Neuronal nicotinic acetylcholine receptors(nAChRs) are widely distributed in the central nervous system,and they participate in complex neuronal signal transduction and regulation in the brain.The high Ca~(2+) permeability of nAChRs,plays a pivotal role in shaping nAChRs-induced neuromodulatory effects.The nAChRs-mediated Ca~(2+) signals may regulate diverse neuronal activities such as the improvement of learning and memory,and are closely related to drug addiction or neuroprotection mechanism.

Objective To study the phenotype profiles of bone marrow micrometastatic tumor cells in patients with breast cancer. Methods Immunocytochemical staining was used to study the biologic markers of disseminated tumor cells, and U-PA activity of the plasma in the bone marrow was assayed in breast cancer patients.Results Judged by positive immunocytochemical staining of EMA and CK19, bone marrow micrometastasis was found in 30 out of 72 (41 67%) breast cancer patients. The primary tumor size (? 2=6 417, P=0 040) and p53 protein expression (? 2 =5 930, P=0 025) were significantly correlated with the incidence of micrometastasis. Disseminated tumor cells expressed low protein cyclinD1, p53,Ki-67, EGFR, and high protein p21. Tumor size and axillary lymph node status were found to be significantly correlated with the u-PA activity level. Conclusion Disseminated tumor cells in the bone marrow was in a status of low-cycling and low-proliferation.

Objective To study the correlationship among plasma lipoprotein associated phospholipase A2 (Lp-PLA2),hypersensitive C reactive protein (hs-CRP) and D-dimer levels of coronary heart disease (CHD) patients.To evaluate their relationship and prognostic values.Methods Case control study.This study enrolled 150 CHD patients,130 age and gender compared health control.The clinical data were recorded and plasma Lp-PLA2,hs-CRP and D-dimer were test.Lp-PLA2 was test by enzyme kinetics method,hs-CRP and D-dimer were test by immunonephelometry method.The blood were also test low density lipoprotein cholesterol and myocardial injury markers such as troponin I,myoglobin,creatin kinase MB.The numbers of lesion coronary arteries were confirmed by percutaneous coronary intervention operation.Student t-test and mann-whitney U test were used to compare two groups datas,analysis of variance was used to compare more than three groups datas,spearman test was used for correlationship analysis between groups.Results The Lp-PLA2,hs-CRP and Ddimer of CHD group were (43.62 ±13.23) g/L,5.09 mg/L(95％ CI:0.56-74.15),173.0 mg/L(95％ CI:103.0-966.5),respectively.They were higher than healthy control group whose values were (21.49 ± 7.63)g/L,2.61 mg/L(95 ％ CI:0.37-45.12),68.0 mg/L (95％ CI:48.0-540.6),respectively.The difference were statistical significant (t =2.69,U =67.52,U =37.84,P ＜ 0.001).After correlation with clinical data,the Lp-PLA2 was correlated to LDL(r =0.487,P ＜ 0.001) and the numbers of coronary artery lesion (r =0.260,P =0.018).D-dimer was correlated to hs-CRP(r =0.263,P =0.011),it was also correlated to NYHA heart function level (F =23.78,P =0.001).Conclusions Lp-PLA2 is important in lipid metabolize,especially in LDL metabolize.Lp-PLAz can be used to predict severity of coronary artery lesion.Hs-CRP correlated to D-dimer level and cardiac injury markers.It stated link between inflammation and thrombosis.D-dimer is increasing with NYHA level.Lp-PLA2,hs-CRP and D-dimer are correlated to coronary artery lesions,they are good prognostic indices for coronary heart disease patients.

