1.Effects and safety of citalopram combined with low-dose risperidone on refractory depression

Objective To explore the effects and safety of citalopram combined with low-dose risperidone on refractory depression. Methods All of 54 patients with the refractory depression were ran-domly divided into two groups: augmented treatment group (taken citalopram 20 mg/d, risperidone 0.5-2.0mg/d) and mono-thempy group (taken citalopram alone 20 mg/d). The treatment lasted for 6 weeks. They were estimated with Hamilton depression scale (HAMD), Hamilton anxiety scale (HAMA), and treatment emergent symptom scale (TESS) at baseline and every two weeks subsequently. Results In the augment-ed treatment group, total effective rate was 60.7% and recovery and excellence rate was 50.0%, while that was 26.9% and 15.4% in mono-therapy group (P< 0.05). The adverse effect of two groups was minor. Conclu-sion Rispefidone may be a useful and safe adjunct to citalopram in treatment of refractory depression.
2.Isolation and identification of four isoflavones and one pterocarpan from BUYANGHUANWUTANG

Object To study the antithombotic portion and bioactive fraction of BUYANGHUANWUTANG (BHT), a decoction of astragalus in combination with peony and several other ingredients. Methods The EtOAc portion of BHT extract was repeatedly separated by chromatography and identified by means of spectral analysis. Results One pterocarpan, (6aR, 11aR) 9, 10-dimethoxypterocarpan-3-O-?-D-glucopyanoside (Ⅴ) and four isoflavones, formononetin (Ⅰ), calycosin (Ⅱ), formononetin-7-O-?-D-glucopyanoside (Ⅲ), and calycosin-7-O-?-D-glucopyranoside (Ⅳ) were identified. Conclusion All five compounds were isolated for the first time from BHT.
3.Alkaloids isolated from Buyang Huanwu Decoction and their structure identification

Objective To study the antithrombotic compounds and bioactive fraction from Buyang(Huanwu) Decoction(BHD).Methods Four alkaloids have been isolated from BHD by repeated chromatographies.Their structures were elucidated by means of spectral and single-crystal Xray diffraction analysis.Results They are N-(3′-maleimidyl)-5-hydroxymethyl-2-pyrrole formaldehyde(Ⅰ),4-carbamoyl-2-pyrrole carboxylic acid(Ⅱ),N-(1′-D-deoxyxylitolyl)-6,7-dimethyl-1,4-dihydro-2,3-quinoxalinedione(Ⅲ),and 2,3,4,9-tetrahydro-1H-pyrido indole-3-carboxylic acid(Ⅳ).Conclusion All these compounds are isolated from BHD for the first time and compounds Ⅰ-Ⅲ are new alkaloids.
4.Isomers of amygdalin in BUYANG HUANWU TANG and its production

Object To study the isomers of amygdalin in BUYANG HUANWU TANG and its production Methods Amygdalin was isolated from both peach seeds and BUYANG HUANWU TANG by using various column chromatography Their structures were identified by the various spectral data Results Amygdalin had been isolated from n BuOH fraction of aqueous extract of BUYANG HUANWU TANG and found to be a pair of D , L epimers and their ration was 1∶1 It was also found that the structures and the ration of D , L epimers of amygdalin in decoction of single peach seed were similar to that in BUYANG HUANWU TANG. The peach seed only gave D amygdalin when it was extracted in 95% EtOH at reflux temperature, and D amygdalin cannot be isomerized when it was treated in water at 100 ℃ Conclusion Isomerization of D amygdalin results from interaction between it and other compounds of peach seed in water at high temperature, and has no evident relation to other constituents in BUYANG HUANWU TANG L amygdalin is a new compound generated due to decoction of peach seed
5.Effect of differential electroacupuncture frequencies on behavior of acute stress depression model of C57BL/6 mice

