1.Real time contrast-enhanced ultrasound in diagnosis of liver localized fatty deletion and fatty infiltration

Objective To explore the clinical value of real time contrast-enhanced ultrasound in diagnosis of liver localized fatty deletion and fatty infiltration which couldn't be confirmed by conventional ultrasonography. Methods Contrast-enhanced ultrasound was performed on 94 patients with liver localized diseases confirmed by baseline sonography, among them 34 patients with liver localized fatty deletion and fatty infiltration were enrolled. Results Contrast-enhanced ultrasonography didn't show apparent occupancy nidus in 34 patients. Enhancement mode of lesions was the same as liver parenchyma, which was distinctly different from liver occupancy diseases. Conclusion Real time contrast-enhanced ultrasound can display typical signs of liver localized fatty deletion and fatty infiltration and has significant value in diagnosis and differentiating diagnosis of them.
3.THE EFFECT OF FERMENTATION ON CELLULOLYTIC ENZYME ACTIVITY

Four cellulolytic strains, which can be used as feed additive, were studied under the conditions of various temperature, incubation time, and anaerobic process, and examined the changes of their cell protein content, cellulase and hemi-cellulase activity. The results show: 1) The maximum cellulolytic enzyme activities were observed incubation 20h; 2) Constant medium temperature 28℃ was adequate to the growth of the 4 strains ; 3) anaerobic condition, 39℃±2℃ and fermentation 12h, 24h, 36h, the tested strains can growth well in PDA plate, however, the cellulolytic enzyme activities and growth of the tested strains were influenced adversely when fermentation 48h. The experiment provide many important basis for the strains production, storage and utilization.
4.Study on Relationship between Red Blood Cell Distribution Width Changes and Prognosis of Acute ;Myocardial Infarction

Objective To explore the value of? red blood cell distribution width (RDW) changes in acute myocardial infarction (AMI) prognosis by analysing relationship between red blood cell distribution width, cardiac troponin I (cTnI) and high-sensitivity C-reactive protein. Methods 140 hospitalized patients with AMI were selected from May 2014 to October 2014 and devided into 2 groups. 70 patients without heart failure were AMI1 group and the other 70 patients with heart failure were AMI2 group. 70 cases of healthy persons were selected as control group. RDW was compared between three groups and the relationship between the level of RDW, cTnI and hs-CRP was analyzed. Evaluating value of RDW in predicting heart failure in AMI patients by using univariate logistic regression analysis and ROC curve analysis. Results In the comparison of AMI1 group, AMI2 group and control group, either of RDW, cTnI and hs-CRP showed a trend of increasing (P < 0.01). In the AMI patients , RDW level and cTnI level were positive correlation , and there was no linear correlation between RDW level and hs-CRP level. RDW≥14.55% was the independent risk factors in predicting heart failure in patients with AMI. Conclusion RDW≥14.55% can be an available independent predictor of the prognosis of AMI.
5.Implication of expression of Nanog in prostate cancer cells and their stem cells.

Recent studies suggested that the prostate cancer may arise from prostate cancer stem cells that share some same characteristics with normal stem cells. The purpose of this study was to detect the differences of Nanog expression between PC3 prostate cancer cell line and its tumor stem cells, and the relationship was preliminarily examined between Nanog and prostate cancer and its tumor stem cells. By using magnetic active cell sorting (MACS), we isolated a population of CD44(+)/CD133(+) prostate cancer cells that display stem cell characteristics from PC3 cell line. Immunohistochemistry revealed positive expressions of CD44, CD133 and α(2)β(1)-integin in the isolated cells. CCK-8 analysis showed that isolated cells had a strong proliferative ability. The formation of the cell spheres in serum-free medium and holoclones in serum-supplied medium showed that the cells were capable of self-renewing, indicating that the isolated cells were a population of cancer stem-like cells derived from PC3 cell line. Western blotting exhibited that the isolated cells had higher experession of Nanog, an embryonic stem marker, as compared with PC3 cells. Our study showed that Nanog might be helpful in sustaining the self-renewal and the undifferentiation of prostate cancer stem cells, and may serve as a marker for prostate cancer stem cells for isolation and identification.
6.MR imaging during arterial portography: an experimental study

