1.Effect of jujube on the expression of CYP2E1 and TNF-alpha in the hepatic tissue of mice with alcoholic liver disease.

OBJECTIVETo observe the effect of jujube pretreatment on serum levels of AST and ALT, liver pathology, and the expression of cytochrome P4502e1 (CYP2E1) and tumor necrosis factor-alpha (TNF-alpha) in the liver tissue of alcoholic liver disease (ALD) mice.
METHODSTotally 88 Kunming mice were randomly divided into the control group (n = 28), the model group (n = 32), and the jujube treatment group (n = 28). The animal model was prepared using intragastric alcoholism for mice in the model group and the jujube treatment group, while distilled water was intragastrically given to those in the control group. Extraction of jujube was intragastrically given to mice in the jujube treatment group at week 4, while equal volume of distilled water was intragastrically given to mice in the rest two groups. The therapeutic course lasted for 12 weeks. Serum levels of AST and ALT, liver pathology, and the expression of CYP2E1 and TNF-alpha in the liver tissue of ALD mice were observed after administration of jujube.
RESULTSCompared with the model group, serum levels of AST and ALT decreased, the liver pathology was improved, and the expression of CYP2E1 and TNF-alpha in the liver tissue decreased, showing statistical difference (P < 0.05).
CONCLUSIONJujube had certain effect in treating ALD.

BACKGROUND: Maintenance and activation of cascade reaction influence T cell proliferation or transformation into nonreactive state even apoptosis. B7 binding to CD28 effectively activates T cells in combination with T cell receptor pathway, and enhances T cell proliferative activity. OBJECTIVE: To investigate the costimulated activation of peripheral blood mononuclear cells (PBMC) with CD28 and CpG containing oligodeoxynucleotides (CpGODN) MoAb combined with CD80, and its killing effect on human gastric cancerous cell line MKN45 in vitro.METHODS: PBMCs were isolated by Ficoll density gradient centrifugation method, and cocultured with interleukin-2, CD28 and CpGODN MoAb for 1-5 days. MKN45 cells were divided into 4 culture conditions: CD28/CpGODN, CD80 plus CD28/CpG ODN, CD80 alone, and blank control.The killing efficiency was measured by MTT method.The ultramicrostructure of cells was observed by electron microscope. Apoptosis was verified by a flow cytometery. RESULTS AND CONCLUSION: CD80 alone did not display killing effect on MKN45 cells. By MTT method, combination of costimulated activation of PBMC with CD28/CpGODN and CD80 showed enhanced killing effect compared with single therapy (P < 0.05), and the ratio of effector cell and target cell at 15: 1 resulted in half killing efficiency. Electron microscope and flow cytometery verified necrotic or apoptotic cells after 24 hours exposure to costimulated activation. Compared with blank control group, CD80 alone elevated the apoptosis rate of MKN45 cells (P < 0.01). Results from the present study show that CD80 can elevate the killing effect of costimulated activation of PBMC with CD28/CpGODN on MKN45 cells in vitro.
6.Efficacy of postauricular injection of shuxuening combined with lidocaine for intermediate and high-frequency sudden hearing loss

[ABSTRACT]OBJECTIVETo assess the efficacy of postauricular injection of shuxuening combined with lidocaine in treating intermediate and high-frequency sudden hearing loss.METHODS48 patients with intermediate and high-frequency sudden hearing loss were randomly divided into postaurical injection group (n=24) and control group (n=24). The control group was given hormone, microcirculation improvement, nutrition of nerve and other conventional treatment. The postauricular injection group received 3 doses of postauricular inject of 0.5ml shuxuening and 0.3ml lidocaine tertian injection in addition to conventional treatments.RESULTSTwo weeks after administration, the total effective rates of postauricular injection group 87.5%were significantly higher than that of the control group 62.5%. Pure tone average improved (20.74±15.19)dB on average in the postauricular group, and (12.44±9.12)dB in the control group. The difference was statistically significant (P<0.05). CONCLUSIONPostauricular injection of shuxuening combined with lidocaine treatment for intermediate and high-frequency sudden hearing loss has better curative effect than that of the conventional treatment.

