Objective To evaluate the effects of restraint stress on behaviors of mice with focal cerebral ischemia. Methods Male mice were first subjected to left middle cerebral artery occlusion/reperfusion. Repeated restraint stress (2 hours/day for 2 weeks) was given from 1 week after surgery. At the end of stress scheme, sucrose solution consumption test, open-field test and forced swimming test were performed. AIM: To investigate the effectiveness and safety of intravitreal injection of anti-vascular endothelial growth factors (VEGF) drugs to the patients with proliferative diabetic retinopathy before vitrectomy treatment.METHODS: A Meta-analysis.A comprehensive retrieval was conducted using the database including EMbase,the Cochrane Library,Pubmed,CBM,WanFang Database,CNKI and so on.The retrieval time was limited from the building time of database to Jan.2017.The randomized controlled trial was adopted with no requirements on languages.The Jadad scale and Cochrance cooperation were used as the tool of the risk and bias evaluation to analyze the literature quality.Quality estimation of evidence-based medicine on the parameters of each evaluation index was made via GRADEpro Software.The publishing biases of enclosed documents were inspected with funnel plot.At last,the Meta analysis was conducted with Review Manager 5.3.RESULTS: Totally 16 literatures published from 2008-2016 were finally put into randomized controlled trial.A total of 923 cases were included,among which 493 cases were grouped as intravitreal injection of anti-VEGF before the combined operation of PPV group (the experimental group),and 430 cases were involved in simple PPV group (the control group).The results of Meta-analysis show: (1) The probability of intraoperative bleeding was remarkably lower than the control group [OR=0.06,95%CI (0.02,0.15),P<0.01],with statistically significant differences.(2) The duration of operation was obviously shorter than that of the control group[WMD=-29.13,95% CI (-36.95,-21.30),P<0.01],with statistically significant differences.(3) The probabilities of both early and late postoperative vitreous hemorrhage were lower than those of the control group[OR=0.34,95%CI (0.20,0.58),P<0.01],with highly statistically significant differences.(4) The best corrected visual acuity after surgery was no better than that of the control group,[WMD=-0.51(LogMAR),95%CI(-1.10,0.08),P=0.09] with no statistical significance.(5) The occurrence of iatrogenic retinal rupture was lower than that of the control group[OR=0.24,95%CI(0.14,0.40),P<0.01],with statistically significant differences.(6)Among the 493 patients included in the 16 literature,no case of ocular and general adverse effects due to anti-VEGF injections was reported.CONCLUSION: It is effective and safe for the patients with proliferative diabetic retinopathy to inject anti-VEGF drugs into vitreous cavity before vitrectomy.And it can reduce the occurrence of complications during and after surgery,improving the general treatment effects.However,the specific implementation plans,such as the injection does,the interval between the injection and the operation and so on,still need further exploration and perfection.

The corneal reconstruction is tosurgial recover the structure integrity and corneal function after suffered from various trauma, inflammation and degenerative diseases.The corneal diseases caused millions of people worldwide suffering from eyesight damages and even blindness.At present, the corneal transplant is the main therapy for corneal blindness.However, the shortage in donor corneal issue is a worldwide problem and the failure due to the immunologic rejection of host is common.Nowadays, with the development of tissue culture and bioengineering technology, the application prospect of autologous stem cell transplantation is becoming more and more popular which might replace the allogeneic transplantation, becoming an important clinical treatment of regenerative medicine.Human dental pulp stem cells (hDPSCs) is a class of adult stem cell divided from the third molar teeth.Both hDPSCs and corneal cell are from the cranial nerve in embryonic ectoderm.Extensive researches show that the hDPSCs have the potentialities in corneal cell differentiation without causing immunologic rejection of the recipient.These findings manifested the potentials of hDPSCs in the clinical applications related to ocular surface reconstruction.In this paper, the features and current investigation status of hDPSCs in ocular surface reconstruction are reviewed.

