1.Effects of CO_2 pneumoperitoneum on levels of interleukin-1? and interleukin-6,and peritoneal macrophages in tumor-bearing rats

Objective To investigate effects of CO_2 pneumoperitoneum on cytokines and peritoneal macrophages in a rat model with implanted liver tumor.Methods A total of 32 Wistar rats with implanted liver tumor were randomly divided into 4 groups((n=8)): Control Group(anesthesia only),Laparotomy Group,Gasless Group(gasless laparoscopy),and Pneumoperitoneum Group(laparoscopy under CO_2 pneumoperitoneum).Serum samples were collected at the 2nd and 24th hours after the procedure respectively for the detection of levels of interleukin-1?(IL-1?) and interleukin-6(IL-6).Samples of peritoneal macrophages were collected and incubated for the detection of levels of tumor necrosis factor-?(TNF-?),a product of macrophages.Results At the 2nd and 24th hours after surgery,levels of serum IL-6 in the Laparotomy Group(57.92?2.06 pg/ml and 35.49?1.15 pg/ml) were significantly greater than those in the Pneumoperitoneum Group(14.64?0.34 pg/ml and 15.39?0.86 pg/ml),the Gasless Group(24.75?1.53 pg/ml and 17.10?0.97 pg/ml),and the Control Group(17.75?1.60 pg/ml and 14.55?0.25 pg/ml)(P
2.Clinical study of orthokeratology lens for correction effects of adolescent myopia

Objective To investigate the correction effect and influence of orthokeratology lens and aspheric spectacles on adolescent myopia.Methods A total of 124 cases of juvenile myopia in our hospital from January 2012 to January 2013 was divided into experimental and control groups according to the principle of random.Patients in both groups were followed for a period of 2 years,patient's vision,binocular refraction,and eye axis length were recorded.Results Unaided visual acuity of patients in the experimental group was increased significantly,the trend was clear with statistical significance (P < 0.05).For the control group of patients,visual acuity was significantly decreased after 2 years (P < 0.05).For the experimental group,patients'diopters were decreased from (4.29 ± 1.23)D to (4.23 ± 0.04)D after wearing glasses.The difference was statistically significant (P < 0.05).For the control group,patients' diopters were increased from (4.22 ± 1.19)D to (5.68 ± 1.13)D after wearing glasses.The difference was statistically significant (P < 0.05).The difference between two groups at the same time was statistically significant (P < 0.05).In the experiment group,for patients wearing glasses,their axis lengths were (24.7 ±0.19)mm after six months.After 1 year,their axis lengths were (24.5 ±0.16) mm,and after 2 years,their axis lengths were (24.3 ± 0.20) mm.Compared to non-wearing glasses before,the difference was not statistically significant (P > 0.05).In the experiment group,for patients wearing glasses,their axis lengths were (24.7 ±0.19)mm after six months.After 1 year,their axis lengths were (24.5 ±0.16)mm,and after 2 years,their axis lengths were (24.3 ± 0.20) mm.Compared to non-wearing glasses before,which was (24.3 ± 0.21) mm,the difference was not statistically significant (P > 0.05).In the control group,for patients wearing glasses,their axis lengths were (24.8 ± 0.20) mm after six months.After 1 year,their axis lengths were (24.9 ±0.17) mm,and after 2 years,their axis lengths were (25.7 ±0.15) mm.Compared to non-wearing glasses before,which was (24.5 ± 0.18)mm,the difference was statistically significant (P < 0.05).Conclusions Orthokeratology lenses can reduce myopia in adolescent patients,reduce the degree of anisometropia,and prevent the increase of the axial length.It can achieve the purpose of myopia control.
3.Up-regulated expression of costimulatory molecule CD80 on dendritic cells in mice with acute lung injury

