Objective To explore the influence of XTP4 on genomic expression profile of hepatocyte through screening and cloning of genes trans-regulated by XTP4-expressing plasmid. Methods The expressive vector pcDNA3.1(-)-XTP4 was constructed by routine molecular biological methods, and suppression subtractive hybridization (SSH) method was employed to detect the mRNA differentially expressed by the HepG2 cells transfected with pcDNA3.1(-)-XTP4 and pcDNA3.1(-), respectively, using lipofectamine. The twice enriched PCR products were subcloned into T/A vectors to set up the subtractive library. Amplification of the library was carried out in E. coli strain JM109. The screened cDNA were sequenced and analyzed in GenBank with Blast search after PCR. Results The amplified subtractive library containd 21 positive clones. Colony PCR analysis showed that there were 16 clones containing 200-1000bp inserts. Sequence analysis was performed and 9 kinds of encoding sequences were achieved. These genes trans-regulated by XTP4 protein involved in hepatic fibrogenesis, tumorgenesis, mitochondrial function, and cell growth regulation. Conclusions The findings obtained by SSH provide significant data for a preliminary understanding of the biological function of a new identified gene-XTP4. These results will pave the way for the study of the molecular mechanism of the transactivating effects of HBxAg and the development of new therapy for chronic hepatitis B.

OBJECTIVE:
To evaluate the vocal quality of patients with Reinke's edema before and after the endolaryngeal phonosurgery combined with micro-suture techniques.
METHOD:
Twenty male patients with bilateral Reinke's edema were submitted to surgery. All patients received the endolaryngeal phonosurgery combined with micro-suture techniques on vocal fold and completed voice evaluation preoperative and postoperative 2 weeks, 3 months and 6 months. Acoustic voice parameters were compared with the control group without voice pathology.
RESULT:
The result showed that at postoperative 2 weeks, compared with the preoperative evaluation the patients' subjective sound quality improved obviously. Acoustic voice parameters improved significantly (P<0.05). At postoperative 3 months, vocal form kept well, vocal cord vibration amplitude and mucous wave seemed normal. Acoustic voice parameters had no statistical difference compared with the control group and tended to reach normal values (P>0.05). At postoperative 6 months, voice quality still remain stable, all of the analyzed parameters had no obvious difference.
CONCLUSION
The phonosurgery combined with micro-suture techniques treat on the Reinke's edema, vocal cord postoperative recovery time is short, the sound quality receive basic recovery after 3 months.

Acta Pharmaceutica Sinica 2021;56(12):3431-3440
In order to solve the problems of erratic drug absorption and low bioavailability after oral administration for poorly-water soluble drugs due to low solubility, a series of novel pharmaceutical dosage forms as solid dispersion, liposome, microemulsion, vesicle, cyclodextrin inclusion complexes and drug nanocrystal have been developed in recent years. Among which drug nanocrystal attracts more attentions for its simpler preparation method, higher drug loading and easier manufacturing technology in the design of dosage forms suitable for different administration routes. In this paper, the nanocrystals of the poorly-water soluble drugs prepared based on bottom-up and top-down technologies were introduced. The characteristics and applications of the nanocrystal-based dosage forms as suspension, tablet and capsule were also introduced and carefully evaluated with the focus on their pharmacokinetics, pharmacodynamics and tissue targeted drug distribution after delivery by oral administration, intravenous injection and pulmonary inhalation. The advantages of drug nanocrystals in their therapeutics effects over the bulk drugs were discussed together with the inherent mechanism. Finally, the problems existing in basic research and scaled-up manufacture of drug nanocrystal as well as the possible ways of solution were listed out so as to make the nanocrystal-based preparations exert their maximum therapeutic effect after clinical application.

