A suitable ultra-high performance liquid chromatography coupled with tandem mass spectrometry (UPLC-MS/MS) method was developed for the determination of 11 mycotoxins with isotope internal standard in malt. The mycotoxins in malt were extracted and purified by one-step ultrasonic extraction procedure using acetonitrile/water/acetic acid (80 : 19 : 1), and then detected and confirmed by UPLC-MS/MS, and quantified by isotope labeled AFB1 ([13C17]-AFB1) and ZEN ([13C18]-ZEN) internal standards. Rapid separation of the 11 mycotoxins was successfully achieved on a Phenomenex Kinetex C18 column (100 mm x 2.1 mm, 2.6 μm) with gradient elution using the mobile phase of methanol containing 0.1% formic acid and 2 mmol x L(-1) ammonium acetate in water. Simultaneous acquisition was performed in multiple reaction monitoring (MRM) mode with electrospray ionization (ESI) source operated in both positive and negative ionization modes. The established method provided a good linearity for the 11 mycotoxins within their respective linear ranges with correlation coefficients all higher than 0.999 1. The average recoveries ranged from 75.0% to 117.0% with relative standard deviations (RSDs) below 5.1%. The limits of detection (LODs) and quantitation (LOQs) ranged from 0.05 to 30 μg x kg(-1) and 0.15 to 87.5 μg x kg(-1), respectively, which were below the maximum residue levels (MRLs) set by the European Union. Twenty malt samples were analyzed and nine samples were detected with mycotoxins, which were confirmed according to the same fragment ions found in positive samples and the standards at the same retention time. This study has demonstrated that the one-step extraction procedure of mycotoxins from complex matrices coupled to UPLC-MS/MS method is simple, quick, accurate and sensitive for quantitative and qualitative analysis of multiple mycotoxins in malt.

Objective To investigate effects of different doses of recombinant human bone morphogenetic protein-2 (rhBMP-2) on vascular endothelial growth factor (VEGF) expression in fetal mouse osteoblasts. Methods Calvaria osteoblasts of fetal mice of 19 days were cultured. The effects of rhBMP-2 at different doses and different action times on VEGF expression patterns in fetal mouse osteoblasts were observed with reverse transcrip- tion-polymerase chain reaction (RT-PCR) and Western blot staining. Results In the present study,RT-PCR detected a steady expression of VEGF mRNA in the control fetal mouse osteoblasts. The levels of VEGF mRNA increased in an apparent biphasic manner with a maximum stimulation (about 2-fold above the control,P<0.05 ) in both VEGF mRNA species observed at 300 ng/mL of rbBMP-2.After 48 h of rhBMP-2 treatment, the VEGF mRNA levels approached those in the control. The VEGF mRNA levels appeared to be biphasic in rhBMP-2-treated cultures, showing peak induction at 3 and 24 h and remaining elevated at 48 b.Compared with the individual control value at each time point,an apparent maximum increase (about 2.5-fold above the control,P<0.05) occurred at 6 h.The second peak induction, about 2-fold above that in the control, occurred at about 36 h.Conclusions The expression of VEGF mRNA is steady in the control fetal mouse osteoblasts.RhBMP-2 can promote the expression of VEGF in dose-dependent and time-dependent manners.

Objective To investigate the effect of percutaneous vertebroplasty and kyphoplasty (PKP) on bone density and serum leptin in patients with osteoporotic thoracolumbar compression fractures. Methods One hundred and two patients with osteoporotic thoracolumbar vertebral compression fractures and 100 cases of healthy control were treated with PKP surgery. The serum leptin and adiponectin, and the bone mineral density of the lumbar spine were determined. Results patients Levels of the serum leptin and adiponectin in patients with osteoporotic thoracolumbar vertebral compression fracture were significantly higher than those with non osteoporotic fracture patients (P < 0.05). One month post-PVP, the levels of serum leptin and adiponectin in the two groups were significantly decreased (P < 0.05). postoperative group of The lumbar bone density at 1 month post-operation was is significantly higher than that before operation in each group (P < 0.05, respectively), with no significant difference between the two groups. The levels of serum leptin and adiponectin were negatively correlated with the bone mineral density (BMD) (P < 0.05). Conclusions tLevels of serum adiponectin and leptin were both increased in osteoporotic or non bone osteoporosis thoracolumbar vertebral compressor fractures. The percutaneous vertebroplasty forming operation could effectively reduce the serum level of leptin and adiponectin, and improve the bone mineral density of the lumbar vertebrae.
