Autophagy is a dynamic process which subcellular membranes undergo morphological changes that lead to the degradation of cellular cytoplasmic organelles and macromolecules.It is regulated by themammalian target of rapamycin ( mTOR ) -dependent or -independent signaling pathways.It has been demonstrated that autophagy is induced or inhibited in various tumor cells,and it is closely related with cell survival and drug resistance.Because of the complex relationships with cell death,the roles of autophagy in cancer developnent,treatment,and drug-resistance are not the same,and thus controlling autophagy properly may become one of new means of cancer therapy.

Objective To explore the application value of susceptibility weighted imaging (SWD) in the Parkinson disease (PD) diagnosis by analysis the changes of brain iron content of the basal ganglia and red nucleus.Methods Totally 56 cases of patients with PD and 40 cases of normal control group were diagnosed by SWI with the 3.0 T magnetic resonance imaging system,the corresponding phase image by using SWI processing software was obtained,the signal values of the various regions of the brain basal ganglia and red nucleus were measured and compared.Results In comparison of PD group with normal control group,the signal values showed differences in caudate (0.05±0.01 vs.0.04±0.01,t=1.991),putamen(-0.42±0.13 vs.-0.25±0.07,t=2.544) and globus pallidus(-0.23± 0.07 vs.-0.11±0.02,t=2.893) (all P＜0.05).Conclusions Determining the iron deposition signal of the various regions of the brain basal ganglia by SWI is valuable in the clinical diagnosis of Parkinson's disease.

Objective To evaluate the efleet of continuous subcutaneous insulin infusion (CSII)combined with mefformin hydroehloride( Met H)on type 2 diabetic patients. Methods 240 patients with type 2 diabetes melli-tus (T2DM) were divided into 3 groups randomly: CSII combined with Met H ( A group: n=80) ; simple CSII ( B group:n=80) ; multiple deliver insulin (MDI) ( C group: n=80). The levels of blood glucose, HbA1 e, therapeutic time, a-mount of insulin, Fins ,C-P, HOMA-IR and IAI were compared among them before and after 10d of treatment. Results GroupA, B eornpared with group C, the levels of HbA1 e significantly deeroased in other two groups [ ( 8.0±3.2)%vs. (6.9±1.4)% and(7.0±1.2)% ] (aU P<0.05),so did the therapeutic time[(6.0±1.9)d vs. (3.0±1.4) and(3.5±1.5) d] and insulin dose[ (55.0±17.0) U/d vs. (38.0±15.0) U/d and(45.0±16) U/d] (all P<0.05). The insulin dose was much less in group A than in group B. Versus group B and C,the HOMA-IR of group A (0.8±0.3 vs. 1.5±0.4 and 1.6±0.4)decreased(P<0.05) ,and IAI(-2.0±4.3 vs. -3.6±4. 2 and -4.1±4.3) increased (all P<0.05), without the function impairments of heart, liver or kidney. Conclusion CSII in combi-nation with mefformin hydreehloride can significantly enhance the treatment effect of the patients with type 2 diabetes mellitns; reduce blood glucose and HbA1 e, improve insulin resistenee,lessen insulin need.

Most patients with allergic conjunctivitis in children are vernal keratoconjunctivitis.Dry eye accompanied with al- lergic conjunctivitis in children is correlated with the decline of tear film stability.However,tear film stability decline is correlated with eosinophilic activation and concomitant release of inflammatory mediators.Moreover,conjunctival epithelial,goblet cells and tear film are damaged,then dry eye occur.Main symptom of dry eye in children is twinkling frequency increasing,followed by eye itch,eye red and photophobia.When allergic conjunctivitis in children is diagnosed,dry eye should be considered at the same time.Proper manage- ment for those allergic conjunctivitis can decrease ocular surface damages.

Objective: To investigate the predictors of subjective well-being (SWB) in older adults. Methods: 236 old people in Beijing were assessed on 7 potential predictors chosen from 2 taxonomies of resources-internal vs. external and physical vs. psychological.Results: There was no relationship between age and SWB. Perceived social support, health status and general self-efficacy correlated significantly with subjective well-being(?=0.27,0.19,0.15); perceived social support, health status and general self-efficacy have direct impact on subjective well-being, perceived social support and health status have secondary impact on subjective well-being through the factor general self-efficacy as well.Conclusions:The external factors can influent the SWB both directly and indirectly.

