1.Effects of different doses of simvastatin on endothelial function in elderly patients with coronary artery disease

Objective To analyze the effect of 10 mg,20 mg simvastatin on vascular endothelial function in elderly patients with coronary heart disease. Methods 96 cases withcoronary heart disease admitted in the First people's Hospital of Xiaoshan District from Janary 2009 to December 2010 were selected and divided into three groups,each had 32 cases. Control group were given expanding coronary artery,blood,anticoagulation therapy.Observation group 1 and observation group 2 were separately taking 10 mg,20 mg simvastatin on the basis of control group. Endothelium-dependent vasodilation (FMD)functional,non-endothelium-dependent vasodilation (NMD)functions,nitric oxide (NO),and endothelin (ET)in three groups before and after treatment were detected and compared. Results Before treatment,the differences of FMD,NMD,NO,ET in three groups were not significant. After treatment,FMD were increased in all three groups,but the difference between two observation group was not statistically significant.NO was also increased in all three groups,but value in two observation group was higher than control group. NMD were unchanged in all three groups.Compared with before treatment,ET in two observation groups after treatment were decreased significantly (P<0.05 ). Conclusion simvastatin can effectively rectify the metabolic imbalances of vascular endothelial cells in patients with coronary heart disease and promote the improvement of FMD,but there is no significant difference between 10 mg~20 mg dose.
2.The impact of anastomotic level and preventive ileostomy on postoperative anastomotic fistula in rectal cancer patients

Objective To evaluatate the impact of different anastomotic location and preventive ileostomy on postoperative anastomotic fistula in rectal cancer patients.Methods The clinical data of 316 cases of rectal cancer patients after total mesorectal excision from January 2015 to January 2016 were analyzed retrospectively.Patients were divided into anterior resection (AR) group,low anterior resection (LAR) group,uhra-low anterior resection (ULAR) group according to the anastomotic location.There were 33 cases of anastomotic fistula with preventive ileostomy or without and anastomotic leakage varied from mild to severe as grade A,B,C.Results There was no statistically significant difference in the incidence of anastomotic fistula between the different anastomotic location (x2 =3.829,P =0.147).In anterior resection group and low anterior resection group,there was no statistically significant difference in the incidence of anastomotic fistula whether or not a preventive ileostomy was performed (x2 =0.606,1.096,P =0.436,0.326).While in uhralow anterior resection group,preventive ileostomy helped dicrease the incidence of anastomotic fistula (x2 =11.667,P =0.001),and once happened the severity of the anastomotic fistula tended to be less severe.Conclusion The incidence of anastomotic fistula does not vary with anastomotic level in our study.Preventive ileostomy reduces the incidence of anastomotic fistula in patients with ultralow anterior resection.
3.Study of State Speech Anxiety of Participants for College Speech Contest

Objective:To explore the psychological and behavioral pattern of state speech anxiety of participants of college speech contest under two conditions.Methods:Narrow band measurements of both Subjective Uncomfortable Degree(SUD)and objective behavioral assessments were used under two conditions.Results:Under lab condition,SUD rat-ings differed among four time bands,while the behavioral ratings failed to show any difference.Under field condition,for the prepared theme speech,SUD ratings differed among four time bands while they failed to show any difference for the improvisational speech.For both speech forms,several behavioral items displayed a pattern of habituation.Conclusion:Under both conditions,the psychological and behavioral pattern of participants' state speech anxiety display a pattern of habituation while the behavioral pattern is affected more by different conditions.
4.Indoleamine 2,3-dioxygenase and forkhead boxP3 on immune tolerance of gallbladder carcinoma cells

