1.The docking prediction of baicalin and BMP receptor using SwissDock

Objective To introduce the application of SwissDock in prediction of protein and small molecule docking , and provide reference for usage of SwissDock.Methods Chooseing baicalin (small molecule) and BMPRII (target protein) as study objects, operation processes of SwissDock system were described in detail and results were analyzed.Results The prediction result of SwissDock systew demonstrates that ASN338 is found as the binding site of docking, and the distance between ASN338 and baicalin is 3.5?and G=-13.17 kcal/mol.Conclusion SwissDock can help usersto submit dockings and retrieve predicted complexes.The system including simple operation interface and good human-computer interaction, it will be a powerful tool for researches of molecular recognition on the atomic scale .
2.Isolation and Degrading Characteristics of a Phenol-degrading Bacterial Strain with High Efficiency

A high efficiency phenol-degrading bacterial strain PS1 was isolated from the drainage ditch of chemical laboratory of East China Institute of Technology.PS1 is a coccus,Gram negative and can live on phenol as its sole carbon and energy source.PS1 to identified as a strain of Raoultella sp.by 16S rRNA gene sequence analysis,which can degrade and tolerate more than 3500mg/L phenol.When phenol concentration is 500mg/L and 1000mg/L,PS1 can completely degrade it in 22 h and 32h,respectively.And while it is between 1500mg/L~3000mg/L,all phenol can be degraded by PS1 in 32h~50h.When phenol concentration is 2500mg/L,the phenol-degrading rate is the biggest and can reach to 78.1mg/h.The optimum growth and phenol-degrading conditions were obtained by orthogonal experiment,which are 25℃,pH6.5,glucose concentration 500mg/L and 20℃,pH7.0,glucose concentration 500mg/L,respectively.
3.Diversity of Endophtic Bacteria Isolated from Glycyrrhiza

120 strains of endophytic bacteria identified by ERIC-PCR were isolated from wild and cultivated Glycyrrhiza uralensis plants which collected from Erdos Innermongolia province.The identified results indicated that Glycyrrhiza uralensis plants has plenty of endophytic bacterium in density and population,and the density is higher in root and leave than in stem.Partial sequence analysis of 16S rDNA gene of 82 strains indicated that these strains were in a high similarity with 19 known genus which belong to?、?、 ?-Proteobacteria、Firmicutes and Actinobacteria.The dominant genus were Bacillus sp.,Pseudomonas sp., Pantoea sp.and Serratia sp..
4.Epidemiological characteristics of epidemic mumps in Chongqing Wanzhou district from 2011 to 2014

Objective To analyze the epidemiological characteristics of epidemic mumps in Chongqing Wanzhou district,and provide scientific basis for taking effective prevention and control measures.Methods Epidemic data of mumps in Wanzhou district from January 2011 to December 2014 were investigated retrospectively.Results A total of 2 785 cases of epidemic mumps were reported,there was no death case,the incidence of mumps was 44.02/100 000.Incidence of mumps in population under 15 years old was the highest,accounting for 84.45% (2 352/2 785 ).Mumps mainly occurred among students (1 674 cases,60.11 %).The peaks were from April to July and Oc-tober to December,the number of mumps increased significantly from October to December in 2013.Difference in incidence among each year of 2011 -2014 was statistically different(χ2 =40.52,P <0.01).Incidence of mumps was statistically different between the city streets and villages/towns(63.99/100 000 vs 32.13/100 000,χ2 =20.168,P<0.01).Incidence was statistically different between the city streets and villages/ towns each year of 2011 -2014 (all P < 0.01 ),except 2012,the incidence of mumps of city streets was significantly higher than villages/ towns. Conclusion It is important to conduct “Five early”for the prevention and treatment of infectious diseases,strength-en the vaccination of measles-mumps-rubella (MMR)vaccine or vaccine against epidemic mumps,especially the vaccination for students and children in nurseries,popularize the knowledge about prevention and control of mumps, improve diagnosis and treatment level,and strengthen the epidemic monitoring.

Nissl stain and immunocytochemical methods were used to observe the structure of the subnuclei of interpeduncular nucleus in human neonatal brain and their localization of substance P-, leu enkephalin-, and serotonin (5-HT)-like immunoreactive elements. It was detected that the human neonatal interpeduncular nucleus could be divided into five subnuclei, including: the dorsal, dorsolateral, lateral, central, and intermediate subnuclei. The immunocytochemical results showed that the substance P-containing cell bodies and fibers or terminals were distributed chiefly in the dorsolateral, lateral subnuclei and the ventral part of the intermediate subnucleus; the enkephalin-containing cell bodies and fibers or terminals were concentrated in the central subnucleus; and meanwhile the serotonin-containing cell bodies and fibers or terminals were found mainly in the dorsal subnucleus.

