1.Association of Human Leukocyte Antigen Allele Polymorphism and Susceptibility to Children with Acute Lymphocytic Leukemia

Objective To explore the possible association between human leukocyte antigen(HLA) allele polymorphism and susceptibility to chlidren's acute lymphocytic leukemia(ALL).Method HLA alleles polymorphism in 38 cases with ALL and 35 healthy controls were determined by polymerase chain reaction(PCR) with sequence specific olignuleotide probe(PCR/SSO).Results The allele frequency of HLA-A01 significantly increased than control group(?2=4.947 P=0.026,RR=10.20),the frequence of A02 significantly decreased(?2=4.187 P=0.041,RR=3.13),the frequcnce of A33 significunt decreased than control group(?2=4.403 P=0.036,RR=0.21).Conclusion These results show that susceptibility to chlidren's ALL is positively related to HLA-A01,A33,especially A01 allele,while HLA-A33 to its genetic resistance.

3.Communication between clinicians and laboratorians in semen analysis.

By direct communication between clinicians and laboratorians, obstacles between clinical work and laboratory diagnosis were unequivocally found at an academic conference. Many clinical physicians feel disappointed and frustrated at the laboratory diagnosis by semen analysis and frankly express their expectations, as to recognize the role and significance of laboratory diagnosis, understand the value of routine semen analysis, work out the puzzles in the analysis of sperm morphology, fulfil the requirements for the identification of cells in semen, and obtain responses from and initiate discussions among laboratorians about the relevant questions. Both laboratorians and clinicians are appealing for strengthened management and earlier establishment of national standards and operating specifications for laboratory diagnosis by semen analysis.

4.Clinical research of canalicular intubation combined external dacryocystorhinostomy for chronic dacryocystitis and upper lacrimal duct stenosis

AlM: To evaluate the clinical efficacy of canalicular intubation combined with external dacryocystorhinostomy ( ext-DCR ) for treatment of chronic dacryocystitis and upper lacrimal duct stenosis.
METHODS:Thirty-three patients (33 eyes) with chronic dacryocystitis and upper lacrimal duct stenosis who underwent canalicular intubation combined ext-DCR were retrospective analyzed. The silicon tube was indwelt for 6mo. All cases were re-examined 1wk;1, 3, 6, 9mo post-operation, flow of tears, pus excretion and lacrimal duct clearance were observed.
RESULTS: Clear lacrimal duct ratio was 100% in all cases during 1wk~6mo post-operation; Silicon tube was removed 6mo post-operation, 3mo after tube removal, rechecking reported 32 cases with clear lacrimal duct (97%) and 1 case with resistant duct (3%). Two cases ( 6%) with minor lacerations, no other complications were observed.
CONCLUSlON: Canalicular intubation combined ext-DCR is an effective treatment for chronic dacryocystitis and upper lacrimal duct stenosis.
5.Polymorphism within the tumor necrosis factor receptor H ( TNFR Ⅱ ) in patients with ankylosing spondylitis in hunan province

Objective To investigate the association between ankylosing spondylitis (AS) in Han population from Hunan province of China and polymorphism located at the position nt587 in the tumor necrosis factor receptor Ⅱ ( TNFR Ⅱ ) gene.Methods Correlation analysis was performed in a case-control of 100 AS cases and 100 healthy control.The SNPs located in the TNFR Ⅱ gene ( TNFR Ⅱ nt587 ) was examined by polymerase chain reaction restriction fragment length polymorphism (PCR-RFLP).With the SPSS software,the relationship between AS and frequencies of genotype and alleles in TNFR Ⅱ nt587 were analyzed.Results There were 43 cases with TNFR Ⅱ nt587 T/T genotype,32 cases with TNFR Ⅱ nt587 T/G genotype and 25 cases with TNFR Ⅱ nt587 G/G genotype in the AS cases.While in the healthy control,there were 56 cases with TNFR Ⅱ nt587 T/T genotype,34 cases with TNFR Ⅱ nt587 T/G genotype and 10 cases with TNFR Ⅱ nt587 G/G genotype.At the same times,the allele frequencies of G in AS group was significantly higher ( x2 =8.734,P=0.003 ) than the control group (41.0% vs.27.0% ).The odds ratio in AS cases with TNFR Ⅱ nt587 G/G genotype was 3.256,which higher than those with G/G ( OR =1.226) genotype and T/T genotype compared the healthy control.Conclusion The study demonstrates that polymorphism within the TNFR Ⅱ at the position nt587 is associated with AS and TNFR Ⅱ nt587 G may play an important role in AS susceptibility,and TNFR Ⅱ nt587 G/G genotype may increase the sicken risk of AS in Hunan population.
6.Effect of curcumin on tumor necrosis factor-alpha-induced expression and release of monocyte chemoattractant protein-1 in rat astrocytes

Objective To investigate the effect of curcumin on tumor necrosis factor-alpha (TNF-α)-in-duced expression and release of monocyte chemoattractant protein-1 (MCP-1) in rat astrocytes.Methods The primary astrocytes were prepared from the cerebral cortex of 5 neonatal Sprague-Dawley rats and cultured.The cultured cells were identified by immunofluorescence staining with glial fibrillary acid protein.The cells were then divided into 6 groups (n =15 each):control group (group C),TNF-α group (group T),TNF-α+ different concentrations of curcumin groups (Cur5,Cur10 and Cur20 groups),and TNF-α+ solvent control group (D group).TNF-α with the final concentration of 20 ng/ml was added and the cells were incubated for 2h in T group.In Cur5,Cur10 and Cur20 groups,curcumin with the final concentrations of 5,10 and 20μmol/L was added,respectively,the cells were incubated for 24h and then the culture medium was abandoned,TNF-α with the final concentration of 20 ng/ml was added and the cells were then incubated for another 2h.In group D,dimethyl sulfoxide with the final concentration of 1 μl/ml was added,the cells were then incubated for 24h,then TNF-α with the final concentration of 20 ng/ml was added and the cells were incubated for another 2h.After treatment in each group,the expression of MCP-1 was determined by immunohistochemistry and the release of MCP-1 was determined by ELISA.Results Compared with group C,the expression and release of MCP-1 was significantly increased in the other five groups (P < 0.05).Compared with group T,the expression and release of MCP-1 was significantly decreased in group Cur20 (P < 0.05),and no significant changes in the expression and release of MCP-1 were found in Cur5,Cur10 and D groups (P > 0.05).Compared with group Cur5,the expression and release of MCP1 was significantly decreased in group Cur20 (P < 0.05),and no significant change in the expression and release of MCP-1 was found in group Cur10 (P > 0.05).Conclusion Curcumin can inhibit TNF-α-induced expression and release of MCP-1 in rat astrocytes and the effect is dose-related and may be one of the mechanisms of curcumin-induced reduction of neurophathic pain.
