Tem + Temra) =9.843,all P < 0.001].Cell culture did not alter the proportion of IFN-γ and CD107a secreting Vγ9Vδ2T cells in the peripheral blood in both HCC patients and healthy people (all P > 0.05).Conclusions While the percentage of Vγ9Vδ2T cell of HCC patients in peripheral blood was lower than healthy people,its matured subtypes are similar to those of healthy people,and functions of expressing IFN-γ and CD107a are not different with healthy people.Applying ZOL + IL-2 can amplifyVγ9Vδ2T cells of patients with HCC.
8.Identification of cimicifugae rhizoma and its adulterants using ITS2 sequence.

In order to identify Cimicifugae Rhizoma from its adulterants and to ensure its safe use, the internal transcribed spacer 2 (ITS2) sequence of Cimicifugae Rhizoma and its adulterants were amplified and bidirectionally sequenced by DNA barcoding technology. Sequence assembly and consensus sequence generation were performed by the CodonCode Aligner V3.7.1. The genetic distances were computed by MEGA 5.0. Identification analyses were performed using neighbor-joining (NJ) methods. The length of ITS2 sequence of the three origin plants of Cimicifugae Rhizoma include Cimicifuga heracleifolia, C. foetida, C. dahurica was 217, 219 and 219 bp, respectively. Their intraspecific genetic distance was much lower than the interspecific genetic distance with their closely related species. The NJ tree of ITS2 indicated that the three origin plants of Cimicifugae Rhizoma formed a monophyletic clade, Cimicifugae Rhizoma and its adulterants could be distinguished clearly. The authors proposed that ITS2 sequence was suitable for the authentication of Cimicifugae Rhizoma and its adulterants.

9.Study of the relationship between apical rotation and apical myocardial structure in dogs by echocardiography

Objective To detect the relationship between apical rotation and apical myocardial structure.Methods To observe the change of apical rotation,14 mongrel dogs were heated at apexes,and half and three quarter of apical wall thicknesses were destroyed according to their corresponding heating time.Ultrasonic tissue Doppler imaging was used to record the velocities of apical rotation.Computer programming was applied to calculate apical rotation according to the formulas of apical rotation calculation.Apical rotations were compared between before apical heating,half and three quarter of apical wall thicknesses destruction,respectively.Results Compared with the apical rotation before heating,apical rotation after half of apical wall destroyed decreased [(5.2±3.1)°vs (12.4±2.9)°,P<0.001];but after three quarter of apical wall destroyed,the apical rotation increased [(7.3±3.6)°vs (5.2±3.1)°P<0.01].Conclusions Outer part of apical myocardium was outer layer myocardium of which the damage leads to the decrease of apical rotation;inner part of apical myocardium was inner layer myocardium of which the damage leads to the increase of apical rotation to some extent.
10.Serum proteomic analysis of patient with severe acute pancreatitis before and after treatment

Objective To evaluate the different expression of the serum proteome of patient with severe acute pancreatitis (SAP) before and after treatment,and search for the protein that can predict SAP improvement.Methods SELDI-TOF-MS was used to analyze the expression of serum protein spectrum of 20 SAP patients before treatment and 7 days after treatment,and make the APACHE Ⅱ score at the same time.Results The APACHE Ⅱ score was 10.70 ± 3.47 in patients with SAP 7 days after treatment,which were significantly lower than that before treatment ( 13.00 ± 2.21,P <0.05 ).One hundred thirty-two spectral peak clusters were detected before and after treatment,there were 27 proteins that had significantly different signal intensities had been detected from 2000 Da to 50000 Da (P < 0.05 ).The peak cluster at 47 000 Dawas significantly down-regulated after SAP attenuation.While the peak clusters at 1l700 Da and 15900 Dawere significantly up-regulated after treatment.Conclusions Serum analysis with SELDI-TOF MS can detect the different expression of the SAP serum proteomics before and after treatment,the increased expression of the protein with molecular weight of 11500 Da and 15900 Da and the decreased expression of 4700 Da may be asignal of improvement of SAP.
