1.Diagnosis and treatment of primery breast lymphoma

Objective:To explore the clinic and pathologic characteristics of primary breast lymphoma(PBL) for its diagnosis and treatment.Methods: Five cases of PBL that has been diagnosed during 1998-2005 were analyzed.Results: One patient died 1 month after her final diagnosis.Tow patients have survived 4 and 3 years,respectively,after radical operation plus chemotherapy,although inguinal lymphatic metastasis was found in the later one 2 years after her operation.Another two patients have been accepting chemotherapy for 3 months after their local resection.Conclusion: PBL is diagnosed basically by methods of pathology and immunohistochemistry,and treated mainly by chemotherapy combining with other therapies.
3.Survey of Relationship between Symptoms and Dietary Factors in 54 Outpatients with Irritable Bowel Syndrome

Background:Irritable bowel syndrome(IBS)is a commonly seen functional intestinal disorder. Dietary factors are important factors affecting the abdominal symptoms of IBS patients. Aims:To investigate the effect of dietary factors on abdominal symptoms of IBS and the application of dietary adjustment strategy in patients lived in Xian,Shaanxi Province and the surrounding areas. Methods:Fifty-four outpatients with abdominal complaints who met the Rome Ⅲ criteria of IBS from Feb. 2015 to Jul. 2015 at the Second Affiliated Hospital of Xian Jiaotong University were enrolled. Twenty healthy volunteers were served as controls. All subjects completed a questionnaire containing issues mainly on IBS symptoms, eating habits,dietary factors causing abdominal symptoms,and the patients’coping styles. Results:Compared with healthy controls,more rice and less spicy food,fatty food,raw/ cold food,acidic beverages and alcohol drinking were observed in diet of IBS patients(P all < 0. 05). Abdominal symptoms of IBS patients could be caused or exacerbated predominantly by cold food(55. 6% ),fatty food(55. 6% ),raw food(48. 1% ),meat(48. 1% )and spicy food (40. 7% ). More than half of the IBS patients(63. 0% ) adopted an elimination diet to minimize their abdominal symptoms. Spicy food(59. 3% ),raw food(48. 1% ),cold food(44. 4% ),fatty food(44. 4% )and meat(37. 0% ) were the most frequently eliminated classes of food. Conclusions:Dietary factors can affect the abdominal symptoms of IBS in patients lived in Xian,Shaanxi Province and the surrounding areas and elimination diet based on foods they perceive as problematic is widely used. The classes of food and eating habits causing abdominal symptoms are quite different between domestic and overseas IBS patients.
4.Effect of Traditional Chinese Drug Combined with Training of Musculus Quadriceps Fexoris on Knee Osteoarthritis

Objective To observe the effect of traditional Chinese drug combined with training of musculus quadriceps fexoris on knee osteoarthritis(KOA).Methods70 out-patient clinic KOA patients were divided randomly into the Chinese drug group and control group with 35 cases in each group.The Chinese drug group was treated with Shentongzhuyu medicinal broth PO bid;the control group was treated with Sulindac 0.2 g PO bid.Two groups were combined with the training of musculus quadriceps fexoris,having 10 times per course and 3 courses in total with a 3 days interval between two courses.The therapeutic effect was evaluated with footplate pressure gait analysis and modified JOA marks.ResultsAfter treatment,the effect of the Chinese drug group was superior to the control group(P<0.05),especially 12 weeks post treatment.There maximum weight loading,time integral and weight loading intergral of affected limb of the Chinese drug group significantly improved after treatment(P<0.01),but for the control group,only weight loading intergral improved(P<0.05).ConclusionTraditional Chinese drug combined with training of musculus quadriceps fexoris has better curative effect on the pain and functional disturbance of KOA.
5.Post-stroke dysphagia in chronic stage treated with magnetic-ball sticking therapy at the auricular points: a randomized controlled trial.