8.Predictive and intelligent control of sevoflurane closed circuit anesthesia

Objective To evaluate the efficacy of servo controlled sevoflurane closed-circuit anesthesia and the feasibility of the predictive and intelligent control.Methods One hundred and forty-three ASA Ⅰ-Ⅲ patients (90 males, 53 females) aged between 3-77 yrs undergoing elective surgery were studied. Anesthesia was induced with intravenous fentanyl 2-3 ?g ?kg-1 , midazolam 0.12 mg?kg-1 and vecuronium 0.1 mg?kg-1 . After tracheal intubation the tracheal tube was connected to the servo-controlled closed-circuit system composed of IBM computer, O2 mass flow controller and electrically controlled sevoflurane injection pump and multifunctional monitor. The fresh gas flow of O2 = [body weight (kg)3/4 ? 10 + 20] ml?min-1 . The end-expired sevoflurane concentration was maintained at 1.3 MAC by predictive and intelligent control.Results The average wash-in time to reach the target concentration was (5.2 ?2.4) min. The O2 flow rate was(0.22?0.04) L?min-1 . The cumulative uptake of sevoflurane was 5.16 ml, 7.74ml, 9.17ml, 11.08ml, 12.57ml, 13.00ml, 14.18ml, 15.60ml, 18.56ml and 24.6 ml at 30, 60, 90, 120, 150, 180, 210, 240, 300 and 420 min respectively. The uptake rate of fluid sevoflurane was equivalent to (0.2673e-0.0598t + 0.2269e-0.0597t + 0.1150e-0.002t) ml?min-1 . Conclusion The servo controlled sevoflurane closed-circuit system can effectively control the pre-set end-tidal sevoflurane concentration in spite of the influence of multiple factors and is safe and effective.

Objective:To explore the value and significance of transcranial color-coded duplex (TCCD) parameters, serum insulin-like growth factor-1 (IGF-1), neuron-specific enolase (NSE) levels in the evaluation of the condition of cerebral infarction.Methods:One hundred and ten patients with cerebral infarction in Anhui Wanbei Coal-Electricity Group General Hosptial were selected from April 2018 to February 2020, and TCCD examination was performed after admission. Blood samples were taken to determine the levels of serum IGF-1 and NSE, and corresponding treatment was given. The TCCD parameters and serum IGF-1 and NSE levels were compared in patients with different disease severity, plaque nature and different prognosis after 3 months′ follow-up. The correlation between the above indicators and the condition and plaque nature and the predictive value of the prognosis of patients with cerebral infarction were analyzed.Results:The level of IGF-1 were negatively correlated with the severity of disease ( r=- 0.650) and the nature of plaques ( r=- 0.711); and the level of NSE and resistance index (RI), pulsatility index (PI) were positively correlated with the severity of the disease ( r=0.609, 0.613, 0.645) and the nature of plaques ( r=0.589, 0.579, 0.608), and the differences were statistically significant ( P<0.05). After 3 months′ follow-up, the level of IGF-1 in the favourable prognosispatients was higher than that in the unfavourable prognosis patients, the levels of NSE, RI and PI in the favourable prognosis patients were lower than those in the unfavourable prognosis patients: (9.01 ± 2.64) μg/L vs. (25.13 ± 3.82) μg/L, 1.05 ± 0.19 vs. 1.32 ± 0.22, 0.69 ± 0.06 vs. 0.89 ± 0.07, and the differences were statistically significant ( P<0.05). The area under the curve (AUC) of RI, PI, IGF-1 and NSE in predicting the prognosis of cerebral infarction was less than that of combined diagnosis. Conclusions:The early use of TCCD parameters, serum IGF-1 and NSE combined detection can provide evidence-based guidance for evaluating the condition and prognosis of cerebral infarction.