Objective To evaluate the optimization of perfusing artery and contrast agent dose, and to determine the effect of precontrast injection of papaverine into the perfusing artery or not on the degree of contrast enhancement of the hepatic parenchyma during MRAP in dog models. Methods Sixteen dogs were included in this study and distributed to three study groups (perfusing artery, dose of Gd-DTPA and use of papaverine or not). Animals were anesthetized with Thiopental Sodium. After placement of a 5F end hole angiography catheter with no metal parts into the perfusing artery, the dogs were transferred immediately to the MR imaging unit. Unenhanced GRE MR and MRAP images were performed with a 2.0 T superconducting magnet. Signal intensity of liver parenchyma was measured with 6 different regions of interest. Relative value (%) of liver parenchymal enhancement was measured and compared. Time-enhancement curves were created and compared. Results MRAP images with good quality were obtained in all 16 dogs. The peak relative value of liver parenchymal enhancement and the time to the peak enhancement was similar in the superior mesenteric artery (SMA) group and in the splenic artery (SA) group. The peak relative value of liver parenchymal enhancement was the lowest in the 0.025 mmol/kg group among all groups. There was no statistical difference in the time to relative peak liver enhancement across the four groups. The time to relative peak liver enhancement in the group with papaverine injection was statistically earlier than that in the group without papaverine injection. Conclusion MRAP is a new and feasible imaging technique to detect hepatic masses before operation. When a 20 ml mixed solution of Gd-DTPA(0.050 -0.100 mmol/kg)was injected into perfusing artery at the rate of 1 ml/s, MRAP image of best liver parenchymal enhancement in dogs could be obtained at 21-28 s after the start of injection of Gd-DTPA. We can place catheters in SMA or SA and there was no statistical difference on MRAP images and time-enhancement curves. The dose of 0.050-0.100 mmol/kg Gd-DTPA was sufficient for MRAP. Application of papaverine precontrast advanced the time to peak liver enhancement, but the peak value of liver enhancement was not affected.
7.Analysis on correlation between quantitative results and semi-quantitative scores from ultrasound elastography and distribution of myofibroblasts in breast tumor

Objective To investigate the correlation between the quantitative results,the semi-quantitative scores from ultrasound elastography and the distribution of myofibroblasts (MFS)in breast tumor,and to analyze the value of quantitative results and the semi-quantitative score from ultrasound elastography in the diagnosis of breast tumor.Methods Thirty eight patients with breast lesions underwent ultrasound elasticity examinations,tissue dispersion quantitative analysis technique was used to assess the 11 characteristic quantities and the corresponding strain ratios in all lesions,and the score of ultrasonic elastography was evaluated.38 cases were divided into benign and malignant group according to pathological diagnosis results.The expression levels of CD34 andα-SMA protein in breast tissue were examined by immunohistochemistry.The distribution patterns of MFS in breast tumor were analyzed. The correlation between the quantitative results, the semi-quantitative scores from ultrasound elastography and the distribution of MFS in breast tumor was studied.Results The expression level of CD34 in malignant group was significantly higher than that in benign group, while the expression level of α-SMA was significantly higher than that in benign group;the differences in the expression levels of CD34 andα-SMA between benign and malignant breast tumor patients were statistically significant (P < 0.01 ).The masses observed in malignant group by elastography were shown in blue,while most of the masses in benign group were shown in green.There were statistically significant differences between two groups in elastography scores (P < 0.05 ). There was a anegative correlation between the CD34 expression in tumor tissue and the score of ultrasound elastography (r=-0.423 7,P =0.027 3).There was a negative correlation between the CD34 expression and the score of ultrasound elastography (r=-0.423 7,P =0.027 3),while the positive correlation was found between theα-SMA expression and the score of ultrasound elastography (r=0.397 0,P =0.014 2).The CD34 expression was significantly correlated with the average relative strain value,entropy,area ratio of low-strain region,kurtosis, skewness and inverse difference moment (P <0.05).Among the 11 characteristics,CD34 was positively correlated with the average relative strain value (r=0.385 6,P =0.016 8)and entropy (r=0.380 5,P =0.018 5);CD34 was negatively correlated with area ratio of low-strain region (r = - 0.351 7,P = 0.030 4),kurtosis (r =-0.427 7,P =0.007 4),skewness (r=-0.394 6,P =0.014 2),inverse difference moment (r = -0.344 3, P =0.034 3),angular second moment (r = - 0.484 9,P = 0.002 0)and strain ration (r = - 0.379 0,P =0.047 5);CD34 was not correlated with standard deviation,complexity and contrast (P > 0.05).The α-SMA espression was positively correlated with kurtosis (r = 0.356 9,P = 0.027 8),skewness (r = 0.323 0,P =0.047 9),area ratio of low-strain region (r=0.382 0,P =0.021 6)and strain ratio (r=0.403 3,P =0.012 0). Conclusion The features and its scores of ultrasound elastography are correlated with the distribution of MFS in breast tumor,suggesting that the ultrasound elastography is very informative and helpful in the diagnosis of breast tumor.
8.The effects of rehabilitation training on cardiac function in cerebral infarct patients

