1.Study on paeoniflorin-6'O-benzene sulfonate's physicochemical property

Objective To investigate the physicochemical property of Pae-6’O-benzene sulfonate ( CP-25 ) . Meth-ods The CP-25 physicochemical property was evaluated by appearance, Lieberman-Burchard reaction, thin-layer chromatogram, Ultraviolet Spectrophotometry ( UV) , solubility and stability. The content of CP-25 was assayed by high performance liquid chromatography. Results The CP-25 had color response featured by terpenoid, and its maximum UV absorption wavelength was 220 nm. CP-25 was slightly soluble in water and petroleum ether. The main influence factor of CP-25 stability was humidity. Conclusion The present study provides experimental basis for quality standard and formulation design of CP-25 .
2.Studies on degradation kinetics of paeoniflorin-6-O’- benzenesulfonate in vitro

Objective To investigate the degradation kinetics of paeoniflorin-6-O’- benzenesulfonate ( CP-25 ) in vitro. Methods The homogenates of liver and intestine were prepared in vitro, and concentrations of CP-25 in ho-mogenates were detected by HPLC. Results CP-25 was obviously degradable in liver and intestine homogenates, and half life of degradation was decreased when levels of homogenates increased;the metabolisms of CP-25 in dif-ferent homogenates of intestine were diverse, the metabolic actions in duodenum and colon were weaker than those of jejunum and ileum. Conclusion Oral administration of CP-25 suffers first pass elimination from intestine and liver, which suggests the absorption of CP-25 could be further improved by appropriate pharmaceutical preparations.
3.Relationship of epigenetic and Dao-di herbs.

Dao-di Herbs is specificity and locality, and its unique phenotypic features is closely related to the growth and development of medicinal plants. In addition to traditional genetic, epigenetic play an important role in formation of Dao-di herbs. This paper introduces the concept of epigenetic and the role of DNA methylation in the gene expression regulation. We further prospects epigenetic mechanism in study of Dao-di herbs formation from specific phenotype and regional analysis. And study on the relationship of epigenetic and Dao-di herbs will provide a basis for quality assessment and identification of Chinese drugs.

5.Effect of the rotation angle of femoral prosthesis on the knee joint function after total knee arthroplasty: a 3-year follow-up

BACKGROUND: Femoral prosthesis malrotation will lead to various complications, such as femoro-tibial and patellofemoral instability, and maladjustment of patellar movement trajectory, further affecting the recovery of the knee function.OBJECTIVE: To evaluate the influence of the rotation angle of the femoral prosthesis on the knee joint function after total knee arthroplasty (TKA).METHODS: Totally 146 patients (186 knees) undergoing TKA in the Department of Joint Surgery, Zhenjiang First People's Hospital from January 2010 to January 2014 were selected. Afterwards, the femoral condyle torsion angle was measured on CT to evaluate the rotational position of the femoral prosthesis. According to the condyle torsion angle, all the patients were divided into three groups: normal group (n=74, extorsion angle 3°-intorsion angle 3°), intorsion group (n=15, intorsion angle > 3°) and group (n=58, extorsion angle > 3°). All the patients were followed up for 3 years.RESULTS AND CONCLUSION: (1) The order of the activity function score was as follows: the normal group > extorsion group > intorsion group (P < 0.05). The standard activity, free activity and senior activity scores were highest in the normal group, followed by the extorsion group, and lowest in the intorsion group (P < 0.05). However, the standing and walking scores, as well as the satisfaction and expected scores did not differ significantly among groups. (2) That is to say, the rotational position of the femoral prosthesis influences the long-term knee joint function after TKA, so we should pay attention to the femoral prosthesis malrotation.

Objective To analyze the prognostic factors of nasopharyngeal carcinoma treated with radiotherapy.Methods 135 preliminary diagnosed patients were selected and treated with radiotherapy to a total dose of 68~80 Gy in 7~8 weeks. 8.Impact of TLR5 and NLRC4 activation on innate immune cells in mice