Objective To explore the effect of different frequencies interference of electoracupuncture on acute stress of C57BL/6 mice. Methods Thirty male C57BL/6 mice were randomly divided into model group,2Hz electroacupunture group and 100 Hz electroacupunture group.After interfering 7 days by electroacupuncture,all the mice were carried forced swimming tests and tail suspension test,and the immobility time in 5 minutes were taken down. Results The immobility time of forced swimming test and tail suspension test of 2 Hz eletroacupuncture group(32.00±14.73)s,(72.50±20.44)s was shorter than those of 100 Hz electoracupuncture group(145.60±34.62)s,(124.22±21.67)s( P <0.05); and the immobility time of 2 Hz eletroacupuncture group was shorter than that of model group(129.50±33.94)s,(142.23±20.05)s( P <0.05); but the immobility time of 100 Hz electroacupuncture group was no difference with that of model group( P >0.05). Conclusions The effect of 2 Hz eletroacupuncture could improve the behaviors of the acute stress of C57BL/6 mice and had the more effectiveness on anti-depression.
6.Effects of Relieving Fatigue Decoction on MDA,GSH,GSH-PX,SOD,Testosterone and Corticosterone in Fatigue Rats

Objective To investigate the anti-fatigue mechanism of Relieving Fatigue Decoction(RFD). Methods 33 male SD rats were evenly randomized into control, model and RFD groups. 21d exhausted swim exercise was used to make an exercise-induced fatigue rat model. RFD group was given RFD in routine doses by gastric perfusion for 21d, once a day. After decapitation, the rats’ testis, liver and blood were taken, detecting MDA with thio-barbituric acid method, GSH and GSH-PX with chromatometry method, SOD with xanthine oxidase method, testosterone and corticosterone with ELISA method respectively. Results (1)Compared with control group, the serum MDA in model group increased significantly(P0.05); the testis, liver and serum GSH, GSH-PX reduced significantly(P0.05); the serum testosterone and corticosterone increased significantly(P0.05); the testis, liver and serum GSH increased significantly(P0.05); the liver SOD increased significantly(P0.05); the serum corticosterone decresed significantly(P0.05).Conclusion RFD may improve the activity of antioxidant enzymes in liver, testis and serum, and raise serum T/C level, which may underlie its action in impoving fatigue.

8.Research progress of p70 ribosomal protein S6 kinase inhibitors.

p70 ribosomal protein S6 kinase (p70S6K), an important member of AGC family, is a kind of multifunctional Ser/Thr kinases, which plays an important role in mTOR signaling cascade. The p70 ribosomal protein S6 kinase is closely associated with diverse cellular processes such as protein synthesis, mRNA processing, glucose homeostasis, cell growth and apoptosis. Recent studies have highlighted the important role of S6K in cancer, which arose interests of scientific researchers for the design and discovery of anti-cancer agents. Herein, the mechanisms of S6K and available inhibitors are reviewed.

9.Effects of exercise on behavior and peripheral blood lymphocyte apoptosis in a rat model of chronic fatigue syndrome.

This study examined the effects of exercise on behavior and peripheral blood leukocyte apoptosis in a rat model of chronic fatigue syndrome (CFS). Thirty-six healthy male Sprague-Dawley rats were equally randomized into 3 groups: the control group, CFS model group and the exercise group in terms of body weight. A total of 25 rats entered the final statistical analysis due to 11 deaths during the study. CFS model was established by subjecting the rats in CFS model group and exercise group to electric shock, chronic restraint stress and cold water swim. Besides, rats in the exercise group took running wheel exercise. After a week of conditioning feeding, model construction and running wheel exercise were performed simultaneously, and lasted for 23 consecutive days. The behavior experiments, including running wheel exercise, open-field test, tail suspension test and Morris water maze test, were conducted, either before or after the model establishment. Rats were sacrificed and peripheral blood was obtained for the assessment of lymphocyte apoptosis index by flow cytometry (FCM). It was found that as compared with those in the control group, the weight of the rats was decreased obviously (P<0.01), the mobility time in the open-field and the tail suspension tests was shortened significantly (P<0.01), the time to locate the platform was enhanced (P<0.01) and the cell apoptosis index was increased substantially (P<0.01) in the CSF model group. Meanwhile, in comparison to the model group, the behavior in the open-field and the tail suspension tests was improved significantly (P<0.05), and the apoptosis index decreased remarkably (P<0.01) in the exercise group. It is concluded that sport intervention can prevent lymphocyte apoptosis and improve animal behavior rather than the memory.