Using immunohistochemical technique, the distribution and development of substance P (SP)-, L-enkephalin (L-Enk)- and serotonin (5-HT)-like immunoreactivity in Gudden's dorsal tegmental nucleus (GDTN) of human foetus (fetal age 11.5 to 35 weeks)and neonate (two days) were demonstrated. Gudden's dorsal tegmental nucleus in the human might be divided into pars centralis and pars peripheralis; and the time of initial appearance for SP-, L-Enk- and 5-HT-like immunoreactivity in GDTN was different. SP-like immunoreactive fibers and terminals appeared first and were distributed mainly in the pars centralis and their density was very high. L-Enk-like imunoreactive cells, fibers and terminals were distributed in the pars peripheralis. 5-HT-like immunoreactive cells, fibers and terminals were distributed in the medial part of the GDTN; As the foetus developed further, SP-, L-Enk- and 5-HT-like immunoreactivity in the GDTN showed their own special ontogenetic changes respectively

Aim To investigate the influence of down-regulating adenosine A1 receptor and adenosine A2 A receptor gene expression on proliferation and activation of acetaldehyde-induced hepatic stellate cell-T6 cells through siRNA. Methods Alcoholic liver fibrosis in vitro model was constructed by inducing HSC-T6 cells with acetaldehyde. siRNA targeting A1R and A2AR were designed and synthesized according to its mRNA. The siRNA was transfected into rat HSC-T6 cells by li-posome LipofectamineTM 2000. HSC cell proliferation was measured by MTT. The mRNA levels of A1R, A2AR, α-SMA, Collagen I in the supernatant of the cell culture were measured by Quantitative Real-Time PCR. The protein levels of A1R, A2AR, α-SMA, Collagen I were measured by Western blot. Results A1 R and A2 AR siRNA effectively inhibited the cell proliferation, and they also significantly decreased the levels of A1R, A2AR,α-SMA, Collagen I, suggesting that A1 R and A2 AR might be potential target genes in the alcoholic liver fibrosis. Conclusions Silencing A1 R or A2 AR by RNAi can significantly inhibit the HSC proliferation, A1R and A2AR may be potential therapeutic target genes for alcoholic liver fibrosis.
8.Inhibitive effect of cremophor RH40 or tween 80-based self-microemulsiflying drug delivery system on cytochrome P450 3A enzymes in murine hepatocytes.

This study examined the effect of self-microemulsiflying drug delivery system (SMEDDS) containing Cremophor RH40 or Tween 80 at various dilutions on cytochrome P450 3A (CYP3A) enzymes in rat hepatocytes, with midazolam serving as a CYP3A substrate. The particle size and zeta potential of microemulsions were evaluated upon dilution with aqueous medium. In vitro release was detected by a dialysis method in reverse. The effects of SMEDDS at different dilutions and surfactants at different concentrations on the metabolism of MDZ were investigated in murine hepatocytes. The cytotoxicity of SMEDDS at different dilutions was measured by LDH release and MTT technique. The effects of SMEDDS on the CYP3A enzymes activity were determined by Western blotting. Our results showed that dilution had less effect on the particle size and zeta potential in the range from 1:25 to 1:500. The MDZ was completely released in 10 h. A significant decrease in the formation of 1'-OH-MDZ in rat hepatocytes was observed after treatment with both SMEDDS at dilutions ranging from 1:50 to 1:250 and Cremophor RH 40 or Tween 80 at concentrations ranging from 0.1% to 1% (w/v), with no cytotoxicity observed. A significant decrease in CYP3A protein expression was observed in cells by Western blotting in the presence of either Cremophor RH40 or Tween 80-based SMEDDS at the dilutions ranging from 1:50 to 1:250. This study suggested that the excipient inhibitor-based formulation is a potential protective platform for decreasing metabolism of sensitive drugs that are CYP3A substrates.
9.Clinical analysis of esophageal foreign bodies of button cell in childhood.

OBJECTIVE:
To improve diagnosis and treatment level of esophageal foreign bodies of button cell.
METHOD:
Reported 4 clinical cases of esophageal foreign bodies of button cell.
RESULT:
Because of different diagnosis time and different surgical time, one case died and the other were cured.
CONCLUSION
Esophageal foreign bodies of button cell has been reported rarely before. Because button cell is caustically basic, drastically dangerous, it should be emphasized clinically. The key point to cure this disease is diagnosing timely and operating a surgery as soon as possible.

10.Analysis on risk factors of emergence agitation in adult patients during recovery period after general anesthesia in postanesthesia care unit

Objective To analyze the risk factors of emergence agitation (EA)in the adult patients during the recovery peri-od after general anesthesia in postanesthesia care unit(PACU).Methods 2 056 adult patients undergoing elective surgery under general anesthesia in PACU were selected,aged more than 18 years old,sex unlimited The following factors of age,gender,body mass,ASA grade,preoperative combined disease,education level,premedicatiou,general anesthesia mode,midazolam use for induc-tion,anesthesia time,operation site,operation mode,operation time,intraoperative blood loss,intraoperative fluid replacement,re-covery time in PACU,body temperature,urinary catheter,urine volume,etc.were recorded.According to whether EA occurring during recovery from general anesthesia,the patients were divided into the quiet group and the EA group.The multivariate Logistic regression was performed to analyze and screen the risk factors for the occurrence of EA.Results 127 cases developed EA during recovery from anesthesia.The occurrence rate of EA was 6.18%.The Logistic regression analysis indicated that high grade ASA, large quantity of intraoperative fluid replacement,placing urinary catheter and postoperative pain were the risk factors for EA(P <0.05).Conclusion Highgrade ASA,large quantity of intraoperative fluid replacement,placing urinary catheter and postoperative pain are the risk factors for EA during the recovery period after general anesthesia in PACU.