Objective:To investigate the impact of recombinant flagellin targeting TLR5 and NLRC4 simultaneously or respectively on innate immune cells in mice. Methods: Induction,expression,purification and identification of recombiant FliC,which were FliC(activating both TLR5 and NLRC4);FliCΔ90-97(unable to activate TLR5),FliC-L3A(unable to activate NLRC4),FliCΔ90-97:L3A(unable to activate both TLR5 and NLRC4). The mice were divided into five groups,namely group FliC,FliC-L3A,FliCΔ90-97,FliCΔ90-97:L3A and PBS,which were injected with 100μl PBS or 10μg recombinant flagellin intraperitoneally,three mice in each group. 12 h later,the mice were executed using dislocation of cervical vertebra and the splenic and peritoneal cells were isolated. The spleen was grinded into single-cell suspension. The proportion of neutrophils,NK cells,DCs and the expression level of CD80 and CD86 on DCs were evaluated with flow cytometry. Results:Group FliC,group FliC-L3A and group FliCΔ90-97 shared the similar proportion of neutrophils in peritoneal cavity ( P>0. 05 ) , and all of which were significantly higher than group PBS and group FliCΔ90-97 ( P<0. 01),and NK cells also showed the similar trend. Compared with group FliCΔ90-97 and FliCΔ90-97:L3A,the mean fluorescence intensities(MFIs) of CD80 and CD86 in group FliC and FliC-L3A increased significantly(P<0. 01). The proportion of Treg in spleen was highest among all groups. Conclusion:Activation of TLR5 and NLRC4 had similar chemotaxis of neutrophils and NK cells. The ex-pression of CD80 and CD86 on DCs were upregulated after stimulation by flagellin and TLR5-dependent. Activation of TLR5,but not NLRC4,increased the proportion of Treg in spleen.
9.Impact of TLR5 and NLRC4 activation on proliferation of different breast cancer cell lines

Objective:To explore the impact of TLR5 and NLRC4 activation on the proliferation of different breast cancer cell lines,MCF-7 and MDA-MB-23 i.Methods:Induction,expression,purification and identification of recombiant flagellin,including FliC (activating both TLR5 and NLRC4),FliC△90-97 (unable to activate TLR5),FliC-L3A (unable to activate NLRC4),FliC△90-97:L3A (unable to activate both TLR5 and NLRC4).Using different concentration of recombinant flagellin to stimulate MCF-7 and MDA-MB-231 cell lines,72 h later,the proliferation of tumor cells were detected with CCK8.We also used soft AGAR forming experiments to detect the inhibition ratio of recombinant flagellin on breast cancer cell lines.Briefly,1 000 cells were plated in the 6-well plate,then stimulated with 1 μg/ml recombinant flagellin,14 days later,the number of cloning were counted after crystal violet staining.Results:After stimulation with four recombinant flagellins at the concentration of 0.1 μ,g/ml,the inhibition ratio on MCF-7 reached 30%,and FliC△90-97 were dose-dependent on the inhibition of MCF-7 proliferation.At the concentration of 1 μg/ml,FliC-L3A which only activated TLR5 showed stronger inhibition ratio than FliC.FliC△90-97:L3A which did not activate both TLR5 and NLRC4 also inhibited the proliferation of MCF-7.After adding transfection reagent,four recombinant flagellins showed inhibition effect on MDA-MB-231.Conclusion:Flagellin can inhibit the proliferation of MCF-7 and MDA-MB-231,and the mechanism of inhibition on the proliferation were not TLR5 and NLRC4 pathway dependent.There might exist new mechanisms to explain this phenomenon.
10.Quantitative assessment of left ventricular systolic synchrony in patients with chronic renal failure using real-time three-dimensional echocardiography

Objective To quantitatively evaluate the left ventricular systolic synchrony in patients with chronic renal failure (CRF) by real-time three-dimensional echocardiography (RT-3DE). Methods Thirty patients with CRF and twenty-five normal subjects were enrolled in this study.The M-mode ejection fraction (M-EF), left ventricular end diastolic internal diameter (LVIDd), interventricular septum diameter (IVSd), left ventricular posterior wall diameter (LVPWd) were obtained on two-dimension. The global and regional volume-time curves were obtained on three -dimensional. The end diastolic volume (LVEDV),end systolic volume (LVESV) and ejection fraction (LVEF) of left ventricule, the time to minimal systolic volume (Tmsv) of 16,12,6-segmental standard deviation (Tmsv16, 12, 6-SD), maximal difference (Tmsv16, 12, 6-Dif) were derived from Qlab software . The above parameters as a percentage of the cardiac cycle with different heart rates between patients were also calculated from the Qlab software, which were Tmsv16, 12, 6-SD% and Tmsv16, 12, 6-Dif %, rspectively. Results The indices of HR, LVIDd, IVSd, LVPWd and LVESV were significantly higher while LVEF was significantly lower in the CRF group than those of the control group (P<0.05 or P<0.01); The indices of systolic asynchrony of Tmsv16,12-SD%, Tmsv16,12-Dif, Tmsv16 ,12 ,6-SD/R-R%,Tmsv16,12,6-Dif/R-R% were significantly larger in the CRF group than those of the control group(P<0.05 or P<0.01). Conclusions RT-3DE provides a simple, intuitional and noninvasive approach to assess the systolic synchrony of all the LV segments simultaneously and LVEF in patients with chronic renal failure.