The study of quality control of Chinese medicine has always been the hot and the difficulty spot of the development of traditional Chinese medicine (TCM), which is also one of the key problems restricting the modernization and internationalization of Chinese medicine. Multivariate statistical analysis is an analytical method which is suitable for the analysis of characteristics of TCM. It has been used widely in the study of quality control of TCM. Multivariate Statistical analysis was used for multivariate indicators and variables that appeared in the study of quality control and had certain correlation between each other, to find out the hidden law or the relationship between the data can be found,.which could apply to serve the decision-making and realize the effective quality evaluation of TCM. In this paper, the application of multivariate statistical analysis in the quality control of Chinese medicine was summarized, which could provided the basis for its further study.

5.Killing effect of costimulated activation of peripheral blood mononuclear cells with CD80 and CD28/CpG ODN on gastric cancerous cells MKN45

BACKGROUND: Maintenance and activation of cascade reaction influence T cell proliferation or transformation into nonreactive state even apoptosis. B7 binding to CD28 effectively activates T cells in combination with T cell receptor pathway, and enhances T cell proliferative activity. OBJECTIVE: To investigate the costimulated activation of peripheral blood mononuclear cells (PBMC) with CD28 and CpG containing oligodeoxynucleotides (CpGODN) MoAb combined with CD80, and its killing effect on human gastric cancerous cell line MKN45 in vitro.METHODS: PBMCs were isolated by Ficoll density gradient centrifugation method, and cocultured with interleukin-2, CD28 and CpGODN MoAb for 1-5 days. MKN45 cells were divided into 4 culture conditions: CD28/CpGODN, CD80 plus CD28/CpG ODN, CD80 alone, and blank control.The killing efficiency was measured by MTT method.The ultramicrostructure of cells was observed by electron microscope. Apoptosis was verified by a flow cytometery. RESULTS AND CONCLUSION: CD80 alone did not display killing effect on MKN45 cells. By MTT method, combination of costimulated activation of PBMC with CD28/CpGODN and CD80 showed enhanced killing effect compared with single therapy (P < 0.05), and the ratio of effector cell and target cell at 15: 1 resulted in half killing efficiency. Electron microscope and flow cytometery verified necrotic or apoptotic cells after 24 hours exposure to costimulated activation. Compared with blank control group, CD80 alone elevated the apoptosis rate of MKN45 cells (P < 0.01). Results from the present study show that CD80 can elevate the killing effect of costimulated activation of PBMC with CD28/CpGODN on MKN45 cells in vitro.

Objective To analyze the factors affecting quality of life (QOL) in patients with stroke. Methods Eight hundred and six stroke patients were recruited by using the stratification and random sampling method, and QOL questionnaire was applied. Results It was shown that QOL was worse comparatively in stroke patients with poorer education, divorce or lose of spouse, non-free medical service, unemployment, serious deficit of nervous function, while it was indicated by the results of multiple factor analysis that QOL was influenced mainly by such factors as education background, marriage and neurological function. Conclusion In order to improve QOL of patients, we should pay more attention to the health of elders, social security system, education and medical technology.
7.Designing and implementing of linkage demonstrating system of join support of medical equipment in the field

In the light of requirements of medical equipment simulation system, the overall framework of joint control system of medical equipment integrated support in the theater is designed in this paper. Such modules are developed with Delphi as communication module, module for equipment basic information, module for equipment video information and module for medical faculty information. A joint control system suiting integrated support simulation sand table for medical equipment is complteted,which provides a multidimensional informational platform for integrated demonstration of medical equipment.
8.Inhibition of Proto-oncogene Pokemon Expression by siRNA Expression Vector in SW480 Cells