Objective To investigate the expressions of indoleamine 2,3-dioxygenase (IDO) and transcription factor forkhead box P3(FOXP3)in gallbladder adenocarcinoma; and to evaluate the relationship between the expressions of IDO and FOXP3 protein,and clinicopathology and lymph node metastasis of gallbladder carcinoma.Methods The expressions of IDO and FOXP3 in 51 cases of primary gallbladder cancer,90 cases of chronic cholecystitis,and 20 cases of normal gallbladder tissues were studied by immunohistochemical staining. Results The positive expression rates of IDO and FOXP3 in gallbladder carcinoma were 72.5% (37/51) and 60.8% (31/51),in normal gallbladder mucosa were 5% (1/20) and 20.0% (4/20),in cholecystitis and gallstone were 11.1% (10/90) and 32.2% (29/90),respectively.There were significant differences among the three groups in IDO and FOXP3 expressions (P<0.01).The positive expression rates of IDO and FOXP3 were 7.1% and 14.3% for simple hyperplasia,17.7% and 35.3% for atypical hyperplasia,40% and 35% for gallbladder carcinoma (stages Ⅰ-Ⅲ),and 77.4% and 64.5% for gallbladder carcinoma (stages Ⅳ-Ⅴ ).There were significant differences among the four groups in IDO and FOXP3 expressions (P<0.01).There were significant differences among the Nevin stage groups,lymph node metastasis group and gallbladder stone in IDO and FOXP3 expressions.However,there were no significant differences among the sex groups and the age groups in IDO and FOXP3 expressions (P>0.05).In gallbladder carcinoma,the expression of IDO had a positive correlation with FOXP3 expression in lymphocyte (γ=0.406,P=0.003).Conclusion In gallbladder cancer,a high-expression of IDO and an expression of FOXP3 in lymphocyte are closely related with immune tolerance of gallbladder carcinoma.The results provide a reference for clinical use of immunotherapy in gallbladder cancer.
5.Clinical Analysis of Extra-pulmonary Complications in 118 Cases with Mycoplasm Pneumoniae Infection

Objective To analyze the clinical features,curative effect and prognosis of extra-pulmonary infections by mycoplasma pneumoniae(MP).Methods ELISA was used to detect anti-MP IgM and IgG.Case histories of 226 patients,whose nasopharyneal lotione and respiratory secretions were positive of virus and bacteria respectively,was analysed retrospectively.Results One hundred and eighteen of the 226 (52.2%) cases suffered from extra-pulmonary infection.Of these infections 38.98%,33.89%,21.11% and 17.79% were found in digestive, urinary, cardiovascular systems and serous membrane respectively.All of the cases were improved after treatment with macrolides antibiotics.All cases were MP IgM positive,35.5% cases were IgG positive.Spatum MP positive rate was 32.2%.Positive rate of cold-agglutination test was 47.46%.Conclusions MP infection may cause many extra-pulmonary complications.When multi-organ infections can not be explained with bacterial and viral infections,MP infection should be considered.
6.Evaluation of different methods in eliminating Mycoplasma contamination in cell culture of human hepatoma-derived cell line C3A

Objective To evaluate the effectiveness of several methods in eliminating Mycoplasma contamination in cell culture of human hepatoma-derived cell line C3A. Methods PCR was performed to detect the Mycoplasmas contamination in cell cultures. The contaminated samples were treated by ciprofloxa-cin, heating, Plasmocure or co-culturing with macrophages. Transmission electron microscope ( TEM) and Q-PCR were used to comparatively analyze the cell morphology and gene expression before and after Plas-mocure treatment. Results Plasmocure succeeded in eliminating Mycoplasma contamination, while cipro-floxacin showed temporary efficacy. Heating and co-culturing with macrophages failed to eliminate Mycoplas-ma contamination. No Mycoplasma contamination in the Plasmocure-treated group was observed under TEM and the expression of ALB, TF and CYP3A4 genes were higher than the genes expressed in the contaminated group (P<0. 01). Conclusion Plasmocure treatment was effective in eliminating Mycoplasma contamina-tion in cell culture. Moreover, the cell morphology and gene expression in Plasmocure-treated group were re-stored to normal.

In the field of artificial muscle(AM) research,it is very difficult for humans to make the biological muscles,but there are experiments trying to put the function of muscles into practice of technology.With the development of materials and models of mathematics theories,people have made the AM of different materials and models true,however,as far as the characteristics of them and the widely used are concerned,they need deeply scientific research.And,in the near future,AM will play an important role in medical equipments,rehabilitation medicine,bionics,intelligent robots,military weapons,aerospace,etc.The authors review the types,characteristics,application in rehabilitation,as well as the function evaluation of AM.