Objective To analyze the levels and speciation of arsenic metabolites in urine of rats treated with sodium arsenite and sodium arsenate in order to investigate the different aspects of metabolism between sodium arsenite and sodium arsenate,thus to understand further the basic data about relationship between it's metabolism and mechanism of toxicity. Methods Seventy Wistar rats,weighting 80-120 g,were divided into 7 groups of 10 each,such as normal control group,high,middle and low sodium arsenite group and high,middle and low sodium arsenate group. After the animals were fed for one month,the urine was collected by metabolic cage in 12 hours. Applying the high efficiency liquid chromatography and hydride genesis atomic fluorescence spectroscopy (HPLC-HGAFS),the levels and speciation of arsenic metabolites were determined in urine of rats. Meanwhile,the recovery rate of dimethyl arsinic acid(DMA) would be determined to estimate the degree of accuracy of results. Results The levels of iAs~(3+),iAs~(5+) and DMA in middle sodium arsenite group[(121.66±1.26),(10.26±2.68),(200.91±0.56) μg/L]were higher than the high sodium arsenite group[(113.20±0.75),(5.16±1.32),(147.70±μ0.77)μg/L,all P < 0.05]and low sodium arsenite group[(79.35±2.12),(5.13±2.25),(56.35±1.23)μg/L,all P < 0.05]. The levels of iAs~(3+) and DMA in middle sodium arsenate group[(315.81±1.69),(245.12±1.18)μg/L]were higher than the high sodium arsenate group[(85.03±0.56),(110.34±1.04)μg/L,all P< 0.05]and low sodium arsenate group[(22.97±2.67),(15.75±2.15)μg/L,all P < 0.05]. Compared with sodium arsenate group,the levels of iAs~(3+) and DMA in high and low sodium arsenite group were higher(all P < 0.05) ; and the levels of iAs~(3+) and DMA in middle sodium arsenite group were lower(all P < 0.05). Meanwhile,the average urinary recovery rate of DMA of rats in different sodium arsenite group were 94.80%-102.70%,and the average urinary recovery rate of DMA of rats in different sodium arsenate group were 95.33%-108.40%. Conclusion The speciation and levels of arsenic are influenced by the external exposure dose,and some distinction appeared in the metabolism and metabolic path between sodium arsenite and sodium arsenate in urine in vivo.

Objective:To investigate the regulation mechanisms of the bone marrow mesenchymal stem cells on lymphocyte pro -liferation of type I diabetic rats .Methods:The rat bone marrow mesenchymal stem cells were isolated , cultured and identified and the effect on lymphocyte proliferation of type Ⅰdiabetic rat was observed by MTT assay , and analyze the CD 4 +CD25 +regulatory T cell ra-tio, cell cycle and apoptosis of type I diabetes rat by flow cytometric .Results:B and C groups was significantly lower than the absor-bance values of group A,the differences between the data were statistically significant (P<0.05), C group was significantly lower than group B absorbance values, the difference was significant (P<0.05);the CD4 +CD25 +regulatory T cells of B and C groups were sig-nificantly higher than group A, the differences of the data were statistically significant (P<0.05), the CD4 +CD25 +regulatory T cell ratio of C group significantly higher than that group B , the differences were statistically significant (P<0.05);the apoptosis levels of B and C groups were significantly higher than group A , the differences were statistically significant (P<0.05), the apoptosis levels of C group were significantly higher in group B , the differences were statistically significant (P<0.05).Conclusion:Bone marrow mesen-chymal stem cells can significantly inhibit lymphocyte proliferation of type Ⅰdiabetic rats, and it may regulate CD4 +CD25 +regulatory T cells, promote apoptosis, thereby affecting the immune function of T lymphocytes , and play its rejection.

Objective To investigate the changes of blood coagulation indices during the percutaneous cor-onary intervention(PCI), so as to provide evidence for anticoagulation therapy. Methods 34 acute coronary syn-drome patients were enrolled,whose blood sample was taken at baseline,1 h,4 h,24 h,and 48 h after PCI operation. Thrombin-antithrombin complex(TAT), vWF, protein C, antithrombin, fibrinogen, D-dimer were tested. The result was processed by statistical software. Results After PCI, TAT continued to rise, protein C and antithrombin drop down,the vWF and D-dimer rose with TAT,and fibrinogen transiently drop down. Conclusion The thrombin pro-duces more quickly after PCI,its time phrase is coordinated with vWF and D-dimer,at the same time the anticoagula-tion system drops down. High anticoagulation at least maintains within 48 h and then tends to be normal.

