OBJECTIVE: To study the effects of high-protein diet (HPD) and high-grade proteinuria in aggravating the progression of chronic renal failure (CRF) in rats. METHODS: CRF with high-grade proteinuria was induced by supplying HPD in five sixth nephrectomy rats, and the changes of serum creatinine (Scr), blood urea nitrogen (BUN), endothelin-1 (ET-1), thromboxane B(2) (TXB(2)) and 6-keto-prostaglandin F(l alpha) (6-Keto-PGF(l alpha)) were observed. At the same time, the content of malondialdehyde (MDA), and the activities of glutathione peroxidase (GSH-Px) and superoxide dismutase(SOD)were measured in blood and renal tissue of the rats. RESULT: HPD and high-grade proteinuria could accelerate the damage of kidney through increasing the levels of ET-1 and TXB(2), reducing the level of 6-Keto-PGF(l alpha), and attenuating the activities of SOD and GSH-Px. CONCLUSION: HPD can accelerate the damage of kidney through inducing the high-grade proteinuria in five sixth nephrectomy rats, influencing the expression of kidney vasoactive substance, and reducing the anti-oxidation.

Objective:To observe effects of Fuzheng Huoxue Recipe on blood growth hormone (GH) and insulin growth factor(IGF-1) in chronic renal failure(CRF) rats.Methods:Platt method was adopted to make 5/6 kidney excision CRF model rat,and the rats were divided into model group,Huangqi group,Enalapril group,Danshen group,Fuzheng Huoxue Recipe group and normal control group according to the blood creatinine level after modeling.Effects of Fuzheng Huoxue Recipe on GH and IGF-1 levels in CRF rats with large quantity of urinal protein were investigated.Results:GH and IGF-1 levels in the model control group were significantly higher than that in normal control group,and after treatment of Fuzheng Huoxue Recipe,blood GH and IGF-1 levels were significantly decreased.Conclusion:Fuzheng Huoxue Recipe can decrease blood GH and IGF-1 levels in CRF rats,and can delay advances of CRF.

To study the level of blood selenium(Se)and nitrogen monoxide(NO)of oral lichen planus patients.Tested the contents of blood selenium(Se)and nitrogen monoxide(NO)of oral lichen planus patients and healthy cases.The level of blood selenium(Se)of oral lichen planus patients was lower than that of the control group,the situation of nitrogen monoxide(NO)was opposite,which indicated that Se and oxyradical may play an important role in the mechanism of oral lichen planus.

Objective The purpose of this study was to determine the value of K-ras mutation in DNA extracted from the plasma as clinical indicator of colorectal cancer. Methods Point mutation at codon 12 of K-ras gene was assayed by polymerase chain reaction of sequence-specific primers in DNA extracted from the plasma and tumors from 32 patients with colorectal cancer. The mutation was further confirmed by dideoxy-mediated chain-termination method of DNA sequencing. Results Fourteen patients (44%) had a codon 12 K-ras mutation within their primary tumors and identical mutations were found in the plasma DNA of 13 patients (93%). Mutant DNA was not detected in the plasma specimens of 18 patients whose tumors tested negative for K-ras alterations or in 5 healthy control subjects. Conclusion Preliminary results suggest the detection of K-ras mutations in circulating DNA extracted from the plasma specimens may have some clinical uti- lity in the detection of colorectal cancer.

Objective To establish a high performance liquid chromatography (HPLC) method for the determination of geniposide in Diaojinyao powder. Methods The HPLC system consisted of the Fortis-C18 (4.6 mm × 250 mm, 5μm) column, the mobile phase consisted of HCN∶H2O (12∶88) , the flow rate was 1.0 ml/min, the column temperature was 30℃, The UV detector was set at 238 nm. Results The linear response range was 0.049~0.980mg/ml ( r=0.999 9). The average recovery of geniposide was 99.1%, and RSD was 1.66%. Conclusion The method is simple, rapid, accurate and repeatable. It can be applied in determination of Geniposide in Diaojinyao Powder.

Objective To explore the utilization of laparoscopy in the diagnosis and treatment of Meckel’s diverticulum in children. Methods A total of 16 children with Meckel’s diverticulum underwent laparoscopic procedures between June 1999 and February 2003. With the help of celioscopy, the focus of the intestines is detected and pulled out through the umbilical port. The wedge resection of diverticulum or enterectomy with anastomosis was then performed. Results The operative time was 75 ～ 150 min, with a mean of 92 min. The patients began to take food on the third postoperative day. No operative complications occurred. The postoperative hospital stay was 5～7 d, with a mean of 5.8 d. Follow-up for 4～44 months (mean, 19.4 months) in the 16 patients showed no intestinal adhesion. Conclusions Laparoscopy is safe and effective in the diagnosis and treatment of Meckel’s diverticulum.

Objective To explore clinic manifestations and laboratory investgation of virus encephalopathy.Methods The clinical course,cerebrospinal fluid(CSF),hepatic dysfunction,computerized tomography of 7 cases treated in our hospital from October 1999 to March 2005 were retrospectively reviewed.Results Seven cases of virus encephalopathy were typically associated with a suddent onset of high fever,severe convulsion,rapidly progressive coma,marked elevations of alanine aminotransferase(AST) and aspartate transaminase(ALT).Four cases died,3 cases had severe sequelae.Blood ammonemia was normal,brain CT scans revealed peripheral or basal nuclei low-density areas.Conclusion Children with a sudden onset of high fever,severe convulsions,rapidly progressive coma may have a poor prognosis.

OBJECTIVE:To establish a method for the determination of the dissolution of Qingyangshen dispersible tablets so as to provide method and parameters for quality control of this kind of tablets.METHODS:With water as dissolution medium in a rotating speed of 100 r?min-1,the dissolution of Qingyangshen dispersible tablets was determined by HPLC at a wavelength of 259 nm.RESULTS:Similar dissolution rates were obtained for 3 batches of Qingyangshen dispersible tablets,with accumulative dissolution rates of all samples at 45 minutes reached above 90%,and a good reproducibility was noted.CONCLUSION:The quality of Qingyangshen dispersible tablets is up to the standard stated in Chinese Pharmacopoeia(2005 Edition).
