Objective To investigate the causes of reduced function of neutrophils in neonates by measuring superoxide, one of the reactive oxygen species, produced by neutrophils following live bacterial phagocytosis in the whole blood. Methods Twenty-three cord blood samples were collected from normal full-term newborns and cultured with live Staphylococcus aureus and Escherichia coli. Blood cells were stained with Hydroethidine, an indicator of superoxide. The intensity of the superoxide of neutrophils were analyzed by flow cytometry. Fifteen adult blood samples were collected as control. Plasma replacement was used to study the function of body fluid in active oxygen metabolism of neutrophils. Results Most full-term neonatal neutrophils (PMNs) could produce superoxide anion following bacterial phagocytosis in the blood as that in adults. 4.Risk factors for allergic disease in preterm infants during the first year of life

Objective To analyze the risk factors associated with allergic disease in preterm infants during the first year of life. To investigate whether there was any association between peripheral blood eosinophils and allergic di-sease during the first year of life benefit of hydrolyzed formula feeding in preterm infants. Methods A nested case -control study was conducted in a cohort of artificial feeding preterm infants admitted to Neonatal Intensive Care Unit, Shanghai Children's Medical Center from January 2012 to April 2013. The preterm infants were monitored from birth up to 1 year of age and the findings were related to development of allergic disease. Individuals with allergic disease during the first year of life(cases)were matched with gender,as previously described with two individuals(controls)who re-mained event - free during the study. The risk factors associated with allergic disease in preterm infants during the first year of life were analyzed. And whether there was any association between peripheral blood eosinophils and allergic di-sease during the first year of life benefit of a hydrolyzed formula feeding in preterm infants was investigated. Different variables in control and case individuals were compared with t test for normal distribution measurement date,χ2 tests for categorical variables and Mann - Whitney U test for non - normal distribution measurement date,and multiple conditio-nal Logistic regression were used to investigate the risk factors associated with allergic disease in preterm infants. Results Thirty - four individuals were in cases and 68 individuals were in controls. In a conditional multivariable Logistic model,peripheral blood eosinophils of preterm infants at full enteral feeding(EOS - 2)(OR = 5. 941,95% CI:1. 165 - 41. 375,P ﹤ 0. 05),and family history of allergy(OR = 3. 316,95% CI:1. 201 - 9. 152,P ﹤ 0. 05)were the two independent risk factors for allergic disease in preterm infants during the first year of life. Individuals fed with standard preterm formula after birth,the association between EOS - 2 and allergic disease was significantly enhanced (OR = 21. 459,95% CI:1. 686 - 273. 152,P ﹤ 0. 05). By contrast,in individuals fed with hydrolyzed formula,the risk of EOS - 2 was substantially attenuated(OR = 1. 708,95% CI:0. 148 - 19. 743,P ﹥ 0. 05). Conclusions Peripheral blood eosinophils of preterm infants at full enteral feeding EOS - 2 and family history of allergy were the 2 independent risk factors for allergic disease in preterm infants during the first year of life. Contrast to individuals fed with standard preterm formula after birth,individuals fed with hydrolyzed formula had lower association between factors of peripheral blood eosinophils and family history of allergy,and allergic disease during the first year of life.

Objective To observe the effects of Angong Niuhuang Bolus(ANB) on vital organ injury and mortality in rats with sepsis.Methods SD rats were randomized into normal control group,model group,and low-,middle-and high-dose ANB groups(in the dose of 1.0,2.0 and 3.0 g?kg-1?d-1 respectively).Except that the normal control group,the rats in other groups received cecal ligation and puncture(CLP) method to induce sepsis through abdominal infection.ANB groups were given gastric gavage of ANB 0.5 hours before CLP and 6 hours after CLP.After CLP for 24 hours,the mortality was observed,serum levels of alanine aminotransferase(ALT),total bilirubin(TB) and creatinine were detected,and pulmonary myeloperoxidase(MPO) activity in all of the rats was examined.Results Serum ALT and TB levels,MAO activity and 24-hour mortality were increased in the model group(P

7.Near-infrared fluorescence imaging with methylene blue for detecting sentinel lymph node in early breast cancer

Objective To explore the clinical value of indocyanine green near-infrared fluorescence imaging with methylene blue for the sentinel lymph node biopsy (SLNB) in early breast cancer.Methods SLNB was performed in forty patients by fluorescence navigation with methylene blue.Frozen section was performed,followed by conventional histopathology.Results The identification rate of SLN was 100%.100% of SLNs were fluorescent,with the average number being 2.88.92.5% of SLNs were stained blue,with the average number being 1.75.The presence of lymph node metastasis was observed in intraoperative biopsy in 7 patients,and a total dissection of the axillary lymph nodes was performed immediately.Metastatic lesions were not found in the identified lymph nodes by intraoperative biopsy and permanent pathological diagnosis in the other 33 patients.Conclusions Sentinel lymph nodes biopsy by fluorescence navigation with methylene blue was a feasible minimally invasive technology with high detection rate.
8.The curative effect of ursodeoxycholi acid on retinopathy of prematurity in newborn rats

Objective To explore the curative effect of ursodeoxycholi acid on retinopathy of prematurity (ROP) caused by high concentration of oxygen in newborn rats. Methods The model of ROP was established. Neonatal rats were divided into normal control group, ROP model group, low dose ursodeoxycholi acid treatment group (10mg/kg) and high dose ursodeoxycholi acid treatment group (40mg/kg). Rats were sacrificed at days 17. The new retinal vessels were observed and counted under lfuorescence microscope. Results The new retinal vessels in ROP rats were hyperplastic, twisted and unevenly distributed. There was signiifcant difference in the number of new retinal vessels among different groups (P=0.000). The number of new retinal vessels of rats in ROP group, low-dose group and high-dose group was signiifcantly more than that in control group (P=0.000). The number of new retinal vessels in low-dose group and high-dose group was significantly less than that in ROP group (P<0.05). The number of new retinal vessels in high-dose group was less than that in low-dose group (P>0.05). Conclusions Ursodeoxycholi acid could inhibit the angiogenesis of retina and could have curative effect on ROP.
9.Determination of curcumol and germacrone in Zedoary Turmeric Oil and Glucose Injections by RP-HPLC

AIM: To establish a HPLC method for determination of curcumol and germacrone in Zedoary Turmeric Oil and Glucose Injections. METHODS: RP-HPLC was applied for quantitative analysis of curcumol and germacrone. Hypersil ODS2 column (250 mm? 4.6 mm, 5 ?m) and gradient elution was used. Mobile phase A was acetonitrile and mobile phase B was water. Phase A time-percentage composition was as follows:0-3 min,45%;3-30 min,45%→65%;30-38 min, 65%; 38-45 min, 65%→90%;45-55 min,90%→100%;55-65 min,100%. The flow rate was at 1.0 mL?min -1 . Diode array detector was set at 214 nm and column temperature was at 25 ?C. RESULTS: The calibration curves of curcumol and germacrone were linear in the range of 0.26 - 6.5 ?g(r= 0.999 9 ) and 0.112 5 - 2.812 5 ?g(r=1), The average recoveries of them were 102.1% and 98.8% (n=5), respectively. CONCLUSION: Curcumol and germacrone can be determined by the method, and this will improve the quality control of Zedoary Turmeric Oil and Glucose Injections.