Objective To investigate the effects of children's mentality on mental symptoms during recovery period of ketamine anesthesia. Methods According to the mentality, forty-five children undergoing smalminor surgery were assigned into active group, stable group and fretful group with 15 cases each. Child was anaesthetized by ketamine alone. RP, BP, HR and SpO_2 were recorded at the end of operation, 5,10 and 30 min after operation. Meanwhile the expression, feeling, behavior, talking and activity were observed. Results RR, BP, HR in active group and fretful group were significantly higher than those in stable group. There was a significant difference in the expression, feeling, behavior, talking and activity among three groups. Conclusion Children's mentality has a significant effect on mental symptoms and hemodynamics during recovery period of ketamine anesthesia.

Objective To investigate the effects of rubella virus R16 infection on heat shock protein 70 (HSP70) and heat shock transcription factor 4 (HSF4) in the human embryonic lens epithelial cells in vitro. Methods The human embryonic lens epithelial cells were infected by rubella virus R16 for 3, 7 and 14 days respectively in vitro. Then the mRNA levels of HSP70 and HSF4 were measured by Real-time PCR assays, the HSP70 protein level was detected by Western blot assay, and the DNA sequence of HSF4 was also identified by DNA Sequencing. Results Both mRNA and protein levels of HSP70 were increased dramatically at 3 and 7 days after rubella virus R16 infection in vitro. But mRNA levels of HSF4 were decreased significantly. The DNA sequence of HSF4 had no change in the human embryonic lens epithelial cells at 14 d after rubella virus R16 infection. Conclusions Rubella virus R16 could directly induce the increased expression of HSP70 in the human embryonic lens epithelial cells infected by rubella virus R16 in vitro, which may prevent human embryonic lens epithelial cells from infecting with virus. The heat shock transcription factor may serve as a negative regulator at transcription level. However, the DNA sequence of HSF4 had no change in the human embryonic lens epithelial cells within 14 days after rubella virus R16 infection.

Obejective To evaluate the accuracies of ten commercial total 25-hydroxyvitamin D [25(OH) D] immnoassays.Methods NIST 25 (OH) D reference material SRM 972a,which consisted of four vials of frozen serum with different concentration levels of different 25 (OH) D types,and two human serumsamples provided by our lab (BIMT),which had different concentration levels of 25 (OH) D3,were analyzed by ten total 25-hydroxyvitamin D immnoassays from Biomerieux,Mindray,Maccura,Chemclin,Abbott (2),Siemens,SNIBE (2),Roche,and by isotope-dilution liquid chromatography-tandem mass spectrometry(ID-LC/MS/MS) founded by BIMT.For the measurements of SRM 972a,the biases between tested values and certified values were calculated as a evaluating indicator,meanwhile the test biases between immnoassays and ID-LC/MS/MS were used as a evaluating indicator for the measurements of BIMT 25(OH) D3 serum samples.Test bias lower than 10％ was deemed acceptible.Results The ID-LC/MS/MS exhibited low biases at (1.6％-2.8％) in the measurements of all levels of SRM 972a.8 immnoassays showed low biases at(1.5％-3.5％) in the measurements of level 1 of SRM 972a,which had a high 25(OH) D3 concentration level,but only 2 immnoassays gave low biases at (3.6％-3.7％)in the measurements of high 25 (OH)D2 concentration sample (level 3).While,5 immnoassays gave low biases at (-0.3％-9.0％) in the measurements of high 3-epi-25 (OH) D3 concentrationsample (level 4).It seems that,when SRM 972a were analyzed,only one of the ten commercial total 25 (OH)D immnoassays were in good accuracy and analytical specificity agrements with ID-LC/MS/MS.When two human serum25(OH) D3samples from BIMT were tested,most immunoassays were overall in relative good agreement with ID-LC/MS/MS at high 25 (OH) D3concentration level.Conclusion The test biases in the total 25 (OH) D measurements are differences between differentimmnoassays and ID-LC/MS/MS,and the specificities of current commercial total 25 (OH) D immnoassays should be improved,especially the performance on the equivalent recognition of 25 (OH) D2 and 25 (OH) D3.