Objective To study the effects of rehabilitation(RT)on cardiac function in cerebral infarct (CIF)patients with cardiac insufficiency(CIS).Methods Fifty-nine CIF patients with CIS were randomly divid- ed into a treatment group(T group,n=29)and a control group(n=30),and all patients were treated with routine pharmacotherapy for 2 months.In addition,RT was administrated in the T group at the same time.The grading of the New York Heart Association(NYHA)and the changes in cardiac function associated index were observed in both groups before and after treatment.Results Compared with the control group,NYHA grades,left ventricle ejection fraction(LVEF),the levels of brain natriuretic peptide(BNP)in the blood plasma,and the 6min walking range of the T group patients were all significantly improved after treatment(P<0.05).Conclusion RT can improve car- diac function in CIF patients with CIS.
9.Index components contents in honeysuckle change trend at different time in a day and different stubbles in a year.

The index components contents of different time and different stubbles in honeysuckle were measured by HPLC, and were analysis by using the method of SPSS. Results showed that the content of index ingredients of different time had differences, and firstly decreased, then increased with time. The content of index ingredients of different stubbles had significantly differences, and firstly decreased, then increased with time. The chlorogenic acid contents were 2.059%-3.593%. The luteolosid contents were 0.110%-0.171%. Results indicated that the best picking buds time is before seven o'clock in the morning and evening at before and after seven o'clock, the index component content is higher. Picking buds in spring and at autumn index component content is higher; Picking buds in summer index component content is low. The experiment provides theoretical support for quality control in the whole process of the honeysuckle harvested and comprehensive utilization of honeysuckle.

10.The effect of fluorine on proliferation of osteoblast through extra cellular signal-regulated protein kinase signaling pathway

ObjectiveTo study the effect of fluorine on proliferation of osteoblast through extra cellular signal-regulated protein kinase(ERK) signaling pathway.MethodsMouse osteoblasts(MC3T3-E1) were cultured in vitro with different concentrations of fluoride for 24 and 48 h (the concentrations of Fˉ were 0,200,400,600,1000,2000,4000,8000,10 000 μmol/L,respectively).The optimum concentration for promotion of cell proliferation was determined by methylthiophene tetrazolium(MTT) assay.According to the optimum concentration,the cells were randomly divided into three groups:control group (0 μmol/L Fˉ); fluorine group (400 μmol/L Fˉ); fluorine and MAPK inhibitor PD98059 group(400 μ mol/L Fˉ + 10 μ mmol/L PD98059).Cell cycle was detected by flow cytometry after 48 h culture.The expression of P-ERK protein was determined by Western blotting and immunofluorescence.ResultsThe optimum concentration of fluorine for proliferation of osteoblasts was 400 μ mol/L.Compared with the control group[(76.12 ± 10.08)%,(2.06 ± 0.31)%],the number of cells in G0/G1 phase[(63.04 ± 8.12)%] reduced and the number of cells in S phase[(9.13 ± 2.08)%] increased in fluorine group (all P < 0.05) ; but the number of cells in G0/G1 phase [(92.11 ± 9.01 ) %] in fluorine and mitogen-activated protein kinases (MAPK) inhibitor PD98059 group was significantly increased(P < 0.05 ).Western blotting results showed that:compared with the control group[(100.00 ± 0.00)%],the expression of P-ERK protein in fluorine group[(131.24 ± 13.88)%] was significantly higher(P < 0.05 ),but the expression of P-ERK protein in fluorine and MAPK inhibitor PD98059 group [(91.33 ± 9.68 )%] was not significantly changed(P > 0.05).The results of immunofluorescence were similar to that of Western blotting.ConclusionsFluorine at the concentration of 400 μmol/L can promote the proliferation of osteoblasts.ERK signaling pathway has played a key role in the proliferation of osteoblasts.