OBJECTIVETo assess the effectiveness and safety on post-stroke dysphagia in chronic stage treated with magnetic-ball sticking therapy at the auricular points.
METHODSNinety cases of post-apoplexy dysphagia in chronic stage were randomized into an auricular points group and an acupuncture group. In the auricular points group, the magnetic-ball sticking therapy was applied to subcortex (pizhixia, AT4), brainstem (naogan, AT(3,4i)), mouth (kou, CO1), cheek (mianjia, LO(5,6i)), tongue (she, LO2) and throat (yanhou, TG3) on one ear each time, and were changed on the other ear once every 3 days. In the acupucnture group, acupuncture was applied to Feng-chi (GB 20), Yifeng (TE 17), Shanglianquan (Extra), Jinjin (EX-HN 12), Yuye (EX-HN 13), Shuigou (GV 26) and Tongli (TH 5), etc. The needles were retained for 30 min in each treatment. The treatment was gi-yen once a day in the two groups and the treatment of 6 days made one session. There was 1 day at an interval among the sessions. Totally, 3 sessions of treatment were required. The video fluoroscopic swallowing study (VFSS) was performed for 4 kinds of food with different properties and shapes in each patient. The main indices were Rosenbek penetration-aspiration score, oral-retaining score and throat-retaining score. The efficacy, and the incidences of aspiration pneumonia and malnutrition were compared between the two groups. The nutrition indices were compared before and after treatment between the two groups, such as the skinfold thickness of triceps brachii muscle, serum albumin and peralbumin.
RESULTSIn 21 days of treatment, in the auricular points group, the 1 mL liquid loversol Rosenbek penetration-aspiration score (1.51 +/- 0.69), oral-retaining score (1.17 +/- 0.38) and throat-retaining score (1.30 +/- 0.66) were all lower than those (2.51 +/- 0.67, 1.63 +/- 0.72, 1.67 +/- 0.7) in the acupuncture group separately. The 10 mL liquid loversol Rosenbek penetration-aspiration score (2.27 +/- 0.65), oral-retaining score (1.60 +/- 0.50) and throat-retaining score (1.49 +/- 0.51) were all lower than those (4.19 +/- 0.73, 2.30 +/- 0.51, 2.41 +/- 0.50) in the acupuncture group separately. The 10 mL paste loversol Rosenbek penetration-aspiration score (1.68 +/- 0.81), oral-retaining score (1.11 +/- 0.31) and throat-retaining score (1.10 +/- 0.31) were all lower than those (3.91 +/- 0.68, 1.63 +/- 0.76, 1.60 +/- 0.76) in the acupuncture group separately. The 1/4 cake-form loversol Rosenbek penetration-aspiration score (2.60 +/- 0.65), oral-retaining score (1.40 +/- 0.50) and throat-retaining score (1.74 +/- 0.49) were all lower than those (4.14 +/- 1.10, 2.40 +/- 0.73, 2.30 +/- 0.83) in the acupuncture group separately. The incidence of aspiration pneumonia was 14.9% (7/47) in the auricular points group, which was lower than 55.0% (22/40) in the acupuncture group (P < 0.01). The incidence of malnutrition was 8. 5% (4/47) in the auricular points group, which was lower than 50.0% (20/40) in the acupuncture group (P < 0.01). In 21 days of treatment, the results of the skinfold thickness of triceps brachii muscle and serum albumin in the auricular points group were better than those in the acupuncture group (both P < 0.05).
CONCLUSIONThe magnetic-ball sticking therapy at auricular points achieves the definite efficacy on post-stoke dysphagia in chronic stage and decreases the incidences of aspiration pneumonia and malnutrition. The efficacy of this therapy is better than acupuncture.

6.Curcumin induces apoptosis by PTEN/PI3K/AKT pathway in EC109 cells.