Objective To observe the regularity of expression changes in CD80 in peripheral blood, lung and splenic dendritic cells (DCs) in mice with acute lung injury (ALI) induced by lipopolysaccharide (LPS) and its correlation with lung inflammation. Methods Twelve C57BL/6 mice were randomly divided into two groups, namely control group and ALI group, with 6 mice in each group. LPS (2 mg/kg) was intra-tracheal instilled in mice to reproduce ALI model, while the control mice received intra-tracheal administration of phosphate buffer saline (PBS) instead. The mice were sacrificed 6 hours after model reproduction, lung tissues were collected, and lung coefficient was calculated (lung wet weight/body weight ×100%). The pathological changes were examined under optical microscope after hematoxylin and eosin (HE) straining, the severity of lung injury was evaluated by the Smith score. Interleukin-6 (IL-6) level was determined by enzyme-linked immunosorbent assay (ELISA). After single-cell suspensions were isolated from the lung and spleen, the level of CD80 on DC in peripheral blood, lung and spleen was assessed by flow cytometry (FCM). The correlation between CD80 positive DC number and the severity of lung injury was analyzed by Spearman correlation method. Results Compared with control group, LPS-induced ALI resulted in a significant increase in lung coefficient [(0.67±0.04)% vs. (0.52±0.02)%, P < 0.05], and the levels of IL-6 were significantly enhanced (pg/mg: 2 712±475 vs. 335±168, P < 0.05). Histologically, widespread alveolar wall thickening caused by congested, severe hemorrhage in the interstitium and alveolus, and marked and diffuse interstitial infiltration with inflammatory cells were observed in ALI group. In contrast, no histological alteration was observed in the control group. In addition, the lung injury scores were markedly higher in ALI group than those in the control group (5.10±0.24 vs. 0.60±0.12, P < 0.05). It was shown by FCM analysis that there was a gradual but significant increase trend of CD80 positive dendritic cells number in blood, lung, and spleen in both control mice and ALI mice. However, no significant differences in the number of peripheral blood CD80 positive DCs were found between control group and ALI group [(3.3±1.5)% vs. (5.1±2.1)%, P > 0.05]. In contrast, there were a low but significantly higher percentage of CD80 positive DCs in the lung and spleen in ALI group than that in control group [Lung: (9.6±2.5)% vs. (3.6±1.2)%, spleen: (25.2±4.7)% vs. (9.0±3.6)%, both P < 0.05]. It was shown by the Spearman correlation analyses that respiratory CD80 positive DCs number was significantly positively correlated with IL-6 level in the lung (r = 0.761, P = 0.042) and the Smith score (r = 0.752, P = 0.047). Conclusions There is a significantly higher percentage of CD80 positive DCs in the lung and spleen in ALI mice, and a significantly positively correlation between respiratory CD80 positive DCs number and IL-6 level in the lung or the Smith score, which suggests elevated expression of CD80 on dendritic cells seems to play an important role in the pathogenesis of acute lung injury.
4.Effect of Wenyang Huoxue Lishul Recipe Containing Serum on Expression of Cathepsin L in Puromycin Aminonucleoside-induced Injury of Mouse Glomerular Podocytes.

OBJECTIVETo observe the effect of Wenyang Huoxue Lishui Recipe (WHLR) containing serum on the expression of cathepsin L (CatL) in puromycin aminonucleoside-induced injury of mouse glomerular podocytes.
METHODSMouse podocyte cells (MPCs) in vitro cultured were divided into the normal control group, the model group, the dexamethasone (DEX) group, 10% WHLR containing serum group, 20% WHLR containing serum group, the vehicle serum control group. MPCs in the normal control group were cultured at 37 degrees C culture solution for 24 h. 45 mg/L puromycin was acted on MPCs in the model group for 24 h. On the basis of puromycin intervention, 1 limol/L DEX was co-incubated in MPCs of the DEX group for 24 h; 10% or 20% WHLR containing serum was co-incubated in MPCs of the 10% WHLR containing serum group and 20% WHLR containing serum group for 24 h. The vehicle serum control group was also set up by incubating with WHLR containing serum alone for 24 h. The expression of CatL and its substrate Synaptopodin in podocytes were detected by cell immunofluorescence staining. FITC-conjugated phalloidin was used to stain F-actin. A cortical F-actin score index (CFS index) was designed to quantify the degree of cytoskeletal reorganization in cultured podocytes.
RESULTSCompared with the normal control group, the expression of synaptopodin significantly decreased and the expression of CatL significantly-increased in the model group. F-actin arranged in disorder, gradually forming pericellular F-actin ring. CFS index was obviously elevated (P < 0.01). Compared with the model group, the epression of synaptopodin increased, the expression of CatL decreased, and CFS index also decreased in the DEX group, 10% WHLR containing serum group, and 20% WHLR containing serum group (P < 0.05, P < 0.01). Compared with the DEX group, the expression of synaptopodin decreased in 10% WHLR containing serum group, CFS index also decreased in 20% WHLR containing serum group (P < 0.05).
CONCLUSIONSWHLR could up-regulate the expression of synaptopodin, down-regulate the expression of CatL, and alleviate cytoskeletal reorganization of F-actin. It was helpful to stabilize the cytoskeleton of F-actin and improve the merging of podocytes.

