Objective To investigate the apoptosis of cultured normal human hepatocyte HL-7702 cells induced by glycodeoxycholate(GCDC)and to explore its possible mechanism. Methods HL-7702 cells were incubated with various concentrations(100,150,200 and 250 ?mol/L)of GCDC.The changes of cellular morphology were observed under optical microscope. The apoptosis rate of HL-7702 was determined by Annexin V-FITC/PI double staining. The changes of HL-7702 cell intracelluar \[Ca2+\]i were determined with Fluo-3/AM load technique. The mRNA expression levels of Bcl-2/Bax in HL-7702 cells were analyzed by RT-PCR.Results Typical apoptotic morphological changes were observed after HL-7702 cells had been treated with 150 ?mol/L GCDC for 24 h;HL-7702 cells could be induced to undergo apoptosis in a concentration-dependent manner after 100,150,200,and 250 ?mol/L GCDC treatment for 24 hours. The apoptosis rates were(13.16?2.9)%,(20.3?3.0)%,(25.02?2.1)% and(45.02?3.5)%,which were markedly higher(P

To study still further the activity of CNTF mutant designed by computer molecular modling, the methods of dissociated cultures of chick dorsal root ganglion、TF-1 prolification and the normal mice'weight loss tests weve used. The results indicated that the mutant protein promoted the survival of dorsal root ganglion、induced TF-1 prolification and made the normal mice lose weight, decrease appetite and reduce fat index. The weight loss effect was dependant with its administration dosage,ED50 was 150.986?g/kg/d.To TF-1,the specific activity reached 2.0?106U/mg against international reference reagent. In a word,CNTF mutant had excel bioactivity. So it provided clues for its development and application.

98 endophytic bacteria strains were isolated from different internal tissues of Glycyrrhiza uralensis plants collected from Innermongolia region. Results indicated that the population densities of endophytic bacteria ranged from 5.0?104cfu/g~2.9?107cfu/g fresh weight although it varied depending on tissue of the plant. Among these strains, Bacillus sp. was the most prevalent endophytic bacterium, which was amount to 30%.Of the 98 isolates, 6 strains exhibited extensive antagonistic activities against pathogenic bacteria. Characterization showed that these bacteria were Bacillus atrophaeus、Paenibacillus polymyxa、Bacillus subtilis、Paenibacillus ehimensis. This study indicated that selected 6 endophytic bacteria strains have potential for biological control of plant disease.

AIMTo study the protective role of endogenous carbon monoxide to lung and kidney tissues during septic shock and its mechanism.
METHODSA rat model of CLP was built by using the method of CLP. The malondialdehyde (MDA) content and the activity of superoxide dematase (SOD) in blood, lung and kidney were detected by immunohistochemical technique and light microscope.
RESULTSPathological changes of lung and kidney in CLP + Hemin group were lighter than CLP group, inflammatory reaction and lipid peroxidation were also lighter.
CONCLUSIONEndogenous CO can protect lung and kidney from the oxidative injury. It can suppress in flammation and the oxidative injury caused by activated inflammatory cells, it is probably an important mechanism of its protective effects.

The difference between doctor selection needs of the public and medical service at present was analyzed according to the questionnaire investigation of doctor selection habit of the public. The association between the focus on doctor personal information and the importance of different factors influencing doctor selection was analyzed ac-cording to the information searching methods used by the public when they selected their doctor. The effective pre-cision search approaches and methods on the Internet medical platform were proposed, including information searching methods on innovation platform, standardizing the weight of different kinds of information, and perfecting the pushing and recommending mechanisms for doctor selection searching results.

Objective To observe the expression of stromal cell-derived factor-1α(SDF-1α)and glial cell line-derived neurotrophic factor(GDNF)genes in bone marrow stromal cells(BMSCs)of rhesus monkey. Methods With gene recombination technique,SDF-1α and GDNF genes obtained from cDNA were subcloned into pBudce4.1 vector to get pBudCE4.1-SDF-1-GDNF, evaluated by restriction enzyme analysis and sequencing analysis. The pBudCE4.1-SDF-1α-GDNF was transfected into BMSCs with lipofectamine2000.After 48 h,the expression of SDF-1α and GDNF was measured by RT-PCIL,Western blotting and immunohistochemistry. Results Correct construction of pBudCE4.1-SDF-1α-GDNF was identified by enzyme restriction analysis and sequencing analysis.Western blotting and immunohistochemistry confirmed the expressions Of SDF-1α and GDNF genes in the transfected cells. The protein expressions of GDNF and SDF-1α in the GDNF and SDF-1α transfected BMSCs were 6 times higher than those in the negative control cells. Conclusion The pBudCE4.1-SDF-1α-GDNF can successfully express SDF-1α and GDNF in BMSCs of rhesus monkey, which may be used in gene therapy for Parkinson's disease.

BACKGROUNDPatient characteristics may be an internal factor influencing patient complaints, but in China patient characterization is restricted to patient satisfaction surveys, and few studies have considered the relationship between patient characteristics and patient complaints. The aim of this research was to determine the reasons for complaints.
METHODSIn this study, we analyzed the characteristics of hospitalized patients and explored their relationship with medical complaints.
RESULTSThe significant factors were age (P = 0.045), hospital cost (P = 0.003), household nature (P < 0.001), and education (P < 0.001). The complaint rate decreased when the patients' age increased (regression coefficient, -0.606; OR, 0.545, and 95%CI, 0.301 - 0.987). The complaint rate increased with an increase in hospital cost (regression coefficient, 0.818; OR, 2.266; and 95%CI, 1.320 - 3.889). Patients from non-agricultural households had a higher complaint rate (regression coefficient, 1.051; OR, 2.861; and 95%CI, 1.611 - 5.082). Patients with higher education levels had lower complaint rates (regression coefficient, -0.944; OR, 0.389; and 95%CI, 0.234 - 0.647).
CONCLUSIONThe survey confirms that older patients and patients with higher education levels had lower complaint rates, while non-agricultural population and patients with higher hospital expenses had higher complaint rates.

Objective:To better understand the pathogenesis of obstructive jaundice (O J),a variety of rat OJ and biliary drainage models have been tried;however, complications are still common. We aimed to establish a stable rat model of OJ using microsurgical techniques, and to assess its reversal by internal bile drainage(IBD).Methods:After the pilot study,we developed a standardized surgical procedure. All operations were carried out under an operating microscope. In the first laparotomy, the proximal common bile duct (CBD) of the rat was ligated and transected.A tube was introduced into the distal end of the duct, and the other end of the tube was sealed and fixed. In the second laparotomy, the drainage tube was inserted into the (by now markedly dilated) proximal CBD,and ligated into position. We evaluated the general condition of the rats, the status of the liver and pancreas before and after IBD.Results:Complications such as intestinal reflux and bile duct blockage, were not found.Pancreatic injury was not evident by day 4 after the first laparotomy. After biliary drainage, the serum glucose and albumin concentration rapidly returned to normal levels.Liver weight/body weight ratio increased. The biochemical indicators and ultrasonographic elastography results for the liver gradually returned to normal. Conclusion:Using microsurgical techniques,we have developed a stable rat model of OJ reversed by IBD.