OBJECTIVETo study the molecular mechanism of curcumin in human esophageal carcinoma cell line (EC109).
METHODSEC109 cells were cultivated in vitro. When 80%-90% confluence was reached, they were treated with curcumin in different concentrations (15-120 µmol/L). The effects on cell proliferation were examined by CCK-8 colorimetry. The ultrastructure of EC109 cells were detected with transmission electron microscope(TEM). The cells apoptosis was observed with laser confocal microscope(LCM) by AnnexinV-FITC/PI double staining. The proteins level of PTEN, AKT, GSK3β and Caspase 3 were tested by flow cytometry(FCM) .
RESULTSCCK-8 test showed that curcumin could inhibit the proliferation of EC109 cells in a time- and concentration-dependent manner. TEM and LCM examinations indicated that curcumin could make EC109 cells apoptosis. The data of FCM showed that curcumin could increase the expression of PTEN, GSK3β and Caspase 3, decreased the expression of AKT.
CONCLUSIONThe effects of curcumin on inhibiting proliferation and promoting apoptosis of EC109 cells were related with increased expression of PTEN and inhibition of PI3K/AKT signaling pathway.

7.Apoptosis in Jurkat cells induced by bortezomib combined with adriamycin.

OBJECTIVEThe aim of this study was to observe the suppressive effect of bortezomib alone and the synergistic suppressive effect of bortezomib and adriamycin on the proliferation of the cell line Jurkat cells, and to discuss the mechanism of apoptosis induced by bortezomib.
METHODSThe suppressive effect of bortezomib and adriamycin on the proliferation of Jurkat cells in vitro was detected by MTT colorimetry, and the morphology of the cells was examined by histology. The cell apoptosis was measured by flow cytometry with Annexin V/PI staining and cell cycle analysis. The effect of bortezomib and adriamycin on the expression levels of caspase-3, caspase-8 and PARP were measured by Nestern blot.
RESULTSThe proliferation of Jurkat cells was significantly inhibited by bortezomib treatment (between 10 - 320 ng/ml) for 24 h, 48 h and 72 h, and the growing inhibition ratio showed a positive correlation with the drug concentration (r(24 h) = 0.900, P < 0.01; r(48 h) = 0.849, P < 0.01; r(72 h) = 0.679, P < 0.01), in a concentration-dependent manner. The IC(50) of Jurkat cells treated with bortezomib in a dose of 10 - 320 ng/ml was 137.64 +/- 6.82 ng/ml, but the IC(50) of Jurkat cells treated with bortezomib combined with adriamycin (125 ng/ml) for 24 h was significantly decreased to 20.44 +/- 2.85 ng/ml. The apoptosis rate had a positive correlation with the concentration of bortezomib (P < 0.01). After the Jurkat cells were treated with bortezomib, apparent shear bands of caspase-8, caspase-3 and PARP proteins were observed.
CONCLUSIONThere is an effect of Bortezomib to induce apoptosis in Jurkat cells, and the extrinsic pathway is one of the apoptosis-inducing mechanisms. There is a synergistic suppressive effect of the combination of bortezomib and adriamycin on the proliferation of Jurkat cells and enhances their chemosensitivity.

8.Effects of hIFN-λ1 recombinant adenovirus on proliferation of gastric adenocarcinoma cell line and its mechanism