OBJECTIVETo study the relationship between Chinese medical syndrome types and metabolomics of non-small cell lung cancer (NSCLC) patients.
METHODSTotally 120 NSCLC patients were assigned to asthenia syndrome group and sthenia syndrome group, 60 in each group. Meanwhile, 60 cases of benign pulmonary nodules in physical examinations were recruited as the control group. Tumor tissues or benign pulmonary nodules tissues were obtained by thoracoscope. Changes of their metabolites were observed using gas chromatography-mass spectrometry (GC-MS). Their differences were studied using principal component analysis (PCA) and partial least-squares discriminant analysis (PLS-DA). ROC curve analysis was performed in different metabolic compounds of sthenia and asthenia syndromes groups. The area under the curve (AUC) was calculated to evaluate the sensitivity of diagnosing syndrome types.
RESULTSCompared with the control group, difference existed in 16 compounds. Of them , contents of citric acid, pyruvic acid, alanine, choline phosphate, glycerol phosphate choline, linoleic acid, oleic acid, lactic acid, inositol were more in the two tumors group than in the control group. Difference existed in 10 compounds between the sthenia syndrome group and the asthenia syndrome group. Of them, citric acid, pyruvic acid, alanine, choline phosphate, glycerol phosphate choline, lactic acid, and inositol were more in the asthenia syndrome group than in the sthenia syndrome group. Contents of valine, glucose, and glutamine were more in the sthenia syndrome group than in the asthenia syndrome group. ROC curve analyses of different compounds indicated that AUC of lactic acid and glucose was more than 0.8 (P < 0.01); AUC of inositol, choline phosphate, and glycerol phosphate choline was more than 0.7 (P < 0.01); AUC of valine, citric acid, glutamine, alanine, and pyruvic acid was more than 0.6 (P < 0.05).
CONCLUSIONSThere existed certain correlation between CM syndrome types and metabolomics of lung cancer. Lactic acid, glucose, inositol, choline phosphate, glycerol phosphate choline, valine, citric acid, glutamine, alanine, pyruvic acid were sensitive diagnostic compounds, and the first four kinds were most sensitive compounds.

Chinese Journal of General Surgery 2010;25(10):826-828
Objective To study the relationship between graft injury and expression of redox factor-1 in early period after liver transplantation in rats. Methods One hundred and fifty adult male Wistar rats were randomly divided into three groups: liver transplant group, sham surgery group and control group. Animals were sacrificed in each group at different time points: 3,6,9, 12,24 hour after liver transplantation. The changes and significance of the expression of Ref-1 were explored by immunohistochemistry, serology and histopathology. Results Compared with sham surgery group and control group, the expression of Ref-1 protein in transplant group was higher than that in other two groups in early period after liver transplantation ( P < 0. 05 ). In pathology there were lots of inflammation cells infiltration around the portal veins, and cell damage of hepatic tissue, while that in sham surgery and control group was comparatively slight. Serum ALT and AST values reached the peak at 6 ~ 12 h, and decreaced significantly after 12 h ( P < 0. 05 ). Conclusions Graft injury after liver transplantation during early period reaches peak at 6 hour and then decreased. Hepatic ischemic reperfusion injury promotes Ref-1 expression, which in turn compensates for programme cell death induced by the injury.

Objective To investigate the effects of pancreatic kininogenase on pressure, myocardical fibrosis and serum nitric oxide (NO) level in spontaneously hypertensive rats (SHR). Methods Twenty-four (fifteen weeks) male SHR were randomly divided into three groups:SHR group, pancreatic kininogenase group and captopril group(n=8 ),8 male Wistar kyoto rats with normal blood pressure were considered as control group. Pancreatic kininogenase was given by peritoneal injection (7.2 U?kg~ -1 ?d~ -1 ), captopril was given by intragastric administration(10 mg?kg~ -1 ?d~ -1 ), the rats in SHR group and rats with normol blood pressure in control group were treated with 0.9% NaCl(2 mL?kg~ -1 ?d~ -1 )administered through peritoneal injection. After four-week experiment, the pressure was measured in rats througth carotid artery, then the rats were sacrificed , and LVMI,CVF,PVCA and serum NO level were measured. Results In SHR group, SBP,LVMI,CVF and PVCA were higher, serum NO level was decreased obviously than those in control group (P0.05). Conclusion Pancreatic kininogenase can obviously reduce the blood pressure and reverse myocardial fibrosis, its mechanism may be concerned with the increasing of NO level in SHR.
