Objective:To investigate the feasibility,safety and superiority of transvagina endo- scopic cholecystectomy used in clinical application.Methods:31 patients with gall-bladder diseases were treated in transvagina endoscopic cholecystectomy.Through a 5mm cut below the umbilicus, inserted laparoscopic guide gastroscope and NOTES manipulator enter abdominal cavity through posterior fornix 15mm cut.After the gastroscope can capture stable vision,withdraw the laparoscope and insert normal laparoscopic manipulator.With the gastroscope monitoring,accomplish cholecystectomy using umbilical and pudendum manipulator.Results:All of patients were accepted this new type operation.The operation time was 60～150 minutes,and the average time was 85 minutes.No postoperative draining,bleeding and biliary fistula occurred in those patients. There was no obvious scar in belly,and all of patients felt no pains after operation.The average inpatient time was shorten,the medical cost decreased.Conclusion:Transvagina endoscopic cholecystectomy is feasible and safety.Comparing to traditional laparoscopic operation,it has obviously superiority.Transvagina endoscopic cholecystectomy should be widely applied in good condition hospitals.

BackgroundRetinal pigment epithelial (RPE)cell senescence damage the metabolism of photoreceptor,leading to retinal dysfunction and loss of vision.To understand RPE cell senescence mechanism will contribute to the study of age-related macular degeneration ( AMD).ObjectiveThe present study was to prepare the ageing RPE cell model with passage and explore its potential mechanisms.MethodsThis study was approved by the Ethic Committee of Qingdao University Medical College,and the informed consent was obtained from each gravida.Six human eyeballs were obtained from artificial labor fetusl with the gestational age 16-28 weeks.RPE cells were isolated,cultured and passaged in vitro to establish the cell replicative aging model.The third to twelfth cells were collected to be used to this experiment.Human keratin was used to identify the cells by immunochemistry,and MTT method was utilized to assess the proliferation and viability of different generations of cells as the A490 value.The cellular cycles and transmembrane potential (△ψm)of mitochondrion (△ψm) with passage were detected and compared using Flow Cytometry. Results Cultured and passaged cells showed the hexagon in shape with the melanin in 1-2 generations of cells and presented with the brown staining in cytoplasm for human keratin.The melanin was absent in the third generation cells.Vibrant growth statues were seen from the 3-6 generations cells and thereafter the proliferation ability reduced.The cells of G0/G1 phase were gradually elevated with the passage from 3 - 12 generations with a percentage of 68.40％ in the third generation of cells to 87.33％ in the twelfth generation of cells,showing a significant difference among various generations ( F =180.43,P =0.00),and that of the sixth,ninth and twelfth generation of cells was significant higher than the third,sixth and ninth generation respectively (t =4.002,P＜0.05 ; t=12.885,P＜0.01 ;t=16.387,P＜0.01 ).MTT assay showed that of RPE cells were significantly declined with the passage ( F =38.77,P =0.00),and the A490 value of the ninth,twelfth generations of cells was considerably lower in comparison with sixth and ninth generation respectively ( t =5.991,11.983,P＜0.01 ).From 3 through 12 generations of cells,the staining intensity of rhodamine 123 was gradually decreased ( F =121.68,P =0.00 ),and the staining intensity in the sixth,ninth and twelfth generation of cells was significant lower than that the third,sixth and ninth generation respectively(t=6.918,7.620,11.207,P＜0.01 ).Conclusions A replicative aging model can be successfully created by the passage in vitro using human fetal RPE cells.The reduce of transmembrane potential and damage of mitochondria might be one of mechanisms of senescence of RPE cells.

Objective To study the value of bone marrow MR imaging in mornitoring chronic myeloid leukemia(CML).Methods 12 cases of CML underwent twice MR examination of pelvis and femur respectively at different time.MRI appearances were compared with the results of morphology and blood routine at the same time.The relation between MR findings and course was analysed.Results MR appearances of bone marrow were changed with the progression of disease.The condition of patients was improved in 2 cases and worse in 4 cases,the signal intensity of bone marrrow were increased and decreased respectively.The signal intensity of bone marrow showed no change in 6 patients whose condition kept stable.Conclusion The characteristics of MRI can reflect the progression of CML,it is of important value in mornitoring this disease.

AIM:To investigate the effects of ?v?6 integrin-mediated cell adhesion on 5-fluorouracil (5-FU) induced apoptosis in colon carcinoma cell lines.METHODS: The expression of the ?v?6 integrin in colon carcinoma cell lines HT-29 and WiDr cells was analyzed by flow cytometry. The apoptosis induced by 5-FU and the effects of ?v?6 integrin-mediated cell adhesion on 5-FU induced cell apoptosis were measured by enzyme-linked immunosorbent assay (ELISA) method and acridine orange-ethidium bromide (AO-EB) double fluorescent dye staining.RESULTS: Both the colon carcinoma cell lines HT-29 and WiDr cells expressed the ?v?6 integrin. Objective To investigate CT features of subconjunctival fat prolapse and choristoma. Methods Thirty-eight patients, including 18 with subconjunctival fat prolapse and 20 with choristoma, were retrospectively analyzed. Transverse plain CT scan was performed in all the patients, and then coronal plane images was reconstructed at CT workstation. Results CT features of subconjunctival fat prolapse revealed a crescent-shaped or horn-shaped fat density mass continuous with the orbital fat in the superior temporal quadrant of the orbit. CT features of choristoma revealed a crescent-shaped or triangle fat density mass incontinuous with the orbital fat in the temporal side. Conclusion Subconjunctival fat prolapse typically occurs in bilateral superior temporal quadrant of the orbit in elderly males. Subconjunctival choristoma occurs in the unilateral temporal side in adolescent. CT is helpful to distinguish subconjunctival fat prolapse and choristoma.

Objective To observe bone marrow MR imaging of adult acute leukemia patients in first diagnosis and to reveal the rule of bone marrow infiltration and the role of MRI in diagnosing and predicting the prognosis of untreated acute leukemia adult patients. Methods Fifty-four adult acute leukemia patients received MRI after diagnosis relying on FAB subtype and immunophenotyping including 28 cases with AML and 26 cases with ALL. MR imaging was obtained by the short time inversion recovery and T 1W spin echo technique of pelvis and femur at one time. The examining results of morphology and blood routine were collected at the same time. 15 age-matched volunteers were selected as controls. Results MRI showed that bone marrow of all patients were infiltrated by leukemia cells. The MRI appearance was classified into five patterns based on scope of focus. MRI patters from grade 1 to grade 3 were observed in most of patients with AML and in none with ALL, however, all patients with ALL distributed in grade 4 to grade 5.
