1.The value of electromyography in diagnosis of myotonic dystrophy

Objective To explore the value of electrophysiology in diagnosis of myotonic dystrophy .Methods Electromyogra-phy(EMG) and nerve conduction were detected in 26 myotonic dystrophy patients(myotonia group) .Myotonic discharges were de-tected in 25 patients with non-tonic neuromuscular disease(control group) .Results The main features of EMG in myotonia group were myotonic discharges ,myogenic damage ,and normal nerve conduction .There was no myotonic discharges in control group .The positive rate of myotonic discharges of abductor pollicis brevis was 87 .5% .The positive rate of myotonic discharges of extensor digitorum communis was 77 .8% .The positive rates of myotonic discharges of anterior tibial muscle ,quadriceps femoris and trian-gular muscle are 57 .1% ,45 .5% and 45 .5% respectively .Conclusion Myotonic dystrophy distinguished from other neuromuscular disease might be mainly based on myotonic discharges and myogenic damage .Myotonic discharges were mainly observed in the small hand muscles .However ,the positive rate of abductor pollicis brevis ,which has the highest chance of myotonic discharge ,was less than 100% .
2.Demethylation stduy of decitabine in acute leukemia patients with Apaf-1 gene methylated

Objective To investigate the effectiveness of decitabine demethylation in treatment of acute leukemia.Methods Methylation specific PCR (MSP) was used to detected the methylation status of Apaf-1 gene promoter.10 cases entering the group.MSP was used to detected the 10 cases methylation status of Apaf-1 promoter between pre-and post-treatment of dicitabine.RT-PCR method used was to detect the differential expression levels of Apaf-1 mRNA in acute leukemia bone marrow mononuclear cell between preand post-treatment of decitabine.Results In post-treatment of decitabine,6 cases Apaf-1 gene promoter was demethylated.The loss expression of Apaf-1 mRNA re-expressed in 4 cases.6 cases Apaf-1 mRNA still express deletion.6 cases patients have Apaf-1 mRNA exprssion deletion,However,4 cases Apaf-1 gene was demethylated,2 cases methylated in post-treatment,maybe related to allele deletion or allelic varriants.Conclusion Post treatment of decitabine.Apaf-1 gene promotor was demethylated and repress the expression of Apaf-1 mRNA,play a key role in apoptosis maybe a new method for treatment of acute leukemia.

[Summary] Postoperative cognitive dysfunction ( POCD) is a common central nervous system complication after anesthesia and surgery.The mechanisms of POCD are not clear yet.The article reviewed several potential mechanisms that have been reported. Further researches are needed to identify the pathophysiologic progress of POCD, which have great clinical and economic values.
6.Isolation,cultivation and identification of dental pulp stem cells from canine

Objective:To culture canine dental pulp stem cells(cDPSCs)in vitro.Methods:Canine pulp cells were isolated and cultured by enzyme digestion and explanted tissue culture respectively.Cell morphology was observed under phase-contrast micro-scope.The clone forming unit(CFU)of the cells was examined by plate clone formation assay.Cell markers and protein-expression were examined by flow cytometry(FC)and immunofluorescence.Odontogenic and adipogenic potential were evaluated by alizarin red staining and oil red O staining.Results:Short spindle fibroblast-like and steadily growing cells were obtained by both methods.The clone assay showed that CFU was 1 5.1 7% ±2.79%.FC observasion showed that the CD90,STRO-1 and CD24 positive cells were 24.43% ±7.1 0%,20.67% ±1 .42% and 2.03% ±0.06% respectively,but CD34 was negative.Immunofluorescence analysis showed positive expression of Nestin,Vimentin,weak expression of ALP and negative expression of DSP of the cells.Differentiation ex-periment confirmed the odontogenic and adipogenic differentiation potential of the cells.Conclusion:cDPSCs can be cultured in vitro.
7.Diagnosis of Congenital Dextrocardia with Complex Anomalies by Echocardiography