Objective To explore the effects of Ad-hIFN-λ1 recombinant adenovirus on prolifera-tion of gastric adenocarcinoma cell line SGC-7901 and its mechanism .Methods Ad-hIFN-λ1 recombinant adenovirus and empty plasmid Ad-LacZ were respeetively transfated to human gastric adenocarcinoma SGC-7901 cells.The proliferation of transfected cells was detected by MTT assay .IFN-λ1 gene expression at mR-NA and protein levels in the cells were measured by RT-PCR analysis and immunofluorescence assay , re-spectively .Tunel assay and flow cytometry were used to analyze the rate of cell apoptosis .Results The proliferation of gastric adenocarcinoma SGC-7901 cells were significantly inhibited with Ad-hIFN-λ1 inter-vention in accordance with highly expressed IFN-λ1 at mRNA and protein levels , respectively .The apoptosis rate of Ad-hIFN-λ1 transfected cells was markedly higher than that of the empty plasmid Ad-LacZ group and PBS blank control group .Conclusion The expression of hIFN-λ1 could be detected after transfection of Ad-hIFN-λ1 recombinant adenovirus into gastric adenocarcinoma SGC-7901 cells.Ad-hIFN-λ1 could signifi-cantly inhibit the proliferation of gastric adenocarcinoma SGC-7901 cells by promoting the apoptosis of cancer cells.
9.Hypertonic solution does not decrease mortality from traumatic hemorrhagic shock: a systematic review and Meta-analysis based on randomized control trials

Objective To evaluate systematically whether administration of hypertonic saline transfusion affects clinical outcomes with compared to standard fluid in the early stage of resuscitation for traumatic shock patients.Methods Seven English and Chinese routine biology and medicine databases were searched for randomized controlled trials (RCTs) published from January 2002 to August 2012,and established inclusion and exclusion criteria to evaluate these RCTs.The quality assessment was based on the Cochrane Handbook for Systematic Reviews of Interventions Version 5.0.1 and Jadad' s score scale.RevMan 5.0 statistical software was used for meta-analysis.Results After evaluated 211 related literatures,five RCTs met all the inclusion criteria and were enrolled for meta-analysis.The meta-a nalysis demonstrated that early hypertonic transfusion did not decrease short-term (first 48 hours after admission) mortality (RR =1.04,P =0.74); nor did it decrease later-term (7day to 3month after injury) mortality (RR =0.97,P =0.72).It also did not decrease the total volume of fluid and blood transfusion required during the first day (P =0.38).Similarly,it did not affect the incidents of infections (RR =1.04,P =0.70),the length of stay in ICU (P =0.2) and total length of stay in the hospital.Conclusions Compared to standard fluid,there was no advantage on mortality and hospital infection by using hypertonic supplement transfusion in the early stage of resuscitation for traumatic shock patients.Hypertonic transfusion did not have any significant effect on the volume of total fluid and blood transfusion required the first day,and no trend of reduction for the length of ICU and hospital stay.Further well-designed randomized controlled trials are needed to demonstrate the cost effectiveness of hypertonic transfusion to traumatic shock patients while in ICU.
10.Analysis of Myocilin gene regulatory network using a genetic genomics approach

Background The pathogenesis of primary open angle glaucoma(POAG) and high myopia are very complex.To construct the regulatory network of virulence genes and relevant genes that involved in pathogenicity are helpful for reveal of the pathogenesis.Objective The aim of this study was to investigate myocilin(Myoc),a gene that contributes to POAG and high myopia in eyes of BXD Recombinant Inbred(BXD RI)mice and construct the regulatory network of Myoc.Methods The affymetrix microarray system was used to detect the differential expression of Myoc in the eyes of C57BL/6J(B6),DBA/2J(D2) and BXD RI mice.Expression quantitative trait loci (eQTL) mapping was performed to construct the regulatory network of Myoc gene.Results The average expression level of the Myoc gene in the BXD strains was 10.83,and the gene exhibited expression levels ranging from 8.39 in BXD55 mice tol 1.43 in B6 mice.The eQTL mapping for the Myoc gene showed a significant likelihood ratio statistic (LRS) of 21.78.The QTL was mapped in chromosome 2,and Myoc was located on chromosome 1,indicating that the Myoc gene was a trans-acting QTL.Olfml2a was identified to be a candidate upstream gene of Myoc by analysis of bioinformatics.Genetic regulatory network analysis demonstrated that a series of genes associated with Myoc probably played roles in the pathogenesis and development of POAG and high myopia.Conclusions The genetical genomics approach provides a powerful tool for constructing pathways that contribute to complex traits,such as POAG and high myopia.