5.The early enteral nutrition in severe acute pancreatitis

Objectives: To evaluate the feasibility, safety and validity of the early enteral nutrition in severe acute pancreatitis(SAP). Methods: 15 patients of SAP during April 2002 and June 2003 had received early enteral nutrition through naso jejunal tube. The nutrition and immune index and the rates of complications were analyzed. Results: 2~3 days after nutrition tube placed to stomach, the tube heads in 11 cases reached the jejunum automatically, while 3 cases needed the help of X ray and 1 case needed the help of gastroscopy. All of 15 cases tolerated the enteral nutrition well, and there was no relapse of SAP. The nutrition and immune measurement were improved after 2 weeks' enteral nutrition, without infection of pancreatic necrosis. Conclusions: It is safe, efficient and feasible of the early enteral nutrition in severe acute pancreatitis(SAP) through naso jejunal tube. Early enteral nutrition can improve the nutrition, immune function and prognosis.
7.Bacteria Contamination on Hands of Medical Staff in Tianjin and Their Antibiotic Resistance

OBJECTIVE To investigate and analyze hand carrier state of medical staff in Tianjin and the antibiotic resistance of the pathogens.METHODS Bacteria or pathogens were isolated from the samples using a sterilized cotton swab on the whole hand before hand washing or hand disinfection,and drug sensitivity test was performed.RESULTS In 16.72% of all samples(50/299),there were 64 kinds of bacteria or pathogens isolated.A.baumannii,Staphylococcus aureus and A.lwoffii were the most common bacteria,40.6% resistant to more then 3 kinds of drugs.Four Meticillin-resistant S.aureus(MRSA) strains were detected out.CONCLUSIONS The high rate of hand carrying bacteria and resistance in Tianjin should pay attention to.
8.Advance in studies on long-circulating tumor targeted compound preparations.

The effect of the conventional drugs is very limited because of the intervention of multiple micro-environmental factors during the occurrence and progression of tumors. With the characteristics of high efficiency, low toxicity and multi-targets synergistic effect, the long-circulating tumor targeted compound preparations show its unique advantages in improving tumor microenvironment and enhancing the therapeutic effect of treatment, thus it has gradually become a hotspot of studies both at home and abroad. Through consulting a great number of professional literatures at home and abroad in recent years, the authors summarized the current studies in vitro and in vive on long-circulating tumor targeted compound preparations in different carriers, in the expectation of providing new ideas and methods for the development of long-circulating tumor targeted compound preparations.

9.Dynamic alteration of spleen dendritic cells in acute lung injury mice

Objective To investigate the kinetics and phenotype of spleen dendritic cells (DC) in lipopolysaccharide (LPS)-induced acute lung injury (ALI) mice.Methods Thirty-six C57BL/6 mice were randomly (random number) divided into two groups:control group and ALI group.Spleens were harvested at the following intervals of 6,12,and 24 h after LPS or PBS administration.Lung wet weight / body weight ratio (LW/BW) was recorded to assess lung injury.Meanwhile,pathological changes were examined under optical microscope.The IL-6 level in the lung was measured by using ELISA (enzymelinked immuno sorbent assay).The DC in the spleen was measured by flow cytometry (FCM).Results (1) LPS-ALI resulted in a significant increase in LW/BW ratio.(2) Histologically,extensive alveolar wall thickening resulted from edema,severe hemorrhage in the interstitium and alveolus,and marked and diffuse interstitial infiltration with inflammatory cells were observed in the ALI group.(3) Meanwhile,the levels of IL-6 in lung tissue were significantly increased in the LPS-induced ALI mice.(4) LPS-induced ALI led to divergent kinetics of spleen DC in ALI mice.In ALI mice,spleen DC only showed a transient increase at 12 h.(5) All DC within spleens had a modest maturation in ALI mice.Conclusions LPS-induced ALI provokes a transient increase as well as modest maturation of spleen DC.
10.Development of seismic trauma database version 1.0

Objective To develop a special seismic trauma database that could record and analyze the data including injuries, trauma cares and outcomes. Methods (1) The items and content of the database were determined based on the method of evidence-based medicine. (2)The fields, tables, items and options of the seismic trauma database were designed. (3) The database software was developed based on dot net framework platform, with C# as programming language. (4)The database was used to record and manage data of injuries caused by 5.12 earthquake. Results The Seismic Trauma Database Version 1.0 was developed with 728 fields (including 380 inner fields) and normative content and structure. The database software could record 10 aspects of the seismic patients including basic information, injuries, transfer and evacuation, clinical medical care, outcome and discharge. The database software had combination, custom queries and meta-analysis functions. Conclusions This database software can conveniently and normally record and manage the information of the seismic patients, can systemize and analyze the data conveniently and is an excellent data platform for trauma care research and epidemiologic study of seismic injuries.