0.05),the specificity of transthoracic echocardiography in diagnosing the atrial situs,ventricular structure,the connection of the ventricle and arteries or auricle,arterial situs was more than 90%.In mirror-image dextrocardia,majority had concordant segmental connection and could present anomalies with simple anomalies(42.9%) including atrial septal defect and ventricular septal defect,in addition,the major complex anomalies such as pulmonary stenosis were tetralogy of Fallot,double outlet of right ventricle(50%);In dextroversion,majority had discordant segmental connection and mostly presented anomalies with pulmonary stenosis or pulmonary atresia,including single ventricle,double outlet of right ventricle,transpo sition of great arteries,pulmonary atresia and so on(88.6%).Conclusion Echoca rdiography can exactly diagnose the most congenital dextrocardia with complex cardiovascular anomalies,and can be the first or alternative screening choice for diagnosing dextrocardia.
8.Study on dynamic changes of myocardial perfusion after coronary artery occluded with myocardial contrast echocardiography

The minimum DA%s after occlusion were 22.19 ? 6.21 , 16.26 ? 3.88 , 6.24 ? 2.48 , 7.97 ? 2.78 and 8.80 ? 3.45 at the time instances of occlusion,1,2,3,and 4 hours after occlusion,respectively. The minimum DA%s were related to the infarcted area (IA) in the pathology within 2-4 hours of occlusion,with the values of r equal to 0.76 , 0.80 ,and 0.85 ,respectively. Conclusions Within the different time intervals after the LAD were occluded,the maximum DA%s have no significant difference,however the minimum DA%s change and relate to the IA% after 2 hours of occlusion.
9.Assessment of left ventricular function of pulmonary hypertension patients by real-time three dimensional echocardiography

Objective To assess the changes of left ventricule and analyze the influential factors in pulmonary hypertension (PH) by comparing the changes of left ventricular volume and function using single cardiac cycle real-time three dimensional echocardiography ( sRT-3DE ) and two-dimensional echocardiography (2DE).Methods In case group,there were 30 inpatients diagnosed as PH,systolic pulmonary artery pressure(PASP)≥40 mmHg estimated by echocardiography,excluding left heart disease or congenital systemic-pulmonary circulation shunt.There were 30 healthy people as control group.Conventional 2DE and sRT-3DE were performed in both groups and obtained 2D and 3D parameters.Results ①In case group,the 2D-LVEF were significantly larger than 3D-LVEF (P < 0.01 ).There was less agreement of 2DE and 3DE in case group than in control.②Comparing with the control group,left ventricular EDV,ESV and SV in ease group decreased ( P <0.01 ),so as to 3D-LVEF ( P <0.05).The left ventricular spherical index(SIs,SId)and eccentric index (EId,EIs ) in case group were significantly larger than those in control group ( P <0.01 ).③LVEDV in case group was negatively correlated with PASP (r =- 0.47),and positively correlated with RVSV and RVEF ( r =0.84,0.66) ;3D-LVEF in case group was negatively correlated with PASP ( r =- 0.54),and positively correlated with RVSV and RVEF ( r =0.58,0.53); there was no significant correlation between LVEDV and RVEDV.LVEDV was negatively correlated with SIs and SId ( r =-0.65,-0.61),so as to EId and EIs ( r =-0.67,0.67).LVEF was negatively correlated with SIs and SId ( r =-0.64,-0.61),so as to EId and EIs ( r =-0.66,-0.68),and the corresponding P all above were less than 0.01.Conclusions PH can result in reductions of left ventricular volume and systolic function,and the variations of which have the correlations with the severity of PH and the increase in SI and EI.sRT-3DE can provide a method to get more accurate information of left ventricle.
10.Analysis and determination of irone in Iris tectorum

Object To establish an analysis method for determ in ation of irone in Iris tectorum Maxim. f. alba Makino. Methods The three isomers of irone were quantitatively analyz ed by GC-MS. Irone in I. tectorum extract was determinated by GC. Results The three isomers of irone, ?, ?, ?- irone were s p eculated according to the MS splitting decomposition law. Irone contents in the extract were 687, 238 ?g/g (n=6), which h ad much difference. Conclusion The analysis method for irone by GC-MS and GC is hig her efficiency, precise, and the analysis time is acceptable.