Methods:
A total of 10 specific pathogen free (SPF) grade female DBA/2J mice were randomly divided into model group and QGA II group (n = 5 for each group), while additional 5 SPF-grade female C57BL/6J mice were assigned to control group. Mice presented spontaneous high IOP and showed elevated approximately at the age of seven months. The high IOP was maintained until week 38, when gavage was initiated. Mice in control group underwent the same intragastric treatment, while those in QGA II group were gavaged with QGA II (9.67 g/kg), once a day for four weeks. Retinal morphology was examined using hematoxylin and eosin (HE) staining, with the number of retinal ganglion cells (RGCs) counted. The expression level of Brn3a protein, a specific marker for RGCs, was detected by immunofluorescence, with the mean optical density (OD) measured for quantitative analysis. In addition, 16S rDNA sequencing was leveraged to analyze changes in the diversity of gut microbiota, including their α-diversity (Chao1, Shannon, Pielou’s evenness, and observed species index) and β-diversity. Venn diagrams and linear discriminant analysis effect size (LEfSe) analysis was employed to investigate the number of amplicon sequence variants (ASVs), the abundance of differential gut microbiota species, and the classification of species at both the phylum and genus levels within the three groups of mice.
Results:
HE staining revealed that compared with control group, model group showed significant reduction in the number of RGCs (P < 0.01), with intracellular vacuolar degeneration and nuclear pyknosis. After QGA II treatment, the number of RGCs was significantly increased compared with model group (P < 0.01), with notable improvements in intracellular vacuolar degeneration. Immunofluorescence analysis showed that the mean OD of Brn3a protein was significantly decreased in model group compared with control group (P < 0.01), while QGA II treatment significantly elevated its expression level (P < 0.01). Analysis of α-diversity showed that after QGA II intervention, the Chao1, Shannon, and Pielou’s evenness indices were significantly increased (P < 0.01), and the observed species index was elevated (P < 0.05). β-Diversity analysis demonstrated distinct clustering among the three groups, indicating relatively low similarity in bacterial community structures. ASV clustering identified a total of 14 061 ASVs across all groups, with 9 514 ASVs shared between model and QGA II groups. At the phylum level, the abundance of Bacteroidetes was significantly decreased in model group compared with control group (P < 0.01), while Firmicutes and the Firmicutes/Bacteroidetes (F/B) ratio were significantly increased (P < 0.01). QGA II treatment significantly reduced both Firmicutes abundance and the F/B ratio (P < 0.01). At the genus level, Lactobacillus was dominant across all groups, with its abundance significantly increased in model group (P < 0.01) and subsequently decreased following QGA II intervention (P < 0.05).
Conclusion
QGA II restructured the gut microbiota of DBA/2J mice with chronic high IOP, bringing changes in their diversity and abundance of components. Firmicutes, Bacteroidetes, Lactobacillus, along with their associated microorganisms, are likely critical components of the gut microbiota that contribute to the optic neuroprotective effects of QGA II on chronic high IOP mice.

10.C-arm X-ray machine displacement measurement of acetabular anteversion angle and abduction angle during total hip arthroplasty

BACKGROUND:In total hip arthroplasty, the accurate placement of the acetabulum is needed to guarantee the survival rate of the prosthesis and improve the prognosis. In order to ensure the accurate placement of the acetabulum, the accurate measurement of the abduction angle and the anteversion angle of the acetabulum is needed. OBJECTIVE:To investigate the application value of C-arm X-ray displacement measurement of acetabular anteversion angle and abduction angle in total hip arthroplasty. METHODS:Total y 63 cases undergoing total hip arthroplasty were divided into two groups according to their wil . Patients in the control group (n=30) were implanted with traditional acetabular prosthesis locator. Patients in the observation group (n=33) were implanted with acetabular prosthesis after C-arm X-ray displacement measurement. Acetabular anteversion angle, abduction angle and pelvic inclination were measured before, during and after arthroplasty. Acetabular anteversion angle and abduction angle were measured in both groups after arthroplasty. Pain score and hip function Harris score were recorded in both groups at different time points. RESULTS AND CONCLUSION:(1) No significant difference in acetabular anteversion angle, abduction angle and pelvic inclination was detected before, during and after arthroplasty (al P>0.05). (2) No significant difference in abduction angle was determined between the two groups after arthroplasty (P>0.05), but acetabular anteversion angle was significantly smal er in the observation group than in the control group (P<0.05). (3) At 7 days after arthroplasty, Visual Analog Scale scores were less in the observation group than in the control group (P<0.05). (4) Compared with that before arthroplasty, Harris score was significantly higher after arthroplasty in both groups (P<0.05). Harris score was higher in the observation group than in the control group at 3 and 12 months after arthroplasty (P<0.05). (5) Results indicated that during total hip arthroplasty, C-arm X-ray machine can measure acetabular anteversion angle and abduction angle, and effectively correct pelvic inclination, internal and external rotation, abduction and adduction, improve the accuracy of acetabular cup placement and the quality of replacement surgery, and improve the prognosis of the patients.