To investigate the inhibitory effects of siRNA expression vector on proto-oncogene Pokemon expression in SW480 cells and to provide experimental basis for further research about the biological function of Pokemon.siRNA expression vectors were constructed to express a short hairpin RNA against Pokemon.The recombinants were transfected into SW480 cells with liposome.Cellular morphology and transfection efficiency were observed.The expression of Pokemon were checked by real-time fluorescence quantitative PCR and Western blot.MTT assay was used to detect the effect of siRNA on the growth of SW480 cells and Fluorescence Activated Cell Sorter was applied to study cell apoptosis of SW480 cells.The transfection efficiency was about 36% and the cellular morphology changed greatly at 48h after transfection.siRNA expression vectors could specifically reduce the expression of Pokemon mRNA and protein in SW480 cells.Compared with negative control,the inhibition ratio of Pokemon mRNA expression was 34.2% and 67.7%,in 24th hour and 48th hour,the inhibition ratio of Pokemon protein was 48.3% and 73.6%,in 48th and 72th hour,respectively.Transfection with Pokemon siRNA expression vectors could inhibit cell growth and induce cell apoptosis of SW480 cells.siRNA expression vectors were successfully established that could effectively inhibit the expression of Pokemon in SW480 cells.Pokemon gene silencing could decrease growth and increase apoptosis of SW480 cells.
9.The diagnostic significance of single or combination lung cancer-related serum biomarkers in high risk lung cancer patients

Objective The aim of this study was to assess the clinical value of pro-gastrin releasing peptide (ProGRP) , squamous cell carcinoma antigen (SCC-Ag), cytokeratin 19 fragment antigen 21-1 (Cyfra21-1) and carcino-embryonic antigen (CEA) in the diagnosis and clinical stage of lung cancer in Chinese patients.Methods Patients with lung cancer and benign lesions confirmed by pathology were enrolled in Peking Union Medical College Hospital from January 2013 to October 2014.The serum levels of four tumor markers (ProGRP, SCC-Ag, Cyfra21-1 and CEA) were measured using immunoassays before treatment.The sensitivity, specificity, positive predictive value (PPV), negative predictive value (NPV) and the areas under the receiver operating characteristic curve (AUCROC) of these four tumor biomarkers were analyzed for the diagnosis of lung cancer.Results A total of 134 patients were finally analyzed, including 73 patients with lung cancer and the other 61 patients with benign lung disease.The diagnostic sensitivity of serum Cyfra21-1 to lung cancer was 67.1%, the specificity 45.1%, the AUCROC 0.658.The diagnostic sensitivity of the panel including ProGPR, Cyfra21-1 and CEA to lung cancer was 75.3% , the specificity 57.4% , the AUCROC 0.702.In the lung cancer group, the AUCROC of ProGRP over 65 ng/L to diagnose small cell lung cancer was 0.954;the AUCROC of SCC-Ag over 1.5 μg/L to diagnose squamous cell lung cancer was 0.788;the AUCROC of Cyfra21-1 to diagnose non-squamous-non-small-cell lung cancer was 0.716.In small cell lung cancer patients, the level of ProGRP in limited-disease small cell lung cancer (LD-SCLC) were significantly higher than that in extensive-disease small cell lung cancer (ED-SCLC) (P =0.005).Conclusion This panel of serum tumor markers including ProGRP, Cyfra21-1 and CEA improves the diagnostic specificity and sensitivity in patients with high-risk lung cancer.The serum CEA level of advanced lung cancer patients is significantly increased.The high level of serum ProGRP predicts the ED-SCLC.

Objective To explore the change of serum level of neuron specific enolase (NSE) in neonates with asphyxia before and after head mild hypothermia.Methods Eighty-two asphyxial neonates were selected,including 39 mild asphyxial neonates and 43 severe asphy-xial neonates,and 29 healthy neonates were selected as control group.Forty-three severe asphyxial neonates were randomly assigned into mild hypothermia treatment group and traditional treatment group.Neonates in traditional treatment group were just given traditional treatment.While neonates in mild hypothermia treatment group received head mild hypothermia therapy and their nasopharyngeal temperature were maintained at(34.0 ? 0.5) ℃ for 72 h.Before treatment and 72 h after treatment,2 mL blood was collected,and the serum NSE was determined by radio immunoassay.Results NSE levels in mild asphyxial neonates group[(34.83?6.17) ?g/L] and severe asphyxial group[(59.58?8.87) ?g/L] were significantly higher than that of control group[(30.57?4.88) ?g/L](t=3.07 P0.05).The level of NSE at 72 h in severe asphyxial neonates with head mild hypothermia therapy[(40.97?6.55) ?g/L] was significantly lower than that of traditional treatment group [(48.15?5.57) ?g/L](t=3.86 P