Objective To study the changes of serum protein spectrum in patients with systematic lupus erythematosus (SLE) in order to screen specific protein markers. Methods Serum samples from 72 patients with SLE and 85 age- and sex-matched controls were assessed using surface-enhanced laser desorp-tion/ionization time-of-flight mass spectrometry (SELDI-TOF-MS) with weak cation exchange (CM10) pro-rein chip. Forty samples from the patients and 50 control samples were randomly selected to serve as a pre-liminary training set; significantly different protein peaks were automatically chosen for the system training and development of a decision classification tree model. The validity of the model was then challenged with a blind test set (including another 32 samples from patients and 35 from human controls). Results A total of 73 effective protein peaks were detected within the mass/charge ratio (m/z) interval 2000 - 50000, among which, 15 protein peaks significantly differed between patients with SLE and controls (P < 0.01). Three pro-tein peaks with an m/z value of 4001, 6305 and 7356 were automatically chosen as a biomarker pattern in the training set that discriminated patients with SLE from controls with a sensitivity of 90.0% (36/40), speci-ficity of 92.0% (46/50) and accuracy rate of 91.1% (82/90). When the SELDI marker pattern was tested with the blinded test set, it yielded a sensitivity of 87.5% (28/32), specificity of 91.4% (32/35) and accuracy rate of 89.6% (60/67). Conclusions SELDI-TOF-MS protein chip could be used to screen serum protein for SLE, and the decision classification tree model with these biomarkers may favor the diagnosis of SLE.

Aim The protective effect of total salvianolic acids (Sal) on cerebral hypoxia inmice was studied. Methods Acute cerebral hypoxia was induced by sodium nitrite scand decapitation.The effect of Sal on acute cerebral hypoxia in mice and neuronalhypoxia injury induced by sodium dithionite in primary cultures were ob-served. Results Sal in the doses of 10, 20 mg?kg-1 iv protected mice against theacute cerebral hypoxia and inhibited the production of lipid peroxidation in brain tis-sue of mice caused by cerebral hypoxia. Sal in the doses of 1~10 ?g? L-1 reduced therate of cell death and the content of MDA and lowered LDH content in extra-cellularbathing media in oxygen deprived cortical cultures.Conclusions Sal protects miceagainst cerebral hypoxia by suppressing the generation of lipid peroxide.

Objective To investigate gene polymorphisms and distribution features of glutathione Stransferase Omega-1 (GSTO 1 ) gene in Ala 140Asp site in 16 Chinese populations.MethodsA total of 1369 samples were from the human genome project(HGP)-the establishment and preservation program of Chinese minority genetic resources.The phenotypes of Ala/Ala (C/C),Ala/Asp (C/A),and Asp/Asp (A/A) of GSTO1 Ala140Asp were determined by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP).With analysis of molecular variance(AMOVA),the genetic variation levels among nations and regions were analyzed by estimating the evolutionary distance of alleles or genotypes.ResultsOf the 1369 individuals analyzed,979 (71.51％) were carriers of the wild homozygous allele Ala/Ala(C/C),365 (26.66％) were heterozygotes Ala/Asp(C/A) and 25 (1.83％) were mutant homozygotes Asp/Asp(A/A),with an overall frequency of the GSTO 1 mutant allele A 15.16％ [ (365 +50)/( 1369 × 2)].AMOVA analysis showed that the difference was statistically significant(P ＜ 0.05) of genetic variations of GSTO1 gene Ala140Asp among the 14 ethnic groups,and was significant between the northem and southern populations (P ＜ 0.05 ).Conclusion In different regions and populations the GSTO1Ala140Asp mutant allele frequencies are different.
